Formadicins, new monocyclic beta-lactam antibiotics of bacterial origin. II. Isolation, characterization and structures.

New monocyclic beta-lactam antibiotics, formadicins A, B, C and D, were isolated from the culture filtrate of Flexibacter alginoliquefaciens sp. nov. YK-49 by various types of column chromatography and preparative reverse-phase HPLC. Their structures were determined by spectroscopic analyses and degradation studies. They have a nocardicin-type skeleton and a formylamino group at the 3- or 12-position. Formadicins A and B each have a D-glucuronide moiety and give formadicins C and D, respectively, upon hydrolysis using beta-D-glucuronidase.

Cephabacin M1-6, new 7-methoxycephem antibiotics of bacterial origin. II. Isolation, characterization and structural determination.

Six components of new cephem antibiotics, cephabacin M1-6, were isolated from the culture filtrate of Xanthomonas lactamgena YK-431 by various types of column chromatographies and preparative reverse-phase HPLC. Their structures were determined by spectroscopic analyses and degradation studies. They consist of 7-methoxydeacetylcephalosporin C as a nucleus and a tri- to heptapeptide including a new amino acid, which is bound at the 3-position with an ester bond.

Cephabacins, new cephem antibiotics of bacterial origin. III. Structural determination.

The structures of 15 new cephem antibiotics, cephabacin F1-9 and H1-6, were determined by their spectroscopic analyses and decomposition studies. They are consisted of a cephalosporin nucleus and a di, tri or tetrapeptide including a new amino acid which is bound at the position 3 with an ester bond. The components, F1-9, showed unique biological activities by the presence of a formylamino group at the position 7.

Retinal detachment in Morning Glory syndrome.

A 9-year-old girl with bilateral Morning Glory syndrome developed bilateral bullous retinal detachment. There was a small break on the temporal margin of the disc in the left eye. The retinal detachment in this eye was successfully treated by drainage of the subretinal fluid, vitrectomy, intravitreal air injection, and photocoagulation around the edge of the anomalous disc. It is suggested that some retinal detachments in the Morning Glory syndrome may be rhegmatogenous in nature.

Possible inactivation of part of chromosome 13 due to 13qXp translocation associated with retinoblastoma.

Chromosome examination of a female patient with 13/X translocation associated with retinoblastoma was carried out using peripheral blood lymphocytes and cultured skin fibroblasts. The constitutional karyotype was 46,X,t(13;X) (q12;p22). Q-banding analysis showed that the translocated chromosomes were of paternal origin. Studies on DNA replication pattern with Giemsa banding using the bromodeoxyuridine substitution technique revealed that the derivative X chromosome was late replicating, and the translocated chromosome 13 was affected by the spreading of lyonization. Such a functional monosomy of 13q14 may also be involved in retinal blasts, and be related to the development of retinoblastoma.

The interrelation between the latero-dorsal tegmental area and lumbosacral segments of rats as studied by HRP method.

Satoh et al. (1978a) demonstrated that Barrington's nucleus in the rat corresponds to the nucleus laterodorsalis tegmenti (TLD), immediately ventromedial to the rostral pole of the locus coeruleus (LC). To confirm this finding and to identify the course of the tracts and their terminals, we injected small amounts of HRP into the TLD, LC and the lumbar and sacral segments followed by the DAB and TMB reaction. Injection of HRP into the TLD resulted in the anterograde staining of descending fibers in the latero-ventral part of the lateral funiculus, which mainly terminate in the intermediate area of the sacral segments, and in the retrograde labeling of a large number of cells in the intermediate area of the sacral segments. After injection of HRP into the sacral segments, marked labeling of many TLD cells was demonstrated in contrast to that of only a small number of LC cells. On the other hand, after injection of HRP into the LC, a moderate number of descending fibers were found in the ventral part of the lateral funiculus, with small numbers in the apex of the dorsal horn and ventral funiculus, mainly ending in the intermediate gray of the lumbar segments. Moreover, moderate numbers of retrogradely labeled cells were revealed in the intermediate area of the lumbar segments. From these results it may be concluded that the reciprocal interrelation between the TLD and intermediate gray of the sacral segments is more important for the micturition reflex than that between the LC and lumbo-sacral segment.

The fine structural organization of the locus coeruleus in the rat with reference to noradrenaline contents.

Combination of glyoxylic acid perfusion and postfixation in permanganate was used in an electron microscopic study of the locus coeruleus (LC) of the rat to give good preservation of fine structure and a reproducible demonstration of noradrenaline (NA) storage granules. Medium-sized LC cells (18 x 30 microns) contained a moderate number of small granular vesicles (SGV) and a few large granular vesicles (LGV), mainly near the Golgi apparatus. Dendritic branches were identified by their SGV content up to the tip. Dendrites were occasionally in close contact with each other or with the soma of LC cells, forming dendro-dendritic or dendro-somatic contacts. Numerous axon terminals containing many SGV and some LGV were observed in the neuropil, and they tended to contact dendrites and somata of LC cells or dendrites of unknown origin. These neuronal contacts were devoid of synaptic specializations except for an array of dense perpendicular lines between the juxtaposed membranes. Small oval cells (10 x 15 microns) devoid of SGV occurred frequently in the peripheral part of the nucleus, and they were occasionally in direct contact with LC cells.

[The effects of sphincteroplasty on manometric profile of common bile duct and the duodenum (author's transl)].

A simultaneous manometric monitoring of the common bile duct (CBD) and the duodenum were performed 2 weeks after choledocholithotomy on 15 patients whose common bile ducts were explored without sphincteroplasty and 30 patients with sphincteroplasty. These manometric studies were carried out by open-tip catheters intubated into the CBD and duodenum through the T-tube at the operation. In patients without sphincteroplasty, no effects of the duodenal pressure on a CBD pressure profile were recognized, while a synchronized pressure profile of the CBD and the duodenum was obtained in patients with sphincteroplasty. By stimulation with morphine (Morphine sulfate; 0.17 mg/Kg iv bolus), waxing and waning of the pressured in the CBD without sphincteroplasty were observed with 20 cmH2O in maximum at about 13 minutes after injection. However, in the CBD with sphincteroplasty, scale-over increase of the pressure curve was seen immediately after duodenal contraction caused by morphine stimulation. A direct infusion of 5 ml of 0.1 N hydrochloride to the duodenum causes hyperperistalsis of the duodenum, which made a synchronized pressure profile in the CBD with sphincteroplasty but made no remarkable change in a pressure profile of the CBD without sphincteroplasty. These findings conclude that the sphincter of Oddi plays an important role as a "pressure barrier" between the CBD and the duodenum, and that with the destruction of this sphincter by sphincteroplasty, a pressure profile of the CBD becomes close to that of the duodenum. This simultaneous manometric study of the CBD and the duodenum might be one of most valuable methods for evaluation of completeness of the sphincteroplasty.
