Immunization with cholinergic cell bodies induces histopathological changes in rat brains.

We have previously shown that sera from patients with Alzheimer's disease (AD) contain antibodies to the cell bodies (perikarya; PK) of purely cholinergic Torpedo neurons, and that repeated immunization of rats with this neuronal preparation for over a year induces learning and memory impairments. In the present study, we examined the brain morphology of cholinergic PK immunized rats relative to controls. Immunohistochemical studies of the brains of these rats revealed the accumulation of IgG in specific areas, such as, the hippocampus. Parallel histochemical studies demonstrated significant changes in the hippocampus, and in white matter areas. They included large vacuoles and necrotic nuclei in the granular layer of the dentate gyrus, tangle-like appearance in some pyramidal neurons of the hippocampus, and vacuolar degeneration accompanied by oligodendroglia hypertrophy in white matter tracts, such as, the corpus callosum and fimbria. In contrast, immunization with Torpedo cholinergic nerve terminals, that has no cognitive effects on the rat, also did not induce brain morphological changes. These findings suggest that the learning and memory deficits induced by immunizing rats with cholinergic PK are related to the observed brain morphological changes, and support the hypothesis that the antibodies to cholinergic neurons found in the sera of AD patients may play a role in neuronal degeneration in this disease.

Aging does not alter the binding characteristics to voltage dependent calcium channels in the brain of CW1 mice.

Structural age-related changes in cholinergic regions within the central nervous system of CW1 mice have been described previously. Since elevated calcium concentration has been suggested to play a role in the brain aging processes, the possible involvement of voltage dependent calcium channels in this degeneration was investigated. The binding characteristics of the calcium channel antagonist [3H](+)PN 200-110 to brains of aged CW1 mice were studied. This ligand exhibited high affinity binding (Kd values in the range of 50-70 pM) to a single type of sites with a density (Bmax) of 150-200 fmol/mg protein. No significant differences were observed between the binding parameters measured in young (3 months), mid-aged (9 and 15 months) and old (20 months) mice. Autoradiographic study confirmed these results and extended them to specific brain regions. It is concluded that age-dependent degeneration processes, observed histologically in specific brain regions of these mice, are probably not related to alterations in voltage-dependent calcium channels.

[Change in the level of vasoactive intestinal peptide in the cerebral cortex and hypothalamus and behavioral reactions of rats after frontal lobectomy].

The effect of a change in the level of the vasoactive intestinal peptide (VIP) on the behavior of rats in frontal lobectomy was studied. It was shown by means of the radioimmunological method that damage to the frontal cortex leads to a decrease of the VIP level in the cortex and increase of its content in the hypothalamus on the 9th postoperative day. Daily injection of VIP in small doses into the lateral ventricles of the brain is conducive to more rapid and complete restoration of the conditioned bilateral avoidance reflex and the animals' exploratory behavior under conditions of an "open field" as compared to that in control rats which received a physiological solution. The possibility of using VIP in experimental complex pathogenetic therapy of disturbed functions of the central nervous system is discussed.

[Effect of neurohypophyseal peptides on the formation of a conditioned feeding reflex in the rat].

The influence of intraperitoneal injection of vasopressin (LVP), oxytocin (OXY) and their fragments (DGAVP, PLG) on the acquisition and extinction of conditioned food reflex was studied in rats. It was found that vasopressin and its fragments had a more pronounced specific effect on the higher nervous activity of the animals. This effect consisted in impairment of the performance of the conditioned food reflex while oxytocin had a tendency to improve its performance. On the ground of the obtained data it is suggested that administration of vasopressin may facilitate the memory function only under specific environmental conditions.

[Effect of cholecystokinin octapeptide on brain monoamine levels during feeding and drinking motivational excitation].

The paper presents studies of the influence of octapeptide cholecystokinin (ChCK-8) on the level of dofamine, noradrenaline and serotonine in the hypothalamus, septum, amygdala, striatum and mesencephalon of the rats brain during 24-, 48- and 96-hour alimentary and water deprivation. A change of catecholamines content under the influence of ChCK-8 was observed for the most part in conditions of thirst and only in the hypothalamus. ChCK-8 action in conditions of water deprivation was selective and directed mainly to the restoration of the changes in transmitter's level. ChCK-8 did not have a significant influence on the level of the brain monoamines during hunger. It may be assumed, that "satiation" hormone ChCK-8 is to a great extent connected with the mechanisms of reinforcement, and its "satiating" effect would appear at catecholamines' level in the process of satisfying of corresponding need.
