The enzymatic properties of plasma membrane-bound Na+, K+-ATPase [EC 3.6.1.3], isolated with high specific activity and in good yield from pig thyroid cells, were examined. The enzyme activity required the presence of both Na+ and K+ at physiological concentrations; it exhibited high sensitivity to K+ and an absolute requirement for Na+. It showed highly specific requirement for Mg2+ and ATP. The apparent Km for ATP was 0.14 mM under the assay conditions. Arrhenius plots had a point of inflection at about 22 degrees C, activation energies being 24.2 kcal/mol at 5-22 degrees C and 19.0 kcal/mol at 22-40 degrees C. In addition to ouabain, the ATPase was strongly inhibited by fluoride and the SH-blocking reagent, PCMB. Iodide and TSH had no appreciable effect on the enzyme activity.
The testosterone-binding globulin (TeBG) from bovine serum was purified by affinity chromatography and hydroxylapatite chromatography. The affinity column used was prepared by coupling 17 alpha-carboxyethynyl-17-hydroxy-4-androsten-3-one to aminoethyl-Sepharose. The compound was replaceable by 17alpha-carboxyethynyl-17-hydroxy-5alpha-androstan-3-one, but not by testosterone 17-hemisuccinate, estradiol 17-hemisuccinate, or testosterone 3-(O-carboxymethyl)oxime. The TeBG isolated was homogeneous on analytical polyacrylamide gel electrophoresis and equilibrium centrifugation. The protein was a glycoprotein having a molecular weight of 89,500 and a carbohydrate content of 17%. The association constant (M-1) at 4 degrees C was 1.1 X 10(8) and the number of binding sites per molecule was 0.8. Treatment with guanidine-HCl dissociated the protein into subunits having a molecular weight of 28,400 (about one-third of that of the original molecule). SDS-gel electrophoresis showed that two of the three subunits were slightly larger than the other. The dissociation into subunits could also be accomplished by GEDTA treatment with concomitant loss of testosterone-binding activity. The activity and molecular size were reversibly restored by incubation with excess Ca2+.

The total and specific activity of thyroid homogenate of normal rat was 0.95 times 10- minus 3 GU (guaiacol unit) per rat and 0.61 times 10- minus 3 GU per mg protein. These were increased 3.4 fold and 1.6 fold by chronic administration of thyroid stimulating hormone (TSH) and 5.1 fold and 2.5 fold by treatment with methylthiouracil (MTU), respectively. However, the soluble fractions, which were obtained by centrifugation at 105,000 xg for 120 min from the homogenates, manifested only 2-3% of the activity of the original homogenates, and the specific activity was scarcely affected by the administration of TSH and MTU. Moreover, even the low activity of the soluble fraction might be due to contaminated microsomes. None of the results obtained favors the view that the peroxidase is released from the cell to the colloid lumen. The number of peroxidase molecule per cell of thyroid was roughly estimated to be 1.5 times 10- minus 5 for normal and 3.9 times 10- minus 5 for MTU-treated rat.

Three-dimensional magnetic resonance imaging (3D MRI) using a multiplanar reconstruction technique with gadolinium diethylenetriamine pentaacetic acid (DTPA) enhancement was used for assessment of a 20-year-old woman who had sudden complete restriction of abduction in the right eye. The 3D MRI revealed abducens nerve enhancement in the cisternal portion. An enhanced lesion was also observed in the right lower pons at the pontomedullary junction. These clinical and 3D-MRI findings were diagnosed as right abducens palsy due to abducens neuropathy. Observation of the entire pathway of the cisternal portion of the cranial nerves can be extremely useful in patients with ophthalmoplegia.
PURPOSE: To study the radiation scattering associated with the digital subtraction angiography (DSA) unit in angiographic procedures and to design an effective radiation protection shield based on these data. MATERIALS AND METHODS: The number of scattered photons was measured at three points relative to the operator's position. Anteroposterior abdominal and lateral cranial fluoroscopy were evaluated. As protective devices, a lead curtain, sliding shields, and a brim-shaped image intensifier (II) hood were designed. RESULTS: In abdominal fluoroscopy, radiation was found to scatter to the operator's lower limbs from the underside of the catheter table, to the abdomen from the side of the patient's body, and to the head and neck from the table surface adjacent to the patient. The use of protective devices reduced exposure from 2.89 to 0.058 mR/min for the operator's lower limbs, from 0.987 to 0.069 mR/min for the operator's abdomen, and from 0.696 to 0.139 mR/ min for the operator's head and neck area. With lateral cranial fluoroscopy, radiation was detected to scatter to the operator's lower limbs from the underside of the catheter table, to the abdomen from the patient's temporal area, and to the head and neck from the patient's face. The use of protective devices reduced exposure from 0.248 to 0.010 mR/min for the operator's lower limbs, from 0.129 to 0.010 mR/min for the operator's abdomen, and from 0.162 to 0.018 mR/min for the operator's head and neck area. CONCLUSIONS: The characteristic directions of scattering to the operator were identified. An effective modified radiation protection system was designed based on this information.
