Coccinin, an antifungal peptide with antiproliferative and HIV-1 reverse transcriptase inhibitory activities from large scarlet runner beans.

An antifungal peptide, designated coccinin, with a molecular mass of 7kDa and an N-terminal sequence resembling those of defensins, was purified from the seeds of large scarlet runner beans (Phaseolus coccineus cv. 'Major'). The peptide isolated was unadsorbed on DEAE-cellulose, and adsorbed on Affi-gel blue gel and Mono S. The peptide excerted antifungal activity on a number of fungal species including Botrytis cinerea, Coprinus comatus, Fusarium oxysporum, Mycosphaerella arachidicola, Physalospora piricola, and Rhizoctonia solani. It also inhibited proliferation in the leukemia cell lines HL60 and L1210, and reduced the activity of HIV-1 reverse transcriptase. However, it did not affect proliferation of mouse splenocytes.

A napin-like polypeptide with translation-inhibitory, trypsin-inhibitory, antiproliferative and antibacterial activities from kale seeds.

A heterodimeric napin-like polypeptide with translation-inhibiting and antibacterial activities has been isolated from kale seeds. The purification procedure entailed ion-exchange chromatography on dielthylaminoethyl (DEAE)-cellulose, affinity chromatography on Affi-gel blue gel, ion-exchange chromatography by fast protein liquid chromatography (FPLC) on Mono S, and gel filtration by FPLC on Superdex 75. The napin-like polypeptide was unadsorbed on DEAE-cellulose but adsorbed on Affi-gel blue gel and Mono S. Its 7-kDa large subunit differs in N-terminal amino acid sequence from the 4-kDa small subunit. The polypeptide inhibited translation in the rabbit reticulocyte lysate system with an IC50 of 37.5 nM. This activity was preserved between pH 5 and pH 11, and between 10 and 40 degrees C. It fell to a low level at pH 3 and pH 13 and at 70 degrees C. Antibacterial activity against Bacillus, Megabacterium, and Pseudomonas species and antiproliferative activity against leukemia L1210 cells were observed. However, the polypeptide did not exert antifungal, ribonuclease, or protease activity.

A gallic acid derivative and polysaccharides with antioxidative activity from rose (Rosa rugosa) flowers.

In this study, the major antioxidant components of rose flower were identified. An aqueous extract of rose flowers was chromatographed on CM-cellulose in ammonium acetate buffer (10 mM, pH 4.5) to yield three un-adsorbed peaks F1, F2 and F3. Each of these peaks was subjected to gel filtration on Sephadex G75. F1 yielded two peaks, whereas both F2 and F3 gave rise to only a single peak. Spectroscopic studies using NMR and FTIR revealed that F3 is a gallic acid derivative. It exhibited the highest antioxidative potency. F1-a derived from F1 by gel filtration is mainly a polysaccharide-peptide complex with less potent antioxidative activity. F2 is a polysaccharide also with reduced antioxidant activity. This study demonstrates, for the first time, the presence of both gallic acid derivatives and polysaccharides as major antioxidant principles of the aqueous extract of rose flowers.

Dendrocin, a distinctive antifungal protein from bamboo shoots.

An antifungal protein, with a molecular weight of 20 kDa and an inhibitory action on mycelial growth in the fungi Fusarium oxysporum, Botrytis cincerea, and Mycosphaerella arachidicola, was isolated from fresh bamboo shoots. The protein, designated dendrocin, was unadsorbed on DEAE-cellulose and adsorbed on Affi-gel blue gel and CM-Sepharose. Dendrocin showed only limited similarity in N-terminal sequence to thaumatin-like proteins, unlike other thaumatin-like proteins which closely resemble each other. Its molecular weight was also lower than those of the previously reported thaumatin-like proteins. The protein was devoid of hemagglutinating and ribonuclease activities found in some antifungal proteins.

Isolation of a large thaumatin-like antifungal protein from seeds of the Kweilin chestnut Castanopsis chinensis.

A protein with an N-terminal sequence showing a much lesser extent of homology than French bean and kiwi fruit thaumatin-like proteins (TLPs) to other TLPs, and possessing a molecular mass of 30 kDa which is considerably higher than those of previously reported TLPs, has been purified from the seeds of the chestnut Castanopsis chinensis Hance. The protein was unadsorbed on DEAE-cellulose in 10 mM Tris-HCl buffer (pH 7.3), and adsorbed on Affi-gel blue gel in the same buffer, on CM-cellulose in 10 mM ammonium acetate buffer (pH 4.5), and on Mono S in 20 mM ammonium acetate buffer (pH 5.5). A highly purified protein preparation was obtained after fractionation on the first three chromatographic media. Castanopsis TLP appeared as a single band (30 kDa) in sodium dodecyl sulfate-polyacrylamide gel electrophoresis and as a single peak (30 kDa) in gel filtration on Superdex 75 by fast protein liquid chromatography. The TLP exerted antifungal activity against Botrytis cinerea, Fusarium oxysporum, Mycosphaerella arachidicola, and Physalospora piricola, with an IC(50) of 0.5 microM against F. oxysporum. Castanopsis TLP was more potent than French bean and kiwi fruit TLPs in its antifungal activity toward F. oxysporum and M. arachidicola. The antifungal activity of Castanopsis TLP remained essentially unaltered after incubation at 40 degrees C for 10 min, was reduced after incubation at 60 degrees C, and disappeared after treatment at 80 degrees C. The antifungal activity underwent a decline after treatment with trypsin (enzyme:substrate ratio 1:100) at 37 degrees C for 1h but some activity remained. Castanopsis TLP exhibited a much more potent inhibitory activity on HIV-1 reverse transcriptase (IC(50) = 1.6 microM) than kiwi fruit TLP (IC(50) > or = 27 microM). Castanopsis TLP was obtained with a yield of 20 mg from 1 kg chestnut seeds.

Purification of a trypsin-stable lectin with antiproliferative and HIV-1 reverse transcriptase inhibitory activity.

A lectin, with a molecular mass of approximately 60 kDa and two different subunits exhibiting an N-terminal sequence manifesting considerable homology to phytohemagglutinin from Phaseolus species, was isolated from the ground bean (Vigna sesquipedalis cv ground bean). The lectin was unique in hemagglutinating activity was inhibited by polygalacturonic acid and not by galacturonic acid and other simple monosaccharides. The lectin was isolated by affinity chromatography on Affi-gel blue gel, ion exchange chromatography by fast protein liquid chromatography (FPLC) on Mono S, and gel filtration by FPLC on Superdex 75. It was adsorbed on both Affi-gel blue gel and Mono S. Ground bean lectin exhibited mitogenic activity on murine splenocytes with the maximal response achieved at a concentration of 156 nM, as similar to the dose required for Con A. The viability of hepatoma (HepG2), leukemia (L1210), and leukemia (M1) cells was reduced in the presence of ground bean lectin, which also exerted an inhibitory activity toward HIV-1 reverse transcriptase IC(50) of 73 microM. The hemagglutinating activity of the lectin was unaffected by trypsinization and the presence of a number of divalent cations, but was augmented by 500 mM K(+) ions. The activity was unstable above 40 degrees C although some activity remained after heating and at 100 degrees C for 30s.
