Comparative study on the structure of the light chains of human immunoglobulins. II. Assignment of a new subgroup.

The primary structure of the variable region of the human lambda type Bence Jones protein NIG-48 was determined by analysis of the N-terminal sequence of the completely reduced and aminoethylated protein, as well as of five cyanogen bromide fragments. The variable region of NIG-48 contains 112 amino acid residues. The protein NIG-48, having a unique sequence of the variable region, a low degree of homology (about 50%) with lambda chains of the five other subgroups and the addition of two residues around 65, may represent a new subgroup, namely V lambda VI.

Primary systemic amyloidosis. A case permitting pathological and biochemical investigations.

In this 72-year-old male patient, large amounts of amyloid substances were deposited in the mesenchymal organs, but not in the parenchymal organs. Histochemical and electron microscopic studies revealed large amounts of amyloid substances and amyloid fibrils in the skeletal and smooth muscle tissues. In the skeletal muscle tissues, where large amounts of amyloid substances were deposited, glycosaminoglycan was also markedly deposited, especially chondroitin-4-sulfuric acid. The concentrations of silicon were also markedly increased in the amyloid fibrils. The concentrations of nickel and cobalt were increased but those of zinc were decreased in all organs measured, although both nickel and cobalt in another case of amyloidosis experienced were within normal values.

The amino-acid sequence of S-100 protein (PAP I-b protein) and its relation to the calcium-binding proteins.

The amino acid sequence of a brain-specific protein, S-100, has been determined by the analysis of four cyanogen bromide peptides and fourteen tryptic peptides derived from the protein. The protein chain consists of 91 amino acid residues and has a molecular weight of 10507. The sequence shows regions of stong clustering of hydrophobic, basic and acidic amino acids, with an apparent calcium-binding site in the acidic cluster.

Extramedullary plasmacytoma of the oropharynx producing kappa-type Bence-Jonas protein.

A case of extramedullary plasmacytoma in the oropharynx of a 41 year-old woman is reported. Histologically, the tumor was plasmacytoma consisting mainly of matured plasma cells and showing a deposit of amyloid in the interstitial structure. Immunoelectrophoretic studies of the concentrated urine specimen revealed a kappa-type Bence-Jones protein even though there was no increase in blood M-protein. Lineac electron radiation of 5,100 rad was given over a period of eight weeks but no effect was noted. After the tumor was surgically removed, follow up of the patient was made for 2 years without evidence of recurrence or metastasis.

Formation of interchain disulfide bonds in Bence Jones proteins and Fab(t) fragments of immunoglobulin G through thiol-disulfide interchange.

The formation of interchain disulfide bonds from partially reduced Bence Jones protein (Nag, type lambda) and Fab(t) fragments of IgG1 myeloma proteins was studied in the presence of various disulfide reagents. The results could be well explained in terms of the scheme proposed previously (Kishida et al. (1976) J. Biochem. 79, 91-105). In this scheme, it was assumed that two kinds of intermediate, which form mixed disulfides with either of the paired thiol groups, are produced. For type lambda Bence Jones proteins, only one of the two intermediates can form the inter L-L disulfide bond. The fraction of intermediate having the ability to form the inter L-L disulfide bond was estimated to be 72% of the total Nag protein and was the same irrespective of the kind of disulfide reagent examined. For Fab(t), on the other hand, both intermediates equally can form the inter Fd-L disulfide bond. On the basis of the results with cystamine, it was shown that the formation of an inter Fd-L disulfide bond from the intermediate proceeds about 100 times as rapidly as that of an inter L-L disulfide bond.

Leukocyte migration inhibition test (LMIT) in systemic lupus erythematosus.

A leukocyte migration inhibition test (LMIT) utilizing the agarose gel technique was performed with native DNA as an antigen in ten patients with systemic lupus erythematosus (SLE) and five normal subjects. Irrespective of disease activity, supernatants obtained at different time intervals during lymphocyte culture in eight patients with SLE showed significant alteration of migration, either enhancement or inhibition, of normal leukocytes. However, supernatants in the control experiments produced no significant alteration of migration. Polyacrylamide gel electrophoresis of supernatants obtained from the SLE group revealed that the inhibitory activity was present in the albumin region, whereas the enhancement activity was found in the beta-globulin region. These results indicate that the hitherto employed estimation of the leukocyte migration inhibition test based on the total activity of these two factors is insufficient for accurate evaluation of chemical mediators from sensitized lymphocytes and that the separation of these two factors may be important for a greater understanding of cellular immunity.
