A method was developed for the discrimination of 8 Eimeria species of chickens, i.e., E. acervulina, E. brunetti, E. mitis, E. maxima, E. necatrix, E. praecox, E. tenella, and E. hagani using the 2-step polymerase chain reaction (PCR). In the first PCR, the small subunit ribosomal RNA (srRNA) gene was amplified from the parasite genome using conserved sequences for the Apicomplexa srRNA gene as the primers. The srRNA gene amplified from the parasite genome was discriminated in the second step by random-amplified polymorphic DNA (RAPD) PCR using 10 arbitrary primers. Each arbitrary primer produced species-specific RAPD patterns that provided a simple method for species identification from the srRNA genes of the 8 Eimeria species. This method should be useful for discrimination of the parasite species for diagnosis or epidemiological surveys of chicken coccidiosis.
A 31-year-old man was admitted to our hospital complaining of epigastric discomfort and severe lumbago. An upper gastrointestinal endoscopy revealed several submucosallike tumors. Histologic examination of biopsy specimens confirmed the presence of endocrine cell carcinoma. Gallium scintigraphy and CT revealed multiple bone metastasis. He was treated with 6 cycles of combination chemotherapy consisting of CDDP, etoposide, CPA, EPI and VCR. Both gastric tumors and bone metastasis completely disappeared. After 7 cycles of the chemotherapy, he was treated with HDCT with PBSCT. There was no severe complication. This result suggested that the combination of conventional chemotherapy and HDCT with PBSCT was useful in cancer patients with poor prognoses, such as advanced gastric endocrine cell carcinoma.
This study was undertaken to assess blood vessel invasion (BVI) and other histologic features to determine the best method of histologic prognosis in node-negative breast cancer patients. The prognostic significance of the clinico-pathological findings was evaluated in 70 patients with node-negative breast cancer among 135 patients operated on between 1971 and 1981. The prognostic factors investigated included BVI, peritumor lymphatic invasion, clinical tumor size, nuclear grade, histological grade, mitotic grade, and tumor necrosis. BVI was detected by factor VIII-related antigen and elastica van Gieson staining. BVI-negative patients had a 20-year cumulative survival of 93.7%, versus 74.7% for BVI-positive patients (P = 0.0294). The clinical tumor size also correlated well with prognosis (P < 0.001). However, the other histologic features did not correlate with a poor prognosis. Moreover, we retrospectively examined the effect of postoperative chemotherapy for patients with BVI and T3, and the prognosis of those given chemotherapy seemed to be better than that of those who were not. Tumors measuring more than 51 mm and BVI may thus represent adverse prognostic factors in node-negative breast cancer patients.
A 60-year-old woman presented with chronic active hepatitis C whose HCV-RNA genotype was II according to Okamoto's classification and serum HCV-RNA concentration was 10(4) copies/mL. Agranulocytosis was induced 13 days from the commencement of interferon (IFN)-alpha 2b (6 MU/day) therapy, so the IFN therapy was immediately discontinued. The agranulocytosis improved rapidly with the administration of a granulocyte colony stimulating factor (G-CSF). The possibility that IFN was associated with maturational arrest of myeloid progenitor cells was considered. During the course of 3 years of follow-up, her liver function has remained normal and serum HCV-RNA remains negative.
Transovarial transmission of a newly isolated large intraerythrocytic parasite, Babesia sp. 1 by Haemaphysalis longicornis was experimentally demonstrated. Larvae of H. longicornis were transovarially infected with the parasite by feeding as adults on the calf which had been experimentally infected with B. sp. 1. Piroplasms of B. sp. 1 were observed in peripheral blood of the calf which was infested with the parasite-infected larvae. Based on the transmissibility of the parasite with vector ticks, this parasite was suggested to be a variety of B. ovata. Thus, we propose a new variety name for B. sp. 1 as B. ovata oshimensis n. var.
The estimation of intraductal lesions of the breast has become possible due to the careful examination of ultrasonic ductal images. The diagnostic accuracy ratio of the lesions was 94.4% by using our classification of dilated ducts by ultrasonography. The characteristics of the findings of intraductal spreading of breast cancer are that the wall of the duct is thick and uneven because of the proliferation of cancer cells, the tumor usually has a broad base all along the walls, and, when the duct is filed with cancer cells, the circumference becomes so varied that it appears outwardly irregular in size and shape. Microcalcifications and irregular nodules along the ductal network are also sometimes apparent. The diagnosis of the area of intraductal spreading of cancer cells could come about from examining the continuous spreading throughout the network by ultrasonography. We have succeedingly obtained successful results from using this method for the past eight years. The diagnostic accuracy ratio to determine the indication for breast conservative surgery and the extent of the breast excision was 94.2% in cases in which the tumor size was less than 3.0cm and the distance from the nipple was over 3.1cm, and in nonpalpable cases. In breast conservative surgery, the firm establishment of a proper diagnostic method is needed to prevent any cancerous tissue from being left. At this point in the evolution of the diagnostic method for intraductal spreading, ultrasonography is one of the key elements to achieve this goal. From now on, ultrasonography will be in the spotlight and take a more leading role in the continuing development of this diagnostic procedure.
Benign Theileria species in cattle. Theileria sergenti and T. buffeli, are morphologically indistinguishable. The polymerase chain reaction (PCR) was used to amplify the genes encoding the 33- and 34-kDa major piroplasm antigens (p33/34) of T. sergenti and T. buffeli from cattle blood infected with these parasites and tick salivary gland infected with T. sergenti. Following amplification, the p33 gene from T. sergenti and the p34 gene from T. buffeli were clearly differentiated using the restriction enzyme sites that were not shared between them. The oligonucleotide primer set, designed from the p33/34 genes, was specific for these Theileria species, since no amplification was detected with DNA from Babesia ovata, B. bovis, Anaplasma marginale, A. centrale, Eperythrozoon wenyoni, bovine white blood cells, and uninfected tick salivary glands. One tenth vol of the template prepared from either 25 microliters of blood with 0.5% parasitemia or individual tick salivary glands with six infected acini allowed sufficient amplification for differentiation of the two parasite species by restriction enzyme digestion. In addition, this system could be used to demonstrate the simultaneous, experimentally induced infection of cattle with T. sergenti and T. buffeli. The PCR-based marker system therefore provides a means to differentiate T. sergenti from T. buffeli in infected cattle blood and infected tick salivary glands. This system may also be useful for the characterization of other benign Theileria species in cattle.
To evaluate the clinicopathological significance and the objective methods for angiogenesis, we immunohistochemically stained all of the regions around the cancer nests with Factor VIII-related antigen staining. We also identified microvessels in the many areas all along the border between cancer nests and the stroma by 200 x power field. We then examined the relationship between the average microvessel count (AC/mm2) and clinicopathological factors and between a 10-year cumulative survival rate in 109 cases of primary breast cancer from 1971 to 1979. There were an average of 35 fields counted among all the cases, and AC was 39.1 +/- 16.1. AC without blood vessel invasion was 36.4 +/- 15.2, while AC with blood vessel invasion was 44.7 +/- 16.6 (p < 0.02). AC without node metastasis or with n1 alpha was 37.6 +/- 15.4, while AC with n1 beta or n2 or n3 was 45.2 +/- 17.4 (p < 0.05). There was no relationship between AC and clinical tumor size, histological classification, lymphatic invasion, nuclear grade, and histological extension. A 10-year cumulative survival rate in AC (> or = 39) was 62.4%, while that in AC (< 39) was 82.7% (p < 0.03). Thus AC was considered a useful prognostic factor.
The classification of the benign species of Theileria of cattle is very confusing Representatives of this group of parasites appear worldwide, and are known as T. sergenti in Japan, T. buffeli in Australia and T. orientalis elsewhere. Consequently, these parasites are frequently referred to as the T. sergenti/buffeli/orientalis group. Kozo Fujisaki, Shin-ichiro Kawazu and Tsugihiko Kamio here argue that the Australian T. buffeli and British T. orientalis belong to one and the same species, and that the Japanese T. sergenti is a separate species.
Schizogony of Japanese Theileria sergenti of cattle was studied by light and electron microscopy. Schizonts were detected in the draining lymph node between 4 and 8 days after sporozoite inoculation. Macroschizonts (the phase of nuclear division having invaginations) were formed 6 days after inoculation. Subsequently, microschizonts (the phase of merozoite formation displaying rosette-like appearance) were observed 8 days after inoculation. Multiple infections of a host cell with sporozoites were suggested to occur since different stages of schizonts were simultaneously detected in the same cell. Host cells of schizonts were considerably enlarged by parasitism. However, morphological characteristics of the developmental stages of T. sergenti schizonts resembled those of malignant Theileria species (e.g. T. parva). Schizogony of T. sergenti observed in this study seems to be the primary generation.

Histological observations were performed on the schizonts of Japanese Theileria sergenti in three calves necropsied 8 and 10 days after application of nymphal Haemaphysalis longicornis infected with T. sergenti. In all the three calves, schizonts were observed in the cytoplasm of huge cells formed in the drainage lymph nodes, liver and spleen. The huge cells were 50 to 200 microns in diameter. Schizonts had granular appearance and an irregular shape and were 1 to 7 microns in diameter. Ultrastructurally, schizonts had more than one nuclei and were formed in the unit enlarged cell. Schizonts showed a specific reaction against anti-T. sergenti anti serum, therefore, it was concluded that the schizonts were those of T. sergenti histologically.
Invagination or transformation of a zygote of Theileria sergenti into a kinete was examined with Giemsa-stained smears of the midgut of vector ticks, Haemaphysalis longicornis and H. mageshimaensis. Zygotes invaginated almost simultaneously with the moulting of host ticks. They were round and oval, and measured 5.4 to 6.7 microns in diameter on average. The kinetes developing in the zygotes were situated in the periphery of cytoplasm and were spindle- or club-shaped.
The effects of atrial natriuretic factor (ANF) on the adrenal steroidogenesis were examined in the zona glomerulosa of rats in vivo. ANF administration provoked a significant decrease in the width of zona glomerulosa (P < 0.01) and a significant enlargement of nuclei of zona glomerulosa cells (P < 0.01). Ultrastructurally conspicuous change was observed in mitochondria of the cells of the zona glomerulosa treated with ANF, representing elongated mitochondria with "parallel arrays" of tubules with pipe-like structure. In vitro autoradiography 125I-labelled ANF binding displayed a significant decrease of a specific receptor site in the zona glomerulosa treated with ANF. A significant lowering in plasma levels of aldosterone was observed in ANF-treated rats. These findings could be interpreted as an inhibitory effect of ANF on adrenal steroidogenesis of the glomerular zone by action through a specific ANF-receptor.
Recently, the effectiveness of preoperative chemotherapy for cancer treatment has attracted much attention. The authors conducted a study to investigate the influence of 5'-DFUR administration on the cell cycle, based on DNA analysis using FCM in patients with breast cancer. A total of 53 patients with breast cancer (Stages I, II and III) were investigated concerning tumor PyNPase activity and cell cycle phase. Subjects were stratified according to tumor diameter, histologic type and ER in the group receiving preoperative 5'-DFUR administration (25 patients) and the group not receiving 5'-DFUR (28 patients). The subjects in the group on 5'-DFUR preoperative administration were classified into three sub-groups according to total dosages of 400 mg (8 patients), under 4,800 mg (8 patients) and 4,800 mg or over (9 patients). In terms of tumor diameter, significant increases (p = 0.027) of S-phase were observed in patients with the diameters of 5 cm or more in the sub-group of under 4,800 mg dosage. Similar tendencies were seen in all drug administration groups (p = 0.05). However, no influence was observed on S-phase in the group without 5'-DFUR administration. In terms of ER stratification, ER(-) patients in the dosing groups showed increasing tendencies in S-phase and significant differences of G1- as well as G2+M phases. No differences in PyNPase activity were noted in each stratified group. Preoperative administration of 5'-DFUR resulted in alterations of cell cycle in breast cancers. In patients with a total drug dosage of under 4,800 mg and with tumor diameters of 5 cm or more, increases of S-phase and decreases of G1-phase were observed. It was suggested that 5'-DFUR acted in DNA synthesizing stage to accumulate S-phase inhibiting cell cycles. Consequently, this drug is considered useful for neoadjuvant chemotherapy.
Serological comparisons among piroplasm antigens of the benign Theileria species of cattle from Japan, Australia and Britain, which are frequently referred to as T. sergenti, T. buffeli and T. orientalis, were carried out. The results obtained from comparative enzyme-liked immunosorbent assay (ELISA) using sera from infected cattle suggest that T. sergenti could be differentiated from both T. buffeli and T. orientalis by their serological dissimilarities. Western blotting combined with two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) revealed that both the 33-kDa piroplasm protein of T. sergenti and the similar 34-kDa protein of T. buffeli and T. orientalis corresponded to immuno-dominant antigens against cattle. The other 32-kDa proteins of . T. buffeli and T. orientalis also represented immunodominant antigens. Cross-reactivities of the 32-and 34-kDa proteins were observed between T. buffeli and T. orientalis, whereas the 33-kDa protein of T. sergenti could be differentiated from the similar 34-kDa proteins of T. buffeli/orientalis. The present study suggests that T. sergenti should be separated from T. buffeli and T. orientalis on the basis of their serological dissimilarities.
