Inbred guinea pig aortic endothelial cell clones. Model for studying the vascular endothelium under totally isologous conditions.

Aortic endothelial cell clones were established from adult inbred strain 2 guinea pigs by using a simple method requiring neither feeder layers nor growth factors other than isologous serum. One clone has been maintained in continuous culture for 26 passages over a 9-month period. Cloned endothelial cells grew in a cobblestone pattern at confluence, formed intercellular junctions, and expressed factor-VIII-related antigen by immunofluorescence microscopy. The cloned cells also contained high levels of angiotensin-converting enzyme activity, although the level of enzyme activity expressed by different cloned cell lines varied. Cloned endothelial cells contained numerous Weibel-Palade bodies which appeared larger and less electron-dense than those of strain 2 guinea pig aortic endothelial cells in vivo. These clones may serve as an in vitro model to investigate the formation and function of this organelle. Cloned populations of strain 2 guinea pig vascular endothelial cells maintained in medium supplemented only with strain 2 guinea pig serum also provide an immunologically isologous system ideal for investigating interactions between the vascular endothelium and immunocompetent leukocytes in vitro.

Surgical treatment of double-outlet left ventricle in situs inversus [I,D,D].

A case of double-outlet left ventricle [I,D,D] with a subaortic ventricular septal defect and pulmonary stenosis is reported. The diagnosis was made at operation and was reconfirmed postoperatively by two-dimensional echocardiography. Repair was achieved by patch closure of the ventricular septal defect and enlargement of the functional right ventricular outflow tract using a valved patch. The postoperative course was uneventful except for a transient complete heart block. To our knowledge, this represents the second instance of surgical treatment of double-outlet left ventricle [I,D,D] reported in the literature.

[Immunocytochemical localization of carcinoembryonic antigen (CEA) and transport of CEA into the blood vessels in gastric cancer].

CEA was localized in the luminal border, cytoplasm, but not in mucus, in Signet ring cell carcinoma (sig) and mucinous carcinoma (muc). Electronmicroscopically, CEA was localized in the glycocalyx of the microvilli and microvesicles of the cytoplasm. The histologically different cancer types showed no difference in the localization of T-CEA. We also studied P-CEA elevating factors in 38 CEA-positive (++) patients manifesting subserosal (ss) or deeper invasion. No remarkable findings were obtained. When P-CEA elevating factors were studied in 20 patients with (+) CEA reaction and ss or deeper invasion, we found that the incidence of por was high in P-CEA negative cases. In particular, the por incidence was significantly low (p less than 0.01) in patients with scirrhous type. High P-CEA levels were detected in blood adjacent to the cancer. Among 6 cases with negative P-CEA in the vessels adjacent to the cancer whose T-CEA reactions were (++) or (+), and who manifested ss or deeper invasion, there was a high incidence of por (5 cases) and scirrhous type (4 cases), histologically.

[Valvuloplasty for congenital mitral valvular diseases].

Valvuloplasty is first applied as the surgical treatment for congenital mitral regurgitation (MR). Follow up study after the operation around the valvuloplasty was performed in the patients with congenital MR. In the patient under 12 years old, there were isolated MR in 3; MR associated with VSD in 14; MR and ASD in 3; MR with PDA in 10; and MR with ECD in 36. Operative methods were suturing of mitral cleft in 27; mitral annuloplasty in 17; mitral valve a, advancement in 7; cusp plasty in 4; no treatment for the MR in 14. Early death was noted in 6 cases (9.0% of mortality) and late death was noted in 3 cases (mortality rate of 9.0%). Conclusions were as follows: Surgical treatment was required in the patients with MR more severe than Sellers' Grade-II. Single suturing was recommended for the suturing of mitral cleft because shortening of the valve occurred in a few degree and the residual regurgitation reduced. Mitral valve advancement by elongation of the valvular apex was useful for the impediment of the regurgitation caused by incompetence of mitral coaptation. Mitral annuloplasty increased the effect for the impediment of the regurgitation. Ultrasonic cardiogram was useful for the follow up after the operation of the mitral valvuloplasty.

Distribution of albumin- and/or alpha-fetoprotein-positive cells in hepatocellular carcinoma.

Albumin (ALB)-positive cells were identified by an immunoperoxidase technique in 52 of 53 autopsy cases and in all of 13 surgical cases of hepatocellular carcinoma (HCC). The distribution pattern of ALB-positive cells could be classified into three groups: diffuse, localized, and sparse. The diffuse type was the most common pattern and was usually seen in well or moderately differentiated HCC, showing a trabecular growth pattern. The localized or sparse patterns were more frequent in poorly differentiated HCC showing a compact growth pattern. alpha-Fetoprotein (AFP)-positive cells were detected in 37 of the 53 autopsy cases of HCC and 11 of the 13 surgical cases. The number of AFP-positive cells and the intensity of the immunoperoxidase reaction were roughly proportional to serum AFP levels in most cases. In most regions of HCC, there seemed to be an inverse relationship between the number of ALB- and AFP-positive cells, suggesting tht most HCC cells synthesized only one of the two antigens studied. ALB-positive hepatocytes were found in all of the normal or cirrhotic livers examined and in the tumor-free regions of the HCC-containing livers. In contrast, AFP was not detected in nonneoplastic hepatocytes.
