Alteration of tyrosine hydroxylase activity in PC12 cells infected with herpes simplex virus type 1.

During infection with herpes simplex virus type 1 (HSV-1) the activity of tyrosine hydroxylase (TH) in PC12 pheochromocytoma cells was initially depressed reaching a nadir at 6 hours post-inoculation, but recovered rapidly with a return to baseline activity by 8 to 9 hours post-inoculation. Subsequently, TH activity again fell with a second more variable rise in activity occurring at 24 hours post-inoculation. Studies with metabolic inhibitors and 2 temperature-sensitive viral mutants indicated that these alterations of TH activity were dissociated from morphological cytopathology and likely required expression of "late" viral gene products. Immunotitration using anti-TH antibody suggested that early depression of TH activity resulted principally from loss of enzyme protein rather than simple enzyme inactivation, and that reconstitution of activity at 9 hours was related to augmented enzyme synthesis. These observations illustrate the complexity of perturbed cellular metabolism during HSV-1 infection and suggest involvement of two unexpected processes: alteration of a specialized cell function as a result of viral genes expressed late in the replicative cycle, and augmented synthesis of a cell-coded gene product during the course of infection.

Cloning and sequence analysis of a cDNA encoding porcine mitochondrial aspartate aminotransferase precursor.

The primary structure of pig mitochondrial aspartate aminotransferase (mAspATase; L-aspartate:2-oxoglutarate aminotransferase, EC 2.6.1.1) precursor was deduced from its cDNA sequence. A library of cDNA clones was constructed from pig liver poly(A)+ RNA by applying the vector/primer method of Okayama and Berg [Okayama, H. & Berg, P. (1982) Mol. Cell. Biol. 2, 161-170]. The library was screened for pig mAspATase sequences by using a mixture of eight oligodeoxyribonucleotides as a probe. The sequences of the probe were deduced from the known amino acid sequence of pig mAspATase residues 196-201. Two recombinant plasmids containing inserts of about 2500 and 2600 base pairs were selected for sequence analysis. The amino acid sequence predicted from the cDNA sequence shows that the pig mAspATase precursor consists of the mature enzyme of 401 amino acid residues and an amino-terminal segment of 29 amino acid residues called the "presequence" that contains four basic amino acid residues, no acidic residues, and no hydrophobic amino acid stretch. The sequence of this 29-amino acid mAspATase precursor segment was compared with the presequences of other mitochondrial enzymes.

Medullary catecholamine inputs to the anteroventral third ventricular cardiovascular regulatory region in the rat.

Lesions in the tissue at the anteroventral end of the third ventricle (AV3V area) prevent the development of a variety of forms of experimental hypertension. This region receives afferents from several cell groups which have been implicated in cardiovascular regulation, including the nucleus of the solitary tract and neurons in the ventrolateral medulla. About 70% of the nucleus of the solitary tract neurons which could be retrogradely labeled from the AV3V area stained immunohistochemically for tyrosine hydroxylase, but not phenylethanolamine-N-methyltransferase, and were therefore presumably noradrenergic. About half of the neurons in the rostral part of the ventrolateral medulla which innervated the AV3V area stained for both enzymes, and thus were apparently adrenergic. These medullary catecholaminergic inputs to the AV3V area may be of importance in explaining recent data concerning the roles of both regions in experimental hypertension.

Diagnostic significance of early diastolic time intervals in patients with hypertrophic cardiomyopathy and myocardial hypertrophy due to essential hypertension.

In order to differentiate hypertrophic cardiomyopathy (HCM) from myocardial hypertrophy due to essential hypertension (HT), echocardiogram, apexcardiogram and phonocardiogram were simultaneously recorded in patients with HCM (n = 22), HT (n = 34) and normal subjects (N) (n = 22). The following early diastolic time intervals were measured: 1) IIA-O time, 2) IIA-MVO time (IRT) and 3) MVO-O time. The IIA-O time was 119 +/- 9 msec in N, 159 +/- 31 in HT (p less than 0.001, vs N) and 228 +/- 57 in HCM (p less than 0.001, vs N and HT, respectively). The IIA-MVO time was 66 +/- 11 msec in N, 95 +/- 18 in HT (p less than 0.001, vs N) and 102 +/- 24 in HCM (p less than 0.001, vs N). The MVO-O time was 53 +/- 9 msec in N, 66 +/- 21 in HT and 126 +/- 48 in HCM (p less than 0.001, vs N and HT, respectively). These results showed that among the early diastolic time intervals HT was abnormal only in the IIA-MVO time. On the other hand, HCM had abnormalities not only in the IIA-MVO time but also in the MVO-O time. Both the IIA-MVO and the MVO-O times in hypertrophic non-obstructive cardiomyopathy were prolonged significantly as compared to those in obstructive one. Analysis of early diastolic time intervals may be very useful to differentiate HCM, especially of the non-obstructive type, from HT.

[Case of double primary carcinoma of the lung and ureter].

A 73-year-old man visited our hospital complaining of chest distress in September 1979. A chest X-ray film showed a tumor shadow in the left lung and left inferior lobectomy was performed. The tumor measured 50 X 40 X 30 mm; it was diagnosed as large cell carcinoma. In November 1980 the patient suddenly noticed macroscopic hematuria although there had been no microscopic hematuria at his first visit. Ultrasound-guided percutaneous pyelography confirmed complete obstruction of the left ureter and he underwent left ureteronephrectomy. The papillary tumor, measuring 23 cm, was diagnosed as transitional cell carcinoma.

[Carcinoid tumor of the stomach].

A 49-year-old female complaining of hematemesis was examined by emergency endoscopy. Multiple gastric erosions and ulcers which had healed after 6 weeks, except for one ulcer, were found. A biopsy of this unhealed ulcer revealed gastric carcinoma. Enucleation of the tumor was performed because of multiple metastases to the liver, lung and the lymph nodes. The high serum alpha-fetoprotein level (800 ng/ml) and the size of a coin lesion in an X-ray film of the chest before the enucleation decreased post- operatively.

Plasma lipids and lipoproteins in Japanese male patients with coronary artery disease and in their relatives.

Plasma cholesterol (CH), triglyceride (TG) and high density lipoprotein cholesterol (HDL-C) were measured in 92 consecutive Japanese male subjects undergoing diagnostic coronary cineangiography. Sixty-nine of them were classified as having coronary artery disease (CAD), the remaining 23 subjects were classified as having normal coronary arteries (NCA). The CAD group had significantly lower HDL-C and higher TG levels than the NCA group. However, there was no significant difference in plasma CH between the two groups. First-degree relatives of the CAD patients were also investigated. The male blood relatives of the CAD patients also had significantly lower HDL-C and higher TG levels than the non-blood male relatives and healthy control males. The female blood relatives, however, showed no significant differences from the non-blood female relatives and the healthy control females in plasma CH, TG and HDL-C levels. These results suggest that low HDL-C and hypertriglyceridemia are the prevalent coronary risk factors, rather than hypercholesterolemia, in a population with a low fat intake such as the Japanese, and that these lipid abnormalities are related to sex and genetic factors.

Is captopril effective in primary pulmonary hypertension?

Several vasodilating agents have been used for the treatment of primary pulmonary hypertension (PPH). However, no effective therapy is currently available for this distressful disease. We tried to use an angiotensin-I converting enzyme inhibitor, captopril, for one woman suffering from PPH. Her initial treatment with furosemide, digoxin and spironolactone had shown little effect. Further administration of captopril improved her clinical condition from NYHA IV to II and made it possible to treat her at our out-clinic. No adverse effect has been found. We would like to appreciate captopril as one of the effective drugs in the treatment of PPH.

Preganglionic nerve stimulation increases the amount of tyrosine hydroxylase in the rat superior cervical ganglion.

Seventy-two hours after a 90 min period of electrical stimulation of the cervical sympathetic trunk, tyrosine hydroxylase activity was twice as high in homogenates of stimulated ganglia than in homogenates of contralateral control ganglia. When aliquots of the homogenates of the two groups of ganglia were mixed, the tyrosine hydroxylase activity was additive. Immunoprecipitation studies using an antibody prepared against purified tyrosine hydroxylase demonstrated that the doubling in enzyme activity is accompanied by a doubling in the amount of immunoprecipitable material. Thus the increase in tyrosine hydroxylase activity can be totally accounted for by an increase in the amount of tyrosine hydroxylase in the ganglion.

Experimental diabetes: alterations in circulating dopamine-beta-hydroxylase and norepinephrine.

Dopamine-beta-hydroxylase (DBH) activity is increased at least 5-fold in the serum of rats with experimental diabetes produced by Streptozotocin or Alloxan. The increased DBH activity persisted for 7 months. Dialysis did not reduce the elevated activity. Based on a large increase in the maximal velocity of the enzyme reaction and immunoprecipitation studies, there appears to be increased amounts of DBH in the serum of diabetic rats. Thyroxin and triiodothyronine concentrations were reduced in diabetic serum but replacement doses of these hormones did not correct the high DBH activity. In contrast, adrenalectomy or insulin administration prevented the diabetes and elevations of circulating DBH. Circulating norepinephrine levels in arterial and venous blood were measured at 4 and 16 weeks after the onset of diabetes and elevated only in venous blood at the early time interval. In summary, circulating DBH levels were increased in experimental diabetes but did not correlate with circulating norepinephrine levels. Further studes are required to differentiate between the possibilities of increased sympathetic nervous system activity and altered DBH turnover in experimental diabetes.

Purification and properties of angiotensin I-converting enzyme in human lung and its role on the metabolism of vasoactive peptides in pulmonary circulation.

Purification of angiotensin I-converting enzyme from human lung and characteristics of the enzyme was studied. Experimental pneumonitis was produced in rabbits and the change of the activity of angiotensin I-converting enzyme was studied in purpose to clarify the role of this enzyme in the metabolism of vasoactive peptides in the lung. Purification was performed using trypsin treatment, acid treatment, DE52-cellulose column chromatography, hydroxyapatite chromatography and Sephadex G-200 gel filtration. The enzyme after final step showed a single band on disc gel electrophoresis. Experimental pneumonitis was produced by injection of Complete Freund's adjuvant (acute pneumonitis) and of N-nitroso-N-methylurethane (chronic pneumonitis). In acute experiment, angiotensin I-converting enzyme activity in pulmonary tissue and in plasma was significantly decreased. In perfusion experiment, conversion of angiotensin I to angiotensin II and inactivation of bradykinin were also significantly decreased. In case of decreased activity of angiotensin I-converting enzyme in the lung, less angiotensin II will be released into systemic circulation and bradykinin will pass through the pulmonary circulation into systemic circulation, thus this may result in the decrease of systemic blood pressure.

A semi-automated measurement of urinary catecholamines using high-speed ion-exchange column chromatography.

Semi-automated measurement of urinary catecholamines, specific for dopa, noradrenaline, adrenaline and dopamine and other metabolites has been developed. Steps included alumina extraction, separation by high-speed ion-exchange column chromatography and fluorescent development by trihydroxyindole (THI) method without interference by other fluorescent substances. Recovery, reproducibility and sensitivity were satisfactory. alpha-Methyldopa, a metabolite of alpha-methyldopa, and isoproterenol were separated completely without interfering with the measurement of noradrenaline, adrenaline and dopamine.

Percutaneous radio-frequency ablation therapy using a clustered electrode for malignant liver tumors.

GOALS: To examine the safety and effectiveness of percutaneous radio-frequency ablation therapy (PRAT) for malignant liver tumors, using a needle with cluster radio-frequency (RF) electrodes. STUDY: The subjects were 13 patients with solitary malignant liver tumors: 10 had hepatocellular carcinoma and 3 had metastatic liver tumors. One session of PRAT with cluster RF electrodes was performed until roll-off occurred two times. Dynamic computed tomography (CT) and fine needle tumor biopsy under ultrasonographic guidance were conducted to assess the therapeutic efficacy. Aspartate aminotransferase, alanine aminotransferase, lactic dehydrogenase, and total bilirubin were evaluated before and 1, 3, and 7 days after PRAT. RESULTS: There were no serious complications. Aspartate aminotransferase, alanine aminotransferase, and lactic dehydrogenase levels peaked 1 day after PRAT and decreased thereafter. No icterus occurred. Of the 13 tumors, 12 showed complete necrosis on dynamic CT; however, one of them showed histologically incomplete necrosis in the tumor biopsy. In both of the ineffective cases, the tumors were located near relatively large vessels. There was no recurrence in the liver in all cases of PRAT that were effective (observation periods: 6--14 months; mean, 10 months). CONCLUSIONS: Percutaneous radio-frequency ablation therapy using a clustered electrode is a safe and effective treatment of malignant liver tumors, if the tumor is not located near the large vessels.

Molecular biology of adrenergic receptors in the rat and frog central nervous system.

Recent developments in the characterization of the adrenergic receptors have led to the identification and purification of the binding subunits of the various catecholamine receptors. beta-Adrenergic receptors have been identified in a wide variety of tissues by photoaffinity labeling with the antagonist [125I]p-azidobenzylcrazolol and have been purified to apparent homogeneity from several of these tissues. Thus, beta 1- and beta 2-adrenergic receptor binding sites appear to reside on peptides with molecular weights of 60,000 to 65,000. The alpha 1-adrenergic receptor binding subunit has been identified in several peripheral tissues by photoaffinity labeling with a newly developed probe (4-amino-6,7-dimethoxy-2[4(5(3-[125I]-iodo-4-azidophenyl) pentanoyl)-1-piperazinyl]-quinazoline, or [125I]APDQ). This binding site resides on a peptide with a molecular weight of 80,000. These techniques have been applied to the elucidation of the binding subunit structure of these receptors in the rat central nervous system with the result that beta 1-, beta 2-, and alpha 1-adrenergic binding sites appear to reside on peptides of similar molecular weight to those identified in peripheral tissues (i.e., 60,000-65,000 and 80,000). Using immunocytochemical techniques with antibodies raised to the frog erythrocyte, beta 2-adrenergic receptor, beta-adrenergic receptors were identified at the light microscopic level in regions of the rat and frog brain previously found by ligand binding and autoradiography to be richest in beta-adrenergic receptors. At the electron microscopic level, beta-receptor immunoreactivity was found throughout dendritic processes with local accumulations at certain postsynaptic sites. This finding is consistent with the idea that the density of the receptors might be significantly increased at postsynaptic junctions of adrenergic neurons.
