Role of tryptophanyl and tyrosyl residues of flavoproteins in binding with flavin coenzymes. X-ray structural studies using model complexes.

The crystal structures of 7,8-dimethylisoalloxazine-10-acetic acid-tryptamine (1:1) tetrahydrate and 7,8-dimethylisoalloxazine-10-acetic acid-tyramine (1:1) tetrahydrate complexes were determined by the X-ray diffraction method, as models for flavin-tryptophan and flavin-tyrosine interactions in flavoproteins. The observed parallel stackings and the intermolecular spacing distances, which were less than the normal van der Waals separation between the isoalloxazine and indole rings and between the isoalloxazine and phenol rings, suggest the existence of charge-transfer interactions in their ground states. The indole and phenol rings interact with the pyrimindinoid and pyrazinoid portions of the isoalloxazine ring and have short contacts, less than 3.4 A, with the reduction site (N1 and N5 atoms) of this ring. This suggests that the reduction of oxidized flavin to the semiquinone state may be facilitated by charge transfer from the former rings to the N1 and N5 atoms. Absorption difference spectra showed that both complexes associate with equimolar ratios in solution as well as in the crystalline state and that they have the same charge-transfer bands and association constants as flavin mononucleotide (FMN)-Trp and FMN-Tyr complexes, respectively. On the other hand, proton magnetic resonance spectra suggested that in solution, the stacking modes of the indole and phenol rings to the isoalloxazine ring are different from those observed in the crystal structures and both aromatic rings are stacked over the whole of the isoalloxazine ring.

Terpenoids biotransformation in mammals III: Biotransformation of alpha-pinene, beta-pinene, pinane, 3-carene, carane, myrcene, and p-cymene in rabbits.

The biotransformation of (+)-, (-)-, and (+-)-alpha-pinenes, (-)-beta-pinene (nopinene), (-)-cis-pinane, (+)-3-carene, (-)-cis-carane, myrcene, and p-cymene in rabbits was investigated. The major metabolites were as follows: (-)-trans-verbenol from (+)-, (-)-, and (+/-)-alpha-pinenes; (-)-10-pinanol and (-)-1-p-menthene-7,8-diol from (-)-beta-pinene; (-)-alpha-terpineol and (-)-trans-sobrerol from (-)-cis-pinane; (-)-m-mentha-4,6-dien-8-ol, 3-caren-9-ol, (-)-3-carene-9-carboxylic acid, and 3-carene-9,10-dicarboxylic acid from (+)-3-carene; carane-9,10-dicarboxylic acid from (-)-cis-carane; and myrcene-3(10)-glycol, myrcene-1,2-glycol, uroterpenol, and p-cymene-9-carboxylic acid from p-cymene. These metabolisms include allylic oxidation, epoxidation, stereoselective gem-dimethyl hydroxylation and its oxidation, cleavage of a conjugated double bond by epoxidation, and regioselective oxidation, some of which are not found usually in chemical reactions, and due to which various new compounds were determined. This biotransformation of the monoterpene hydrocarbons gave some insect pheromones in high yield.U

Teratoma of the neck in a newborn infant - a case report.

A case of teratoma of the neck in a newborn baby is presented. Teratoma of the neck is a rare lesion and almost always found in the newborn, but also in the fetus and stillborn infant. Although the mass was found prenatally by ultrasonography in the present case, the patient could not be saved because of respiratory distress. Urgent surgical management is the only way to save life in this kind of neonate.

The effect of tolbutamide and hepatic extraction of insulin and glucagon and hepatic glucose output in anesthetized dogs.

The effects of tolbutamide and insulin infusion on hepatic extraction of insulin and glucagon and on hepatic glucose output were compared in anesthetized dogs. The basal hepatic extraction of insulin was not significantly different in the two experiments (62 +/- 7% vs. 49 +/- 8%). The fraction of insulin extracted by the liver was not changed by either tolbutamide or insulin administration. In contrast, hepatic extraction of glucagon significantly increased from a basal value of 12 +/- 8% to 41 +/- 12% 30 min after tolbutamide, coincident with hypoglycemia and increased secretion of glucagon. The percent hepatic extraction of glucagon did not change during insulin infusion despite similar hypoglycemia and an even greater increase in the amount of glucagon reaching the liver. Tolbutamide and insulin produced a transient fall in hepatic glucose output which was associated with a significant increase in the insulin to glucagon molar ratio of the portal vein. Despite the persistence of hypoglycemia, hepatic glucose production returned to control values, and the portal venous insulin to glucagon molar ratio returned toward normal. Thus, the initial hypoglycemia after tolbutamide and insulin treatment reflects decreased hepatic glucose production, while the later effects represent increased peripheral glucose utilization. Hepatic glucose output correlated better with the portal venous insulin to glucagon molar ratio than the ratio of the hormones removed by the liver. These findings indicate that insulin and glucagon extraction by the liver are quite different and are independently regulated. Tolbutamide directly increases the fraction of glucagon removed by the liver. Because of changes in hepatic extraction after tolbutamide, increased pancreatic secretion of glucagon might not be reflected in its peripheral concentration.

Hepatic extraction of plasma immunoreactive glucagon components. Predilection for 3500-dalton glucagon metabolism by the liver.

This study examines quantitatively the extraction of plasma immunoreactive glucagon (IRG) components by the liver. It was shown that the liver has a predilection for removal of the 3500-dalton biologically active IRG component with virtually no extraction of the other IRG fractions. Hepatic extraction of whole plasma IRG was 25.2 +/- 2.5%. Analysis of the hepatic extraction of the four plasma immunoreactive components, separated by gel filtration, revealed variable but quantitatively insignificant extraction of all components other than the 3500-dalton fraction, which was 33.4 +/- 3.2% (P less than 0.001). Hepatic extraction of whole plasma IRG was significantly less than that of the 3500-dalton component during periods of basal glucagon secretion when IRG fractions other than the 3500-dalton fraction contribute substantially to the whole plasma IRG level. However, during periods of stimulation of glucagon secretion by arginine or arginine plus cholecystokinin-pancreozymin, when the 3500-dalton component accounts for virtually all of the whole plasma IRG level, hepatic extraction of whole plasma IRG was similar to that of the 3500-dalton fraction.

Acute demyelination in mice inoculated intraspinally with mouse hepatitis virus, JHM strain.

After intracerebral or intralumbar inoculation of mouse hepatitis virus, JHM strain, into weanling mice, viral growth and lesions in the central nervous system were comparatively studied for 5 days postinoculation. In the intralumbar group the virus titer of the spinal cord exceeded that of the brain until 24 hr postinoculation, and declined later. In the intracerebral group the brain virus titer was always higher, and the cord titer attained the same level as that of intralumbar group at 72 hr. Demyelinating lesions in the spinal cord appeared 48 hr after intralumbar inoculation, that is, 36 hr earlier than after intracerebral inoculation. Virus-specific fluorescence was detected within glia cells in the cord white matter 48 hr intralumbar inoculation. Electron microscopy revealed a number of virions within oligodendroglia cells, suggesting that the acute phase demyelination might be due to viral growth within this type of cells.
