Experimental approaches for the reconstitution of hair in vitro.

When plucked follicles were cultured together with isolated dermal papillae in a collagen gel matrix, outer root sheath cells (ORSCs) grew more rapidly and actively than without the papillae. Cultured dermal papilla cells also activated the colony growth of ORSCs in vitro. These results may suggest the existence of some papilla-derived factor(s) activating the growth of ORSCs. In cultures of excised whole follicles whose dermal papilla had been removed, epithelial cells of bulb matrix origin grew out from the bulbous portion, and formed spikes. When a dermal papilla was implanted close to the follicle, the spikes elongated toward the papilla, and finally reached and surrounded it. This finding suggests that dermal papillae may produce some factor(s) attracting epithelial cells of hair bulb origin. In cultures of excised whole follicles, when the dermal papilla remained originally positioned in contact with the hair bulb matrix, the hair and follicle elongated for more than 1 week. But when the dermal papilla was detached from the bulb matrix, the matrix cells proliferated into the gel and formed a hair-follicle-like structure (folliculoid). In cultures of excised whole follicles whose papilla-matrix junction had been damaged by dispase, elongation of the hair and follicle was almost completely suppressed. Thus, the attachment of the dermal papilla to the bulb matrix appears to be necessary for normal hair and follicle growth.

Effect of fatty acid on the accumulation of thiamine disulfide in rat skin.

The effect of long chain fatty acid (FA) and its analogs on the accumulation of thiamine disulfide (TDS) in rat skin using propylene glycol as a vehicle was studied in vitro. Lauric acid (12:0) increased the accumulation of TDS in skin, while myristic acid and stearic acid caused a slight decrease in accumulation. Lauryl alcohol and lauric acid methyl ester did not change the accumulation of TDS in the skin. The ratio of the amount of TDS accumulated in skin to the solubility of TDS in the vehicle increased dependent on the concentration of 12:0 added in the vehicle. It was suggested that the increase in the permeability coefficient of TDS by 12:0 results from the enhanced transport of TDS from the vehicle to skin.

Laparoscopy-assisted colostomy.

The surgical modality of laparoscopic cholecystectomy is now widely accepted, and other intra-abdominal operations are also being done laparoscopically. This paper presents a case of laparoscopy-assisted colostomy in which a complete obstruction of the sigmoid colon associated with intraperitoneal dissemination and massive ascites in the end-stages of ovarian cancer was encountered. This procedure was found to be less invasive and thus should be considered the treatment of choice for an obstruction of the gastrointestinal tract in patients with a poor general status.

[Caesarean section in a patient with past history of fulminant malignant hyperthermia].

Caesarean section in a 26 year old patient (50 kg, normal weight 36 kg) with past history of fulminant malignant hyperthermia (f-MH) is reported. Hyperthermia over 42 degrees C and subsequent cardiac arrest occurred during general anesthesia with nitrous oxide-oxygen-halothane and succinylcholine when she had been planned to perform hip joint arthroplasty at the age of 7. Prophylactic oral dantrolene administration started 4 days before the operation and the dose went up to 75 mg. Further 25 mg was given as premedication on the day of the operation. Intravenous dantrolene was administered 1.2 mg.kg-1 during the operation and 0.6 mg.kg-1 on the day after surgery. The Caesarean section was performed under spinal anesthesia with hyperbaric tetracaine 20 mg. No clinical symptoms of MH occurred during perioperative period. The delivered baby was a healthy female and no side effects of dantrolene were observed.

In vitro percutaneous absorption of thiamine disulfide from a mixture of propylene glycol and fatty acid.

The in vitro percutaneous transport of thiamine disulfide (TDS), an oxidized dimer of thiamine, from propylene glycol through excised abdominal rat skin was studied. The application of saturated, long-chain fatty acids [stearic acid (18:0), myristic acid (14:0), and lauric acid (12:0)] as enhancers to the system was also studied. TDS permeated through rat skin from propylene glycol with a flux of 2.5 +/- 0.8 micrograms/cm2/min. The flux was enhanced 31 times by 12:0 and 1.4 times by 14:0 and was suppressed to 80% of its original value by 18:0. The absorption of TDS could not be explained by TDS permeating across a dialysis membrane, but the interaction between TDS and fatty acids may influence the system. The results show the possibility of developing a transdermal thiamine delivery system.

In vitro percutaneous absorption of thiamine disulfide through rat skin from a mixture of propylene glycol and fatty acid or its analog.

Percutaneous absorption of thiamine disulfide, (TDS), a lipophilic derivative of thiamine, from a mixture of propylene glycol (PG) and fatty acid (FA) or its analog through rat skin was tested in vitro. Lauric acid (12:0) enhanced the absorption depending on its concentration in PG and showed a maximal enhancement at 10% w/w. At 10% w/v, lauryl alcohol also enhanced the absorption, but less than 12:0, which lauric acid methyl ester suppressed the absorption. The flux of TDS did not depend on the solubility of TDS in the vehicle, but on the permeability coefficient. From these results, it is suggested that FA increases the permeability coefficient not only because FA increases TDS diffusion by disrupting lipid packing in the stratum corneum but also, FA increases TDS partition to lipid phase by interacting with TDS.

Studies of the development of optical fiber sensors for biochemical analysis.

An optical fiber sensor utilizing Thymol blue and an ion-exchange resin complex in a cellulose acetate membrane was developed. By monitoring several different chromophores of Thymol blue, the sensor could measure the pH of the solution from 1.0 to 12.0 with good reproducibility. An optical fiber glucose sensor utilizing a cellulose acetate membrane containing glucose oxidase, 2,7-diaminofluorene dihydrochloride, and sodium N-(3-sulfopropyl)-3,3',5,5'-tetramethylbenzidine was developed. Reflectance changes at 580 nm were large enough to trace changes in glucose concentration in physiological saline solution.

Release kinetics of nicotinamide from fatty acid-nicotinamide equimolar complexes. II. Activation thermodynamic quantities.

The rates of release of nicotinamide (NAA) from fatty acid (FA)-NAA complexes, FA-NAA, were determined at various temperatures, and the thermodynamic quantities for the release of NAA were estimated. The results were compared with the previous results obtained for FA-thiamine disulfide (TDS) complexes, (FA)6(TDS). The values of activation enthalpy (delta H ++) and activation entropy (delta S ++) for the release of NAA from FA-NAA were positive and negative, respectively, indicating that the release of NAA is disadvantageous from not only enthalpic but also entropic viewpoints. The plots of delta H++ against the carbon number (n) in the constituent FA showed a zig-zag line with an upward convex at an odd-numbered position and the plots of the absolute values of (-delta S++) showed a zig-zag line with a downward convex at an odd-numbered position, though the positive value of delta H++ increases and the negative value of delta S++ decreases with an increasing n for either even-numbered or odd-numbered FA. It was found that the release of NAA from FA-NAA formed with odd-numbered FA is more disadvantageous enthalpically but more advantageous entropically as compared with that from FA-NAA formed with even-numbered FA. This phenomenon was similar to that observed for (FA)6(TDS). Furthermore, it is suggested that FA-NAA is formed at least by van der Waals forces and hydrophobic interactions and that van der Waals forces are dominant for the formation of FA-NAA formed with odd-numbered FA and that hydrophobic interactions are dominant for the formation of FA-NAA formed with even-numbered FA.

[A latent form of essential thrombocythemia presented as portal hypertension and associated with acquired von Willebrand syndrome].

A 32 year-old male patient was admitted to our hospital because of abdominal tumor. The examination on admission showed massive splenomegaly and esophageal varices although peripheral blood cell counts were within normal limits. Exploratory laparotomy was performed with the diagnosis of portal hypertension and revealed the multiple thrombus formations in the splenic vein and the extramedullary hematopoietic findings in the spleen by the microscopic examination. In vitro colony forming assay showed the formation of spontaneous erythroid colonies in cultures of progenitor cells (from peripheral blood mononuclear cells) in erythropoietin-poor medium. Increasing thrombocytosis was observed immediately after splenectomy, and hemorrhagic diathesis of nasal bleeding and gastrointestinal bleeding were also detected. The analysis of plasma von Willebrand factor (vWF) revealed the decrease of ristocetin cofactor activity and the lack of large multimeric components of vWF. These abnormal findings observed after splenectomy led to recovery through the administration of busulfan with the improvement of thrombocytosis. Accordingly, the course of the disease clearly indicated it to be the essential thrombocythemia represented as portal vein thrombosis and in latent form with normal cell counts in peripheral blood at the time of diagnosis, and subsequently, to develop into a full-blown form associated with acquired von Willebrand syndrome following splenectomy.

[A histological evaluation of the therapeutic effects and the clinical findings in prostatic carcinomas].

A histological evaluation of the effects of therapy has been conducted on 22 prostatic carcinoma cases, according to the criterion proposed by the committee to establish the general rule for clinical and pathological studies judging the effects of therapy in cases of prostatic cancer. Studied in particular was the relation between the histological effects of therapy and the clinical course. Marked therapeutic effects were observed in cases of low grade tumors. Further, the patients' prognosis tended to be affected by the histological grade rather than by the histological effect of the treatment. The course of the serum total acid phosphatase levels tended to correlate with the histological effect of the treatment.

Multi-component analysis of urine of dogs with liver diseases by high-performance liquid chromatography.

Components of the urine of normal dogs and dogs with liver diseases were analysed by high-performance liquid chromatography (HPLC) using a macroreticular anion-exchange resin with UV detection. The dogs with liver diseases were experimentally prepared by either Eck fistula by portal-caval anastomosis, dimethylnitrosamine-induced liver failure or total hepatectomy by operation. Eighteen chromatographic peaks were numbered and the cross-correlation coefficient of the intensities of every pair of peaks was computed. There were high correlations between several pairs of peaks according to the types and degrees of the hepatic failures, whereas no significant correlation was observed for the peak pairs of normal urine samples. The results showed that HPLC analysis of urine is useful in the diagnosis of the physiological state in liver dysfunction.
