Contribution of RGD sequence to neuronal migration in developing cerebral cortex.

The RGD sequence (Arg-Gly-Asp) is known to bind integrins and the synthetic RGD tripeptide inhibits cell adhesion and migration of cultured neuronal cells. However, it is not known whether migration of neuronal precursors in vivo during the cortical histogenesis of the mammalian telencephalon depends on the RGD sequence. To examine this possibility, the RGD peptide was injected into the telencephalic vesicle of mouse embryos. A hypoplastic cortical plate was induced and histological analysis and BrdU-immunohistochemistry revealed that the RGD sequence is involved in neuronal migration and proliferation in the telencephalon during formation of the cortical plate.

The superiority of exercise testing over spirometry in the evaluation of postoperative lung function for patients with pulmonary disease.

Thoracic surgeons have often been embarrassed by the discrepancy between an improvement in symptoms and the unchanged or even worse results of spirometry in postoperative patients with either bullae or inflammatory lung disease. Forty-four patients with lung diseases, who underwent a total of 47 operations, were categorized as follows: 12 cases of empyema, 16 cases of giant bulla (undergoing surgery a total of 19 times), 4 cases of bronchiectasis, and 12 cases of other miscellaneous diseases. All patients were tested preoperatively and again 4-6 months after surgery on both the spirometer and treadmill exercise tests. The forced vital capacity (FVC) and forced expiratory volume (FEV1.0) results were as follows: the empyema group 1.82 +/- 0.52 liters preoperatively to 1.93 +/- 0.69 liters postoperatively and 1.47 +/- 0.44 liters to 1.56 +/- 0.53 liters, respectively; and the giant bulla group, 3.49 +/- 0.96 liters to 3.35 +/- 0.77 liters and 2.35 +/- 0.96 liters to 2.48 +/- 0.69 liters, respectively. However, the exercise time was prolonged in the empyema group from 6.00 +/- 3.77 min to 8.33 +/- 3.80 min (P < 0.01) and in the giant bulla group from 11.83 +/- 3.71 min to 12.92 +/- 2.84 min (P < 0.05). It was thus concluded that exercise testing should be chosen for the postoperative evaluation of patients with inflammatory pulmonary disease and giant bullae, especially if any discrepancies are seen between spirometry and performance status, because on the basis of our results, it appears that the benefits obtained by surgery are best measured by the dynamic values of exercise testing and not by the static values of spirometry at rest.

Cohort effects in the lateral preference of Japanese people.

Handedness data of the Japanese were collected twice with an interval of 20 years, using an identical handedness inventory. In the second cohort, non-right-handers among male and female students numbered 11.5% and 5.9%, respectively. The results showed that the incidence of non-right-handedness among Japanese females has increased from 20 years ago.

Laterality of mental imagery generation and operation: tests with brain-damaged patients and normal adults.

The relationships between hemispheric function and components of the imagery process were examined in patients with unilateral right and left brain damage and in intact adult subjects. In the image generation condition, subjects were required to mentally generate Katakana letters corresponding to Hiragana letters displayed on a CRT. The results for the intact adults suggested a left hemisphere superiority, but the unilaterally brain-damaged subjects showed no hemispheric difference in this task. In the imagery operation task (transformation or lateral translation), subjects were asked to find a genuine Kanji among distractors (pseudo-Kanji) that were constructed from two Kanji radicals (themselves real Kanji) that were either displayed in reverse order or shifted apart. The results for both intact adults and patients with unilateral brain damage suggest the superiority of the right hemisphere.

Development of the lens in human embryos: a histochemical and ultrastructural study.

The early development of the lens was examined, using 36 externally normal human embryos at Carnegie stages 13-23 (4 to 8 weeks of gestation). Twenty-two embryos were sectioned serially and stained with periodic acid-Schiff and a modified method of PAS. In 14 embryos, not only the differential distribution of glycogen but also the ultrastructural change in the developing lens, with special reference to junctional complexes, were examined electron microscopically. At stage 15, when the lens vesicle was formed, glycogen was observed in the cytoplasm of the lens epithelium, especially in the posterior lens epithelium. From stages 16 to 18, when the posterior lens epithelium was differentiated into the primary lens fibers and elongated toward the anterior lens epithelium, the amount of glycogen increased in the basal cytoplasm of the primary lens fiber, where the intracellular organelles, such as the tubular vesicles, mitochondria and multivesicular bodies, began to aggregate. At stage 20, when the lens cavity was obliterated, glycogen was also present in the anterior lens epithelium. At stage 21, as the formation of the secondary lens fibers proceeded, glycogen was noted in the secondary lens fibers in the equator region. These findings suggest that the distribution of glycogen is associated with the formation of the primary and secondary lens fibers. In addition, we provide additional information that a lot of glycogen is distributed in the region where many intracellular organelles aggregate in the embryonic lens vesicles.

Protection from caseous lymphadenitis in sheep by spraying iodine tincture on shearing wounds.

The effect of spraying shearing wounds with iodine tincture on Corynebacterium pseudotuberculosis infection in lambs was examined. The ELISA-negative lambs which had received some visible wounds during their first shearing were randomly divided into two groups: one was sprayed with iodine tincture on wounds after shearing, and the other was not. Anti-C. pseudotuberculosis toxin titers were measured by ELISA. The seroconversion ratio of the group with iodine tincture treatment 3 months after shearing was smaller than that of the untreated group (P < 0.05). These results suggest that treatment of shearing wounds with iodine tincture is effective in the protection of C. pseudotuberculosis infection in lambs.

[Severe obstructive necrotizing enterocolitis associated with carcinoma of the sigmoid colon: report of a case].

A 68-year-old male diagnosed as carcinoma of the sigmoid colon developed shock and cardiac arrest just 8 hours after the onset of ielus. After resuscitation, exploratory laparotomy was carried out and it revealed gangrene spreading from the sigmoid colon over the distal 150cm of the terminal ileum, separated from the carcinoma by a normal segment of the colon 13cm in length. All the mesenteric vessels were pulsating normally. The whole necrotic lesion including the carcinoma was resected and an ileostomy was constructed. This is a noteworthy report of severe obstructive necrotizing enterocolitis with critical condition and wide involvement of the colon and intestine associated with carcinoma of the sigmoid colon.

Mechanisms of the inhibition of reverse transcription by unmodified and modified antisense oligonucleotides.

We demonstrated that unmodified and modified (phosphorothioate) oligonucleotides prevent cDNA synthesis by AMV or HIV reverse transcriptases. Antisense oligonucleotide/RNA hybrids specifically arrest primer extension. The blockage involves the degradation of the RNA fragment bound to the antisense oligonucleotide by the reverse transcriptase-associated RNase H activity. However, the phosphorothioate oligomer inhibited polymerization by binding to the AMV RT rather than to the template RNA, whereas there was no competitive binding of the phosphorothioate oligomer on the HIV RT during reverse transcription.

Ultrastructure of the developing stomach in human embryos.

Ultrastructural development of the stomach was studied by light, scanning electron and transmission electron microscopy, using 19 human embryos at Carnegie stages from 14 to 23 (6.8-28.0 mm in crown-rump length, 5 to 8 weeks of gestation). The precise time of appearance of differentiated characteristic structures was examined electron microscopically. The first gastric pit, with radially arranged epithelial cells beneath which the basement membrane bulged into the mesenchyme, was observed on the lesser curvature at stage 22. Although the mesenchymal condensation which would develop into the inner circular muscle layer appeared at stage 18 onward, cytoplasmic myofibrils were not observed until stage 22. Nerve fibers were first observed at stage 16, and at later stages they gathered into bundles to form a nerve plexus external to the developing inner circular muscle layer. On the basis of accurate timing of the appearance and the mode of development of these structures, possible relations between developing gastric layers were discussed. Histocytochemically, glycogen or other carbohydrates were demonstrated in the cytoplasm of the gastric epithelium throughout the stages examined. These carbohydrates were localized mainly in vacuole-like spaces in the basal part of the epithelial cells. This subcellular localization, and the amount of carbohydrate, did not change significantly during the observed embryonic period. In the serosa, carbohydrates were not detected at stages 14 and 15, but observed consistently within the vacuoles in the cytoplasm from stage 17 onward. No other layer of the embryonic stomach had detectable carbohydrates.(ABSTRACT TRUNCATED AT 250 WORDS)

Interhemispheric integration of number stimuli: comparison of Arabic with Kanji numerals.

Thirty normal subjects were requested to add two numbers which were presented tachistoscopically. Two numbers were displayed either to one visual field or simultaneously one to the left and one to the right visual field. Arabic or Kanji numerals were used. Bilateral presentation was superior to unilateral presentation with both kinds of stimuli. In unilateral presentation, Arabic numbers were processed more proficiently in the left visual field and Kanji numbers in the right visual field.

Seroepidemiological evidence that shearing wounds are mainly responsible for Corynebacterium pseudotuberculosis infection in sheep.

Anti-Corynebacterium pseudotuberculosis toxin titers were examined by an enzyme-linked immunosorbent assay (ELISA) at 3 to 5 month intervals in sheep of three age groups (lambs, 1- and 2-year-old sheep). A significant increase in the ELISA-positive ratio was found only in July, 3 months after shearing, in 1- and 2-year-old sheep. A decrease in the positive ratio was found in the same month in lambs, possibly due to a decrease in maternal antibodies. Since the 1- and 2-year-old sheep had been sheared in April, it was considered that the increase in the positive ratio in July might be closely related to the shearing in April.

[Clinical characteristics of impacted bile duct stone in the elderly].

Acute cholangitis due to impacted bile duct stone is sometimes fatal and require prompt bile duct decompression. Particularly, AOSC (acute obstructive suppurative cholangitis) is the most serious form of bacterial cholangitis and its prognosis is usually very poor when treated conservatively. We have been treated impacted bile duct stones by decompressing the duct endoscopically. Here, we report the characteristics of our elderly patients with impacted bile duct stones who received urgent decompression treatment mainly by endoscopic techniques. In the endoscopical treatment group, the number of patients with AOSC was 5 out of 46 AC (acute cholangitis) patients (11%) under 69 years old, 14 out of 52 (27%) from 70 to 79 years old, and 12 out of 28 (43%) over 80 years old. Obviously, the severity of acute cholangitis (ratio of AOSC to AC) was higher in the elderly. This tendency was similar to the patients who received urgent transhepatic decompression treatment, although they were small in number. In all the patients treated, 7 were classified into the most serious AOSC category with Reynolds' sign, and notably 6 out of 7 were over 70 years old. More strikingly, 5 of 7 patients were over 80, and their mortality rate was indeed high. Thus, in the elderly, bacterial cholangitis by impacted bile stones was a very serious condition. Especially, the outcome of AOSC over 80 years old with Reynolds' sign was very poor, in spite of emergency endoscopic treatment.

Mechanisms of the inhibition of reverse transcription by unmodified and modified antisense oligonucleotides.

We have demonstrated that the synthetic oligonucleotides, either unmodified or phosphorothioates, prevent cDNA synthesis by the AMV or HIV reverse transcriptases. The RNA was truncated at the antisense oligonucleotide-RNA duplex during the reverse transcription. Th blockage involves the degradation of the RNA fragment bound to the antisense oligonucleotide by the reverse transcriptase associated RNase H activity. However, in the case of phosphorothioate oligomer, the production of cDNA would be inhibited by a hybrid formed between the AMV RT and phosphorothioate oligomer than arrested elongation of the cDNA strands, whereas arrest of a growing cDNA strand by HIV RT can be blocked by an oligonucleotide complementary to a region downstream from the primer.

[A case of simultaneous bilateral spontaneous pneumothorax].

A 18-year-old man was admitted urgently complaining dyspnea, cyanosis and chest pain. The chest x-ray film revealed simultaneous bilateral pneumothorax. Chest drains were inserted immediately to the both chest cavities. On the 3 rd hospital day, radical operation for spontaneous pneumothorax with synchronous bilateral thoracotomies were performed through both axillary incision. Postoperative course was uneventful. The patient was discharged on the 8 th day after operation.

The effects of Kanji attributes on visual field differences: examination with lexical decision, naming and semantic classification tasks.

Using three experimental tasks (lexical decision, naming and semantic classification), the relative contribution of various Kanji attributes in left and right visual field recognition was examined. The results showed that first, factors contributing to the left visual field processing of Kanji differed depending on the level of processing; second, in an early stage of processing, perceptual attributes such as the number of strokes was a strong contributing factor; and third, in a later stage both perceptual- and imagery-related attributes contributed strongly to left visual field or right hemisphere processing.
