Superovulation induction by human menopausal gonadotrophin in rabbits.

Superovulation induction in laboratory animals is an important technique as a means of providing eggs for embryonic research. Superovulation induction by human menopausal gonadotrophin (HMG) has not been fully established in rabbits. In the present study, the relationship between the dosage of injected HMG and the superovulatory response including fertilization rate was studied. The most satisfactory result was obtained in the group in which 30 iu of HMG was injected subcutaneously three times at 24-hr intervals. The superovulatory response by this method was considerably better than that by conventional six injections of follicle-stimulating hormone (FSH) having been widely accepted.

Desensitization to somatostatin analogue (Octreotide) observed in a patient with acromegaly.

We encountered a 33-year-old female patient with a pituitary growth hormone (GH)-secreting macroadenoma. The patient was treated with somatostatin analogue (Octreotide) in combination with bromocriptine for 2 months before a transsphenoidal adenomectomy was carried out. Octreotide (300-800 micrograms/day) in combination with bromocriptine was effective in reducing the size of the adenoma by 36%, but produced only a marginal decrease in serum GH. After the operation, bromocriptine alone (15 mg/day) did not lower the level of GH which was produced by residual adenoma tissue. When octreotide (200 micrograms/day) was resumed along with the bromocriptine one year after the operation, it effectively lowered serum GH for 6 months. Thereafter, octreotide therapy became ineffective with a concomitant rise in serum GH and somatomedin C, which was not accompanied by an increase in tumor size. This was a rare case of acromegaly that showed desensitization to octreotide after long-term treatment.

Effects of TNF-alpha injection into the ovarian parenchyma on luteal blood vessels in rabbits.

It has been reported that tumor necrosis factor-alpha (TNF-alpha) derived from luteal macrophage is concerned with luteolysis. In the present study, to evaluate the correlation between TNF-alpha and regression of luteal blood vessels, recombinant human TNF-alpha (rh-TNF) was injected into the parenchyma of pseudopregnant rabbit ovaries. These injections were performed on day 7 of pseudopregnancy (functional luteal phase). Only Mg++ and Ca++ free phosphate buffer saline (PBS-) as the solvent was injected in the control group. Estimations of conditions in the luteal vessels after the injections was performed by observations of luteal vascular corrosion casts under a scanning electron microscope. Concentrations of serum progesterone before or after the injection were also assayed. In the control group, no change in the structure of luteal vessels was observed after PBS-injection, but regressing blood vessels with strictures, obstructions and rugged surfaces on the vessels were observed, and also concentrations of serum progesterone decreased noticeably after rh-TNF injection. These findings suggest that TNF-alpha plays a role in angiolysis through luteolysis in the rabbit corpus luteum.

Inositol 1,4,5-trisphosphate 3-kinase activity in FRTL-5 cells: regulation of the enzyme activity by TSH.

Inositol 1,4,5-trisphosphate (InsP3) 3-kinase phosphorylates the Ca(2+)-mobilizing second messenger InsP3 to inositol 1,3,4,5-tetrakisphosphate (InsP4). InsP3 5-phosphatase dephosphorylates InsP3 to inositol 1,4-bisphosphate (InsP2). We compared the effects of TSH added to a culture of FRTL-5 thyroid cells on the activity of InsP3 5-phosphatase and InsP3 3-kinase. InsP3 3-kinase activity was decreased at a physiological concentration of TSH. Inhibition of this activity started after 3 h of incubation with TSH and was maximal after 24 h. In contrast, InsP3 5-phosphatase activity was not affected by TSH under the same conditions. The inhibitory effect of TSH on InsP3 3-kinase was characterized as follows: a) inhibition of activity was mimicked by both dibutyryl cyclic AMP and forskolin; b) activity obtained by mixing lysates of TSH-stimulated and non-stimulated cells was the sum of each activity measured separately; c) inhibition persisted after a crude lysate of TSH-stimulated cells had been subjected to SDS/polyacrylamide gel electrophoresis and the extraction of InsP3 3-kinase activity. The data suggest that TSH reduced the activity of InsP3 3-kinase in FRTL-5 cells either by a phosphorylation/dephosphorylation mechanism, or by affecting expression of the enzyme.

Melanotic neuroectodermal tumor of infancy: a molecular approach to diagnosis. Case report.

A case of melanotic neuroectodermal tumor of infancy arising from the transverse sinus is presented. The tumor was located on the outer surface of the dura and extended extracranially through the occipitomastoid suture. Two cell populations were observed: pigmented melanocyte-like cells and small neuroblast-like cells. Ultrastructural analysis revealed epithelial tumor cells and melanosomes at various stages. Expression of melanotransferrin messenger RNA transcripts within the tumor tissue was observed using a reverse transcriptase-polymerase chain reaction method in addition to immunohistological studies. The positive expression of melanotransferrin confirmed that this melanotic neuroectodermal tumor was derived from neural crest cells.

[CT findings of mass-forming pancreatitis: correlation with histopathologic findings].

We attempted to analyse the computed tomographic (CT) characteristics of mass-forming pancreatitis. Thirty inflammatory masses were studied using the combined method of dynamic CT (Dy-CT) and high dose enhancement CT (HD-CT) with a thin-section scan. From the correlation between the CT enhancement effect and the histopathologic changes, injury of the glandular element and proliferation of fibrous tissue were more prominent in inflammatory masses showing a different enhancement effect from the surrounding pancreatic parenchyma. In addition to these changes, constriction of the intrapancreatic artery caused a poor enhancement effect on Dy-CT, whereas marked round cell infiltration with lymphoid follicle formation increased the enhancement effect on HD-CT. This combined CT method is valuable for the diagnosis of mass-forming pancreatitis and adequately reflects its histopathologic condition.

[A case of pancreatic cancer surviving for 7 years 11 months following intra-arterial and portal infusion chemotherapy].

A case of unresectable pancreatic cancer in a 76-year-old female was reported, in which a tumor had liver involvement. She was treated with infusion chemotherapy of mitomycin C (MMC) from gastroduodenal artery and portal vein after palliative operation. A drug was administered via an extracorporeal infusion catheter. MMC was given totally 47 mg from artery and 27 mg from portal vein for 9 months. Then, the serum CA 19-9 level gradually decreased, the metastatic liver tumor disappeared and the pancreatic tumor became smaller. After that, the tumor did not grow. The patient survived for 7 years 11 months until she died from other causes. Thus, it seems that infusion chemotherapy might be tried in some cases of pancreatic cancer.

[Treatment of solid tumor by a small amount of direct current].

Solid tumor treatment was given in our animal laboratory to determine the mechanism of tumor disappearance by direct electric current, and clinical trials were done on 9 far advanced recurrent rectal cancers. Solid tumors of Yoshida sarcoma in Donryu rat were treated by 1 mA of constant direct current for 1 hour a day, for 4 days. The tumors disappeared in 13/16 within 21 days. Positive results of DNA Nick-end labeling and ladder patterns in the gel electrophoresis of DNA were observed in the regressing tumor specimen. It is considered that apoptosis is the one of the mechanisms of a disappearing tumor. As for the clinical trial concern, in 9 cases of rectal cancer, one CR and 5 PR were seen.

[Is CA125 useful in the management of recurrence of endometrial carcinoma?].

It has been considered that there is no useful serum tumor marker to diagnose endometrial cancer. But recently several reports describe how the serum CA125 value decreases in patients who have received bilateral oophorectomy and in postmenopausal women. In this study, we therefore determined a new cutoff level for serum CA125 and examined the usefulness of serum CA125 in the assessment of recurrence in 42 endometrial cancer cases. These cases were divided into 3 groups: one of 22 low risk cases, 10 moderate risk cases and 10 high risk cases cased by prognosis 1) In the low risk group, 5 cases had a higher serum CA125 level within 1 month after operation than before. 2) In 151 measurements, the mean CA125 value was 9.7 +/- 3.6U/ml, so we considered that 17 U/ml is the cutoff value for determining recurrence. 3) In the low risk group, the CA125 value was either lower than 17U/ml or it has not continued to increase. 4) We divided the high risk group into 3 subgroups into those who had died within 6 months, those who died within 13-14 months and those who died 2-5 years after the initial treatment. In the group who died 13-14 months after the initial treatment, the seurm CA125 level had continued to be high and there was no change in the value after the initial treatment. In the group who died 2-5 years after the initial treatment, the mean +/- S.D. for serum CA125 was 157.3 +/- 158.3U/ml, the maximum was 377U/ml and the minimum was 28U/ml.(ABSTRACT TRUNCATED AT 250 WORDS)

[CT findings of pancreatic carcinoma: evaluation with the combined method of early enhancement CT and high dose enhancement CT].

Computed tomographic (CT) findings of pancreatic ductal adenocarcinoma were studied with the combined method of early enhancement CT and high dose enhancement CT in 72 carcinomas. Common Findings were change in pancreatic contour, abnormal attenuation in a tumor and dilatation of the main pancreatic duct. The incidence of abnormal attenuation and dilatation of the main pancreatic duct and bile duct was constant regardless of tumor size. The finding of hypoattenuation at early enhancement CT was most useful for demonstrating a carcinoma. However, this finding was negative in ten cases, five of which showed inhomogenous hyperattenuation at high dose enhancement CT. The detection of change in pancreatic contour and dilatation of the main pancreatic duct was most frequent at high dose enhancement CT. The finding of change in pancreatic contour and/or abnormal attenuation in a tumor could be detected in 47 cases at plain CT, 66 at early enhancement CT and 65 at high dose enhancement CT. Since the four cases in which neither finding was detected by any CT method showed dilatated main pancreatic duct, there was no case without abnormal CT finding. This combined CT method will be a reliable diagnostic technique in the imaging of pancreatic carcinoma.

[Detection of wall motion abnormality in old myocardial infarction with principal component analysis to body surface potential distribution].

The purpose of this study was to examine the correlation between body surface potential distribution and the extent of abnormally contracting segments (ACS) of the left ventricle in patients with old myocardial infarction (MI). Body surface potential distribution was quantitatively analyzed using principal component analysis. The first 6 principal components were extracted from data set obtained from both 25 normal subjects and 100 patients. The z scores were calculated at every 4 or 8 msec in another 81 patients with previous MI. In conclusion, the principal component analysis on body surface maps successfully condensed mapped data without significant loss of total variance and minute differences in the extent of ACS were sensitively detected on z scores of principal components.

[Principal component analysis of body surface potential distribution and that clinical application].

The purpose of this study was to compress a huge amount of body surface potential data with principal component analysis. The first 6 principal components were extracted from data set obtained from both 25 normal subjects and 100 patients. The factor loading from the 1st to the 3rd principal component (PC) displayed dipolar distribution and that from the 4th to the 6th PC displayed multipolar distribution. In conclusion, the principal component analysis on body surface maps successfully condensed mapped data without significant loss of total variance and it was suggested that this method may be useful for detecting the presence of multipolar distribution.

Chemical modulation of 5-HT3 and 5-HT4 receptors affects the release of 5-hydroxytryptamine from the ferret and rat intestine.

In species which vomit, elevated intestinal serotonin (5-hydroxytryptamine, 5-HT) may stimulate abdominal vagal afferent fibers, which in turn evoke the vomiting reflex. The release of 5-HT from intestinal enterochromaffin (EC) cells is regulated by polymodal mechanisms. The object of this study was to evaluate the involvement of 5-HT autoreceptors in the regulation of 5-HT release from the small intestine. Functional studies were carried out using 5-HT3 receptor agonist and antagonists, and 5-HT4 receptor agonist. Ferret and rat ileal tissue were isolated and 5-HT released into the bathing solution was determined using HPLC with an electrochemical detector (ECD). We previously reported that cisplatin produced a significant increase in cumulative 5-HT release and that ondansetron, a selective 5-HT3 receptor antagonist, did not alter the 5-HT release from the ferret ileum. In this study, a selective 5-HT3 agonist, 2-methyl-5-HT, induced a dose-dependent increase of 5-HT from the rat ileum. This release of 5-HT was significantly reduced by granisetron, a selective 5-HT3 receptor antagonist. Furthermore, a selective 5-HT4 receptor agonist, 5-methoxytryptamine induced a concentration-dependent increase of 5-HT in the rat ileum. These results suggest that both 5-HT3 and 5-HT4 receptors may be involved in intestinal 5-HT release.
