Ras oncogene expression and progression in intraepithelial neoplasia of the uterine cervix.

To examine the correlations between ras oncogene expression and the development of cervical cancer, the authors studied the reactivity of cervical intraepithelial neoplasia (CIN) and microinvasive lesions of the human uterine cervix by using anti-ras p21 mouse monoclonal antibody rp35. The frequency of positive p21 staining increased with increased grades of malignancy from 17.9% in CIN 1 to 28.9% in CIN 2 and 53.9% in CIN 3, whereas in microinvasive carcinoma it was 50.0%. Furthermore, ten cases of lesions that regressed during a 1-year follow-up period were positive for ras p21 in 20% of cases, but 14 cases of lesions that progressed and developed into higher graded lesions during the 2- to 5-year follow-up period had a 50.0% rate of positive p21 staining. It was concluded that ras oncogene product p21 correlates with the early phase of carcinogenesis of squamous cells of the uterine cervix.

Properties of membrane currents in isolated smooth muscle cells from guinea-pig trachea.

Tracheal smooth muscle cells were enzymatically isolated from guinea-pig trachea. These cells contracted in response to acetylcholine (0.01-10 microM) in a concentration-dependent fashion. Under current-clamp conditions with 140 mM K+ in the pipette solution, the membrane potential oscillated spontaneously at around -30 mV. Under voltage-clamp conditions, there appeared spontaneous but steady oscillations of outward current (IO). On depolarization from a holding potential at -40 mV, three components of outward current were elicited: transient outward current (IT), steady-state outward current (IS) and IO. These three components of outward current reversed around the K+ equilibrium potential and were abolished by Cs+ in the pipette, indicating that K+ was the major charge carrier of these outward currents. All these three components were completely suppressed by extracellular tetraethylammonium (10 mM). Both IT and IO were depressed by quinidine (1 mM), 4-aminopyridine (10 mM) and nifedipine (100 nM), but IS was not affected. IT and IO were suppressed by a Ca2(+)-free perfusate with less than 1 nM Ca2+ in the pipette, while with 10 nM Ca2+ in the pipette, only IO was suppressed. In both conditions, IS was not affected by the Ca2(+)-free perfusate. Therefore, it is suggested that IO, IT and IS are separate types of K+ current. With Cs+ in the pipette, K+ currents were almost completely suppressed and a transient inward current was observed during depolarizing pulses. The inward current was not affected by tetrodotoxin and increased when the concentration of extracellular Ca2+ was raised, indicating that the current is a Ca2+ channel current. Even with a holding potential of -80 mV, the low-threshold inward current could not be observed. The high-threshold Ca2+ current was abolished by nifedipine (100 nM) and was enhanced by Bay K 8644 (100 nM). The order of permeation of divalent cations through the Ca2+ channel was Ba2+ greater than Sr2+ approximately Ca2+. Cd2+ blocked the Ca2+ current more effectively than Ni2+. These results may indicate that the Ca2+ current of tracheal smooth muscle cells is mainly composed of the current through an L-type Ca2+ channel.

ras oncogene expression and prognosis of invasive squamous cell carcinomas of the uterine cervix.

By using anti-ras p21 mouse monoclonal antibody rp35, the authors studied the reactivity of 170 squamous cell carcinomas of the uterine cervix of different histologic types. The overexpression of p21 was noted in 57.1% of keratinizing type (K type) and 54.2% of large cell nonkeratinizing type (LNK type), but only 38.7% of small cell type (S type). Upon statistical analysis of the correlations between p21 overexpression and patient prognosis with the Kaplan-Meier method and generalized Wilcoxon text, the p21-positive K and LNK types showed poorer prognosis, whereas the positive S type showed better prognosis in comparison with the negative cases. These findings suggest that the expression of ras p21 is one of the parameters related with prognosis of cervical cancers, but the mode of the correlation is dependent on their histologic types.

[Acute myeloblastic leukemia development in a patient with small cell lung cancer in complete remission].

A 67 year-old man was admitted to our hospital because of cough and sputum. He smoke one pack of cigarettes a day for more than twenty years and the chest X-ray film revealed a mass in the left hilum and left sided pleural effusion. The diagnosis of small carcinoma of the lung (limited disease, T4N1MO, stage 3B) was made by trans-bronchial lung biopsy and radiographic studies. Both chemotherapy (nimustine (ACNU), cyclophosphamide, vincristine, and methotrexate) and radiation therapy was started, however, the chemotherapy was discontinued in July 1987 because of severe anemia. The diagnosis of refractory anemia with excess of blasts in transformation (RAEB in T) was made by bone marrow aspiration and the patient was treated by transfusion (400-800 ml/week). In December 1987 transition to acute myeloblastic leukemia was confirmed by another bone marrow aspiration biopsy and the patient was given low dose cytosine arabinoside (Ara-C). The response was favorable in the beginning but in about two months pancytopenia became refractory and the patient died in June 1988. Clinically there was no sign of local or distal recurrences of lung cancer, and the complete remission of small cell lung cancer (SCLC) was confirmed by autopsy. Survival in SCLC remains poor, so that the choice of treatment is still the primary concern, however, development of other malignancies which include acute leukemia is another problem which should be taken into account when the treatment is extensive.

The evaluation of therapeutic efficacy of hachimi-jio-gan (traditional Chinese medicine) to mouse hereditary cataract.

This study was of a series of the evaluation of Hachimi-Jio-Gan (Rehmannia Eight Formula, pa-wei-ti-huang-wan or Bawei dihuang wan) to the various cataracts. In this study, the drug was evaluated for its therapeutic efficacy to mouse hereditary cataract from the delay effect of cataract appearance age and the suppression rates of variation of some biochemical parameters. The dose of 200 mg of Hachimi-Jio-Gan/day/100 g of mouse body weight significantly delayed the cataract appearance age by 4 days as compared to that of non-treated group. We estimated that the delay effect of 4 days in mouse may be corresponded to 13.9 years, when it was converted into the case of human. This drug also suppressed variation of sodium and potassium ions level in the lens with cataractogenesis. Furthermore, the drug dramatically reactivated the sodium-potassium ATPase activity damaged with the cataract formation, and also had a slight action of reducing agent. From these facts, we presumed that the drug may have a prophylactic efficacy to the cataract caused by the inhibition of sodium-potassium ATPase activity and also the oxidation of lens protein.

Relationship between forced expiratory flow and tracheal sounds. Possible effect of vortices on flow.

We measured both tracheal sounds and forced expiratory flows simultaneously in 20 normal subjects and 4 patients with stenosis of the main bronchus. Spectral analysis of the tracheal sounds revealed development of discrete line spectra, the frequencies of which ranged between 488 and 942 Hz. There was a positive correlation between these frequencies and the peak flows (r = 0.601). We speculate that the origin of these line spectra are vortices rather than the vibrations of the wall. It is postulated that the pressure drops caused by vortices make the airways more susceptible to collapse and may lead to flow limitation.

The evaluation of therapeutic efficacy of hachimi-jio-gan (traditional Chinese medicine) to rat galactosemic cataract.

Hachimi-jio-gan (Rehmannia Eight Formula, pa-wei-ti-huang-wan or Bawei dihuang wan) is one of traditional Chinese medicines which has been used for treating various senile disease for a few hundred years. This drug was evaluated for its therapeutic efficacy to rat galactosemic cataract from the suppressive rate of variance of some biochemical parameters, whose variation with cataractogenesis or the advance of cataract have been reported already. The dose of 500mg of Hachimi-jio-gan/day/200g of rat body weight suppressed significantly the variations of hydration rate, Na/K ratios, and calcium ion level in the lens with the advance of galactosemic cataract, especially when the drug was administered by the pre- or concurrent-administration before or with 30% of galactose diet respectively. This drug also could delay the progressive rate of lens opacification. However, the drug had no effect to suppress the galactitol accumulation in the lens. From these results, we presume that a drug action of Hachimi-jio-gan may control the balance of sodium, potassium and calcium ions which are important in relation to the maintenance of lens transparency. Then, we realized, that this drug may have a prophylactic efficacy to diabetic cataract, though a more detail study should be needed to apply this drug to human cataract disease.

The application of microspectrocytofluorometric measurement of Feulgen nuclear DNA content to experimental tumors of rat submandibular gland. 1. Pathogensis and nuclear DNA content.

Pellets containing 9,10-dimethyl-1,2-benzanthracene (DMBA) were inserted into the submandibular glands of 65 male Fischer rats to determine the nuclear DNA content of both metaplastic epithelium during the process of development of squamous metaplasia (noted at 1, 2, 5, 7, 9, 10, 11, 13 and 15 weeks after insertion) and tumor cells of squamous cell carcinoma 24 weeks after insertion. From the second week of insertion onward, squamous metaplasia was found in excretory duct remaining epithelium in the tissues around the pellet. Dysplasia or a lining of metaplastic epithelium with atypia adjacent to pellets (i.e., formation of an epidermal cyst) was observed 5 weeks after insertion. Coincident with this dysplasia or metaplastic epithelium with atypia, variation in the nuclear DNA content was observed, showing the emergence of octaploids and a widespread histogram pattern with many peaks. Between 7 and 15 weeks, 25%-66% of the lining metaplastic epithelium showed variation in the nuclear DNA content due to shift of a peak (major mode) to a triploid as well as a vague major mode with the appearance of octaploids, etc. Tumors of the submandibular gland were recognized at 24 weeks in 11 (55%) of 20 rats. Nine were keratinized squamous cell carcinomas and the remaining 2 were carcinosarcomas. Marked abnormalities in the nuclear DNA content were observed in 5 of the 11 rats (45.5%), 3 with keratinizing squamous cell carcinoma and 2 with carcinosarcomas. In contrast, no marked variation in the nuclear DNA content was found in 5 rats in which the tumor was associated with cysts.

A histochemical study of APUD ability in the taste buds of experimentally induced zinc-deficient mice.

To explore the relationship between taste acuity and zinc deficiency, a histochemical investigation was made into the taste buds of mice fed a zinc-deficient diet. Nine weeks after the start of the diet, the average serum zinc level of the mice was 45% lower than that of a control group of mice. Moreover, growth was arrested significantly. Two-bottle preference tests revealed that the intake ratio of 10(-5) M quinine hydrochloride solutions had increased markedly in the zinc-deficient mice compared with the controls. The circumvallate taste buds showed no morphological changes. Fluorescent histochemical examination showed an uptake of a monoamine precursor (5-HTP) by the gustatory cells in the zinc-deficient mice after the 5-HTP treatment. Upon immunohistological examination, however, no serotonin immunoreactivity appeared in the gustatory cells of the zinc-deficient mice after the 5-HTP treatment. These results suggest that zinc-deficiency may induce hypogeusia and decrease the ability to transform a monoamine precursor to monoamine in the gustatory cells, albeit the monoamine precursor uptake ability is not affected.

The application of microspectrocytofluorometric measurement of Feulgen nuclear DNA content in experimental tumors of rat submandibular gland, 2. The correlation between proliferative ability and nuclear DNA content of tumor tissue in autotransplanted areas.

We have reported that there is a difference in the variation of the nuclear DNA content of tumor cells among cases of squamous cell carcinoma induced by 9,10-dimethyl-1,2-benzanthracene (DMBA) in the rat submandibular gland. In the present investigation, the relationship between the nuclear DNA content of tumor cells in autotransplanted sections collected from primary lesions of DMBA-induced tumors and their proliferative ability in the subfascial area of the rat abdomen was examined. As a result of autotransplantation, proliferation in the autotransplanted area was observed in 6 of 14 (42.8%) cases of autotransplantation. Five of these had a keratinizing squamous cell carcinoma while the remaining one had a sarcomatoid tumor. The histological type of the tumor of the proliferative lesion in the transplanted area was very similar to that of the tumor tissues in the primary lesion or the transplanted section collected from the primary lesions. In the proliferative group, marked variation of the nuclear DNA content was observed in the tumor cells of the transplanted section. The proliferative index (PI) was high for these tumor cells in this group. There was no variation in the nuclear DNA content in the tumor cells of the nonproliferative group, and the PI was also low. These results were considered to suggest that there was a correlation between the nuclear DNA content of these experimental tumor cell and their proliferative ability in the autotransplanted area. Therefore, the determination of nuclear DNA content by this method can be used as an objective index of the proliferative ability of tumor tissue.

Determination of DNA by sub- and super-equivalence method of isotope dilution analysis using enzyme reaction.

Sub- and super-equivalence method of isotope dilution analysis (SSE-IDA) using enzyme reaction was first applied for the determination of biological substance, DNA. Radioactive DNA (pUC18) to be analysed was prepared by incorporating 3H-thymidine in growing E. coli. A part of DNA was cut into the linear form (L-form) DNA under definite conditions using a restriction enzyme HindIII, following the separation of each by gel electrophoresis. Radioactivites of separated L-form DNA of two series were measured. The quantity of DNA was obtained by a graph method of SSE-IDA. As preliminary experiments, it was examined under what conditions the enzyme reaction proceeds as zeroth or first order reaction. We used the enzyme and substrate concentrations near zeroth order, where ordinary subst-IDA seems not to give a satisfactory results. As the results, 0.25 microgram of DNA was determined the error of about 10%.

The relationships between the Ga-67 uptake and nuclear DNA Feulgen content in thyroid tumors: concise communication.

It has been reported that Ga-67 uptake by malignant tumors differs somewhat according to the histologic type. Previously, we reported that uptake of Ga-67 is predictably low in well-differentiated adenocarcinoma of the thyroid gland but high in anaplastic carcinoma and malignant lymphoma. We studied the relationship between Ga-67 uptake and nuclear DNA content in four papillary adenocarcinomas, three follicular adenocarcinomas, three anaplastic carcinomas, and five malignant lymphomas of the thyroid gland. In anaplastic carcinoma and malignant lymphoma, the nuclear DNA content and proliferative index were significantly higher than in well-differentiated adenocarcinoma. These results suggest that there is close correlation between Ga-67 uptake and degree of malignancy of thyroid tumor cells.

Sexual cyclicity and diurnal corticosterone secretion in rats given hypothalamic transection at two days of age.

Female rats given neural transection between suprachiasmatic and arcuate nuclei at 2 days of age showed regular sexual cyclicity and had ovarian corpora lutea. Thus, the ovarian activity seemed identical to that of intact or sham-operated control rats. However, the diurnal rhythm of the blood corticosterone concentration of the neonatally transected rats was not evident at 22 days of age, while it was already evident in controls. At 30 and 50 days of age, they had clear adrenal rhythms although the peak values were significantly lower than those of controls. Thus, the anterior connections of the medial basal hypothalamus, which are necessary for both cyclic gonadotropin discharge and diurnal corticosterone secretion, are still plastic and not yet established at 2 days of age.
