Malonylated anthocyanins from bulbs of red onion, Allium cepa L.

Four cyanidin-based anthocyanins (1-4) were isolated from the red onion, Allium cepa L. Pigments 1 and 3 were identified as cyanidin 3-glucoside (Cy 3-Glc) and 3-malonylglucoside (Cy 3-MaGlc), respectively, by cochromatography with standard pigments. Anthocyanins 2 and 4 were respectively determined as cyanidin 3-laminaribioside (Cy 3-Lam) and 3-malonyllaminaribioside (Cy 3-MaLam), a new anthocyanin, mainly by NMR techniques. Malonylated anthocyanins 3 and 4 were found for the first time in red onions.

The role of cell-mediated immunity in chickens inoculated with the cell-associated vaccine of attenuated infectious laryngotracheitis virus.

The cell-mediated immune responses of the chickens inoculated with the cell-associated (CA) ILT vaccine were studied. Lymphocyte blastogenic response was tested by MTT assay with spleen, thymus, bursa of Fabricius and peripheral blood lymphocytes of the vaccinated chickens. The blastogenic response of peripheral blood and spleen lymphocytes was enhanced by a T-cell mitogen. The CA vaccine induced an immunity satisfactorily in bursectomized chickens. To study the role of immune cells in the vaccinated chickens, the bursectomized chickens were inoculated intravenously with the splenic, thymic, bursal and peripheral blood lymphocytes of the vaccinated chickens. Protective effects were shown in the chickens received the splenic cells and peripheral blood lymphocytes, indicating that these play an important role in eliciting the protective effects of the CA vaccine. From these results, it was found that the immune mechanism with CA vaccine against ILT involves mainly cell-mediated immunity.

Immune response and in vivo distribution of the virus in chickens inoculated with the cell-associated vaccine of attenuated infectious laryngotracheitis (ILT) virus.

Chickens inoculated with the cell-associated (CA) vaccine acquired higher protective immunity to ILT. In chickens vaccinated with CA or cell-free (CF) vaccine, respectively, virus-neutralizing and IgG- and IgM-ELISA antibodies were detected in the serum, but no antibody was detected in the tracheal washes of the vaccinated chickens. More apparent antibody response was seen in chickens vaccinated with the CA vaccine than with the CF vaccine. The antibody titers did not correlate closely with the protection against challenge with ILT virus. After subcutaneous injection of either CA or CF vaccine, ILT virus was isolated from the liver, spleen, thymus, lungs and other organs of the chickens from 1 to 6 days after injection, and there was no correlation of the isolation rate for the CA-vaccinated and the CF-vaccinated chickens.

[52-week oral toxicity study of lactitol (NS-4) in dogs followed by 9-week recovery test].

Five male and 5 female beagle dogs were orally given lactitol, a hepatic encephalopathy drug, for 52 weeks at doses of 0, 0.25, 1.25 or 6.25 g/kg/day. A 9 week recovery test was conducted after the discontinuation of the drug treatment. Soft stool, diarrhea, and vomiting were seen in the 1.25 and 6.25 g/kg groups. In the 6.25 g/kg group, bloody stool and increased water consumption were also observed. Urinalysis showed larger amount of the urine volume in the 6.25 g/kg group. The cecum weight of this group was increased without any morphological changes. There were no drug related effects on survival, body weight gain and food consumption. Electrocardiographic, ophthalmoscopic, hematologic and biochemical examinations failed to show any abnormalities related to the drug treatment. The above mentioned changes were satisfactorily reversible. Based on the results obtained, the NOAEL of this study was suggested to be 0.25 g/kg/day.

[The influence of aging on cerebral blood flow and oxygen metabolism during moderate hypothermic cardiopulmonary bypass--a clinical study by means of transcranial Doppler ultrasound].

The purpose of this study was to determine the influence of aging on cerebral blood flow and oxygen metabolism during moderate hypothermic cardiopulmonary bypass (CPB) in 25 patients undergoing open heart surgery. the patients were subdivided into one of two groups according to their age (the cut off value was 40 years for males and 50 years for females). Ten patients were included in group A (YOUNG group), and 15 patients were included in group B (OLD group). In these 25 patients, blood flow velocity in the middle cerebral artery (MCAv) was measured by means of transcranial Doppler ultrasound, and cerebral oxygen consumption was estimated by relating the arterio-venous oxygen content difference to flow velocity (D-CMRO2) during surgery. MCAv and D-CMRO2 were expressed as percent of the values determined at 30 minutes before CPB. High dose fentanyl anesthesia was employed and alpha-stat strategy (uncorrected for body temperature), moderate hemodilution and nonpulsatile flow pattern were maintained during CPB with moderate hypothermia. In both groups, MCAv and D-CMRO2 changed in parallel in proportion to the change in body temperature during the procedure. Thus, there was a good correlation between flow velocity and D-CMRO2 during CPB (group A: r = 0.648, p < 0.001, B: r = 0.636, p < 0.001). This result suggests that the coupling between cerebral blood flow and oxygen consumption was maintained throughout CPB in spite of aging. In group B, however, the usual of significant increase in the flow velocity and D-CMRO2 from the rewarming to the weaning stage of CPB tended to be delayed and suppressed.(ABSTRACT TRUNCATED AT 250 WORDS)

[A case of lung metastasis from Meibomian gland carcinoma of eyelid with effective chemotherapy].

A 47-year-old man was admitted to Shinshu University Hospital in 1992 because of cough and chest abnormal shadow. In 1987, he had an operation for Meibomian gland carcinoma of his right lower eyelid, and also received radiotherapy. His chest X-ray film on admission showed tumor shadows in the bilateral upper lung fields. The chest CT scan showed tumors in the right S1 and the left S3. Bronchofiberscopic findings demonstrated stenosis of the right upper bronchus and an endobronchial mass occluding the left B3. A biopsy specimen of the endobronchial mass revealed sebaceous carcinoma, which was identical with that of the resected eyelid tumor. He received two courses of chemotherapy in a combination of CDDP 75 mg/m2 and ADM 50 mg/m2. The tumors on chest X-ray and CT scan became small by the chemotherapy. Bronchofiberscopy after the chemotherapy also revealed that the stenosis of the right upper bronchus improved and the endobronchial mass in the left B3 had disappeared. He died 8 months after initial chemotherapy with a response duration of 7 months.

[Capillary endothelial cells in sarcoidosis--electron microscopic findings].

The distribution of von Willebrand (vW) factor, thrombomodulin (TM) and fibronectin (FN) in the capillary endothelial cells of the lung of patients with sarcoidosis was investigated using their monoclonal or polyclonal antibodies. The vW factor showed strong reaction with exudation towards outside and irregular dilatation or occlusion in the capillary walls surrounded sarcoid granulomas and capillaries in the remote area. The area showing alveolitis disclosed also strong reaction with exudation by vW factor. TM having a anti-coagulating function showed a very low reactivity in the lung tissues with or without of granulomas in sarcoidosis at light microscopic level, but in electron microscopically, strong reaction and broad distribution in capillary endothelial cells, involving luminal occlusion and reaction in subendothelial region. FN showed new and many capillaries formation in the growing collagenous tissues between the old sarcoid granulomas suggesting neocapillarization originating from endothelial cells. These findings suggest that the capillary endothelial cells participate strongly in formal genesis of sarcoid granulomas.

[Clinicopathological study of 4 patients with idiopathic BOOP, interstitial type on chest X-ray].

We studied four patients with idiopathic Bronchiolitis Obliterans Organizing Pneumonia (BOOP) diagnosed by open lung biopsy. All of four presented with acute onset and a rapid course with progressive dyspnea. Their chest X-ray films showed ground-glass opacities with reduced volumes in the bilateral lower lobes. Their chest CT revealed marked increases in lung density with air bronchograms. Radiologically, it was difficult to distinguish these cases from patients with acute interstitial pneumonia. Histologically, in the uninflated specimens, the collapse of alveoli and alveolar spaces filled with foamy macrophages and proteinous exudates were observed. The organizing tissues were present predominantly in the alveolar ducts, but varied in extent among cases. We considered the reduced lung volumes on chest X-ray films and the marked increases in lung density on chest CT to reflect alveolar collapse. Proliferation of type II pneumocytes was remarkable in all cases, but reactivity to antisurfactant protein-A antibody staining was decreased in the collapsed alveoli. It was suggested that the alveolar collapse was caused, in part, by decreased pulmonary surfactant in addition to obstruction of the alveolar ducts. The rapid responses to of these patients corticosteroid therapy may be due to improvement of the alveolar collapse.

MR of pulmonary hamartoma: pathologic correlation.

We describe the magnetic resonance (MR) appearance with pathologic correlation of six pulmonary hamartomas. All six tumors showed intermediate signal intensity on T1-weighted images and high signal intensity on T2-weighted images. Four of the six tumors showed a lobulated appearance separated by septa on precontrast T1-weighted or T2-weighted images. Gadolinium-enhanced T1-weighted images showed marked enhancement of the septa that separated the tumors into less well enhanced lobules; this was seen in all six tumors. Comparison between MR images and pathologic specimens showed that the regions with less enhancement corresponded to core cartilaginous tissue and septa; areas of marked contrast enhancement corresponded to cleft-like branching mesenchymal connective tissue that dipped into the cartilaginous core. MR images correlated well with the surgically resected tumors.

Ultrasonography of thymoma with pathologic correlation.

Preoperative ultrasonograms of 11 surgically proved thymomas and ex vivo ultrasonograms of 3 resected specimens were compared with pathologic findings of resected specimens. Among 11 thymomas 7 appeared solid, 3 were solid with several cystic regions, and the remaining one was unilocular cystic in appearance. Cystic regions on ultrasonograms corresponded to cystic changes on pathologic specimens. Six malignant thymomas showed a typical lacy appearance on ultrasonograms, which corresponded to the lobulated configuration separated by fibrous septa shown on the pathologic specimens.

Kinetic characterization of a stably expressed novel Na+/H+ exchanger (NHE-2).

We have recently reported the molecular cloning, sequencing and tissue distribution of a novel Na+/H+ exchanger (NHE-2). The cDNA for NHE-2 was cloned by screening a rat intestinal cDNA library. This clone was unique due to the fact that it lacks the first two transmembrane domains which are present in the other Na+/H+ exchanger isoforms (NHE-1, NHE-3, NHE-4). This structural change in the cDNA offered a unique opportunity to study in detail the properties of this stably expressed cDNA in chinese lung fibroblasts that lack the Na+/H+ exchanger (PS120) cells. Amiloride-sensitive Na+ uptake was linear up to 2 min in PS120 cells transfected with the cDNA. Kinetics of the amiloride-sensitive Na+ uptake showed a Vmax of 24.7 +/- 5 nmol/microliters ICW per min and a Km of 33.1 +/- 2.0 mM. The inhibitory constant (KI) for amiloride and its analogue 5-N-ethyl-N-isopropylamiloride (EIPA) was 0.15 microM and 0.66 microM, respectively. Epidermal growth factor, a known stimulator of NHE-1, also upregulated the expressed NHE-2. These results characterize the kinetic properties of this unique exchanger and suggests that the first two transmembrane domains of the Na+/H+ exchanger isoforms are not essential for the expression of amiloride-sensitive Na+ uptake.

Quantitative analysis of central terminal projections of visceral and somatic unmyelinated (C) primary afferent fibers in the guinea pig.

In guinea pigs, intracellular labeling of the dorsal root ganglion (DRG) cells with Phaseolus vulgaris-leucoagglutinin (PHA-L) was used to demonstrate the central projections of somatic and visceral afferent C-fibers. The terminations of the afferent fibers were analyzed qualitatively and quantitatively with the aid of camera lucida drawings. Terminal branches of C-fibers of both somatic and visceral origin were, in general, distributed in accord with the organization of the neuropil in lamina of the spinal cord. Terminal boutons arranged from longitudinally coursing fibers were distributed in lamina I, while boutons in lamina II were scattered in an apparent random fashion. The synaptic enlargements were counted in gray matter of the spinal dorsal horn and measured on each terminal branch of a fiber. All synaptic boutons (over one thousand) of somatic fibers were found in the superficial dorsal horn (laminae I and II). More than 60% of the synaptic enlargements of the visceral afferents also were localized superficially (lamina I and adjacent dorsal funiculus) while 10-20% of the visceral enlargements appeared in deeper layers of the spinal cord. Boutons of somatic C-fibers were larger than those of visceral origin. Quantitative data of the unmyelinated afferent fibers are discussed in the context of the sensory functions of myelinated afferent fibers.

Molecular cloning, sequencing, tissue distribution, and functional expression of a Na+/H+ exchanger (NHE-2).

The present studies demonstrate cloning, sequencing, tissue distribution, and functional expression of a Na+/H+ exchanger which was isolated from a rat intestinal cDNA library. The cloned cDNA recognizes two transcripts in poly(A)+ RNA from the stomach, jejunum, ileum, liver, large intestine, and uterus. Based on deduced amino acid sequences, this clone shares sequence homology with the other known Na+/H+ exchanger isoforms (NHE-1, NHE-3, and NHE-4) except for its 5' end. Overall, the protein exhibits 47.8%, 41.2%, and 56.2% amino acid sequence identity to NHE-1, NHE-3, and NHE-4, respectively. The hydropathy profile of the predicted protein shows 10 transmembrane domains, suggesting a protein with transport characteristics. The tissue distribution differs from that of the other Na+/H+ exchanger isoforms. The cDNA hybridizes to two closely related transcripts in the mRNA of these tissues, which suggests that the predominant transcript of this clone is alternatively spliced. Transfection of this cDNA into Na+/H+ exchanger-deficient mutant fibroblasts (PS120 cells) results in functional Na+/H+ exchange activity. These data suggest that we have cloned a member of the Na+/H+ exchanger family with tissue-specific expression. We suggest the designation of NHE-2 for this Na+/H+ exchanger.

Involvement of p53 gene mutations in human endometrial carcinomas.

Mutations in the p53 gene are associated with a wide variety of human malignancies. Point mutation in one allele and loss of the remaining one generally lead to inactivation of p53 protein. A high frequency of allelic losses corresponding to the 17p13.3 region that contained the p53 gene sequence was also noted in human endometrial carcinoma. Thus, in order to confirm involvement of the p53 gene in endometrial carcinogenesis, we searched for nucleotide sequence change in this gene in 42 endometrial carcinomas that had been subjected to previous LOH analyses. Using the polymerase-chain-reaction-single-strand conformation polymorphism (PCR-SSCP) method, we detected p53 gene mutations in 4 specimens. Two adenocarcinomas with allelic loss on 17p contained a mutant p53 gene in the allele that was retained. One specimen with a p53 gene mutation contained a 17q deletion but was uninformative for LOH on 17p. p53 gene mutation was also noted in the remaining stage-I carcinoma, though the 17p deletion was not detected in the previous LOH examination. However, 5 specimens with the LOH on 17p retained the wild-type p53 gene. In the remaining 33 specimens, both alleles of p53 gene seemed to be normal. The mutations observed in 2 specimens (cases 10 and 24), involving C-to-T and T-to-G substitutions, were located in a highly conserved region. However, the mutations identified in the remaining 2 cases (29 and 35) were at regions positioned outside conserved stretches.
