Nitric oxide synthase in the glossopharyngeal and vagal afferent pathway of a teleost, Takifugu niphobles. The branchial vascular innervation.

To examine the presence of nitric oxide synthase (NOS) in the sensory system of the glossopharyngeal and vagus nerves of teleosts, nicotinamide adenine dinucleotide phosphate diaphorase (NADPHd) activity and immunoreactivity for NOS were examined in the puffer fish Takifugu niphobles. The nitrergic sensory neurons were located in the ganglia of both the glossopharyngeal and the vagal nerves. In the vagal ganglion, positive neurons were found in the subpopulations for the branchial rami and the coelomic visceral ramus, but not for the posterior ramus or the lateral line ramus. In the medulla, nitrergic afferent terminals were found in the glossopharyngeal lobe, the vagal lobe, and the commissural nucleus. In the gill structure, the nitrergic nerve fibers were seen in the nerve bundles running along the efferent branchial artery of all three gill arches. These fibers appeared to terminate in the proximal portion of the efferent filament arteries of three gill arches. On the other hand, autonomic neurons innervating the gill arches were unstained. These results suggest that nitrergic sensory neurons in the glossopharyngeal and vagal ganglia project their peripheral processes through the branchial rami to a specific portion of the branchial arteries, and they might play a role in baroreception of this fish. A possible role for nitric oxide (NO) in baroreception is also discussed.

Gastrin/CCK-ergic innervation of cutaneous mucous gland by the supramedullary cells of the puffer fish Takifugu niphobles.

The supramedullary cells (SMCs) are spinal neurons lying at the dorsal surface of teleosts. In the present study, we examined whether the SMCs of the puffer fish (Takifugu niphobles) might express gastrin/cholecystokinin-immunoreactivity, as observed in some other teleosts. All the SMCs were immunoreactive for gastrin/cholecystokinin. On the other hand, many immunoreactive varicose nerve fibers were also found terminating in the mucous glands in the skin. In addition, immunoreactive fibers were sparsely distributed in the epidermal layer. No neuronal cells other than the SMCs showed gastrin/cholecystokinin-immunoreactivity centrally or peripherally. The results suggest that gastrin/cholecystokinin-immunoreactive axons in the cutaneous mucous glands and epidermal layer are axons of the SMCs. In view of the present findings, the possible nature of SMCs was discussed.

Characterization of [3H]5-hydroxytryptamine and [3H]spiperone binding sites in clathrin-coated vesicles from bovine brain.

Coated vesicles prepared from bovine brain cerebral cortex exhibited [3H]5-hydroxytryptamine (5-HT, serotonin) and [3H]spiperone binding activities. The binding activities were localized in the inner core vesicles. Binding reached an equilibrium level by 30-45 min at 30 degreesC, and was reversed by the addition of 100 microM 5-HT for [3H]5-HT binding or 10 microM ketanserin for [3H]spiperone binding. The saturation binding experiments indicated a single class of binding sites for [3H]5-HT and [3H]spiperone with apparent Kd values of 2.4 and 1.75 nM, respectively. The binding of [3H]5-HT was displaced by 5-HT and 8-hydroxy-2-(di-n-propylamino)-tetralin (8-OH-DPAT), but not by ketanserin. The binding of [3H]spiperone was displaced by spiperone and ketanserin but not by 5-HT or 8-OH-DPAT even at 1 mM. The coated vesicles were shown by immunoblotting assay to contain alpha-subunits of GTP-binding proteins, Galphas, Galphai2, Galphai3, Galphao and Galphaq/11. Forskolin-stimulated adenylate cyclase activity in the coated vesicles was inhibited to 80% of the control level by 5-HT or 8-OH-DPAT. These results suggested that 5-HT1A and 5-HT2A receptors are present in bovine brain coated vesicles and that the 5-HT1A receptors are coupled to adenylate cyclase activity via GTP binding proteins.

Expression of neuronal type nitric oxide synthase and renin in the juxtaglomerular apparatus of angiotensin type-1a receptor gene-knockout mice.

Angiotensin type-1a (AT1a) receptor gene-knockout (AT1a-/-) mice exhibit chronic hypotension and renin overproduction. In the kidneys of AT1a-/- mice, the activity of neuronal type nitric oxide synthase (N-NOS) was histochemically detected by nicotinamide adenine dinucleotide phosphate (NADPH) diaphorase (NADPHd) reaction combined with N-NOS immunohistochemistry. The localization of renin was detected by immunohistochemistry and the results were analyzed morphometrically. The levels of N-NOS and renin mRNA in the renal cortical tissue were determined by reverse transcription-PCR and Northern blot analysis, respectively. In the renal sections from wild-type mice, NADPHd activity and N-NOS immunoreactivity were localized to the discrete region of the macula densa in contact with the parent glomerulus. In contrast, N-NOS-positive macula densa cells were distributed beyond the original location of the macula densa, occasionally extending to the opposite side of the distal tubules. The mean number of N-NOS positive macula densa cells was significantly increased in AT1a-/- mice (186 per 100 glomeruli) compared with wild-type mice (65 per 100 glomeruli). AT1a-/- mice showed 1.4-times higher N-NOS mRNA levels in the renal cortical tissues than wild-type mice. The plasma renin activity was significantly higher in AT1a-/- mice (205.5 +/- 26.1 ng/ml/hr) than in wild-type mice (8.0 +/- 0.2 ng/ml/hr). The renin-positive areas per glomerulus and renal renin gene expression were 12-times and 2.6-times higher in AT1a-/- mice than in wild-type mice, respectively. These abnormalities, however, were less remarkable in AT1a-/- mice compared with angiotensinogen-knockout mice. When AT1a-/- mice were fed a high-salt diet, the signal intensity of the NADPHd reaction and the number of positively-stained macula densa cells were significantly decreased. The levels of renal cortical N-NOS mRNA were also suppressed by the treatment. Dietary salt loading produced a parallel decrease in plasma renin activity, renal renin-immunoreactive areas, and the levels of renin mRNA without affecting systemic blood pressure. These results provide evidence for the possible involvement of N-NOS at the macula densa in the increased renin production in AT1a-/- mice.

Malignant glioma. Evaluation with 3D time-of-flight MR angiography.

PURPOSE: To investigate the clinical utility of 3D time-of-flight (TOF) MR angiography (MRA) in the evaluation of pathological vessels in patients with malignant glioma. MATERIAL AND METHODS: Sixteen patients with malignant glioma (12 with glioblastoma multiforme and 4 with anaplastic astrocytoma) were examined with 3D TOF MRA before and after the injection of gadopentetate dimeglumine. The 3D TOF MRA study was compared with conventional angiography in all patients. RESULTS: The unenhanced 3D TOF MRAs successfully depicted the feeding arteries in 9 patients but failed to depict small feeding arteries in 2 patients in whom these arteries were shown by conventional angiography. The enhanced MRAs demonstrated draining veins from the lesions in 8 patients but failed to depict a draining vein in 1 patient that was shown by conventional angiography. The stereoscopic view projection effectively depicted displacements of the major cerebral arteries in 14 patients, and deep venous structures in 11 patients which almost accorded with the findings at conventional angiography. CONCLUSION: Our results indicate that 3D TOF MRA can provide an accurate visualization of pathological vascular structures and thus help establish a diagnosis in patients with malignant glioma.

Dietary salt loading decreases the expressions of neuronal-type nitric oxide synthase and renin in the juxtaglomerular apparatus of angiotensinogen gene-knockout mice.

The present study investigates whether neuronal type nitric oxide synthase (N-NOS) in the macula densa participates in the regulation of renal renin expression during altered dietary salt intake in angiotensinogen gene-knockout (Atg-/-) mice. Wild-type (Atg+/+) and Atg+/+ mice were fed a low-salt (0.04% NaCl), normal-salt (0.3% NaCl), or high-salt (4% NaCl) diet for 2 wk. Histochemical staining for NADPH diaphorase (NADPHd) and renin were analyzed morphometrically. Levels of N-NOS and renin mRNA in renal cortical tissues were determined by reverse transcription-PCR and Northern blot analysis, respectively. In animals fed a normal-salt diet, the renal expressions of N-NOS and renin were markedly increased in Atg-/- mice compared with Atg+/+ mice. When mutant mice were fed a high-salt diet, the signal intensity of the NADPHd reaction and the number of positively stained macula densa cells were significantly decreased. The levels of renal cortical N-NOS mRNA were also suppressed by the treatment. These changes were paralleled by decreases in renal renin-immunoreactive areas and the levels of renin mRNA. On the other hand, salt restriction did not produce further significant increases in the renal N-NOS and renin expressions in mutant mice, whereas a parallel inverse relationship was observed between these enzyme expressions and the levels of salt intake in wild-type mice. These results suggest that the N-NOS expression in the macula densa is inversely regulated by salt intake and that the enzyme activity is functionally linked to renal renin production. Salt-modulated renal N-NOS and renin expressions are independent on angiotensin formation in Atg-/- mice.

[Intraarterial chemotherapy using a combination of etoposide and cisplatin for recurrent malignant lymphoma].

Superselective intraarterial infusion of etoposide and cisplatin was performed in 2 patients with recurrent malignant lymphoma which had progressed after operation, radiotherapy and chemotherapy with various anticancer agents. The tip of a microcatheter (Tracker-18) was placed in the left posterior cerebral artery in the first patient with the recurrent tumor located in the left occipital lobe and at the basilar top in the second patient with the recurrent tumor extending from the right thalamus to the pons. Etoposide (60 mg) and Cisplatin (60 mg) were each infused for 1 hour safely with a combination of intermittent infusion of nicardipine and papaverine chloride. The tumor disappeared completely after 2 courses of intraarterial chemotherapy in the first patient and after 1 course in the second patient. Intraarterial chemotherapy using a combination of etoposide and cisplatin was performed safely and showed good results.

NADPH-diaphorase activity in the vagal afferent pathway of the dogfish, Triakis scyllia.

Nicotinamide adenine dinucleotide phosphate (NADPH)-diaphorase activity was examined in the cranial sensory ganglia and brainstem of the banded dogfish, Triakis scyllia. Positive neurons were found in the vagal sensory ganglion projecting to the coelomic organs, but not in those projecting to the gills or the lateral line organs. Nerve terminals in the vagal lobe were also positive. No positive neurons were found in the glossopharyngeal, facial, or trigeminal sensory ganglia. These results suggest that use of nitric oxide in the vagal sensory transmission from the coelomic organs may have been maintained in the evolutionary process from fish to mammals.

Anti-tumor efficacy of paclitaxel against human lung cancer xenografts.

We examined paclitaxel for anti-tumor activity against human lung cancer xenografts in nude mice and compared its efficacy with that of cisplatin, currently a key drug for lung cancer chemotherapy. Five non-small cell lung cancers (A549, NCI-H23, NCI-H226, NCI-H460 and NCI-H522) and 2 small cell lung cancers (DMS114 and DMS273) were chosen for this study, since these cell lines have been well characterized as regards in vitro and in vivo drug sensitivity. These cells were exposed to graded concentrations of paclitaxel (0.1 to 1000 nM) for 48 h. The 50% growth-inhibitory concentrations (GI50) for the cell lines ranged from 4 to 24 nM, which are much lower than the achievable peak plasma concentration of paclitaxel. In the in vivo study, 4 cell lines (A549, NCI-H23, NCI-H460, DMS-273) were grown as subcutaneous tumors xenografts in nude mice. Paclitaxel was given intravenously as consecutive daily injections for 5 days at the doses of 24 and 12 mg/kg/day. Against every xenograft, paclitaxel produced a statistically significant tumor growth inhibition compared to the saline control. Paclitaxel at 24 mg/kg/day was more effective than cisplatin at 3 mg/kg/day with the same dosing schedule as above, although the toxicity of paclitaxel was similar to or rather lower than that of cisplatin, in terms of body weight loss. In addition, paclitaxel showed potent activity against 2 other lung cancer xenografts (NCI-H226 and DMS114). Therefore, paclitaxel showed more effective, wider-spectrum anti-tumor activity than cisplatin in this panel of 6 lung cancer xenografts. These findings support the potential utility of paclitaxel in the treatment of human lung cancer.

The neuronal isoform of constitutive nitric oxide synthase is up-regulated in the macula densa of angiotensinogen gene-knockout mice.

Angiotensinogen gene-knockout (Atg -/-) mice lacking angiotensin II exhibit chronic hypotension and an increase in renal renin gene expression. The present study was designed to provide evidence for the possible involvement of neuronal type nitric oxide synthase (N-NOS) at the macula densa in the increased renin production in Atg -/- mice. The enzyme activity of N-NOS was histochemically detected by NADPH diaphorase (NADPHd) reaction combined with N-NOS immunohistochemistry. N-NOS mRNA expression in the renal cortical tissue was determined using reverse transcription-PCR in a semiquantitative manner. The levels of renal renin mRNA were evaluated by Northern blot analysis. In the kidneys of wild-type (Atg +/+) mice, N-NOS activity was localized to the macula densa as reported previously. On the other hand, N-NOS-positive macula densa cells of Atg -/- mice were distributed beyond the original location of the macula densa. They often occupy the entire cross-sectional profiles of the tubules. In addition, Atg -/- mice showed a stronger signal intensity for the enzyme reaction than Atg +/+ mice. The mean total number of N-NOS-positive cells per 100 glomeruli was 6 times higher in Atg -/- mice than in Atg +/+ mice. Semiquantitative reverse transcription-PCR revealed an increase in the N-NOS mRNA level in renal cortical tissue of Atg -/- mice compared with Atg +/+ mice. Furthermore, the selective inhibition of N-NOS activity by 7-nitrondazole significantly decreased the level of renal renin mRNA in Atg -/- mice. These results suggest that increased N-NOS activity at the macula densa is involved a renal renin overproduction in Atg -/- mice.

[Successful TSH suppression therapy with triiodothyronine in a patient with pulmonary metastases from differentiated thyroid carcinoma in the absence of 131I uptake].

A 28-year-old woman was referred to us to undergo 131I therapy who had multiple pulmonary metastases from papillary thyroid carcinoma after total thyroidectomy. There was no increased accumulation of a tracer in the pulmonary metastatic foci on whole-body scanning using a 111 MBq diagnostic dose of 131I. However, the pulmonary metastases were gradually decreased in size, and then clearly reduced 8 months after the start of TSH suppression therapy, which was maintained by T3 instead of T4 to bring down the serum TSH level below 0.1 microU/ml. Reduction rates of the foci were 33-76% on chest X-ray. The reduction was confirmed and no new lesions were found on the serial CT scans. Serum thyroglobulin level was lowered 80 to 25 ng/ml by this suppression therapy and progression of disease was not observed under a 54 months' T3 treatment. Thus, TSH suppression therapy might improve survival of patients with differentiated thyroid cancer.

[Unruptured intracranial aneurysms: evaluation with high-resolution MR angiography with magnetization transfer contrast (MTC) and tilted optimized nonsaturating excitation (TONE)].

Forty-one patients with suspected intracranial aneurysms were evaluated by conventional three-dimensional (3D) time-of-flight (TOF) MR angiography (MRA) or high-resolution 3D TOF MRA with magnetization transfer contrast (MTC) and tilted optimized nonsaturating excitation (TONE). Correlative study was done with conventional angiography in all patients. 3D TOF MRA detected 30 of 36 aneurysms depicted at conventional angiography. The overall rate of sensitivity for the detection of aneurysms was 83%. Unruptured aneurysms were unlikely to occur in the anterior communicating artery (3%), but were frequently found at the bifurcation of the middle cerebral artery (33%). Conventional MRA showed 7 of 9 aneurysms demonstrated at catheter angiography. The rate of sensitivity of conventional MRA for aneurysms smaller than 5 mm and greater than or equal to 5 mm were 0% and 100%, respectively. High resolution MRA with MTC and TONE demonstrated 23 of 27 aneurysms detected by catheter angiography. Sensitivities of high resolution MRA with MTC and TONE for aneurysms smaller than 5 mm and greater than or equal to 5 mm were 71% and 100%, respectively. Our results indicate that high-resolution 3D TOF MRA with MTC and TONE can provide precise, accurate depiction of intracranial aneurysms larger than or equal to 5 mm.

Developmental profiles of synaptophysin in granule cells of rat cerebellum: an immunohistocytochemical study.

The expression of synaptophysin was immunohistocytochemically examined at the protein level in the cerebellar cortex of the neonatal rat with special reference to the development of granule cells. In the premigratory zone of the external granular layer, these cells expressed synaptophysin at an early stage of development, where it was localized in the trans-side cisterns of the Golgi apparatus and in nearby small vesicles. When the granule cells began to horizontally extend their axons in the upper molecular layer, the growth cones and expansions of parallel fibers already contained many small vesicles similar in shape and size to synaptic vesicles. These small vesicles were labeled with antibody against synaptophysin. Tubular structures were often found among the accumulations of synaptic vesicles, and their membranes were also labeled with synaptophysin. Both growth cones and expansions of parallel fibers were often observed to approach and to contact with the small process of Purkinje cell dendrites. The contact between the two components formed a junction with the characteristics of a synapse. The profiles of synaptogenesis began to be observed in the deeper molecular layer during a period from the 7th to the 9th days after birth.

Expression of dopamine transporter at the tips of growing neurites of PC12 cells.

The four kinds of oligopeptides specific in amino acid sequence to a rat dopamine transporter (DAT), peptide-1-peptide-4, were chemically synthethized. An attempt to produce antipeptide antibodies against these oligopeptides was made with an in vitro immunization method. Two monoclonal antibodies, MAbs H-1a and H-1b, were produced against one of the oligopeptides, peptide-1. Western blot analysis confirmed that the two antibodies recognized an approximately 85,000 Da protein in a synaptosomal fraction prepared from the rat striatum but none in the fraction from the cerebellum. The specificity of the antibody to DAT was also confirmed by an antibody absorption test using two synthetic oligopeptides, one of which is specific only to DAT. These results have confirmed the specificity of the present antibody to DAT. The expression and subcellular localization of DAT were immunohistochemically examined with MAbs H-1a and H-1b in PC12 cells treated with nerve growth factor (NGF). The antibody labeled the surface of PC12 cells. When the cells were treated with NGF, the expression of DAT was significantly emphasized, first in the area mainly including the Golgi apparatus and rough endoplasmic reticulum and then on the surface of growth cones from the beginning of neurite outgrowth. DAT was detected by Western blot analysis in a microsomal fraction prepared from PC12 cells. The activity of DAT in the PC12 cells was pharmacologically confirmed by the uptake of [3H]-dopamine and blockade by uptake inhibitors. The NGF treatment doubled the dopamine uptake activity. GBR12909, a specific inhibitor of DAT, blocked the [3H]-dopamine at a concentration of 10(-7) M. The expression of DAT and norepinephrine transporter (NET) mRNA in the PC12 cells was examined by reverse transcriptase-polymerase chain reaction (RT-PCR). DAT mRNA significantly increased in the NGF-treated cells after 7 days of incubation, whereas NET mRNA markedly decreased.

Hemifacial spasm due to compression of the facial nerve by vertebral artery-posterior inferior cerebellar artery aneurysm and elongated vertebral artery--case report.

A 62-year-old female presented with rapid aggravation of long-standing facial spasm occurring within a few months. Neuroimaging and angiography demonstrated compression of the root exit zone (REZ) of the facial nerve by an ipsilateral saccular aneurysm at the left vertebral artery (VA)-posterior inferior cerebellar artery bifurcation, in addition to the elongated VA. Neck clipping of the aneurysm and decompression of the REZ from the elongated VA and clipped aneurysm resulted in complete disappearance of the facial spasm. The aneurysm had very thin walls and was apparently about to rupture. Aggravation of long-standing hemifacial spasm may be a warning sign for rapid growth and rupture of a causative aneurysm.

Intramedullary spinal cord meningioma--a case report.

A 54-year-old female presented with a spinal intramedullary meningioma manifesting as extremely slow development of dysesthesia in the extremities. Sagittal T1-weighted resonance imaging showed fusiform enlargement of the cervical spinal cord with an area of low-signal intensity suggestive of syringomyelia extending bidirectionally up to the medulla oblongata and downward from the homogeneously enhanced intramedullary space-occupying lesion. The tumor was confirmed to be intramedullary mass with no dural attachment, and was totally removed en bloc. Histological examination showed transitional type of meningioma. The tumor specimen was positive for epithelial membrane antigen, and included glial fibrillary acidic protein-positive fibers, which were thought to be surrounding neural tissues trapped by the nodularity of the tumor in the growing process.

[Double lumen--coaxial catheter system for combined CT during arterial portography and CT hepatic angiography].

The high sensitivity of CT during arterial portography (CTAP) for hepatic lesions is accompanied with a lack of specificity for diagnosis. Combined CTAP and CT hepatic angiograpy (CTHA) had been proved to improves lesion detection and heightens confidence in interpreting perfusion abnormalities. We describe a new double lumen - coaxial catheter system for performing combined CTAP and CTHA without the need for repeated transfer of the patient or bilateral arterial punctures. This technique was employed in eight patients with liver neoplasms. In all eight patients, CTAP and CTA images were obtained successfully. We concluded that this method was useful for the evaluation of liver tumors.
