Roles for both the RAP30 and RAP74 subunits of transcription factor IIF in transcription initiation and elongation by RNA polymerase II.

Mammalian transcription factor IIF (TFIIF) is a heterodimer composed of approximately 30-kDa (RAP30) and approximately 70-kDa (RAP74) subunits. TFIIF has been shown to bind RNA polymerase II and control the activity of the enzyme in both the initiation and elongation stages of transcription. Although previous studies have established a role for RAP30 in assembly of the preinitiation complex and in transcription initiation, information on the function of RAP74 in these processes has been lacking. Using a highly purified transcription system and assays that permit sensitive measurement of the contributions of both RAP30 and RAP74 to TFIIF function, we have investigated the roles of these TFIIF subunits in transcription initiation and elongation. Results of template competition experiments indicate that both RAP30 and RAP74 contribute to the formation of stable preinitiation intermediates containing RNA polymerase II. Investigation of the role of TFIIF in transcription initiation indicates that both RAP30 and RAP74 function in synthesis of the first few phosphodiester bonds of nascent transcripts and in formation of Sarkosyl-resistant pre-initiation intermediates. Finally, kinetic experiments indicate that both RAP30 and RAP74 function in TFIIF-mediated stimulation of the rate of RNA chain elongation by RNA polymerase II.

A TATA sequence-dependent transcriptional repressor activity associated with mammalian transcription factor IIA.

In the process of characterizing cellular proteins that modulate basal transcription by RNA polymerase II, we identified a novel repressor activity specific for promoters containing consensus TATA boxes. This activity strongly represses TATA-binding protein (TBP)-dependent transcription initiation from core promoter elements containing a consensus TATA sequence, but activates TBP-dependent transcription from core promoter elements lacking a consensus TATA sequence. Purification of this activity to near homogeneity from rat liver nuclear extracts led to the surprising discovery that it co-purifies closely with mammalian transcription factor IIA (TFIIA). The close association of TATA sequence-dependent transcriptional repressor activity with TFIIA adds a new and unexpected dimension to the already complex picture of this factor's function in transcription by RNA polymerase II.

Japanese type of spondylo-metaphyseal dysplasia.

Five members of a Japanese family with a new form of spondylo-metaphyseal dysplasia (SMD) are reported. Another member was also probably affected. The disease was characterised by severe coxa vara, moderately severe metaphyseal changes of the long bones of the lower limbs, mild changes in the long bones of the upper limbs and grossly normal short tubular bones. Platyspondyly, present in the boys, was less marked in their father, whereas two affected aunts had normally shaped vertebral bodies.

Autocrine/paracrine function of transforming growth factor-beta 1 in porcine granulosa cells.

The present study was undertaken to investigate the effects of transforming growth factor (TGF)-beta 1 on ovarian steroidogenesis in immature or moderately mature porcine granulosa cells in vitro. TGF-beta 1 (0.01-10 ng/ml) significantly attenuated progesterone release from the basal and follicle stimulating hormone-stimulated porcine granulosa cells, and significantly increased DNA synthesis. TGF-beta 1 also significantly increased the extracellular accumulation of cyclic AMP, but did not change the production of inositol phosphate or the intracellular calcium concentration in these cells. Thus, TGF-beta 1 appears to have a direct effect on porcine granulosa cell function, regulating the differential synthesis of progesterone mainly via the intracellular signal transduction of the cyclic AMP-protein kinase A pathway. This growth factor may play a physiologically significant role in controlling differentiation of immature and moderately mature porcine granulosa cells via an autocrine/paracrine mechanism.

Endothelin-1 as a local ovarian regulator in porcine granulosa cells.

This study was conducted to investigate whether endothelin-1 (ET-1) has direct effects on the functions of porcine granulosa cells harvested from small or medium follicles. Positive ET-1-like immunoreactivity (ET-1-LI) was observed in the cultured porcine granulosa cells, and Northern blot analysis demonstrated the expression of mRNA for prepro-ET-1 in these cells. The binding study showed the presence of a nonselective, single class of binding sites which predominantly included the subtype ETB. Increased ET-1-LI was detected in human follicular fluid obtained from the patients with stimulated ovarian cycles. ET-1 stimulated basal and follicle-stimulating hormone (FSH)-stimulated secretion of progesterone during short-term incubation (2 h), while it inhibited FSH- and human-chorionic-gonadotropin-stimulated accumulation of progesterone during long-term incubation (48 h) of immature or moderately mature granulosa cells. ET-1 significantly increased DNA synthesis in the cells. These biological actions were induced by ET-1, probably via a cAMP-protein kinase A pathway and intracellular calcium mobilization-protein kinase C pathway. The results suggest that ET-1 exerts autocrine/paracrine effects on porcine granulosa cell functions.

Incomplete androgen insensitivity associated with a thermolabile androgen receptor.

One infant and a cousin with incomplete androgen insensitivity syndrome were reported. The familial pedigree showed that the disorder was inherited in three generations in X-linked recessive fashion. An androgen binding study of cultured genital skin fibroblast from patients showed normal maximum binding capacity and a normal apparent dissociation constant. Heat stability assay showed binding decreased to less than 30% at 41 degrees C compared with the amount at 30 degrees C, indicating that the androgen receptor was thermolabile.

Hypopituitarism with invisible pituitary stalk: two case reports of males with micropenis suggesting fetal onset of hypopituitarism.

The presence of hypopituitarism and invisible pituitary stalk on a magnetic resonance image (MRI) is commonly attributed to birth trauma. Two patients with severe hypopituitarism and invisible pituitary stalk are presented. One was born by breech delivery, the other by Cesarean section. The presence of a micropenis since early infancy in these two patients suggested that their hypopituitarism might have begun during early fetal life thus effecting penile growth during the second and third triministers of gestation. These findings raise the possibility that the association of hypopituitarism and invisible pituitary stalk may have multiple etiologies including hormonal abnormalities during early fetal life.

Clinical utility of insulin-like growth factor binding protein-3 in the evaluation and treatment of short children with suspected growth hormone deficiency.

We have shown previously that serum insulin-like growth factor binding protein-3 (IGFBP-3) levels have good predictive value for complete, but not partial, growth hormone deficiency (GHD). In this study, we compare IGFBP-3 levels in short children previously divided into groups on the basis of their post-stimulation GH levels. Complete GHD (N = 59) included those children with peak post-stimulation GH < 5 micrograms/l. The partial GHD group (N = 49) had post-stimulation GH peaks of > 5 micrograms/l but < 10 micrograms/l. The normal children with short stature (N = 103) had post-stimulation GH peaks > 10 micrograms/l. Partial GHD and normal children with short stature also were divided into either low IGF-I or normal IGF-I subgroups. The clinical sensitivity of IGFBP-3 for complete GHD was 92%, whereas its sensitivity for partial GHD was 39%. For partial GHD, among those with low IGF-I (N = 19) 68% were also low for IGFBP-3, while 80% of those with normal IGF-I (N = 30) were also normal for IGFBP-3. The clinical specificity of IGFBP-3 for normal children with short stature was 69%. For these groups, among those with low IGF-I (N = 22) 73% also were low for IGFBP-3, while 80% of those with normal IGF-I (N = 81) also were normal for IGFBP-3. In addition, we tested whether IGFBP-3 can predict the response to GH treatment in prepubertal children by comparing pretreatment IGFBP-3 with the height gain achieved by 1 year of GH treatment.(ABSTRACT TRUNCATED AT 250 WORDS)

Domain structure of a human general transcription initiation factor, TFIIF.

The structural and functional domains of a general transcription initiation factor, TFIIF (RAP30/74, FC), have been investigated using various deletion mutants of each subunit, both in vivo and in vitro. An in vivo assay showed that the N-terminal sequence containing residues of 1-110 of RAP30 that is located close to a sigma homology region interacts with a minimum sequence of residues 62-171 of RAP74 to form a heteromeric interaction. Reconstitution of in vitro transcription activity by deletion mutants of RAP74 clearly indicated that both N-terminal residues 73-205 and C-terminal residues 356-517 are essential for full activity, the former interacting with RAP30, thus complexing with RNA polymerase II. From these data, the functional significance of domain structure of TFIIF is discussed in terms of its sigma homology sequences and complex formation with RNA polymerase II in the initiation and elongation of transcription.

Assignment of the human GTF2F1 gene to chromosome 19p13.3.

TFIIF (also termed FC) is a general transcription initiation factor that binds to RNA polymerase II and recruits it to a promoter. TFIIF is a heterodimer composed of 74-kDa (RAP74; gene symbol GTF2F1) and 30-kDa (RAP30; gene symbol GTF2F2) subunits. Here we report the mapping of the human GTF2F1 gene to band 19p13.3. Localization was performed by fluorescence in situ hybridization using the human RAP74 genomic cosmid clone as the probe.

Endothelin-1 is an autocrine/paracrine regulator of porcine granulosa cells.

We studied whether a novel vasoconstrictor, endothelin-1 (ET-1), is synthesized by and released from porcine granulosa cells, and whether ET-1 acts directly on granulosa cells in an autocrine/paracrine fashion. The dilution curve of the conditioned medium from cultured porcine granulosa cells was parallel to a standard curve of ET-1 in RIA. Reverse-phase HPLC of the conditioned media from the granulosa cells revealed a major peak of ET-1-like immunoreactivity (ET-1-LI) coeluting with standard ET-1. ET-1-LI was released from cultured porcine granulosa cells as a function of time. Northern blot analysis demonstrated the expression of mRNA for prepro-ET-1 in the granulosa cells. We demonstrated the presence of ET-1 in the follicular fluid of porcine ovaries, and that the concentration of ET-1 in large follicles was higher than that in small-medium follicles. ET-1 dose-dependently stimulated cell growth and DNA synthesis in porcine granulosa cells. ET-1 inhibited the FSH- and hCG stimulated accumulation of progesterone in porcine granulosa cells in long-term incubation. These findings suggest that ET-1 produced by porcine granulosa cells may function as an autocrine/paracrine growth factor and modulator of steroidogenesis in ovarian granulosa cells.

Insulin-like growth factor II in follicular fluid of the patients with in vitro fertilization and embryo transfer.

OBJECTIVE: To investigate whether insulin-like growth factor II (IGF-II) is present in follicular fluids (FF) and whether IGF-II in FF plays an important role on human ovarian function. DESIGN, SETTING, PATIENTS: Insulin-like growth factor II concentrations were measured using an RIA technique in 46 samples of human FF obtained from 20 patients who were undergoing IVF and ET. The distribution profiles of unsaturated IGF-II binding protein were also investigated. Moreover, the effect of IGF-II on steroidogenesis by cultured granulosa cells (GCs) obtained simultaneously in the IVF-ET program was investigated. RESULTS: The IGF-II levels in FF (92.7 +/- 7.5 nmol/L) were approximately eight times greater than those of IGF-I (11.4 +/- 1.0 nmol/L), and significant positive correlations were observed between these IGFs in FF. By Sephadex G-150 gel-chromatography of FF, two apparent peaks of unsaturated IGF-II binding protein could be detected in the high molecular weight (MW) (150 kd) and low MW (approximately 36 to 38 kd) regions. Additionally, IGF-II dose dependently increased the release of P and E2 from the cultured human GCs. CONCLUSIONS: These findings suggest that the large quantity of IGF-II in FF may play possibly some roles in the ovarian steroidogenesis.

High levels of immunoreactive endothelin-1 in human follicular fluids.

Follicular fluids were obtained from 180 follicles of 15 women undergoing follicular aspiration for in-vitro fertilization. Follicular development was induced by a combination of buserelin acetate and human menopausal gonadotrophin. Endothelin-1 (ET-1) concentrations in human follicular fluids were determined by specific radioimmunoassay. ET-1-like immunoreactivity (ET-1-LI) ranged from 338 to 928 pg/ml. ET-1-LI concentrations in follicular fluids obtained from immature (< 15 mm) follicles were significantly higher than those from mature (15-25 mm) and post-mature (> or = 25 mm) follicles. No correlation was found between the concentration of ET-1-LI, on the one hand, and that of oestradiol, progesterone, testosterone, prolactin, luteinizing hormone, insulin-like growth factor-I, prostaglandin E2 or platelet activating factor on the other, in follicular fluids. However, a significant positive correlation was observed between ET-1-LI concentration and follicle stimulating hormone and IGF-II concentrations, respectively. These data suggest that the high concentration of ET-1 found in follicular fluids may play some physiological role in follicular development.

Comparison between insulin-like growth factor-I (IGF-I) and IGF binding protein-3 (IGFBP-3) measurement in the diagnosis of growth hormone deficiency.

To analyze the utility of insulin-like growth factor-I (IGF-I) and insulin-like growth factor binding protein-3 (IGFBP-3) radioimmunoassay in the diagnosis of growth hormone deficiency (GHD) we measured IGF-I and IGFBP-3 in sera from normal children (n = 309), short children (n = 99) and patients with GHD (n = 73). In 80% and 93% of classical GHD (cGHD), IGF-I and IGFBP-3 levels, respectively, were below the age-related cutoff levels (lower limit). In 81% and 88% of normal short children (NS), IGF-I and IGFBP-3 levels, respectively, were above the age-related cutoff levels. Thus, both IGF-I and IGFBP-3 were good parameters for screening GHD. In contrast, in more than half of partial GHD (pGHD), either IGF-I or IGFBP-3 was above the age-related cutoff levels. The poor discrimination between patients with pGHD and NS by using these two parameters may be the result of their relatively similar GH levels, as compared to cGHD, or due to the limitations of GH stimulation tests. In about 80-90% of NS, IGF-I and IGFBP-3 were above the age-related cutoff levels at all ages. A hundred percent of cGHD under 10 years old had IGFBP-3 below the age-related cutoff levels, whereas 79% of cGHD under 10 years old had IGF-I below the age-related cutoff levels. Thus in the younger age groups, IGFBP-3 may be more sensitive than IGF-I. It may be because IGFBP-3 levels are relatively higher than those of IGF-I in younger subjects.(ABSTRACT TRUNCATED AT 250 WORDS)

The effects of chemotherapy including cisplatin on vitamin D metabolism.

Plasma concentrations of three major vitamin D3 metabolites, 25-hydroxyvitamin D3 (25-OHD3), 24R, 25-dihydroxyvitamin D3 (24,25-(OH)2D3), and 1 alpha, 25-dihydroxyvitamin D3 (1,25-(OH)2D3), were determined serially in patients with gynecological malignancies treated by chemotherapy which included cisplatin. While 25-OHD3 and 24,25-(OH)2D3 levels did not change regularly, 1,25-(OH)2D3 decreased significantly (50%) after only one or two courses of the treatment in all cases. In concert with declining 1,25-(OH)2D3, the PTH levels increased two or three times. The results were explained by the impairment by cisplatin of mitochondrial function in proximal tubules in the kidney.
