ROR gamma: the third member of ROR/RZR orphan receptor subfamily that is highly expressed in skeletal muscle.

In this study, we describe the identification and cloning of a novel member of the nuclear receptor superfamily. This orphan receptor, referred to as ROR gamma, belongs to the ROR/RZR subfamily. The open reading frame of ROR gamma encodes a protein of 560 amino acid residues with a predicted molecular mass of 63 kD. The amino acid sequence of ROR gamma exhibits a 50 and 51% identity with those of ROR alpha/RZR alpha and RZR beta, respectively, whereas the DNA-binding domains were 89% identical. ROR gamma was localized on human chromosome 1. Northern blot analysis using RNA from multiple tissues indicated that ROR gamma is expressed in several tissues but is most highly expressed in skeletal muscle.

Structural organisation and chromosomal mapping of the human Id-3 gene.

The helix-loop-helix (HLH) family of transcription factors plays a central role in the regulation of cell growth, differentiation and tumourigenesis. Members of the Id (inhibitor of DNA binding) class of these nuclear proteins are able to heterodimerise with and thereby antagonise the functions of other transcription factors of this family. We report here on the genomic organisation of the human Id3 (HLH 1R21/heir1) gene. Comparison with the two other mammalian Id genes, Id1 and Id2, reveals a highly conserved protein coding gene organisation consistent with evolution from a common, ancestral Id-like gene. In addition, by using a yeast artificial chromosome (YAC) clone of Id3, we have fine-scale mapped the gene to chromosome band 1p36.1 by fluorescence in situ hybridisation (FISH) and, using the same FISH technique, we have detected heterogeneity in tumour-associated 1p36 chromosome translocations.

Differential regulation of CD27 expression on subsets of CD4 T cells.

Our earlier studies showed that although CD27 was stably expressed on the CD45RA(+)-CD45RO-CD29low subset of CD4 T cells, its expression on the CD45RA-CD45RO+CD29high subset of CD4 T cells was gradually lost within 3 weeks after PHA activation. In the present study, we further determined the mechanisms by which the CD27 expression was differentially regulated on the subsets of CD4 T cells. We showed that disappearance of CD27 from the surface of the CD45RA-CD45RO+ subset of CD4 T cells was not solely due to the shedding of the CD27 molecule from the cell surface since the release of a soluble form of the CD27 molecule from the CD45RO+ CD4 T cells was consistently less than that from CD45RA+ CD4 T cells. Although the surface CD27 expression was undetectable on long-term cultured T cell lines originally derived from CD45RO+ CD4 T cells, some of these cells still expressed intracellular CD27 and CD27 mRNA. Moreover, restimulation could not induce CD27 expression on such cells. Further analysis of CD27 protein and mRNA expression at a clonal level showed that cloned cells derived from CD45RO+ CD4 T cells having lost cell surface expression of CD27 were of two types: one expressed intracellular CD27 mRNA and protein whereas the second lacked both intracellular CD27 mRNA and protein.

Frontal analysis of protein adsorption on a membrane adsorber.

The adsorption behaviour of two kinds of proteins, myoglobin and ovalbumin, with a membrane adsorber, DEAE MemSep 1000 (Millipore), was studied in comparison with a bead-based packed-bed adsorber, DEAE Sephacel (Pharmacia-LKB), by means of frontal analysis. Adsorption isotherms were obtained by integrating the breakthrough curves for various feed concentrations. Adsorption isotherms were expressed by the Langmuir equation and the adsorption capacity for the membrane adsorber was smaller than that for the packed-bed adsorber. The breakthrough curves of myoglobin for the membrane adsorber were independent of the flow-rate, but those of ovalbumin were affected by the flow-rate. Abnormal behaviour was observed for the adsorption of ovalbumin on a membrane adsorber. With the packed-bed adsorber, the breakthrough curves for both proteins were significantly affected by the flow-rate. A mathematical model for the membrane adsorber involving axial dispersion and adsorption kinetics was derived. The model simulated the breakthrough curves for myoglobin well. Axial dispersion was dominant for the membrane adsorber whereas intraparticle diffusion was dominant for the packed-bed adsorber.

Characterization of a cDNA encoding a novel type of RNA-binding protein in tobacco: its expression and nucleic acid-binding properties.

A cDNA encoding an RNA-binding protein (ribonucleoprotein or RNP) was isolated from a tobacco (Nicotiana sylvestris) cDNA library. The predicted protein (termed RGP-2) is 259 amino acids in length and consists of an N-terminal sequence of 39 amino acids, a consensus sequence type RNA-binding domain of 82 amino acids, a glycine-rich domain of 83 amino acids and an acidic C-terminal domain of 46 amino acids. It is distinct from the RGP-1 proteins previously reported, which consist of an RNA-binding domain in the N-terminal half and a glycine-rich domain in the C-terminal half. Homology searches revealed that RGP-2 is a novel consensus sequence-type RNA-binding protein. Its RNA-binding domain is structurally related to those of some chloroplast RNPs, while the amino acid composition of its glycine-rich domain (rich in glycine and asparagine) is similar to those in animal heterogeneous nuclear RNPs (hnRNP) A1 and A2/B1. The RGP-2 gene seems to be a single-copy gene, and its transcripts accumulate mainly in cultured cells and roots. A nucleic acid-binding assay using RGP-2 protein synthesized in vitro confirmed that it is an RNA-binding protein. Based on its greater affinity for total tobacco RNA than for poly(G) and poly(U), RGP-2 is suggested to bind to specific RNA sequences, probably G/U-rich regions. Quantitative analysis of the nucleic acid-binding properties of RGP-2 and RGP-1b indicates that they bind differently to nucleic acids. A possible role for RGP-2 is discussed in relation to known functions of animal hnRNP proteins.

The rolC promoter of Agrobacterium rhizogenes Ri plasmid is activated by sucrose in transgenic tobacco plants.

The 5'-upstream region of the rolC gene of the Ri plasmid is expressed specifically in phloem cells of transgenic higher plants. In this study, we demonstrated that the rolC promoter is activated by sucrose in phloem cells of transgenic tobacco seedlings bearing rolC promoter-uidA chimeric fusion gene. Since the rolC promoter is not activated by sorbitol, sucrose metabolism rather than osmotic pressure exerted by the disaccharide may be responsible for induction. Thus, experiments using 5'-upstream deletion mutants, internal deletion mutants, and chimeric constructs with a heterologous promoter (-90 region of the cauliflower mosaic virus 35S promoter) were conducted to define the region of the rolC promoter involved in sucrose activation. The results indicated that a cis-acting sucrose responsive region of the rolC promoter is located between -135 and -94 bp with respect to the transcription initiation site. In phloem cells, high concentrations of sucrose are encountered owing to ongoing translocation of photosynthates from source to sink tissues. Therefore, sucrose as a signal molecule may regulate the phloem-specific expression of the rolC promoter.

Determination of urinary glucuronide conjugates by high-performance liquid chromatography with pre-column fluorescence derivatization.

A simple and highly sensitive high-performance liquid chromatographic method for the direct determination of urinary glucuronide conjugates is described. The method is based on the direct derivatization of the glucuronic acid moiety in glucuronide conjugates with 6,7-dimethoxy-1-methyl-2 (1 H)-quinoxalinone-3-propionylcarboxylic acid hydrazide. The derivatization reaction proceeds in aqueous solution in the presence of pyridine and 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide at 0-37 degrees C. The resulting fluorescent derivatives are separated on a C18 column using methanol-acetonitrile-0.5% triethylamine in water (1:1:2, v/v) as mobile phase, and are detected spectrofluorimetrically at 445 nm with excitation at 367 nm. The detection limits (signal-to-noise ratio = 3) for the glucuronides are 13-48 fmol for an injection volume of 10 microliters (130-480 fmol per 5 microliters of human urine). The method was applied to the measurement of etiocholanorone-3-glucuronide and androsterone-3-glucuronide in human urine. The method is simple and rapid without conventional liquid-liquid extraction of the glucuronides from urine.

Renal cell carcinoma producing alpha-fetoprotein (AFP) with a unique lectins-affinity profile.

Renal cell carcinoma (RCC) producing alpha-fetoprotein (AFP) is a rare entity and merely 7 cases have been reported so far. The present case, a 71-year-old woman, showed a high serum AFP level of 204 ng/ml. The RCC of the autopsied right kidney consisted mainly of spindle-shaped or bizarre sarcomatous tumor cells. AFP was immunolocalized only in the concomitant clear cell component. Concanavalin A (Con A)-nonadsorption rate of serum AFP was 42%, which was an intermediate value between those of yolk sac tumors and metastatic liver carcinomas. Lens culinalis agglutinin (LCA)-affinity study of the patient's AFP showed an unknown peak X, which was eluted between the known peaks 2 and 3. These results suggest a certain structural alteration in carbohydrate moieties of the AFP derived from this RCC. A review of the clinicopathologic features of 8 patients with AFP-producing RCC was made to understand the pathophysiology of AFP-producing neoplasms.

RNA editing of tobacco petB mRNAs occurs both in chloroplasts and non-photosynthetic proplastids.

We found an RNA editing site in the protein coding region of tobacco (Nicotiana tabacum) petB transcripts. This editing (CCA to CUA) leads to an amino acid conversion from proline to leucine. It is observed not only in chloroplasts isolated from tobacco leaves but also in non-photosynthetic proplastids isolated from a tobacco cell culture. Also unspliced pre-mRNA shows complete editing. The editing site is the same as that recently observed in the maize petB transcripts which restores the codon for a highly conserved leucine residue, suggesting that RNA editing at this site is critical for the protein (cytochrome b6) function.

Prognostic value of initial electroretinogram in central retinal vein obstruction.

We retrospectively studied the correlation between the initial electroretinogram (ERG) recordings and the final visual acuities (VA) of 47 patients with central retinal vein obstruction (CRVO) after follow-up of more than 1 year. The a- and b-wave amplitudes and the b/a ratio with white flash were significantly larger in patients with good vision (VA > 20/200) than in those with poor vision (VA < or = 20/200). The b/a ratio and b-wave amplitude with white flash showed sensitivity and specificity of more than 78% for visual prognosis. The latest VA was < or = 20/200 in all 20 patients with a subnormal b-wave amplitude and in all 15 with b/a ratio < 1.05. These results suggest that the ERG recorded soon after (CRVO) onset correlates strongly with visual prognosis.
