Distinction of the shape of Helicobacter pylori using stereo pairs constructed from digitized light microscopic images.

This study was performed to distinguish between the coccoid form or spiral forms of Helicobacter pylori (Hp) and to elucidate the pathologic significance of these shapes of Hp. Specimens obtained from human or Mongolian gerbil stomachs were fixed in Carnoy's solution and embedded in paraffin. Sections 3 or 10 microm thick were stained with polyclonal anti-Hp antibody by the immunoperoxidase method. Stereo pairs were prepared from these thick sections by computer-assisted reconstruction. The two shapes of Hp were easily distinguishable by this method. In the human stomach, the proportions of Hp in the surface mucous gel layer (SMGL) and the gastric pits were 31.5% and 68.5%, and the percentage of the spiral form varied from 31.9% to 66.3%. In Mongolian gerbils, a higher proportion of Hp colonized the SMGL, and the spiral form existed more frequently both in the SMGL and on the surfaces of the surface mucous cells.

[Training of personnel for work in home medical care and social welfare].

Our foundation initiated another home nursing station last year, bringing to 7 the number of social welfare institutions we are running (hospital, elderly, special elderly, day service, home support center, nursing station). We are trying to maintain close communication between the institutions and local community, but service in keeping with the demand is more necessary than ever. Moreover, in supplying medical care and welfare services, we are working harder for very close communication with those on the spot. Among employees the call is also for faster, more sure communication for dealing with matters on a "quality work" basis. From the earlier attitude of local medical and welfare institutions of "Bring them here to our hospital or facility," to "We'll come there," a definite change in thinking has begun to take over now. In the home care and Welfare office end of things, while "business ability and skill" are important as ever, "attitude" has become a large factor in doing the job among employees. The report gives actual examples of training of personnel to handle these various tasks with greater ability.

[Multidrug resistant P-glycoprotein expression on acute nonlymphocytic leukemia cells at diagnosis].

In performing cancer chemotherapy, it is essential to know the expression of multidrug resistant (MDR) P glycoprotein (p-gp) on cancer cells. In the present study, in order to clarify the relationship between MDR of leukemic cells and cytologic, immunological and clinical features of acute nonlymphocytic leukemia (ANLL), leukemic cells in peripheral blood and/or bone marrows obtained from 28 ANLL patients were examined. Each smear was stained with C219 monoclonal antibody against P-gp by the APAAP method, and then 1,000 ANLL cells in each smear were observed. Among the FAB subtypes, M4 showed the highest proportion of leukemic cells expressing P-gp. Concerning the response to chemotherapy, five of seven patients (71%) having 1.0% or more of P-gp positive leukemic cells and 11 of 19 patients (58%) having less than 1.0% of those cells achieved complete remission. However, there was no significant correlation between P-gp expression and clinical outcome. There was also no significant correlation between P-gp expression and CD7 or CD34. Furthermore, no significant correlation between chromosome 7 abnormality and P-gp expression was observed. From these results, if we can clarify the mechanism of MDR and the relationship between MDR and cytogenetic or clinical features of ANLL with further study, P-gp expression may become a useful marker for predicting the outcome of ANLL.

Preoperative measurement of segmental liver volume of donors for living related liver transplantation.

Segmental liver volume determination by computed tomographic scan was carried out preoperatively in nine donors for living related liver transplantation. The calculated volume was compared with the graft size actually obtained by three types of donor hepatectomy. The volume of the left lateral segment (175 to 241 ml) and the left lobe (310 to 490 ml) varied markedly among the donors. The ratio of the left lobar to total liver volume also showed a wide range of values (23.2% to 35.9%). The value of the left lobar volume did not correlate positively with the donor's body weight, suggesting that graft size cannot be predicted only on the basis of the donor's body size. Segmental graft liver volume was estimated by use of computed tomographic scan, with acceptable accuracy on comparison with the graft volume actually obtained. In living related liver transplantation, the type of donor hepatectomy should be selected on the basis of the segmental liver volume of the donor in addition to the recipient's body size so that liver failure can be prevented in recipients and the donor's safety can be assured as far as possible.

[Five cases of paroxysmal sleep disturbances].

Five cases with paroxysmal disturbances occurring during sleep were presented. The diagnosis of epilepsy was made in 3 cases and the diagnosis of non-epilepsy was made in 2 cases. The latter 2 cases had been treated as epilepsy. Clinical differences were not apparent on ictal manifestations between the epileptic and non-epileptic paroxysmal sleep disturbances. Seizures on awakening, which were noticed in only the epileptic cases, appeared 1 to 10 months later than sleep seizures. Final diagnosis was based on ictal EEG. Slow wave bursts were followed by alpha activity in non-epileptic cases. In epileptic cases, EEG revealed frontal dominant localized spike discharges which spread around in 2 cases and fast wave discharges in 1 cases.

The embryo pancreas is a source of increased yolk amylase in the fertilized eggs of domestic fowls.

The amylases were studied in the yolk of fertilized eggs and in the pancreases of the embryos of domestic fowls. The amylase activity in the yolk increased markedly from 13 days of incubation until hatching, but the activity decreased when the embryos were taken out of the eggs. The isoamylases in the yolk and in the pancreas of the embryo were identical electrophoretically. The amylase occurs mainly in the pancreas of the embryo. We think that the increase in amylase activity in the yolk of fertilized eggs during incubation depends upon the accumulation of pancreatic amylase synthesized by the developing embryo in the egg.

Effects of prolonged parotid duct ligation, parotidectomy and acute hepatitis of rats on amylase activity.

Amylase activity in the parotid gland was extremely reduced (to 2.4 per cent) by prolonged duct ligation but that in the pancreas and serum as similar to controls. The serum amylase activity of parotidectomized rats was remarkably reduced within a week of surgery, but recovered to approximately the previous level in 4 weeks. The amylase activities in the pancreas and liver were not different between control and parotidectomized rats. The serum amylase activity of rats with acute hepatitis (1 day after administration of D-galactosamine) decreased to 18.5 per cent compared with controls; however, the activity recovered to its original level after 4-7 days. There was no pancreatic amylase in the serum either of parotidectomized rats or those with acute hepatitis. These observations show that the parotid glands may not be the only source of serum amylase. Serum amylase is mainly derived from the parotid gland, but liver may contribute to serum amylase in compensation for the parotid glands.

Effects of propolis on dental caries in rats.

Propolis, the resinous hive product collected by bees, is important in the defense of the hive. The effects of propolis on growth and glucosyltransferase activity of Streptococcus sobrinus 6715, Streptococcus mutans PS14 and Streptococcus cricetus OMZ61 in vitro, and on dental caries in rats infected with S. sobrinus 6715 were investigated. Propolis had antimicrobial activity against S. sobrinus, S. mutans and S. cricetus, and inhibited both water-insoluble glucan synthesis and glucosyltransferase activity. In rats inoculated with S. sobrinus, about half of their fissures were carious, while dental caries was significantly less (p = 0.01) in rats given propolis. Dental caries was markedly decreased by the multiple actions of propolis which had antimicrobial activity, inhibited water-insoluble glucan synthesis, and inhibited glucosyltransferase activity. No toxic effects of propolis on the growth of rats were observed under experimental conditions in this study. These results suggest that propolis can control dental caries in the rat model system.

Amylase activity increases in the yolk of fertilized eggs during incubation in chickens.

Amylase activity was detected in the egg of chickens. In unfertilized eggs, no change of amylase activity was observed during 22 days incubation, however, in fertilized eggs, amylase activity increased markedly during embryonic development. The increase depended upon an increase of amylase in the yolk, not in the albumen. Isoamylases in the yolk were electrophoretically identical to those in the pancreas of the embryo or the hen.

An electrophoretic polymorphism in salivary amylases (Amy-1) of mastomys (Praomys coucha).

An electrophoretic polymorphism of salivary amylases (Amy-1) in mastomys (Praomys coucha) (MWC, MRJ and MCC strains) was detected. Amylase in MWC or MRJ saliva, which migrated fast toward the anode, was designated as AMY-1A, and that in MCC saliva migrating slowly as AMY-1B. Salivary amylases are controlled by a pair of codominant alleles at a single autosomal locus (Amy-1). No polymorphism was seen in pancreatic amylases (Amy-2). The frequencies of these phenotypes did not differ between the sexes. Some isoamylases were observed and these were different from those in mouse or rat.

Intravenous injection of caerulein or cholecystokinin increases the parotid-type and pancreatic-type amylase in the serum of rats.

Caerulein and cholecystokinin (CCK) injection (i.v.) into rats caused about a 2.3- and 1.9-fold increase in serum amylase activity within 2 h, respectively. The increase of amylase activity in the serum caused by pilocarpine or isoproterenol injection (i.p.) was only of parotid-type amylase. Caerulein or CCK markedly increased pancreatic-type amylase, and significantly increased parotid-type amylase in the serum.

Effects of prolonged duct ligation of the rat salivary glands on the activity of trypsin-like protease.

The effects of ligation on body weight, protease activity and hair kynurenine content were observed. The body weights of parotid gland(PT)-ligated and of combined submandibular (SM)-, sublingual (SL)- and PT-ligated rats were much less than those of sham-operated controls. However, the weights of SM,SL-ligated rats were similar to those of controls. The activity of trypsin-like protease in the duct-ligated submandibular gland was significantly reduced to 0.3-0.7 per cent of that in controls. Protein concentration also decreased in the duct-ligated submandibular gland. Kynurenine content in the hair of duct-ligated rats was higher than that in controls.

Relationship between serum-amylase activity and intraductal pressures in the rat parotid and submandibular salivary glands after administration of pilocarpine or isoprenaline.

Ligation of the parotid ducts induced marked elevation of serum-amylase activity after administration of pilocarpine or isoprenaline. Secretion of saliva and serum-amylase activity increased most rapidly in the first 15 min after injection of pilocarpine, whereas isoprenaline induced a constant rate of secretion and serum-amylase activity over 120 min. Amylase activity in the serum was closely connected with intraductal pressure due to saliva secretion.

Electrophoretic behavior of amylase in mouse: the parotid gland is major source of serum amylase.

Amylase activity in the saliva, salivary glands, serum, liver (perfused and non perfused) and pancreas was assayed and isoamylases were separated by electrophoresis in these organs using C57BR/cdJ and M. m. molossinus (Kor) mice. Amylase isozymes in the saliva, parotid gland, serum and liver were identical in both strains, respectively. Amylase activity in the liver was lower than that in the serum and liver amylase disappeared almost by perfusion. Major serum amylase was released from the parotid gland in intact animals.

The activity of alkaline phosphatase in the sera of DBA/2NCrj and CBA/BrA mice with calcified tongue lesions.

Spontaneously occurring calcified lesions were found in the tongues of DBA/2NCrj and CBA/BrA mice. In the DBA/2NCrj strain, the frequency of the lesion was 80% (males) and 88% (females). The youngest age of a mouse with this lesion was 18 days after birth, and 3-4 lesions were found in the tongue of 6- to 8-week-old mice. In CBA/BrA mice, 20% of females and 48% of males had the lesions. No significant differences were found in the serum calcium concentrations in high and low lesion-developing strains, but the alkaline phosphatase activities in the high-developing DBA/2NCrj, DBA/LiA, and CBA/BrA strains were higher than in strains with no calcified lesions.
