Physical constitution and smoking habits of patients with idiopathic spontaneous pneumothorax.

This is a report on a statistical study comparing the Kaup index and the level of tobacco consumption of patients with idiopathic spontaneous pneumothorax and those of healthy subjects. A statistically significant difference was found between the average Kaup index of the patients with idiopathic spontaneous pneumothorax and that of normal healthy individuals in Japan as published by the Ministry of Health and Welfare. A statistically significant difference was also found in the physical features between 2,433 males with idiopathic pneumothorax and 1,906 healthy males when compared by means of the Kaup index. Again, a statistically significant difference was noted in the level of tobacco consumption between the males with idiopathic pneumothorax and normal males when compared by means of the Brinkmann index. The Kaup index of idiopathic pneumothorax patients was lower than that of healthy subjects, while the level of tobacco consumption of the former was higher. Idiopathic spontaneous pneumothorax is related to thin and tall individuals and the level of tobacco consumption among such patients is higher than among healthy subjects.

Species and strain differences in sensitivity to Toxoplasma infection among laboratory rodents.

Different susceptibilities to Toxoplasma infection were shown among various inbred and outbred laboratory rodents. On the basis of several criteria, the animals can be divided into three groups: (i) highly sensitive (S) mice, Syrian hamsters and Chinese hamsters; (ii) moderately sensitive (M) Mongolian gerbils and mastomyses ; and (iii) resistant (R) rats and Korean striped field mice. S and M hosts supported intraperitoneal growth of the parasites and died of acute infection. M hosts showed prolonged time to death and low fatality as compared with S hosts. Some R hosts died only when they had received an extremely large size of inoculum, accompanying poor intraperitoneal proliferation of the parasite. WM/Ms and WKY rats markedly resisted the infection. These hosts were considered to be useful models for studying infection of and immunity to toxoplasmosis.

Toxicological studies on bestatin. I. Acute toxicity test in mice, rats and dogs.

Studies on acute toxicities of bestatin (NK421) were carried out in both sexes of mice and rats, and male dogs. NK421 was administered subcutaneously, intraperitoneally and orally in mice and rats, and orally in dogs respectively. Mice and rats were observed for 14 days after treatment and LD50 values were calculated by the probit method. NK421 showed very low toxicity and no death occurred in any species following the oral administration of the maximum dose capable of dosing such as 4 g/kg for mice, 2 g/kg for rats and 1.2 g/kg for dog. General toxic signs seen in mice and rats following subcutaneous and intraperitonial injections were as follows; depression, suppressed movement, piloerection, inhibition of spontaneous movement, anorexia and emaciation. Death occurred within 5 days after administration. The toxic target organs of NK421 were found to be kidney, lymphoid tissue and liver based on histopathological examination of dead animals.

Toxicological studies on bestatin. II. Subacute toxicity test and recovery study in beagle dogs.

Subacute toxicity and its recovery of bestatin (NK421) was studied on both sexes of 34 Beagle dogs. At dose levels of 600, 240, 96 and 38.4 mg/kg, NK421 was administered orally to dogs for 90 successive days. The control group was treated orally with 2 g/dog of corn starch. Each group was constituted of 3 males and 3 females, and 2 males and 2 females were added to the 240 mg/kg group for the recovery test for 35 days. As general symptoms, loss of appetite, vomiting, abnormal feces (loose stool, diarrhea, mucous stool), eye mucus, decoloration of the visible mucous membrance and unkempt fur were observed slightly and almost dose-dependently in the group dosed with more than 96 mg/kg. Body weight decreased with the passage of time in the 600 and 240 mg/kg groups, but no death appeared in any group. In correlation with general signs, slight anemia was seen hematologically, and the increased alkaline phosphatase activity and the decreased albumin ratio in serum protein fraction were observed biochemically. The slight abnormal findings of bone marrow, spleen and liver were also demonstrated histopathologically. All the above findings disappeared during the recovery period. The maximum non-toxic dose of NK421 in this study is estimated to be 38.4 mg/kg in dogs.

Toxicological studies on bestatin. III. Chronic toxicity test and recovery study in beagle dogs.

Chronic toxicity and its recovery of bestatin (NK421) was studied in both sexes of 28 Beagle dogs. At dose levels of 96, 38.4 and 15.4 mg/kg, NK421 was administered orally to dogs for 540 successive days. Control dogs were treated orally with 2 g/dog of corn starch. Each group consisted of 3 males and 3 females, and 2 males and 2 females were added to the 38.4 mg/kg group for a recovery test of 35 days. As general signs, anorexia, abnormal feces (loose stool, diarrhea, mucous stool), loss of activity, loss of lustre in fur, decoloration of the visible mucosa and emaciation were transiently observed in a early stage in 1 male and 1 female of the 96 mg/kg group. In correlation with these signs, slight anemia appeared hematologically, and the increased alkaline phosphatase activity and the decreased albumin ratio in serum protein fractions were observed biochemically. Except for the slight abnormal findings observed in the liver of the above 2 dogs, no significant changes were histopathologically noticed in any organ of all the dogs examined. The maximum non-toxic dose of NK421 in this study is estimated to be 38.4 mg/kg in dogs.

Toxicological studies on bestatin. IV. Studies on antigenicity, ocular mucosal irritation and mutagenicity.

The antigenicity, local irritation and mutagenicity tests of bestatin (NK421) were carried out as special toxicity studies. The antigenicity tests in rabbits and guinea pigs were as follows: active and passive anaphylactic shock tests, Schultz-Dale reaction test, PCA (passive cutaneous anaphylaxis) reaction test, agar gel precipitation test, hemagglutination test of tannic acid-treated erythrocyte and the test according to the appraisal in the U.S.A. The ocular mucosal irritation test in rabbits was performed as a local irritation test. The mutagenicity test was made using S. typhimurium TA100, TA98 strains by means of preincubation method. The results were that NK421 had no antigenicity, irritation on the ocular mucosa and mutagenicity.

rec-Assay with spores of Bacillus subtilis with and without metabolic activation.

The rec-assay with Bacillus subtilis strains H17 Rec+ and M45 Rec- was carried out with their spores. The use of spores ensured more than 50 times higher sensitivities with several typical chemical mutagens compared with those with vegetative cells. This improved sensitivity enabled us to detect DNA-damaging activities of typical metabolically activated chemicals. Conditions of improvements and quantitative aspects are analyzed and described.

Augmented followed by suppressed levels of natural cell-mediated cytotoxicity in mice infected with Toxoplasma gondii.

The cytotoxic activity of effector cells from mice infected with Toxoplasma gondii was tested in a 4- to 5-hr (51)Cr release assay, using RL [vertical male] 1 and YAC-1 target cells. They showed enhanced cytotoxicity with a peak on the 3rd day postinfection followed by suppression with a peak on the 12th day. The cytotoxicity seemed to be exhibited by natural killer (NK) cells because: (i) pretreatment of the effector cells with antiasialo GM(1) or antiasialo GM(2) plus complement abolished the cytotoxic activity; (ii) the altered cytotoxicity levels were also induced in nude mice; and (iii) the activity was elicited by nonadherent-nonphagocytic cells. The alteration occurred simultaneously in various lymphoid organs with a similar profile. Neither spleen nor bone marrow cells of 12-day-postinfected mice inhibited NK activity of uninfected mice. Culture fluids of the infected mouse spleen and bone marrow cells did not affect the normal mouse NK activity. The proportion of effector cells capable of binding to target cells was constant during the infection. There was no positive correlation between NK activity and serum interferon level; i.e., interferon was detected in the serum of 12-day-postinfected mice but not in that of 3-day-postinfected or uninfected mice. Passively administered interferon or polyinosinic-polycytidylic acid could not restore the suppressed NK activity of 12-day-postinfected mice. Moreover, in vitro treatment of spleen cells from 12-day-postinfected mice with interferon failed to restore the suppressed NK activity. These results suggested that after toxoplasma infection, defective sensitivity to interferon was induced in NK precursor cells, and differentiation to functionally active NK cells might be blocked.

1H-NMR spectroscopic study of aerobic glucose metabolism in Toxoplasma gondii harvested from the peritoneal exudate of experimentally infected mice.

1H-nuclear magnetic resonance (NMR) was used for the study of aerobic glucose metabolism in Toxoplasma gondii harvested from the peritoneal exudate of experimentally infected mice. Without any pretreatment (i.e., extraction, separation or purification) of the supernatant fluids from the cell suspension incubated with glucose for various time intervals, we were able to identify and quantitate the end-products of glucose metabolism by means of 1H-NMR. The major end-products found were lactic acid and acetic acid.

Adenomatoid odontogenic tumor: light and electron microscopic study.

A case of adenomatoid odontogenic tumor involving the right upper cuspid region of a 17-year-old girl is described. In light microscopy, liminal structures and rosette-like structures were found in the parenchyma of the tumor. Electron microscopic observations revealed that the rosette-like structures were less organized than the luminal structures; the luminal structures are supposed to be associated with secretory activities including the formation of calcified material which show the apatite pattern by electron diffraction. The origin and differentiation of cells constituting the tumor are discussed.
