Clinical pharmacokinetics and tolerance of fleroxacin in healthy male volunteers.

The tolerance and pharmacokinetics of fleroxacin were studied in healthy male adult volunteers. The peak serum concentrations of unchanged fleroxacin were about 1.5, 3 and 5 mg/l at 1-2 h after single oral doses of 100, 200 and 400 mg, respectively. The apparent serum elimination half-life was about 10 h, independent of the dose. Fleroxacin, demethyl fleroxacin and fleroxacin N-oxide excreted in urine over 3 days accounted for about 75%, 5% and 5%, respectively, of the doses. The urine concentrations of unchanged drug were dose-related; the mean concentrations, sustained over 24 h, were about 50, 100 and 150 mg/l after 100, 200 and 400 mg doses, respectively. Food intake did not significantly influence the serum concentration and urinary excretion. Steady state serum concentrations were achieved from day 3 onwards by repeated doses of twice-a-day dosage regimen and were 2-4 and 5-9 mg/l after 200 and 400 mg bid, respectively. The mean concentrations of unchanged drug in urine were about 200 and 300 mg/l at the respective dosages. The pattern of urinary metabolites was not changed by repeated doses and 90% of repeat doses was recovered in urine, including metabolites. The serum protein binding of fleroxacin was 32%. The saliva concentration was 40% of the total serum concentration or 60% of the free serum concentration. The faecal recovery over 3 days was 3% of the dose following a single 200 mg dose after a meal. The unchanged drug concentrations in faeces during 400 mg repeated dosing were 100-150 mg/kg. No severe dose-related side-effects were observed during the study.

Xenogenization of tumor cells by transfection with plasmid containing env gene of Friend leukemia virus.

A rat hepatocellular carcinoma cell line (cKDH-8 cl-11) showed decreased tumorigenicity after transfection with an envelope gene derived from a Friend leukemia virus (FV-env gene). FV-env gene product was found by indirect immunofluorescence staining to be expressed on the cell surface of the FV-env gene-transfected cells. The FV-env-transfected cells (FV-env cKDH-8), however, grew well in X-irradiated immunosuppressed rats, indicating that the reduction in tumorigenicity of the transfected cells is based on immunological reaction in the host. The rats which rejected FV-env cKDH-8 cells showed resistance to rechallenge with the parent cKDH-8 cl-11 tumor cells. These results suggest that the FV-env gene product may elicit antitumor immunity against FV-env cKDH-8 cells in a host with a resultant reduction in the tumorigenicity of these cells.

Xenogenization of a mouse lung carcinoma (3LL) by transfection with an allogeneic class I major histocompatibility complex gene (H-2Ld).

We investigated the tumorigenicity and immunogenicity of tumor cells transfected with an allogeneic class I major histocompatibility complex gene. A single clone (3LL/3) from a Lewis lung carcinoma in the C57BL/6 strain (H-2b) was cotransfected with a BALB/c genomic clone containing an H-2Ld gene and a bacterial neo gene conferring resistance to G418. Three Ld-positive, three Ld-negative, and two Neor clones were selected by means of a 125I-protein A binding assay using an anti-H-2Ld monoclonal antibody. The antigenic expression of the H-2Ld gene products was only 20-40% on the Ld-positive clones compared with Meth-A tumor cells of BALB/c mice. The 50% lethal tumor dose of these clones in C57BL/6 mice was 5.6 X 10(6) in the Ld-positive clones, but only 1.3 X 10(5) in the 3LL/3 parent clone, 1.2 X 10(5) in the Neor clones, and 2.2 X 10(5) in the Ld-negative clones. The tumorigenicity of the Ld-positive clones was, therefore, reduced to less than 1/40 of that of the parent tumor cells. The decreased tumorigenicity of the Ld-positive clones was abrogated in mice irradiated with 600 rads. After inoculation and spontaneous regression of the viable Ld-positive clone cells, the mice acquired transplantation resistance against the challenge of a parental 3LL/3 tumor. However, the immunogenicity variation between Ld-positive, Ld-negative, Neor, and 3LL/3 parent clones showed no statistical difference. These results indicate that tumor cells transfected with an allogeneic class I H-2 gene can express an H-2 foreign antigen, can regress in syngeneic hosts, and can induce antitumor transplantation resistance against the original tumors, although they are not able to enhance their immunogenicity.

Psychomotor performance tests using a microcomputer: evaluation of effects of caffeine and chlorpheniramine in healthy human subjects.

We have developed two kinds of microcomputer programs for evaluation of psychomotor function in man. In one (simple test), two single-digit random numbers are displayed, and the subject inputs the last figure of their sum from the keyboard. The performance during two sequential trial periods of 5 min with a rest interval of 1 min is evaluated. In the other (complicated test), four single-digit random numbers are displayed and the test is performed in the same manner. With the latter we intended to decrease the efficacy of performance by about 50% as compared with the former. A validation study of these programs was conducted in 20 healthy volunteers (14 males and 6 females) with caffeine as a central nervous stimulant. The study was a double-blind, placebo controlled and cross-over one. The subjects performed both of the above tests prior to and 1 h after an oral administration of a capsule containing either placebo or caffeine (230 mg) at an interval of 24 h. The numbers of correct answers during 10 min of trial were used for the evaluation of performance. The relationship between the performance of the simple test and that of the complicated test was linear and, as expected, the efficacy of the latter was about a half (48%) of the former in the total 40 performances of control. The concentration of caffeine in plasma was measured with and HPLC technique to be 4.4 +/- 1.7 micrograms/ml (mean +/- SD) 40 min after administration. Caffeine significantly (P less than 0.05) increased the performances of both tests 1 h after the administration.(ABSTRACT TRUNCATED AT 250 WORDS)

The activity of suppressor cells in the spleen of murine bone marrow chimeras.

An intrasplenic injection (i.s.) of BALB/c bone marrow cells induces a higher survival rate than an intravenous injection (i.v.) in irradiated C3H/He recipients. Coculture experiments revealed the presence of alloantigen-specific and nonspecific suppressor cells in the spleens of mice injected i.s. and i.v. Suppressor activity decreased 50-60 days after bone marrow transplantation in i.v. chimeras, while there was no decrease in i.s. chimeras. In vitro suppressor activity was correlated with in vivo activity. Histopathological changes in the liver were examined. A total of 28% of the i.v. chimeras showed severe changes compared with 6% of the i.s. chimeras. The spleen indices of i.v. and i.s. chimeras were compared. Although there was no statistically significant difference between the spleen indices of i.v. chimeras and those of i.s. chimeras, spleen indices of i.v. chimeras tended to be higher than those of i.s. chimeras. These results show that suppressor cells in i.s. chimeras appear to inhibit graft-versus-host reactions more efficiently. Furthermore, an adoptive transfer assay showed that suppressor cells detected in i.s. chimeras were effective in vivo. We therefore suggest that suppressor cells detected in vitro correlate with in vivo activity and may play some role in the induction and maintenance of transplantation tolerance.

Studies on the synthesis of the hypermodified base isolated from rat liver phenylalanine transfer ribonucleic acid.

Oxidation of methyl (S,E)-4-[4,9-dihydro-4,6-dimethyl-9-oxo-1- (phenylmethyl)-1H-imidazo[1,2-alpha]purine-7-yl]-2-[(methoxycarbonyl) amino]-3-butenoate (3) with osmium tetroxide/N-methylmorpholine N-oxide provided a mixture of diastereomers 4 and 7. Hydrogenolysis of the major dihydroxy compound 4 over Pd-C gave beta-hydroxywybutine [[R-(R*,S*)]-1]. The minor isomer 7 was transformed into [S-(R*,R*)]-1 through the cyclic carbonate 8.

Recognition by a natural cytotoxic antibody of lactoneotetraglycosyl ceramide as an antigenic molecule in a syngeneic rat fibrosarcoma.

Conventionally bred rats possess in their sera an NA which has a cytotoxic effect on a tumor cell line (KMT-17) derived from a fibrosarcoma induced by 3-methylcholanthrene and which can be absorbed by normal rat tissues. We have succeeded in purifying a glycosphingolipid as an antigen reactive to NA. GSLs isolated from the tumor cells were separated into neutral and acidic fractions. The former fraction was judged to be antigenic as detected by its capacity to absorb NA. The antigenic fraction was further separated into 7 fractions (Frs. A to G) by silicic acid chromatography. The antigenic activity was detected in only Frs.D and E, although Fr.D was a more potent antigen than Fr.E. Chemical and immunochemical analyses showed that both fractions are composed of lactoneotetraglycosyl ceramide (paragloboside), Gal(beta 1-4)GlcNAc(beta 1-3)Gal(beta 1-4)Glc(beta 1-1)ceramide, and that the more active GSL, Fr.D, contains larger amounts of long fatty-acid chains. Inhibition studies using disaccharides and monosaccharides indicated that a N-acetyllactosamine moiety, Gal(beta 1-4)GlcNAc, of the GSL is a specific site of NA. These results suggest that paragloboside is an NA antigen and that a sugar chain portion of this GSL is required for defining specificity while the ceramide portion plays a role in potentiating the antigenicity of this GSL antigen for NA.

Anti-IgE autoantibody in patients with bronchial asthma.

The anti-IgE autoantibody in the IgG class was detected in 95.5% of patients with atopic asthma, and in 72.2% of those with non-atopic asthma, using a newly established solid phase enzyme immunoassay (EIA). The specificity of anti-IgE autoantibody was confirmed by both competitive inhibition and absorption experiments, using human IgG, IgA, IgM, IgD, IgE and rabbit anti-human IgG. Significant correlations were observed between the levels of the anti-IgE autoantibody and the serum IgE. Gel filtration studies indicated that the anti-IgE autoantibody in sera from asthmatic patients was present in the immune complex form with self-IgE, in addition to the monomeric antibody. Furthermore, this anti-IgE autoantibody has the ability to induce the reversed type immediate skin reaction, in asthma free individuals. These observations strongly suggest the putative role of the anti-IgE autoantibody in the modulation of IgE-mediated immune systems and the pathogenesis of bronchial asthma.

Methylation of 1-, 3-, 7-, 9-, and N6-methyladenines bearing a methoxyl group at the N6-position: a synthesis of 7,9-dimethyladenine.

The effect of the N6-methoxy group on regioselectivity in methylation of all five possible isomers of N-methyladenine has been investigated. The methoxyl group attached to all these isomers orients methylation to a considerable extent to nitrogen(s) in the same pyrimidine or imidazole moiety that the original N-methyl group is attached to. On the basis of the results of the present methylation study, 7,9-dimethyladeninium perchlorate has been synthesized from 7- or 9-methyl-N6-methoxyadenine.

Natural antibodies carrying a cross-reactive idiotype enhance tumor growth in the rat.

A population of Fab fragments was identified in the papain-solubilized fraction of membranes of a rat chemically-induced sarcoma, KMT 17. The Fab fragments were partially purified by gel filtration and column electrophoresis. A rabbit antiserum against the partially purified Fab fragments was raised and shown to be specific for Fab fragments by immunoelectrophoresis. Furthermore, four lines of evidence indicated that the KMT 17 Fab fragments carry a cross-reactive idiotype: (1) the antiserum could bind only with a restricted population of normal Fab fragments having a pl of 6.3; (2) an unrelated antibody (WKA anti-SRBC) showed a weak cross-reactivity (less than 6%); (3) syngeneic antisera against the KMT 17 could bind with the 125I-KMT 17 Fab preparation that was purified by an immunoabsorbent column with the rabbit anti-KMT 17 Fab antiserum (anti-idiotype antibody); (4) the rabbit anti-KMT 17 Fab antiserum could bind with neither heavy nor light chains of WKA IgG. The natural antibody carrying the idiotype was found in normal serum and in various organs as well, especially in the lung of the conventional rats. In addition, the fractions containing Fab fragments (Frll) were shown to enhance tumor growth when injected into syngeneic animals, whereas Fab-free fractions that were separated from the Fab fragments by column electrophoresis showed no enhancing activity. These results strongly suggested that a population of Fab fragments carrying a cross-reactive idiotype was responsible for the enhancement of tumor growth. Mechanisms for the biological function of natural anti-tumor antibodies carrying a cross-reactive idiotype are discussed.

Relationship between the radioisotopic footpad assay and other immunological assays in tumor bearing rats.

KMT-17, a fibrosarcoma induced by 3-methylcholanthrene in a WKA rat, is a sensitive tumor to various kinds of immunological assays and is a suitable model tumor for the study of the immune status in tumor bearing hosts. The antitumor immune response of KMT-17 bearing rats was studied by a radioisotopic footpad assay (FPA) in comparison with other in vivo and in vitro assays. Delayed hypersensitivity to tumor antigens measured by the FPA was observed from the 8th day after transplantation of KMT-17 cells, reached a peak on the 12-15th day, and then declined in the late stage on the 17th day. The kinetics of the FPA correlated well with those of an in vivo Winn assay and of an in vitro lymphocyte cytotoxicity assay (51Cr-release assay). The appearance of an antitumor antibody detected by a complement dependent cytotoxicity test also correlated well with the kinetics of the FPA. A growth inhibition assay (GIA) for non-specific cell-mediated immunity also showed similar kinetics to that of the FPA. The delayed hypersensitivity footpad reaction to tumor cell extracts measured by this FPA was tumor-specific. These results suggest that the FPA is a simple and reliable in vivo assay for evaluating antitumor immunity in tumor bearing host.

[Cytotoxic natural anti-tumor antibodies against fibrosarcoma and glioma cells in rats (author's transl)].

The presence of natural anti-tumor antibodies (NAA) against fibrosarcoma- and glioma cells was revealed in the normal sera of 10 different strains of rats. By means of a direct cytotoxicity test using guinea-pig complement and an absorption tests, NAA in inbred WKA/Hok rats were observed to be cytotoxically reactive to all investigated syngeneic and allogeneic fibrosarcoma lines and one glioma line, but not to hepatoma, lymphoma, leukemia, and neurinoma lines. Moreover, NAA reactivity to fibrosarcoma cells was significantly absorbed with brain, lung, kidney, skin homogenates, and cultured normal fibroblasts of syngeneic rats, but not with liver homogenates, thymus, spleen, lymph node and red blood cells. NAA were identified as being predominantly IgM and were stables at 56 degree C for 30 min. With the exception of one strain, there were no strain or sex differences in NAA levels among any of the investigated strains of rats. The level of NAA correlated with the in vivo anti-tumor response: when NAA-reactive fibrosarcoma or glioma cells were implanted into syngeneic WKA/Hok rats, groups of rats with high NAA levels suppressed tumor growth and survived longer than groups of rats with low NAA levels, while there was no difference in length of survival days in NAA non-reactive hepatoma or lymphoma cells. When 3-methylcholanthrene was inoculated into these two groups of rats, the tumor incidence in the groups of rats with high NAA level was significantly suppressed as compared to the group of rats with low NAA level. We discuss the mechanism of the induction of NAA in relation to the anti-tumor immunity.
