Cloning of the cDNA encoding human skeletal-muscle fatty-acid-binding protein, its peptide sequence and chromosomal localization.

A cDNA clone for the human skeletal-muscle fatty-acid-binding protein (FABP) was isolated by screening of a human adult muscle lambda gt11 expression library with an anti-(muscle FABP) serum. The identify of the clone was confirmed by transcription/translation in vitro in plasmid pSP6.5, followed by immunoprecipitation. The nucleotide sequence of the 551 bp cDNA insert showed an open reading frame of 399 nucleotides, coding for a protein of 133 amino acid residues with a calculated molecular mass of 14858 Da and a pI of 4.94. Only one cysteine residue was found, at position 125. Peptide sequence analyses of human skeletal-muscle and heart FABP, after carbamoylmethylation and lysyl endopeptidase digestion followed by automatic Edman degradation, showed that both proteins are identical. No evidence was found for the existence of isoproteins in muscle. The chromosomal localization of the human muscle FABP gene was determined by analysing 31 human x rodent somatic-cell hybrid lines. The human muscle FABP gene could be assigned to chromosome 1pter-q31.

Primary structure of a 15-kDa protein from rat intestinal epithelium. Sequence similarity to fatty-acid-binding proteins.

An abundant and novel cytosolic protein was purified from the rat intestinal epithelium by gel filtration, ion-exchange and hydroxylapatite chromatography. The protein was eluted into two different positions (fractions 1 and 2) on DEAE-cellulose chromatography. We have completed the primary structure of the protein of fraction 1 by Edman degradation. The protein (144565 Da) contains 127 amino acid residues and has an acetylated alanine at its NH2-terminus. Comparison of the primary structure of the protein with porcine gastrotropin [Walz, A. D., Wider, M. D., Snow, J. W., Dass, C. & Desiderio, D. M. (1988) J. Biol. Chem. 263, 14189-14195] and rat hepatic fatty-acid-binding protein revealed that identical residues within these proteins are found in 90 and 54 out of a total of 127 positions, respectively. Bioactivity studies demonstrated that neither the protein nor liver and intestinal fatty-acid-binding proteins influence gastric acid secretory activity in rats with gastric fistulas compared to pentagastrin. The protein showed very low affinity for palmitic-acid-binding in vitro assay system and only trace amounts of endogenous fatty acids were detected from the protein. The protein, rat intestinal 15-kDa protein is considered to be a new member of the fatty-acid-binding protein family based on its structural features.

Synapse formation by autonomic nerves in the previously denervated neuromuscular junctions of the feline intrinsic laryngeal muscles.

Nerve terminals of unknown origin at the previously denervated neuromuscular junctions (NMJ) of the feline intrinsic laryngeal muscles were investigated. Until 3 weeks after the transection of the recurrent laryngeal nerve (RLN), no axons were observed in the Büngner's bands and the NMJ, accompanied by a marked decrease of autonomic nerves around the blood vessels. At 5-6 weeks nerve varicosities labeled by 5-hydroxydopamine (5-OHDA) were observed in the Büngner's bands together with an increase of autonomic nerves around the blood vessels. At 9-30 weeks (8 cases) nerve terminals were found at the NMJ in all cases. Even if the ipsilateral vagosympathetic trunk was transected at 17 weeks, nerve terminals were found at all the NMJ 3 weeks after this treatment, indicating that nerve terminals were not from the original RLN. These nerve terminals were considered to be autonomic because nerve terminals labeled by 5-OHDA were observed. Furthermore, in the case of the removal of the superior cervical ganglion (SCG) and the nodose ganglion at 21 weeks, though nerve varicosities were found in the Büngner's bands, nerve terminals were not found 9 days after this treatment, suggesting that the ipsilateral SCG possibly played an important role. In electromyographic findings fibrillation-like activities were recognized in 8/11 cases after 5 weeks. The relationship of this phenomenon to fibrillation and muscle atrophy was discussed.

Ontogenic appearance of three fatty acid binding proteins in the rat stomach.

With the use of specific antibodies against three structurally different fatty acid binding proteins (FABPs), viz, liver FABP (L-FABP), heart FABP (H-FABP), and intestinal FABP (I-FABP), the localization and relative amount of the immunoreactive proteins were determined by immunoblotting and immuno-cytochemistry in the gastric epithelium of rats during prenatal and postnatal development. H-FABP immunoreactivity was first detected at embryonic day 20 (E20), with predominant localization in the parietal cells, whereas I-FABP immunoreactivity was detected at the day of birth in the surface mucous cells. Both immunoreactivities were continuously localized in the same cell types with increasing intensity into adulthood. In contrast, the immunoreactivity for L-FABP showed remarkable changes in intensity and localization during development of the rat stomach. It was first detected in the surface mucous cells of E19. In the first 2 weeks of postnatal life, i.e., the suckling period, L-FABP immunoreactivity reached a peak in intensity and was localized not only in the surface mucous cells, but also in some of the parietal cells, brush cells, and endocrine D cells. In the following few weeks of weaning, the reactivity of surface mucous cells and parietal cells disappeared, leaving only a small amount of total L-FABP immunoreactivity in the adult stomach, which was localized exclusively in the brush cells and D cells. These results revealed that the appearance of the three types of FABPs in the rat stomach is specific to cell types and developmental stages.

Preventive effects of OK432 on murine acute myocarditis due to encephalomyocarditis virus.

OK432, one of the immunomodulators used for cancer treatment in Japan was examined for its effects on murine myocarditis due to encephalomyocarditis (EMC) virus. The survival rate of mice administered with 1 KE of OK432 intraperitoneally every other day starting 2 days before viral inoculation was significantly higher than that of the control mice administered virus alone on days 10-21 (16/20 vs. 4/20, p less than 0.001), and the viral titer in the heart, the heart weight/body weight ratio, and the scores for myocardial inflammation and necrosis were significantly lower. The natural killer (NK) cell activity and interferon (IFN) titer on day 1 after infection were increased in comparison with the control group (NK cell activity: 51.1 vs. 37.9%, p less than 0.05. IFN: 1205 vs 512 U/ml), and the cytotoxicity of peritoneal macrophages was increased (50.6 vs. 29.1%). Thus, OK432 given before viral inoculation significantly improved survival and reduced both the viral titer in the heart and myocardial damage in this experimental model of acute EMC viral myocarditis. Accordingly, the stimulation of the host immunologic response by OK432 may be important for elimination of virus of the heart.

Pseudopseudohypoparathyroidism with recurrent polyneuropathy: an autopsy report with special reference to the peripheral nervous system.

The clinical and pathological findings of a 21-year-old girl suffering from pseudopseudohypoparathyroidism (PPHP) with relapsing neuropathy are described. Episodic exacerbations were accompanied by intracranial hypertension and were relieved by the administration of corticosteroids. At autopsy, pathologic changes were almost restricted to the peripheral axons and showed distal dominant depletion of myelinated fibers without any active myelin breakdown or inflammatory changes. The neuropathy is thought to be similar to chronic inflammatory demyelinating polyradiculoneuropathy (CIDP); however, the relationship, if any, between PPHP and CIDP is unknown.

Gonadal hormones affect the hypothermia induced by serotonin1A (5-HT1A) receptor activation.

Subcutaneous injection of 8-hydroxy-2-(di-n-propylamino) tetralin (8-OH-DPAT) in mice caused hypothermia which was antagonized by pindolol. The hypothermic effect of 8-OH-DPAT was pronounced in female than in male mice. The sex difference in the hypothermic effect was also observed after pretreatment with proadifen. The hypothermic effect of 8-OH-DPAT was attenuated by ovariectomy. In the ovariectomized mice, the effect of 8-OH-DPAT on rectal temperature was enhanced by chronic estradiol and the enhancement was blocked by simultaneous administration of testosterone. In male mice the hypothermic effect of 8-OH-DPAT was enhanced by castration. These results suggest that gonadal hormones affect the 5-HT1A receptor-mediated response.

Human papillomavirus type 16 E6 proteins with glycine substitution for cysteine in the metal-binding motif.

The human papillomavirus type 16 (HPV 16) E6 is a 151 amino acid protein containing four metal-binding motifs, Cys-X-X-Cys. We constructed and characterized three mutants with Gly substitutions for Cys within the motif; for Cys-66, for Cys-136, and for both, respectively. Zinc binding to bacterially expressed E6 was markedly reduced by the substitution for Cys-66, but DNA binding was unaffected by any of these mutations. Immunofluorescence staining showed that, whereas the E6 expressed in monkey COS-1 cells appeared mostly nuclear, the Cys-66 mutant appeared cytoplasmic. Subcellular fractionation followed by immunoprecipitation showed that the E6 in COS-1 cells was located in the membrane, nuclear, and nuclear-wash fractions, but not in the soluble cytoplasmic fraction, and that the nuclear Cys-66 protein was markedly reduced. The mutant proteins in COS-1 cells appeared to be less stable than the wild type, because the immunofluorescent cells were fewer and because the E6 bands in autoradiograms were less dense. The substitution mutants lost their capacity to enhance HPV 16 E7 transformation of rat 3Y1 cells. The data indicate that Cys-66 plays a crucial role for zinc binding and nuclear localization of E6 and that both Cys-66 and Cys-136 are required for a stable or functional structure of E6.

Two immunodominant regions of the human papillomavirus type 16 E7 protein are masked in the nuclei of monkey COS-1 cells.

The human papillomavirus type 16 E7 protein is a 98-amino acid (AA)-long nuclear oncoprotein. We established eight independent mouse hybridoma cell lines producing monoclonal antibodies (MAbs) specific for the E7 protein and characterized the MAbs by competitive binding analysis with the bacterially expressed, nonfusion E7 protein and synthesized oligopeptides. The MAbs were classified into two groups; three and five MAbs recognizing epitopes in probable immunodominant regions AAs 8 to 22 (region I) and AAs 39 to 54 (region II), respectively. These MAbs were capable of immunoprecipitating the E7 protein transiently expressed in monkey COS-1 cells. By immunofluorescence staining of the acetone-fixed E7-producing COS-1 cells, the MAbs for regions I and II detected no E7 protein and only cytoplasmic E7 protein, respectively, whereas a rabbit polyclonal antiserum (anti-lac-E7) detects both nuclear and cytoplasmic E7 proteins. Like anti-lac-E7, the MAbs of the two groups stained both nuclear and cytoplasmic E7 proteins in the COS-1 cells that were denatured with formaldehyde. The results show that two immunodominant regions of the HPV 16 E7 protein are masked in the COS-1 nuclei for binding with the corresponding MAbs. The masking of the intranuclear E7 protein may result from complex formation with cellular proteins (or structures), polymerization, or self-folding.

Parathyroid gland adenoma in primary hyperparathyroidism: report of two cases--chief and oxyphil cell adenoma.

Parathyroid adenomas are classified into two types: chief cell and oxyphil cell variants. In this report two types of parathyroid adenoma in association with hyperparathyroidism were examined. Both cases had suffered from renal calculi, and underwent operation. The laboratory tests showed high serum calcium and parathyroid hormone (PTH) levels. An exploratory surgery revealed a solitary tumor in each case. After extirpation of the parathyroid tumor these data returned to normal values. Electronmicroscopically, oxyphil cell adenoma in this report was characterized by numerous mitochondria and annulate lamellae in the cytoplasm. In some tumor cells secretory-like granules were observed.

Morphological changes in unmyelinated nerve fibres in the sural nerve with age.

Quantitative changes in unmyelinated nerve fibres in sural nerves obtained at autopsy were evaluated in 28 normal adults. The following conclusions were reached. (1) The density of unmyelinated axons showed no significant correlation with age, but the densities of (2) Schwann cell subunits with axons, (3) Schwann cell subunits without axons, (4) single protrusions of Schwann cells and (5) collagen pockets, and (6) the mean number of Schwann cell profiles per axon, all showed positive correlations with age. Additionally, (7) the percentage of subunits containing unmyelinated axons and (8) the mean number of axons in single axon-containing Schwann cell subunits demonstrated negative correlations with age. The density of Schwann cell nuclei related to unmyelinated fibres did not show a significant change with age. The age-dependent changes in unmyelinated nerve fibres thus mainly consist of an increased production of processes by Schwann cells in the absence of cell multiplication. A decrease in unmyelinated nerve fibre density or a compensatory increase of small unmyelinated axons did not occur in these normal adults. In terms of relative sensitivity for the detection of the earliest changes in unmyelinated fibres, the indices (6) and (7) are considered to be useful and superior to the conventional assessment of unmyelinated axon density and diameter distribution. These two indices are not influenced by postmortem swelling of the axons and Schwann cells. Measurements of unmyelinated axon density and size distribution will continue to be useful in the assessment of more advanced pathological conditions.

Detection of human antibody against the human papillomavirus type 16 E7 protein.

We examined 500 human sera for the presence of antibody against the human papillomavirus type 16 E7 protein by enzyme-linked immunosorbent assay with bacterially expressed fusion protein lac-E7, and by radioimmunoprecipitation and immunofluorescence assays with the E7 protein expressed in monkey COS-1 cells. The anti-E7 antibody was detectable in 6 out of 54 cases of cervical carcinoma, but in none of the others, including patients with other gynecologic cancers, those with sexually transmitted diseases, and healthy adults. The data indicate that expression of the E7 protein is closely related to cervical carcinoma.

Increase in cerebrospinal fluid and plasma levels of 3-methoxy-4-hydroxyphenylglycol in acute stroke.

BACKGROUND AND PURPOSE: 3-Methoxy-4-hydroxyphenylglycol is known to be a principal metabolite of brain norepinephrine and to be released into the blood and cerebrospinal fluid in association with activation of the central noradrenergic system. We examined changes in plasma and cerebrospinal fluid levels of 3-methoxy-4-hydroxyphenylglycol during acute stroke to see if there might be a correlation between these and the patient's clinical state. METHODS: We measured plasma levels of 3-methoxy-4-hydroxyphenylglycol in 32 control subjects and in 50 patients with brain hemorrhage and 57 patients with brain infarction who were admitted to the hospital within 72 hours after onset. In addition, we estimated 3-methoxy-4-hydroxyphenylglycol concentrations in the cerebrospinal fluid of 37 patients with brain infarction and eight control patients. RESULTS: Mean +/- SEM values for plasma 3-methoxy-4-hydroxyphenylglycol in the patients with brain hemorrhage and those with brain infarction were 7.3 +/- 0.5 and 6.6 +/- 0.5 ng/ml, respectively. Both values were significantly higher than that obtained in the 32 control subjects (4.6 +/- 0.3 ng/ml, p less than 0.01). Plasma levels of 3-methoxy-4-hydroxyphenylglycol correlated well with state of consciousness and prognosis. The mean +/- SEM level of 3-methoxy-4-hydroxyphenylglycol in the cerebrospinal fluid of the 37 patients with brain infarction (10.9 +/- 0.6 ng/ml) was also significantly higher than that in the eight control patients (7.9 +/- 0.6 ng/ml, p less than 0.01). CONCLUSIONS: The observed increase in plasma and cerebrospinal fluid levels of 3-methoxy-4-hydroxyphenylglycol implies that the activity of the central noradrenergic neurons may be enhanced at the onset of stroke, and these levels may be related to some extent to the clinical state and prognosis of stroke patients.

Ultrastructural and histochemical study of the neuromuscular junctions in the denervated intrinsic laryngeal muscle of the cat.

Morphological changes and acetylcholinesterase (AchE) activity in the neuromuscular junctions (NMJ) of the normal and denervated posterior cricoarytenoid muscle (PCA muscle) of the cat were studied. Two days after denervation, the nerve terminals at the NMJ had almost disappeared. Six weeks after denervation, intensity of AchE activity at the former junctional site (FJS) was unchanged histochemically. At this stage, primary synaptic clefts were distorted and the Schwann's cells covered the FJS. Fourteen weeks after denervation, AchE activity at the FJS had decreased in contrast to that of the non-affected side. Calcitonin gene-related peptide (CGRP) was also investigated at the NMJ of the normal PCA muscles immunocytochemically. The present study shows that CGRP coexists with Ach in the nerve terminals of the PCA muscles and may be involved in the regulation of the contractile function of the intrinsic laryngeal muscle.

Combination therapy of OK432 and recombinant human interferon alpha A/D on viral myocarditis in mice starting after infection.

We investigated the effects of OK432, recombinant human interferon alpha A/D (rIFN alpha A/D) and a combination of these drugs on murine acute myocarditis due to encephalomyocarditis virus. Mice were administered 1 KE of OK432 and 10(4)U/g of rIFN alpha A/D starting 24 hr after viral inoculation for 14 days. The survival rate of mice having the combination therapy was significantly higher than that of untreated mice on day 21 (15 of 20 vs. 6 of 20, P less than .001), whereas the viral titer in the heart, the heart weight/body weight ratio and the scores for myocardial inflammation and necrosis were significantly lower. On the other hand, therapy with OK432 or rIFN alpha A/D individually improved neither the survival rate nor the extent of myocardial damage. The natural killer cell activity in the combination therapy mice on day 3 after infection was significantly increased in comparison with untreated mice (46.2 +/- 5.5 vs. 37.7 +/- 2.8%, P less than .01), and the cytotoxicity of peritoneal macrophages was increased (45.0 vs. 26.7%). Thus, the combination therapy of OK432 and rIFN alpha A/D was more effective than the therapy with OK432 alone or rIFN alpha A/D alone against acute viral myocarditis when the treatment was started early after infection, and the stimulation of host immunologic response by OK432 may be important for this combination therapy.

[The comparison of motor developmental processes between spastic diplegic children and athetotic children who have been treated since infancy].

We studied the correlation between motor developmental processes and prognosis of locomotion at the age of 4 years, among 15 spastic diplegic children (S) and 21 athetotic children (A), who started treatment under 1 year of age. The children of each type were divided into two groups according to the ability of locomotion at the age of 4 years; S- I: 6 walkers, S- II: 9 crawler, A- I: 11 walkers and A- II: 10 crawler. We compared the average corrected ages when the subjects attained the abilities to roll over, creep, sit with hands, crawl, sit up, cruise with support, stand up, and walk alone, and made a statistical analysis by t test among 4 groups. Also we compared the cumulative frequency percentages for the ages about the locomotion mentioned above. The S- I group attained all of the abilities of locomotion significantly earlier than the S- II group. The A- I group could crawl and sit up significantly earlier than the S- II group. The A- I group could crawl and sit up significantly earlier than the A- II group. The S- I group began to creep and sit with hands significantly earlier than the A- I group. We found no significant differences between the S- II group and the A- II group in motor developmental processes. These studies revealed that it would be possible to expect the prognosis of ambulation at the age of 4 years according to the early motor developmental processes among the spastic diplegic children.(ABSTRACT TRUNCATED AT 250 WORDS)

[A case of inflammatory pseudotumor of the lung that extended to involve the mediastinum].

A 17-year-old woman was referred to our hospital because of cough and fever. The chest X-ray films revealed an anterior mediastinal mass shadow. Steroid drugs and antibiotics were administered and the shadow diminished slightly. To confirm diagnosis, open thoracotomy was performed. The tumor was extended to the mediastinum, thymus and heart as well as the right middle lobe and right lower lobe. Microscopic findings of the resected tumor showed proliferation of histiocytes with infiltration by inflammatory cells, lymphocytes and fibrous tissue.
