The effect of Abeta conformation on the metal affinity and aggregation mechanism studied by circular dichroism spectroscopy.

The conformational change and associated aggregation of beta amyloid (Abeta) with or without metals is the main cause of Alzheimer's disease (AD). In order to further understand the effects of Abeta and its associated metals on the aggregation mechanism, the influence of Abeta conformation on the metal affinity and aggregation was investigated using circular dichroism (CD) spectroscopy. The Abeta conformation is dependent on pH and trifluoroethanol (TFE). The binding of metals to Abeta was found to be dependent on the Abeta conformation. The aggregation induced by Abeta itself or its associated metals is completely diminished for Abeta in 40% TFE. Only in 5% and 25% TFE can Abeta undergo an alpha-helix to beta-sheet aggregation, which involve a three-state mechanism for the metal-free state, and a two-state transition for the metal-bound state, respectively. The aggregation-inducing activity of metals is in the order, Cu2+ > Fe3+ > or = Al3+ > Zn2+.

Adsorption and thermal reactions of alkanethiols on pt(111): Dependence on the length of the alkyl chain.

The adsorption and thermal decomposition of alkanethiols (R-SH, where R = CH3, C2H5, and C4H9) on Pt(111) were studied with temperature-programmed desorption (TPD) and X-ray photoelectron spectroscopy (XPS) with synchrotron radiation. Dissociation of sulfhydryl hydrogen (RS-H) of alkanethiol results in the formation of alkanethiolate; the extent of dissociation at an adsorption temperature of 110 K depends on the length of the alkyl chain. At small exposure, all chemisorbed CH3SH, C2H5SH, and C4H9SH decompose to desorb hydrogen below 370 K and yield carbon and sulfur on the surface. Desorption of products containing carbon is observed only at large exposure. In thermal decomposition, alkanethiolate is proposed to undergo a stepwise dehydrogenation: R'-CH2S --> R'-CHS --> R'-CS, R' = H, CH3, and C3H7. Further decomposition of the R'-CS intermediate results in desorption of H2 at 400-500 K and leaves carbon and sulfur on the surface. On the basis of TPD and XPS data, we conclude that the density of adsorption of alkanethiol decreases with increasing length of the alkyl chain. C4H9SH is proposed to adsorb mainly with a configuration in which its alkyl group interacts with the surface; this interaction diminishes the density of adsorption of alkanethiols but facilitates dehydrogenation of the alkyl group.

Systematic structure-based design and stereoselective synthesis of novel multisubstituted cyclopentane derivatives with potent antiinfluenza activity.

The design and synthesis of novel, orally active, potent, and selective inhibitors of influenza neuraminidase differing structurally from existing neuraminidase inhibitors are described. X-ray crystal structures of complexes of neuraminidase with known five- and six-membered ring inhibitors revealed that potent inhibition of the enzyme is determined by the relative positions of the interacting inhibitor substituents (carboxylate, glycerol, acetamido, hydroxyl) rather than by the absolute position of the central ring. This led us to design potential neuraminidase inhibitors in which the cyclopentane ring served as a scaffold for substituents (carboxylate, guanidino, acetamido, alkyl) that would interact with the four binding pockets of the neuraminidase active site at least as effectively as those of the established six-membered ring inhibitors such as DANA (2), zanamivir (3), and oseltamivir (4). A mixture of the isomers was prepared initially. Protein crystallography of inhibitor-enzyme complexes was used to screen mixtures of isomers in order to identify the most active stereoisomer. A synthetic route to the identified candidate 50 was developed, which featured (3 + 2) cycloaddition of 2-ethylbutyronitrile oxide to methyl (1S,4R)-4[(tert-butoxycarbonyl)amino]cyclopent-2-ene-1-carboxylate (43). Structures of the synthetic compounds were verified by NMR spectroscopy using nuclear Overhauser effect methodology. Two new neuraminidase inhibitors discovered in this work, 50 and 54, have IC(50) values vs neuraminidase from influenza A and B of <1 and <10 nM, respectively. These IC(50) values are comparable or superior to those for zanamivir and oseltamivir, agents recently approved by the FDA for treatment of influenza. The synthetic route used to prepare 50 and 54 was refined so that synthesis of pure active isomer 54, which has five chiral centers, required only seven steps from readily available intermediates. Further manipulation was required to prepare deoxy derivative 50. Because the activities of the two compounds are comparable and 54 [RWJ-270201 (BCX-1812)] is the easier to synthesize, it was selected for further clinical evaluation.

Excess Kondo resonance in a quantum dot device with normal and superconducting leads: the physics of Andreev-normal co-tunneling.

We report on a novel Kondo phenomenon of interacting quantum dots coupled asymmetrically to a normal and a superconducting lead. The effects of intradot Coulomb interaction and Andreev tunneling give rise to Andreev bound resonances. As a result, a new type of co-tunneling process, which we term Andreev-normal co-tunneling, is predicted. At low temperatures, coherent superposition of these co-tunneling processes induces a Kondo effect in which Cooper pairs directly participate formation of a spin singlet, leading to four Kondo resonance peaks in the local density of states, and enhancing the tunneling current.

Direct determination of sinigrin in mustard seed without desulfatation by reversed-phase ion-pair liquid chromatography.

Reversed-phase ion-pair liquid chromatography has been investigated for directly analyzing sinigrin in mustard seed without desulfatation. After extraction by phosphate buffer (pH 7.0) from the grind-pastes of inactivated-myrosinase mustard seeds, sinigrin was first isolated through deproteinization and centrifugation, followed by filtration and injection into the chromatographic system. A reversed-phase C18 column was used to separate the sinigrin with an eluent of acetonitrile (ACN)-water (20:80) containing 0.02 M tetrabutylammonium (TBA) as the counter ion at pH 7.0. Detection was carried out with an UV detector operated at 227 nm. Factors affecting the chromatographic separation and quantitative determination, such as concentrations of TBA and ACN, and pH, were studied. The linear dynamic range is larger than three orders of magnitude and the detection limit is 0.045 mg/L. The RSD is around 3% and the recovery is 85% (3% RSD, n = 3).

Effects of nickel chloride on human platelets: enhancement of lipid peroxidation, inhibition of aggregation and interaction with ascorbic acid.

This study was undertaken to examine the effects of nickel (Ni) on human platelet function. In a (concentration-dependent manner, Ni significantly inhibited the function of platelet aggregation induced by collagen. The phenomenon of lipid peroxidation was involved as Ni significantly increased malondialdehyde (MDA) levels with reduction in platelet reduced glutathione (GSH) and alpha-tocopherol content. Further, platelet thromboxane B2, formation was markedly inhibited and the levels of cyclic AMP were significantly elevated by Ni. Treatment with ascorbic acid (Vit C) significantly lowered the levels of MDA and increased the content of alpha-tocopherol and reduced GSH. Vit C also significantly increased platelet aggregation and thromboxane B2 when coincubated with Ni. Data show that Ni is toxic as evidenced by lipid peroxidative damage and inhibition of human platelet aggregation, but that ascorbic acid provides protection, at least partially, against this metal.

In vitro effects of metal ions (Fe2+, Mn2+, Pb2+) on sperm motility and lipid peroxidation in human semen.

The effects of divalent manganese ion (Mn2+), ferrous iron (Fe2+), and lead ion (Pb2+) on human sperm motility and lipid peroxidation were examined. Human semen from healthy male volunteers was incubated with 0, 5, 50, or 500 ppm divalent metal ions, and the sperm motility was determined at 0, 2, 4, 6, or 8 h by microscopy. Malondialdehyde (MDA) levels in seminal plasma was measured by high-performance liquid chromatography after 8 h of exposure. The results showed that 500 ppm Mn2+ or Pb2+ significantly inhibited sperm motility without an accompanying change in seminal MDA levels. Incubation with Fe2+ significantly inhibited sperm motility at 5 ppm, associated with a marked rise in MDA levels. Our results suggested that Fe2+ may induce lipid peroxidation to inhibit sperm motility. In the case of Mn2+ and Pb2+ there is an absence of seminal lipid peroxidation and the observed inhibition of sperm motility at high concentrations is not biologically or environmentally relevant.

Antibody to the nonstructural protein NS1 of Japanese encephalitis virus: potential application of mAb-based indirect ELISA to differentiate infection from vaccination.

An indirect enzyme-linked immunosorbent assay (ELISA) was developed to detect and differentiate the antibody responses to Japanese encephalitis (JE) virus nonstructural protein NS1 between infected and vaccinated individuals. The results showed that all convalescent sera from JE patients contained NS1-specific IgG antibodies, while 65 and 40% of these sera showed detectable NS1-specific IgM and IgA antibodies, respectively. Specificity analysis showed that NS1-specific IgM and IgA antibodies from JE patients do not cross-react to dengue virus NS1 glycoprotein, while IgG antibodies from 10% of JE patients showed significant cross-reaction to dengue virus NS1 glycoprotein. To differentiate infection from vaccination, the immune sera from 24 children vaccinated with inactivated JE vaccine were analyzed. The data showed that none of these immune sera had detectable NS1-specific IgG antibodies. The results demonstrated the potential application of JE NS1-specific indirect ELISA to differentiate infection from vaccination.

Lipid peroxidation in rat adrenal glands after administration cadmium and role of essential metals.

The aim of this study was to investigate the effects of cadmium-induced peroxidative damage to rat adrenals. Cadmium significantly increased adrenal lipid peroxidation in a dose- and time-related manner. Cadmium-induced lipid peroxidation was accompanied by a marked elevation in adrenal iron (Fe) levels, in particular the free elemental form. Chelation of Fe with deferoxamine decreased the cadmium effect on lipid peroxidation. Selenium (Se) was also effective in inhibiting Cd-induced adrenal lipid peroxidation. Data indicate that Cd-induced lipid peroxidation in rat adrenals may be dependent upon Fe and Se levels in this tissue.

Three-dimensional quantitative structure-activity relationship for several bioactive peptides searched by a convex hull-comparative molecular field analysis approach.

Three-dimensional (3D) convex hulls are computed for theoretically generated structures of a group of 18 bioactive tachykinin peptides. The number of peptides treated as a training set is 14, whereas that treated as a test set is four. The frequency of atoms of the same atomic type lying at the vertices of all the hulls computed for all the structures in a structural set is counted. Vertex atoms with non-zero frequency counted are collected together as a set of commonly exposed groups. These commonly exposed atoms are then treated as a set of correspondences for aligning all the other 13 structures in a structural set against a common template, which is the structure of the most potent peptide in the set using the FIT module of the SYBYL 6.6 program. Each aligned structural set is then analyzed by the comparative molecular field analysis (CoMFA) module using the C.3 probe having a charge of +1.0. The corresponding cross-validated r2 values range from -0.99 to 0.57 for a number of 73 structural sets studied. The comparative molecular similarity indices analysis (CoMSIA) module within the SYBYL 6.6 package is also used to analyze some of these aligned structural sets. Although the CoMSIA results are in accord with those of CoMFA, it is also found that the CoMFA results of several structural sets can be improved somewhat for conformations of the structures in the sets that are adjusted by constraint energy minimization and then constraint molecular dynamics simulation runs using distance constraints derived from some commonly exposed groups determined for them. This result further implies that the convex hull-CoMFA is a feasible approach to screen the bioactive conformations for molecules of this class.

Two phenanthraquinones from Dendrobium moniliforme.

Two phenanthraquinones were isolated from the stems of Dendrobium moniliforme. They were identified by interpretation of spectral data as 2,6-dimethoxy-1,4,5,8-phenanthradiquinone and 5-hydroxy-3,7-dimethoxy-1,4-phenanthraquinone, named moniliformin (1) and denbinobin (2), respectively. This is the first report of compound 1, which possesses a novel 1,4,5,8-diquinone moiety in the phenanthraquinone skeleton. Compound 2 showed potent antiinflammatory effects in vitro.

Public health impact of changes in smoking behavior: results from the Tobacco Policy Model.

OBJECTIVES: The relative magnitude of the public health gains from preventing smoking initiation versus encouraging cessation or avoiding relapse in different ages and genders is estimated and compared. METHODS: Health gains are defined as the predicted increase in Quality-Adjusted Life-Years (QALYs) to the US population during a century. To estimate QALYs, we developed the Tobacco Policy Model. The model simulates a 10% reduction in the annual probability of initiation versus a 10% increase in cessation versus a 10% reduction in relapse in males and females in six age groups: 10 to 19, 20 to 29, 30 to 39, 40 to 49, 50 to 59 and 60 to 69. RESULTS: Among youth and young adults, reducing initiation yields far more QALYs than encouraging cessation or averting relapse. In middle-aged adults, cessation yields the most QALYs, followed by averting relapse and reducing initiation. In the oldest age group, averting relapse yields the most QALYs followed by cessation and reducing initiation. In general, increasing cessation and reducing relapse is more beneficial in males than in females whereas reducing initiation is more beneficial in females. CONCLUSIONS: The relative value of preventing initiation, encouraging cessation, and averting relapse differs by age and gender. Reducing initiation in youth is likely to offer the largest public health impact during the next century.

Cystic left atrium myxoma--a rare case report.

Myxoma is the most common primary tumor of heart. The typical picture of myxoma under echocardiography is a solid, dense echo mass and left atrium is the most common site to find it. The cystic form of myxoma is vary rare. We report a patient who received echocardiographic examination under impression of mitral valve stenosis. A multilobulated cystic mass which was like a hydatid cyst was found in the left atrium and atrioventricular flow was affected by this mass. After tumor resection, myxoma with internal hemorrhage was proved by pathology. No further recurrent myxoma was found during follow-up echocardiographic examination.

Protective effects of manganese against lipid peroxidation.

The aim of this study was to investigate the effects of chronic, daily, 30-d administration of manganese chloride (MnCl2) to male Sprague-Dawley rats on lipid peroxidation in various tissues. Rats were intraperitoneally injected with MnCl2 (20 mg/kg) once daily for 30 consecutive days. The Mn accumulated in liver, spleen, adrenal glands, heart, kidneys, lung, and testes. This was associated with decreased lipid peroxidation in liver, spleen, and adrenal glands and a decrease in the levels of Fe in these tissues. In a second group of animals, Mn (20 mg/kg/d) and glutathione (GSH, 15 mg/kg/d) were administered ip for 30 d. GSH counteracted the Mn-induced protective fall in lipid peroxidation, but Fe levels remained lower in liver and spleen. Mn decreases lipid peroxidation in certain tissues, which may involve lowering Fe content, but interaction with Fe is not the sole mechanism.
