Vacuolar myopathy with type 2 A fiber atrophy and type 2 B fiber deficiency. A case of childhood form acid alpha-1,4-glucosidase deficiency.

This report describes a female patient with childhood form of acid maltase deficiency who survived till fifteen years old. Although acid alpha-1,4-glucosidase was deficient in the liver, kidney, skeletal and cardiac muscles, neutral alpha-1,4-glucosidase was present in normal concentrations in those organs. On light microscopic examination, numerous intracytoplasmic vacuoles containing acid phosphatase positive granules and PAS positive materials were present in both type 1 and 2 A fibers, predominantly in the latter. The striking finding in the present case was a selective type 2 fiber atrophy with type 2 B fiber deficiency believed to result from type 2 motor neuron dysfunction in the spinal cord. Electron microscopic study revealed extensive glycogen particle accumulation, autophagic vacuoles and myelin figures in the muscle fibers.

[Difference of steroidogenesis in human ovary at different stage of cycle (author's transl)].

This experiment was undertaken to compare the steroidogenesis in human ovary at different stage of the cycle. Fifteen subjects, 8 in follicular, 7 in luteal phase were selected for this study. On hysterectomy, both ovarian and peripheral venous blood were collected simultaneously and follicle or corpus luteum were obtained. Steroid levels including pregnenolone(P5) 17 alpha-OH-P5, DHA, androstenediol (AD), progesterone(P4), 17 alpha-OH-P5, androstenedione(delta 4A) and testosterone(T) were measured by radioimmunoassay. Tissue were homogenized and incubated with labeled P5, 17 alpha-OH-P5, DHA and AD in the presence of NAD t 37 degrees C. As gas phase, CO was used. Metabolites were extracted and separated by TLC. 3 beta-hydroxysteroid dehydrogenase (3 beta-HSD) activity was expressed as the amounts of delta 4-steroid formed from corresponding delta 5 substrate. The levels of P5, P4, 17 alpha-OH-P4, DHA, delta 4A, T in the ovarian vein and the concentration of P5, 17 alpha-OH-P5, DHA, P4, 17 alpha-OH-P4, T in the ovarian tissue obtained in luteal phase were significantly higher than that in follicular phase. The results suggested that the delta 5 pathway seem to be active in corpus luteum. In follicular phase, highest concentration of 17 alpha-OH-P5 among delta 5 in ovarian vein and 17 alpha-OH-P4 among delta 4 steroid in ovarian vein and tissue was noticed. 3-beta-HSD activity was lower in follicle than that in corpus luteum. Substrate specificity was noticed in 3 beta-HSD in follicle and it was found that 17 alpha-OH-P5 seemed to be the most potent substrate for the enzyme. The difference of steroidogenesis between follicular and luteal phase was demonstrated in human ovary.

Release of prostaglandins evoked by neurohormonal stimuli in the isolated spleen of rabbit.

In the isolated spleen of rabbit perfused with Tyrode solution, sympathetic nerve stimulation (NS) and administration of either noradrenaline (NA), 50--75 ng, or bradykinin (BK), 50 ng, released a material into the venous effluent that caused contraction of rat stomach strip (RSS), rat colon (RC), chick rectum (CR) and bovine coronary artery (BCA). The musculotropic activity of the material released from the spleen was not diminished when it was allowed to stand at either 37 degrees C or at room temperature. The release of this musculotropic material evoked by neurohormonla stimuli was abolished during the infusion of the prostaglandin (PG) synthesis inhibitor, indomethacin, 1 microgram/ml. Application of authentic PGE2 directly on the assay organs caused contraction, whereas PGI2 contracted RSS, RC, and CR but relaxed BCA. In the spleen labeled with [14C]AA, NS, NA or BK released radiolabeled products having the chromatographic characteristics of PGE2 and, to a lesser extent, of PGF2 alpha. Indomethacin abolished the release of these radiolabeled products from the spleen. These observations taken together with the musculotropic characteristics of the material in the venous effluent, suggest that PGE2 is the major product of cyclo-oxygenase relased by NS, NA and by BK from the isolated perfused spleen of rabbit.

Metabolism of [14C] arachidonic acid in the isolated rabbit spleen.

Infusion of [14C] arachidonic acid (AA) into the isolated, Tyrode perfused rabbit spleen resulted in the release of a substance into the venous effluent with the musculotropic activity and chromatographic properties of prostaglandin (PG)E2. Smaller amounts of radioactive materials with the chromatographic properties of PGF2 alpha, 6-keto-PGF1 alpha, and PGD2 were also released. The radiolabeled material released in largest amounts from the spleen was identified as PGE2 on the basis of: 1) Co-chromatography with PGE2 in three solvent systems, 2) Conversion of the radioactive material and of authentic [3H] PGE2 to similar products by treatment with sodium borohydride and with potassium hydroxide, and 3) Stability of the musculotropic activity in Tyrode solution at 37 degrees C. Release of the major and minor radioactive products was inhibited by pretreatment of the spleen with either indomethacin or 5,8,11,14-eicosatetraynoic acid.

Granule formation in rat mast cells. Study with serial ultrathin sections.

The process of granule formation in mast cells was investigated with young mast cells in the milky spot of adult rats, seven days after an intraperitoneal injection of distilled water. Serial ultrathin sections were made and examined with an electron microscope to reveal the following results. The early step of granule formation is observed as a complex which consists of an irregularly shaped narrow vestibular space and several primary aggregate granules which are bowl-like in form, attached to the vestibular space and contain a few subgranules. The vestibular space has microvilli. The precursor materials seem to be secreted from the microvilli and transported through the vestibular space into the primary aggregate granules, being coagulated to become subgranules. As the subgranules increase in number, the primary aggregate granules become aggregate granules which grow further to their maximum size. With further addition of the materials, the aggregate granules become, through a step of ropy cord granule, mature dense homogeneous granules. The complex seems to remain as a unit after the maturation of granules and to react as a whole to any stimulus.

Effects of a new adenosine deaminase inhibitor, isocoformycin, on toxicity, antitumor activity and tissue distribution of formycin A and 9-beta-D-arabinofuranosyladenine.

Single intraperitoneal and intravenous injections of isocoformycin at 1,200 mg/kg did not cause the death of mice. Isocoformycin which inhibited adenosine deaminase enhanced significantly the toxicity of formycin A and ara-A at various combination ratios. Isocoformycin potentiated antitumor activity of formycin A and ara-A against L1210 leukemia. Formycin A and ara-A disappeared rapidly from the blood and tissues and could not be found in any tissues even 0.5 hour after a single intraperitoneal injection. However, when used in combination with isocoformycin both were detected in the blood and tissues, especially at high concentration in liver and kidney. These indicate that the deamination of formycin A and ara-A is blocked by isocoformycin in vivo.

Studies on auromomycin.

A new antitumor antibiotic, named auromomycin, was isolated from the culture broth of Streptomyces macromomyceticus, a macromomycin-producing strain. The antibiotic was recovered from the culture filtrate by salting out with ammonium sulfate and further purified by successive application of ion-exchange chromatography on Amberlite IRA-93 (Cl form) and DEAE-Sephadex (OH form), Gel filtration on Sephadex G-50 and hydrophobic chromatography on Octyl-Sepharose CL-4B. The antibiotic is an acidic polypeptide with a molecular weitht of 12,500 and an isoelectric point of pH 5.4 and consists of 16 different amino acids. It has characteristic absorption maxima at 273 nm and 357 nm in the ultraviolet spectrum and two minima at 280 nm and 350 nm in the optical rotatory dispersion spectrum. Auromomycin exhibits antibacterial activity not only against Gram-positive bacteria, but also Gram-negative bacteria. Antitumor activities of auromomycin were revealed against EHRLICH ascites carcinoma, ascites sarcoma 180, L1210 leukemia and LEWIS lung carcinoma. Auromomycin was found to be converted into macromomycin by adsorption chromatography on Amberlite XAD.

Biological activity of macromomycin.

Macromomycin (MCR) is a polypeptide antimuor antibiotic isolated from the culture broth of Streptomyces macromomyceticus. Antitumor activities of MCR were examined against three different tumor system, i.e., EHRLICH ascites carcinoma, L1210 leukemia and LEWIS lung carcinoma. Daily intraperitoneal treatment with MCR for 5 days showed a strong inhibition against EHRLICH ascites carcinoma. Both single and repeated intraperitoneal injections of MCR were effective over a wide dose range against intraperitoneally inoculated L1210 leukemia and MCR intravenously administered was also active against intravenously inoculated L1210 leukemia. Daily local subcutaneous injections of MCR produced the prolongation of life span of mice to which LEWIS lung carcinoma was subcutaneously inoculated with some cured mice, but daily intraperitoneal injections of MCR showed no activity. Single intravenous administration of MCR inhibited early LEWIS lung carcinoma, but not advanced LEWIS lung carcinoma. The combination of MCR with aracytidine, or cyclophosphamide showed a synergistic activity against L1210 leukemia. MCR was not inactivated by treatment with serum, although neocarzinostatin was markedly inactivated by the same treatment.
