Plasma prolactin level and plasma renin activity were determined in normal subjects and patients with low and normal renin essential hypertension, renal hypertension, renovascular hypertension, primary aldosteronism, Cushing syndrome, pheochromocytoma and malignant hypertension. In both normal subjects and the normal renin essential hypertensives, plasma prolactin was significantly higher in females than in males. Plasma prolactin was also significantly higher in the normal renin essential hypertensives than in normal subjects of both sexes, while no significant difference was found between the low renin group and normal subjects of either sex. A significantly positive correlation was observed between plasma renin activity and the plasma prolactin level in male essential hypertensives, but not in females. Although no significant difference in plasma prolactin level could be detected between patients with secondary hypertension and normal subjects, this level was significantly higher in malignant hypertensives than in normotensives. From these results, it was shown that significant differences of plasma prolactin levels exist between normal renin essential hypertensives, and low renin essential hypertensives or normal subjects, and that these differences may partly depend on renin status which might be related to the central dopaminergic activity. In malignant hypertensives, the high level of plasma prolactin may be caused by diminished renal function, but the suppression of central dopaminergic activity cannot be excluded in the mechanism of plasma prolactin increment.
Changes in plasma ADH levels were investigated in human male subjects whose blood volume was altered under isotonic circumstances. Blood volume was reduced by ambulation and increased by isotonic saline infusion in an overnight dehydrated state, and determinations were made on plasma ADH levels, plasma osmolality and hematocrit values. Plasma ADH levels were clearly affected by the small changes in blood volume, and significantly negative correlation was found between plasma ADH levels and the percent changes in blood volume under isotonic circumstances. From these findings, it was concluded that ADH release in human subjects is also controlled by the changes of the blood volume factor in addition to osmotic stimuli.

In a retrospective review of 32 consecutive patients (20 adults and 12 children) with acute appendicitis, we correlated abnormal urinalysis with the operative findings. Abnormal results on urinalysis were noted in 10 adults and 5 children. All urine specimens were collected by the clean-catch method. Abnormal findings were found more frequently in female patients. The majority of the patients with abnormal urinalysis had a ruptured or inflamed appendix in proximity to the urinary tract.
Endo.SceI which is isolated from cells of Saccharomyces cerevisiae is a eukaryotic site-specific endonuclease active on double-stranded DNA. At each cleavage site, Endo.SceI cuts only a defined phosphodiester bond in each strand of the double helix. We compared nucleotide sequences around five cleavage sites for Endo.SceI using a computer. We could not find any common specific sequence consisting of five base pairs or more among them. However, we found a 26-base pair consensus sequence which included 15 conserved nucleotides, allowing any of the five sequences to include a few nucleotides deviated from the consensus sequence. The consensus sequence is 5'-CAn*PYnnAnnCYYGTTnnnPnYnnYA-3', where P, Y, n, and * denote purine, pyrimidine, any nucleotide, and the center of the cleavage site, respectively. The numbers of sites at which the consensus sequence appears in pBR322 DNA, phi X174 replicative form DNA, fd replicative form DNA, or SV40 DNA are close to those of the cleavage sites for Endo.SceI. We found that a 33-base pair fragment was efficiently cut at the defined phosphodiester bonds by Endo.SceI. This 33-base pair fragment included 25 base pairs out of the 26-base pair consensus sequence. The fragments in which a part of the consensus sequence was missing were not cut by Endo.SceI. These observations suggest that the consensus sequence described above is the major characteristic around the cleavage sites recognized by Endo.SceI and that the mode of recognition of cleavage sites by Endo.SceI is different from that by restriction endonucleases. We found homology between the consensus sequence for Endo.SceI and the sequences around the cleavage sites for two other site-specific endonucleases of S. cerevisiae: Endo.SceII and YZ-Endo which is involved in mating type switching.
It has been found that the fluorescence intensity of 5-(iodoacetamido)fluorescein (5-IAF) attached to the SH1 of myosin subfragment 1 (S-1) increases 3-fold on formation of the rigor complex. On adding Mg2+-ADP, light scattering indicates no dissociation, but the fluorescence increment disappears. Thus, this fluorescence signal can distinguish the rigor state from other states, especially from ternary complexes such as actin-myosin-nucleotide. We demonstrate that by using this signal we can measure spectroscopically several kinetic parameters of acto-S-1-nucleotide interaction: In the presence of 20 mM KCl, 2 mM MgSO4, and 10 mM TES (pH 7.5) at 22 degrees C, Mg2+-ADP binds to acto-S-1(5)* (S-1(5) denotes 5-IAF-labeled S-1) with a Ka = 2 X 10(6) M-1, and Mg2+-PPi binds to acto-S-1(5)* with two apparent affinities, Ka = 8 X 10(4) M-1 and 1.4 X 10(3) M-1; the association rates of Mg2+-ADP and Mg2+-ATP for acto-S-1(5)* are 10(7) s-1 M-1 and 4 X 10(6) s-1 M-1, respectively, and the dissociation rate of Mg2+-ADP for acto-S-1(5)* is 5 s-1. In contrast to the fluorescence intensity of the dye, the lifetime and the absorbance are essentially unaffected by complex formation with F-actin or nucleotides. Therefore, we conclude that there must be a static quencher such as Trp, Tyr, or Met in the neighborhood of the attached dye and that the contact between dye and quencher is modulated by actin-induced or nucleotide-induced conformational changes in S-1.
Venous aneurysm not associated with other cardiovascular disease or trauma is a rare condition. A case is presented of bilateral jugular and mediastinal venous aneurysm, which was detected and depicted to a full extent by means of a blood-pool study with 99mTc -fibrinogen, undertaken because of concurrent thrombocytopenia. The aneurysm was confirmed by subsequent X-ray angiography and surgery. In radioisotopic blood pool study venous aneurysm, unlike arterial or aortic aneurysm, may be filled to the equilibrium state later than the appearance of the cardiac pool.
From phi 29-infected Bacillus subtilis cells, we have isolated a protein fraction which promotes in vitro replication of phi 29 DNA. This fraction catalyses both initiation and elongation, indicating that it contains the product of gene 3 (tp: terminal protein) and the product of gene 2 (gp2: probably a DNA polymerase), since initiation requires the two products (Blanco et al. 1983; Matsumoto et al. 1983). The fractions isolated from cells infected with temperature-sensitive (ts) mutants of gene 2 and gene 3 were thermolabile in both the initiation and elongation assays. When the pre-initiated material from the ts fractions of each mutant was heat-inactivated and mixed no complementation, restoring the elongation activity, was found. These results indicate: (i) tp and gp2 participate not only in the initiation but also in the elongation of phi 29 DNA replication, (ii) they probably function in tight physical association with each other.
