Rapid and large-scale isolation of microsomal fraction of mouse liver by lyophilization and low speed centrifugation.

We could prepare the microsomal fraction of mouse liver, without using an ultracentrifuge but with a low speed centrifuge. The procedure includes 1) lyophilization of post-mitochondrial fraction (9,000 x g supernatant) of mouse liver, 2) powdering of the lyophilized sample, 3) the addition of 1.15 per cent potassium chloride solution or distilled water, which afforded microsomal aggregates, 4) sedimentation of microsomal fraction by low-speed centrifugation (20,000 x g, 20 min). The sedimented microsomal fraction showed normal contents of cytochrome P-450 and cytochrome b5, and gave a normal pattern on SDS polyacrylamide gel electrophoresis and normal electron microscopic feature. This method should be convenient for rapid and large-scale preparation of microsomes, especially for the preparation of cytochrome b5 and cytochrome P-450.

Molecular cloning of a partial cDNA clone encoding the C terminal region of chicken breast muscle connectin.

The cDNA sequence encoding the C terminal region of chicken skeletal muscle connectin was described. Its predicted amino acid sequence had 1,021 amino acids comprising six motif Ils (Immunoglobulin C2 domain) and five interdomains. The sequence showed 70-75% homology with that of human cardiac connectin, but 168 amino acids including one motif II were missing in chicken skeletal muscle connectin. The C terminal sequence of chicken skeletal muscle connectin reported by the previous work (Maruyama et al., 1994) was erroneous due to the accidental ligation of the cDNA clone encoding a N terminal region of connectin with a partial porin cDNA clone.

Effect of 4-acryloxyethyltrimellitic acid in a self-etching primer on bonding to ground dentin.

To develop a self-etching primer for ground dentin, 4-acryloxyethyltrimellitic acid (4-AET) was newly synthesized, and the effect of concentrations varying from 0 (control) to 56.5 wt% 4-AET in the water/ HEMA primer on bonding to the dentin was investigated. Bond strength to the dentin was significantly affected by the inclusion of 4-AET in the primer which also contained N,N-di(hydroxyethyl)-p-toluidine (DEPT) when compared with the control (0% 4-AET) (p < 0.01). The optimum 4-AET-concentration and the mean bond strength (SD) were found to be 6.5 wt%: 24.2 (3.6 MPa, 37.4 wt%: 25.3 (4.4) MPa, 47.2 wt%: 26.9 (11.6) MPa and 54.4 wt%: 29.7 (12.9) MPa. The role of DEPT in the 4-AET/HEMA primer was assessed, and the optimum DEPT-concentration was found to be 0.154 and 0.307 mol%. Regarding the hypothetical bonding mechanism to dentin, it was thought that the ionized 4-AET in water/HEMA would penetrate into dentin substrates, and DEPT as an accelerator in situ would facilitate photo-polymerization at the dentin-resin interface, and result in increased bond strength to ground dentin.

Effects of radical polymerizable spirocyclic monomers with new polymerization initiator systems on adhesion.

To develop a non-polymerization shrinkage adhesive resin, the effects of the contents varying 0-20 wt% radical polymerizable spirocyclic monomers of 2-methylene-1, 4, 6-trioxyaspiro [4, 6] undecane (MTSU) and 2-methylene-8, 9-benzo-1, 4, 6 trioxyaspiro [4, 4] nonane (BTSN) together with three kinds of initiator systems in adhesive resin on bonding to both Ni-Cr alloy and unetched enamel were investigated. Tensile bond strength to either Ni-Cr alloy or unetched enamel was affected by the inclusion of MTSU or BTSN, and indicated that increased contents of MTSU or BTSN resulted in decreased bond strength to the metal in the negative correlations. Regarding the bond strength to Ni-Cr alloy, however, the effective concentration of BTSN was found to be 5 wt%, and the mean bond strength was 58.5 and 58.6 MPa, respectively. The data of differential scanning calorimeter measurement, on the other hand, showed that the copolymerization of MTSU with conventional methacrylate monomers was not completely achieved.

[Renal cell carcinoma associated with circumferential "ring-like" calcification].

A 37-year-old man was found to have large calcification in the left kidney in a health check. This calcification has not changed in size during 3 years with computerized tomographic (CT) studies. However, a suspicious lesion for a malignant tumor with the calcification was suggested by a magnetic resonance image (MRI) study. Thus, the patient underwent tumor enucleation and the histology showed the mixed typed renal cell carcinoma. Although it remains controversial, partial nephrectomy or tumor enucleation would be considered in a case of renal cell carcinoma with calcification, which is usually considered to have a favorable prognosis compared to that of a tumor without a calcification.

Mammographic diagnosis of breast cancer.

It is very important for exact diagnosis of early breast cancer that both the mammography quality and the film reading ability are on a high level. Not only the radiology technician but doctors must know the mammographic apparatus and quality control to obtain high-quality mammograms. The understanding of the mammographic technique will help systematic reading of the mammograms. Knowledge of the correlation with mammographic findings and pathological findings will also be useful to diagnose the mammograms accurately and rapidly.

[Recent improvement of mammographic diagnosis].

The improvement of mammographic diagnosis consists of 3 factors as follows; (1) improvement of mammographic apparatus, (2) technical advancement on photographing and reading of mammograms, and (3) correlation between mammographic findings and pathology. Improvement of apparatus was achieved by molybdenum anode and faster single screen combined with a high-speed single-emulsion film combination system. These system provide the dose reduction as only one day reduction of the average future time if anyone take the mammographic screening of breast cancer once a year over 30 years old. These alterations of apparatus produced the mammograms darkers, and the intensity of illumination of the view system brighter. These some improvements of the mammography have made the mammograms to be very high quality. As natural consequence mammographic diagnosis has some new roles. Screening of the breast cancer is the most important one. The diagnosis the lesion cancer or not is very important, too. If the lesion was found only with mammography, the biopsy under mammographic guidance is necessary. And the diagnosis of the extension of the cancer lesion is very important for surgical treatment. If the lesion has fine calcifications, mammography can diagnose the extent very objectively.

[Adhalin gene mutations in malignant limb-girdle muscular dystrophy and clinical features in adhalin-deficient muscular dystrophy].

We have reported adhalin gene mutations in 4 patients from 3 families with malignant limb-girdle muscular dystrophy (MLGMD), and summarized the clinical features in adhalin-deficient muscular dystrophy (ADMD) reported as severe childhood autosomal recessive muscular dystrophy (SCARMD) in the English literatures. Adhalin cDNA amplified from RNA by reverse transcription polymerase chain reaction (RT-PCR) was sequenced in 3 patients from 2 consanguinous families (Wa. and Ta.) with MLGMD who showed immunohistochemically a complete deficiency of adhalin in the skeletal muscle, and adhalin genomic DNA amplified by PCR was sequenced in 1 patient from a non-consanguinous family (Ma.). In one patient from family Wa., a cytosine to thymine substitution at nt. 229 was identified in the adhalin gene, resulting in the replacement of Arg by Cys at codon 77. In two patients from family Ta., an adenine to guanine substitution at nt. 410 and an insertion of 15 bases between nt. 408 and 409 were identified, resulting in Glu to Gly replacement at codon 137 and insertion of a peptide with 5 amino acids. In one patient from family Ma., a deletion of adenine at nt. 404 or nt. 405 and a thymidine to cytosine substitution at nt. 470 were identified. These amino acid replacements are expected to change the secondary and tertiary structure, which may affect the interaction of adhalin with other dystrophin-associated glycoproteins and basal lamina, and may subsequently cause the degeneration of muscle fibers. Sixty-six cases from 49 families with ADMD have been reported in the literature. Compared with patients with Duchenne muscular dystrophy (DMD), patients with ADMD were older in age at the time of onset or loss of ambulation. Mental retardation and cardiac dysfunction were rarely observed in ADMD patients. On muscle histology, the number of necrotic fibers, opaque fibers and regenerative fibers was less in ADMD. ADMD was classified into two groups; complete and incomplete adhalin-deficient. There was no essential difference between the two groups in clinical features and muscle histology, but the former was characterized by more severe clinical features than the latter. ADMD can be caused by various types of mutations in the adhalin gene.

[Comparative study on the assay for IgE antibodies specific for Chamaecyparis obtusa pollen between AlaSTAT and CAP-RAST].

Titers of IgE antibody specific for the pollen of Chamaecyparis obtusa (C. obtusa) were determined by AlaSTAT and CAP-RAST in 221 patients with Japanese cedar pollinosis. IgE antibody to C. obtusa tested positive by CAP-RAST at a higher rate (80.5%) than by AlaSTAT (52.6%). The results obtained from the two assays were compared with those from intradermal skin test. CAP-RAST had a higher sensitivity than that of AlaSTAT. Because the two methods showed no differences in the determination of IgE antibody specific for Cryptomeria japonica, the above differences between AlaSTAT and CAP-RAST are surmised to be ascribable to the differences of C. obtusa antigen used in the both assays.

[Experimental studies on the cell adhesion molecule E-cadherin and in vitro invasion of endometrial carcinoma cell lines].

The enhancement of the in vitro invasive ability and the morphological changes caused by anti-E-cadherin antibody HECD-1 were investigated by in vitro invasion assay and electron microscopy in three human endometrial carcinoma cell lines. The cell lines were NUE-1 (E-cadherin negative and high in vitro invasive ability), HEC-1BE and HEC-108 (E-cadherin positive and low in vitro invasive ability). In NUE-1 invasive ability was not enhanced by HECD-1, but in HEC-1BE and HEC-108 invasive ability was enhanced to 223 +/- 41.2% and 307 +/- 173% by 5 micrograms/ml HECD-1. Morphologically NUE-1 invaded the extracellular matrix (Matrigel) with a long micro villis. But in HEC-1BE and HEC-108 the villis did not invade the Matrigel, the whole cell invaded it. Together with HECD-1, HEC-1BE and HEC-108 were changed to become similar to the NUE-1 cell line with high invasive ability and the micro villis invaded the Matrigel.
