Establishment of B cell hybridomas with B cell surface antigens.

M12.4, one of several B lymphomas derived from BALB/c mice, was mutagenized with ethyl methanesulfonate in vitro. M12.4.1, a subline of the mutant cells, was sensitive to hypoxanthine-aminopterin-thymidine selective medium and was fused with normal splenic B lymphocytes of C57BL/6ByJ mice. The hybridomas obtained from this fusion were shown to express mu heavy chain, H-2KbDb, Iad, and Iab on the cell surface by analyses of flow microfluorometry and a cytotoxicity assay, although parental M12.4.1 lacked mu heavy chain on the cell membrane. These results demonstrate that the B cell hybridomas with B cell surface antigens have been established in vitro. IgM molecules on the cell surface of the hybridomas were shown to originate from normal B cells of C57BL/6ByJ mice by flow microfluorometry analyses after staining with fluorescein-labeled Bet 1, a monoclonal rat antibody that recognizes Igh-6.5, a mouse IgM allotypic determinant. These hybridomas could generate IgM-secreting cells at the high frequency (more than 10% of the cultured cells) when stimulated with bacterial lipopolysaccharide. On the other hand, parental M12.4.1 did not develop any IgM-secreting cells under the same conditions. These findings suggest that these B cell hybridomas with B cell surface antigens may be a good model for the study of B cell differentiation.

Pluripotent hemopoietic precursors in vitro (CFUMIX) in aplastic anemia.

When human marrow cells were cultured in a medium containing alpha-medium, methylcellulose, fetal calf serum, bovine serum albumin, erythropoietin, and leucocyte-conditioned medium, mixed colonies composed of erythrocytic cells and granulocytes were formed. The clonal nature of the mixed colonies was confirmed by the linear relationship between the numbers of cells plated and the number of colonies, and the absence or presence of Y-chromatin in the mixed colonies in a co-culture experiment with male and female cells. Using the methylcellulose cell culture techniques, the pluripotent hemopoietic precursors (CFUMIX) in marrow cells from 15 patients with aplastic anemia were assayed. In the control subjects of patients with iron-deficiency anemia, lymphoadenitis, reactive leucocytosis or Hodgkin's disease, 8 X 10(5) marrow cells in 4 dishes produced 12.7 +/- 6.9 (mean +/- SD) mixed colonies. On the other hand, 8 X 10(5) marrow cells from patients with aplastic anemia formed only 2.1 +/- 5.5 (mean +/- SD) mixed colonies. Furthermore, the marrow cells from 5 patients who were repeatedly receiving transfusions contained no CFUMIX which give rise to mixed colonies. The present results provided the first direct evidence that pancytopenia in most patients with aplastic anemia results from a reduced influx into the compartment of maturing hemopoietic cells from the compartment of pluripotent hemopoietic precursors.

Studies on the kinetics of hemopoietic stem cells and immune responses to the hapten-carrier conjugate.

The correlation between the kinetics of hemopoietic stem cells and immune responses to the hapten-carrier conjugate was investigated. The numbers of both pluripotent stem cells (CFU-S) and myeloid stem cells (CFU-C) in the spleen from mice immunized with the hapten-carrier conjugate were significantly greater than those of the control and the activity of colony-stimulating factor (CSF) in the serum of these mice was markedly elevated. The supernatant of short-term incubation of splenic T lymphocytes from these mice, when stimulated with carrier protein, had high levels of both activities of CSF and helper T cell factors. The study by gel chromatography showed that these factors are similar m.w. substances of 35,000 to 45,000 daltons. But analysis by ion-exchange chromatography demonstrated that they do not have identical biochemical properties. The present studies suggest that biologically active factors produced by T cells stimulated with carrier protein may induce the enhancing effect on the proliferation and differentiation of hemopoietic stem cells and immune responses to the hapten-carrier conjugate.

Effects of allogeneic stimulations on the proliferation and differentiation of the hemopoietic stem cell.

The effects of allogeneic stimulatins on the hemopoietic system were investigated. Spleen cells of mice activated in vivo with alloantigens were able to produce highly active colony-stimulating factor in the culture medium without any stimulation. But spleen cells of these mice could not release colony-stimulating factor after treatment in vitro with anti-theta serum and complement. The number of granulopoietic progenitor cells (colony-forming unit-culture) in both bone marrow and spleen cells of mice treated with such procedures was significantly greater than that of the control, and the number of pluripotent stem cells (colony-forming unit-spleen) of these mice was markedly increased compared with that of the control, especially in spleen cells. These experiments suggest that T lymphocytes activated in vivo with alloantigens may release active substances in the differentiation and proliferation of hemopoietic stem cells.

Preparation of perfluorodecalin emulsion, an approach to the red cells substitute.

The major problems unsolved in fluorocarbon emulsions have been the long lasting retention of the substance in tissues of organs and acute shock symptom in animals receiving "coarse" emulsion with particles larger then 0.2 mum. This report offers a new, stable and sterile preparation of 25% "fine" perfluorodecalin emulsion with yolk phospholipid that is eliminated from liver, spleen and other organs within a few weeks, and whose acute and subacute toxicity is as low as perfluorotributylamine/Pluronic F68 emulsion. Effects of the perfluorodecalin emulsion thus prepared for maintaining contractile force of isolated guinea pig heart by perfusion and survival of animals receiving repeated circulatory exchange proved similar to perfluorotributylamine/Pluronic F68 emulsion.
