63Cu NQR evidence for spatial variation of hole concentration in La2-xSrxCuO4.

We report experimental evidence for the spatial variation of hole concentration x(hole) in the high T(c) superconductor La2-xSrxCuO4 ( 0.04 < or = x < or = 0.16) via 63Cu NQR using 63Cu isotope enriched samples. We demonstrate that the extent of the spatial variation of the local hole concentration Delta(x)hole is reflected on (63)1/T1 and deduce the temperature dependence. Delta(x)hole increases below 500-600 K, and reaches values as large as Delta(x)hole/x approximately 0.5 below approximately 150 K. We estimate the length scale of the spatial variation in x(hole) to be R(hole) greater, similar 3 nm from an analysis of the NQR spectrum.

Iron lactate induction of pancreatic and endometrial proliferative lesions and a lack of increased tumors in a 104-week carcinogenicity study in F344 rats.

Iron lactate has been used as a food additive for iron supplementation. The present study was conducted to determine whether it might have carcinogenic potential. A total of 150 male and 150 female Fischer 344 rats were divided into three groups and fed basal diet containing 0, 1 or 2% of iron lactate for 104 weeks. No iron lactate-induced tumors were observed in any groups, although the incidences of pancreatic acinar cell and endometrial gland hyperplasias were increased in males and females, respectively, in the 2% group. Thus our in vivo animal data indicate that iron lactate lacks carcinogenicity in male and female F344 rats. However, estrogenic effects might be concluded based on the data for endometrial lesions. In a second experiment, an estrogen responsive rat pituitary tumor cell line, MtT/Se, and a human breast cancer cell line, MCF-7, were therefore employed to examine the estrogenic potential of iron lactate with regard to receptor binding affinity and ERE-reporter gene activation. Results in both cases were negative. Further investigations are needed to elucidate the mechanisms of induction of pancreatic and endometrial proliferative lesions by iron lactate.

Laparoscopic bilateral adrenalectomy for Cushing's syndrome due to ACTH-independent macronodular adrenocortical hyperplasia.

We performed simultaneous bilateral laparoscopic total adrenalectomy in two patients with Cushing's syndrome due to ACTH-independent macronodular adrenocortical hyperplasia (AIMAH). Preoperative serum cortisol in the patients was 29.5 and 53.2 microg/dl, respectively. The clinical symptoms of the latter patient were advanced, and respiration was labored with orthopnea. Laparoscopic adrenalectomies were performed transabdominally in the sequential lateral decubitus positions with extension of the lateral abdominal wall of the affected side. Three 12-mm and three 5-mm trocars were positioned, and two trocar sites in the midline were used on both sides. The flexible fiberscope was inserted through the umbilical port. The adrenal glands were large, fragile, and multinodular. The maximal diameters of the removed glands were 7.8 and 8.7 cm, respectively. In both patients, the adrenal glands were successfully removed without fragmentation. The operation times were 505 and 320 min, and the estimated blood loss was 150 and 5 ml, respectively. Neither intraoperative nor postoperative complications occurred, although the latter patient required muscle training before ambulation on postoperative day 42. The procedures resulted in marked clinical improvements. Compliance with the substitutive therapy remained excellent, and the patients expressed a very high degree of satisfaction with the laparoscopic adrenal surgery. The procedures of bilateral laparoscopic adrenalectomy were successful, and provided increased experience with the laparoscopic techniques.

Biosynthetic pathway for production of a conjugated dienyl sex pheromone of a Plusiinae moth, Thysanoplusia intermixta.

Virgin females of Thysanoplusia intermixta (Lepidoptera; Noctuidae; Plusiinae) produce (5E,7Z)-5,7-dodecadienyl acetate as a main sex pheromone component. GC-MS analysis of the pheromone glands, which were treated with deuterated hexadecanoic, (Z)-11-hexadecenoic, and (Z)-7-dodecenoic acids, showed incorporation of the label into the dienyl component. Their incorporation rates confirmed that its biosynthesis proceeds in the following order: Delta11-desaturation of a C(16) acyl intermediate, chain shortening to a C(12) compound by beta-oxidation, Delta5-desaturation to produce a 5,7-dienyl system, reduction of the acyl group, and acetylation. These deuterated precursors also converted into a minor pheromone component, (Z)-7-docecenyl acetate, which might be prepared by the same pathway except for the step of Delta5-desaturation. While deuterium incorporation into the dienyl acetate was not observed in the extracts treated with other labeled dodecenoic acids with (E)-5-, (Z)-6-, and (E)-7-double bonds, the corresponding dodecenyl acetates were produced. This result showed low substrate specificity of the enzymes for reduction and acetylation. Labeled (Z)-10-hexadecenoic acid was not converted into a dodecenyl acetate, indicating the high substrate specificity of the enzyme for beta-oxidation.

Induction of graft-versus-host disease and a graft-versus-leukemia effect using ubenimex in a patient with infantile leukemia relapsing after an unrelated cord blood transplant.

A 20-month-old boy with infantile leukemia was treated with total body irradiation, etoposide, cyclophosphamide and unrelated cord blood transplantation with a one-antigen mismatch. He relapsed on day 100 and achieved remission after ubenimex administration, and also developed chronic graft-versus-host disease of the skin. He remained in remission for 22 months with repeated courses of ubenimex. Ubenimex may be an alternative to donor lymphocyte transfusion and may be useful for the treatment of a patient who has relapsed after cord blood transplantation.

Evidence that action potential generation is not the exclusive determinant to trigger spontaneous myogenic contraction of guinea-pig urinary bladder smooth muscle.

Urinary bladder smooth muscle (UBSM) exhibits spontaneous contraction. This spontaneous mechanical activity is myogenic and can be closely related to the UBSM cell action potential to facilitate Ca2+ influx through voltage-gated Ca2+ channels. In the present study, to know whether this membrane electrical event is the exclusive mechanism to trigger spontaneous smooth muscle contraction, we compared the inhibitory effects of Ca2+ channel blockers on the spontaneous action potential and mechanical activity in the isolated guinea-pig UBSM. Both action potential and rhythmic contraction were generated spontaneously in the presence of atropine (1 microM), phentolamine (1 microM), propranolol (1 microM), suramin (10 microM) and tetrodotoxin (1 microM), which suggest that both phenomena were myogenic in origin. Nisoldipine (100 nM) and diltiazem (10 microM) completely eliminated the generation of action potential whereas its frequency was dramatically increased by a dihydropyridine Ca2+ agonist, BayK 8644 (1 microM). In contrast to disappearance of action potential in the presence of Ca2+ channel blockers, spontaneous contraction of UBSM was inhibited only partly by nisoldipine or diltiazem and most of the mechanical components persisted in these channel blockers. These results indicate that spontaneous action potential in UBSM cell is generated through the activation of L-type voltage-gated Ca2+ channels. The subsequent elevation of intracellular Ca2+ concentrations during a burst of action potentials can be partly responsible for the induction of UBSM mechanical activity. In addition, the present study provides evidence that UBSM spontaneous mechanical activity is also attributable to the mechanism(s) other than the generation of Ca2+ spike.

Entrapment neuropathy of the palmar cutaneous branch of the median nerve concomitant with carpal tunnel syndrome: a case report.

A case of the entrapment neuropathy of the palmar cutaneous branch of the median nerve, concomitant with carpal tunnel syndrome is presented. This report demonstrates that the Semmes-Weinstein monofilament test and nerve conduction studies can identify entrapment of the palmar cutaneous branch of the median nerve concomitant with carpal tunnel syndrome.

Combined spinal and epidural anesthesia for abdominal aortic aneurysm surgery in patients with severe chronic pulmonary obstructive disease.

BACKGROUND: The presence of chronic obstructive pulmonary disease (COPD) should not be considered a contraindication to operation but should rather identify those patients that require special preoperative and postoperative pulmonary care. The purpose of this paper is to report our experience in the use of combined spinal and epidural anesthesia (CSEA) for open AAA repair in such patients. METHODS: From April 1997 to November 1998, three patients with severe COPD underwent elective open infrarenal AAA repair under CSEA. All the patients satisfied one or more of the following criteria: a room air PaO2 =or<60 mmHg, PaCO2 =or>45 mmHg, FEV1 =or<50% of predicted, and/or FVC =or<75% of predicted, and one of them was on home oxygen preoperatively. Preoperative preparation of the patients consisted of breathing exercises with incentive espirometry, elimination of underlaying pulmonary infections and usage of bronchodilator therapy. RESULTS: All the patients tolerated surgery safely. None of them developed postoperative complications, including pneumonia and other respiratory conditions. No significant changes in room air arterial blood gas or pulmonary function was recognized before and after surgery. CONCLUSIONS: CSE anesthesia is a viable anesthetic option for conventional AAA surgery in patients with severe COPD since it can preserve spontaneous breathing and provide additional respiratory benefits over general anesthesia.

Modeling prediction of membrane bioreactor process with the concept of soluble microbial product.

The existence of soluble microbial products (SMP) produced by microbial cultures involved in biological wastewater treatment process has been widely investigated. This paper aims to establish an available mathematical model by incorporating the SMP concept into the Activated Sludge Model (ASM) No. 3. Prediction of sewage treatment performance in membrane bioreactor process under intermittent aerobic condition by model simulation was presented, and the results provided a more comprehensive image for this process. It was found that SMP could not be ignored and it contributed about 15% of total COD in the reactor under HRT = 12 hr and SRT = 10 days condition. The model also provided reasonable simulation results for nitrogen, biomass concentration, and other treatment behaviors. Furthermore, the treatment performance can be predicted under various operating conditions by this proposed model.

Monitoring of shallow sea environment by using snapping shrimps.

We examine the validity of pulse count of snapping shrimps for sea environmental monitoring. Snapping shrimps, which make a peculiar pulse sound, are found everywhere in the world. Pulse count can be achieved merely by recording their sounds for a few minutes by using a hydrophone without special biological knowledge. From field surveys and laboratory experiments, we found that the pulse count depends on water temperature when sea environment is normal, and it falls due to the occurrence of oxygen-deficient water. The results show that this method can be a useful index of the effect of water pollution on benthic animals in fixed-point observation.

D-amphetamine does not improve outcome of somatosensory training.

BACKGROUND: D-amphetamine has been shown to affect early stages of stroke recovery, and may have a beneficial effect on functions when administered later after stroke. OBJECTIVE: To test D-amphetamine effects on skill acquisition after the acute or subacute stages of stroke, when lesion-related structural changes have consolidated. METHODS: Sixteen healthy subjects were treated with D-amphetamine during a 4-week training of tactile frequency discrimination in a placebo-controlled, double-blind design. RESULTS: All subjects improved significantly in tactile temporal acuity. However, improvement did not differ in subjects treated with or without D-amphetamine. CONCLUSION: No beneficial effect of D-amphetamine on somatosensory training improvements was found in healthy subjects.

(13)C NMR investigation of the superconductor MgCNi(3) up to 800 K.

We report (13)C NMR characterization of the new superconductor MgCNi(3) [T. He et al., Nature (London) 411, 54 (2001)]. We found that both the uniform spin susceptibility and the spin fluctuations show a strong enhancement with decreasing temperature, and saturate below approximately 50 K and approximately 20 K, respectively. The nuclear spin-lattice relaxation rate 1/(13)T(1) exhibits typical behavior for isotropic s-wave superconductivity with a coherence peak below T(c) = 7.0 K that grows with decreasing magnetic field.

17O NMR study of q = 0 spin excitations in a nearly ideal S = 1 / 2 1D Heisenberg antiferromagnet, Sr2CuO3, up to 800 K.

We used 17O NMR to probe the uniform (wave vector q = 0) electron spin excitations up to 800 K in Sr2CuO3 and separate the q = 0 from the q = +/-pi / a staggered components. Our results support the logarithmic decrease of the uniform spin susceptibility below T approximately 0.015J, where J = 2200 K. From measurement of the dynamical spin susceptibility for q = 0 by the spin-lattice relaxation rate 1/T(1), we demonstrate that the q = 0 mode of spin transport is ballistic at the T = 0 limit, but has a diffusion-like contribution at finite temperatures even for T<<J.

Elevated sympathetic nervous activity in mice deficient in alphaCGRP.

alpha-Calcitonin gene-related peptide (alphaCGRP) is a pleiotropic neuropeptide implicated in a variety of physiological processes. To better understand the biological functions of alphaCGRP, we developed an alphaCGRP-null mouse model using a gene targeting approach. Recordings of mean arterial pressure (MAP) and heart rate (HR) showed that basal MAP and HR were significantly higher in both anesthetized and conscious, unrestrained alphaCGRP-null mice than in corresponding wild-type mice. The elevated MAP in alphaCGRP-null mice was shown to be the result of elevated peripheral vascular resistance by alpha-adrenergic blockade with prazosin and by transthoracic echocardiogram, which revealed no significant differences between alphaCGRP-null and wild-type mice in the stroke volume, fractional shortening, and ejection fraction. Moreover, evaluation of autonomic nervous activity by measuring HR after pretreatment of atropine and/or atenolol and by analyzing arterial baroreceptor reflexes showed sympathetic nervous activity to be significantly elevated in alphaCGRP-null mice; elevated levels of urinary catecholamine metabolites and decreased HR variability in mutant mice were also consistent with that finding. These findings suggest that alphaCGRP contributes to the regulation of cardiovascular function through inhibitory modulation of sympathetic nervous activity.

Vascular abnormalities and elevated blood pressure in mice lacking adrenomedullin gene.

BACKGROUND: Adrenomedullin (AM) is a vasodilating peptide involved in the regulation of circulatory homeostasis and in the pathophysiology of certain cardiovascular diseases. Levels of AM are markedly increased in the fetoplacental circulation during pregnancy, although its function there remains unknown. To clarify the physiological functions of AM, we chose a gene-targeting strategy in mice. METHODS AND RESULTS: Targeted null mutation of the AM gene is lethal in utero: the mortality rate among AM(-/-) embryos was >80% at E13.5. The most apparent abnormality in surviving AM(-/-) embryos at E13.5 to E14.0 was severe hemorrhage, readily observable under the skin and in visceral organs. Hemorrhage was not detectable at E12.5 to E13.0, although the yolk sac lacked well-developed vessels. Electron microscopic examination showed endothelial cells to be partially detached from the basement structure at E12.5 in vitelline vessels and hepatic capillaries, which allowed efflux of protoerythrocytes through the disrupted barrier. The basement membrane was not clearly recognizable in the aorta and cervical artery, and the endothelial cells stood out from the wall of the lumen, only partially adhering to the basement structure. AM(+/-) mice survived to adulthood but exhibited elevated blood pressures with diminished nitric oxide production. CONCLUSIONS: AM is indispensable for the vascular morphogenesis during embryonic development and for postnatal regulation of blood pressure by stimulating nitric oxide production.

Significance of serum amyloid A on the prognosis in patients with renal cell carcinoma.

BACKGROUND: Evidence of systemic inflammation, i.e., elevation of serum C-reactive protein, interleukin-6, and/or the erythrocyte sedimentation rate, is correlated to poorer prognosis of patients with renal cell carcinoma (RCC). Serum amyloid A (SAA) has been recognized mainly as acute-phase reactant. METHODS: Serum SAA from 72 patients with RCC were examined. Thirty-eight of 72 patients with RCC had elevated SAA compared with 17 healthy donors. RESULTS: The disease specific survival rate was significantly lower in the elevated SAA group, and SAA level was shown to be an independent prognostic factor by univariate and multivariate analysis. CONCLUSIONS: Evaluation of serum SM level in RCC patients may be a useful prognostic indicator.

Modification of a single tryptophan residue in human Cu,Zn-superoxide dismutase by peroxynitrite in the presence of bicarbonate.

Human recombinant Cu,Zn-SOD was reacted with peroxynitrite in a reaction mixture containing 150 mM potassium phosphate buffer (pH 7.4) 25 mM sodium bicarbonate, and 0.1 mM diethylenetriamine pentaacetic acid. Disappearance of fluorescence emission at 350 nm, which could be attributed to modification of a single tryptophan residue, was observed in the modified enzyme with a pH optimum of around 8.4. A fluorescence decrease with the same pH optimum was also observed without sodium bicarbonate, but with less efficiency. Amino acid contents of the modified enzyme showed no significant difference in all amino acids except the loss of a single tryptophan residue of the enzyme. The peroxynitrite-modified enzyme showed an increase in optical absorption around 350 nm and 30% reduced enzyme activity based on the copper contents. The modified enzyme showed the same electron paramagnetic resonance spectrum as that of the control enzyme. The modified Cu,Zn-SOD showed a single protein band in sodium dodecyl sulfate--polyacrylamide gel electrophoresis (SDS--PAGE) and five protein bands in non-denaturing PAGE. From this evidence, we conclude that nitration and/or oxidation of the single tryptophan 32 and partial inactivation of the enzyme activity of Cu,Zn-SOD is caused by a peroxynitrite-carbon dioxide adduct without perturbation of the active site copper integrity.
