Teletransmission of physiological data.

The routine procedure of EEG recording is recalled to show the advantages of telemetric assessment of this parameter. The procedure for EEG telemetry is shown under different circumstances, e.g., by transmission with the aid of the 'Community Antenna System' of a hospital. Finally, a 16-channel telemetry system, using pulse width modulation is described.

Human lymphocyte cytotoxicity against mumps virus-infected target cells. Requirement for non-T cells.

Subpopulations of human lymphocytes were tested for their capacity to kill mumps virus-infected target cells in a 51-chromium release asaay. Using two different cell fractionation techniques, lymphocytes were fractionated into T cell-enriched (primarily T cells) and T cell-depleted (primarily B cells) subpopulations. Filtration of lymphocytes through columns coated with human immunoglobulin and rabbit anti-human-immunoglobulin (Ig-anti-Ig) rendered the resulting T-cell preparation inactive as effector cells against target cells carrying mumps virus antigens. In the second technique, lymphocytes were fractionated by centrifugation into two fractions according to their ability to form spontaneous rosettes with sheep erythrocytes (E). The E-rosette-forming population (primarily T cells) was shown to lack cytotoxic activity against mumps virus-infected target cells. This activity was present in the nonrosetting population. The results suggest that the effector cells involved in this cytotoxic system are of a non-T variety.

Urinary excretion of 5-S-cysteinyldopa in patients with primary melanoma or melanoma metastasis.

Urinary excretion of 5-S-cysteinyldopa was studied in 9 patients with primary melanoma. All had 5-S-cysteinyldopa excretion in the normal range. In 8 of the patients excretion values decreased after removal of the tumour. Twenty-four patients with clinical signs of melanoma metastasis were examined for 5-S-cysteinyldopa and dopa+dopamine in the urine. 16 of the 24 had pathologically increased excretion of 5-S-cysteinyldopa and 7 of the 24 had pathological excretion of dopa+dopamine. Six of the latter belonged to the group with increased 5-S-cysteinyldopa excretion and one patient had a borderline value of 5-S-cysteinyldopa. Determination of 5-S-cysteinyldopa seems to be of value in the follow-up of patients operated on for primary melanoma.

An in vitro method for study of human lymphocyte cytotoxicity against mumps-virus-infected target cells.

A chromium release assay was used to study lymphocyte-mediated cytotoxicity against mumps virus-infected target cells in vitro. Purified lmyphocytes from randomly selected donors killed significantly more virus-infected Vero cells than non-infected cells. Lymphocyte-target cell ratios of 50 to 100:1 and incubation period from 16 to 20 hr were optimal for determination of cytotoxicity. The lymphocyte induced chromium release was not obviously correlated with serum mumps hemagglutination-inhibition titers of the effector cell donors. However, the lymphocyte reaction against virus-infected target cells seems to have an immunologic basis. Thus, the more pronounced susceptibility of mumps-infected target cells as compared to non-infected cells was not due to a cytocidal effect of the virus, since spontaneous isotope release from both target cells was the same. Also, cord blood lymphocytes which had exhibited good cytotoxicity when induced either with phytohemagglutinin or with antiserum against target cell antigens were not cytotoxic for mumps-infected Vero cells. Moreover, the lymphocyte reaction against virus infected target cells could be inhibited by high concentrations of hyperimmune rabbit antimumps sera. On the other hand, lower concentrations of antisera specifically poteniated the lymphoycte-mediated isotope release from mumps-infected target cells.

Outbreak of giardiasis: effect of a new antiflagellate drug, tinidazole.

We describe here an intensive outbreak of mostly symptomatic (90%) Giardia lamblia infestation in a Swedish student group visiting the U.S.S.R. A new antiflagellate drug, ethyl (2-(2-methyl-5-nitro-1-imidazolyl)ethyl) sulphone, tinidazole, Pfizer, was given in a dosage of 150 mg twice daily for seven days to 10 healthy volunteers and to 24 students infested with G. lamblia. The drug was found to be effective in curing giardiasis and in eradicating G. lamblia from the intestinal tract. All the students with symptomatic giardia infestation became free from gastrointestinal disturbance, usually soon after treatment was started. None of the 24 students had G. lamblia in their stools after tinidazole treatment was discontinued or at follow-up. No side effects of the drug were seen and all of the subjects tolerated it very well.

Immunohistochemical and behaviour pharmacological analysis of rats inoculated intranasally with vesicular stomatitis virus.

A temperature-sensitive mutant of vesicular stomatitis virus was inoculated intranasally into infant Sprague-Dawley rats aged 9 to 17 days. Rats receiving the virus at 9 days of age had an extensive spread of infection throughout the brain and the animals died after a few days. Rats inoculated at day 11 postnatally survived and the infection was limited to the olfactory pathways, hypothalamus, diagonal bands and the anterior raphe nuclei. Stereological measurements showed that the volume of infected neurons constituted 67 +/- 10% of the total neuronal volume in the dorsal raphe nucleus. Double-labelling experiments revealed that both 5-hydroxytryptamine- and substance P-immunoreactive neurons contained the virus antigen. The motor stimulant effect of amphetamine was studied at 3 months post infection. The increase in amphetamine-induced frequency and duration of rearing was significantly attenuated in infected rats and the amphetamine-induced locomotion was slightly reduced.

Pax Vobiscum--a Swedish large PACS project.

The 'Pax Vobiscum' project aims to interlink the five radiology departments of the administrative region Orebro County into one virtual department. Altogether the 48 examination rooms, producing 185000 examinations per year, will convert to digital filmless technology and work as one functional unit although geographically spread out in five different cities within the county. Within the Pax Vobiscum project, a completely integrated application for radiological information systems (RIS) and PACS will be developed and also new image techniques such as direct digital detectors will be integrated in the functional department. Furthermore, the world outside radiology is included in the project while both image distribution and electronic RIS data communication (requests and reports) will be addressed. The paper describes the project regarding background, planning, organization and final realization.

Ca2+-dependent and Ca2+-independent phagocytosis in human neutrophils.

The phagocytic function of neutrophils is a crucial element in host defence against invading microorganisms. Two main specific receptor-mediated mechanisms operate in the phagocyte plasma membrane, one recognizing the C3b/bi fragment of complement and the other the Fc domain of immunoglobulin G (ref. 1). There is evidence that phagocytosis mediated by these receptors differs in the number and nature of the intracellular signals generated. However, the mechanisms by which receptor binding is transduced into a signal that generates the formation of the phagocyte pseudopod is not known, although extensive biochemical evidence has allowed the postulate that calcium ion gradients in the peripheral cytoplasm, by interacting with calcium-sensitive contractile proteins, initiate the process of engulfment. Using the high-affinity fluorescent calcium indicator quin2 both to measure and to buffer intracellular calcium ([Ca2+]i), we show here that in human neutrophils two mechanisms of phagocytosis coexist: a [Ca2+]i-dependent and modulated phagocytosis, triggered by activation of the Fc receptor, and a [Ca2+]i-independent mechanism triggered by the activation of the C3b/bl receptors.

In vivo testing of the protection provided by non-latex gloves against a 2-hydroxyethyl methacrylate-containing acetone-based dentin-bonding product.

In dentistry, allergic contact dermatitis to acrylates and allergic contact urticaria to latex are important occupational hazards. There is a need to identify non-latex gloves which are suitable for dental work but at the same time provide adequate protection against acrylate monomers. In a previous study, a new open-chamber system was used for testing the in vivo protection of 6 different gloves against an acrylate-containing ethanol-based dental adhesive. A nitrile glove gave the best protection among the gloves suitable for dental work. In the present study, the test model was used to investigate the in vivo protection of 7 non-latex gloves against a dental bonding product containing 2-hydroxyethyl methacrylate (2-HEMA) in an acetone/water vehicle. Eight 2-HEMA-allergic patients participated. Two neoprene gloves gave the best protection. The protection of the poorest glove was comparable to that of the positive control (no glove). The study produced in vivo data useful in the implementation of individual preventative measures against contact allergy to acrylates.

Influence of food on the bioavailability of an enteric-coated tablet formulation of omeprazole 20 mg under repeated dose conditions.

The objective of this study was to investigate the influence of food on the bioavailability of omeprazole (20 mg) given as an enteric-coated tablet under repeated dose conditions. This open randomized crossover study consisted of three seven-day treatment periods, each separated by a drug-free period. During each treatment period an enteric-coated tablet of omeprazole was taken once daily either under fasting conditions, or immediately before or after a standardized breakfast. On the last day of each treatment period, blood samples for the determination of plasma omeprazole concentrations were collected at baseline and at predetermined intervals over the 24 h period following drug administration. Fifty-seven male and female subjects, aged 18 to 52 years, completed the study according to the protocol. No statistically significant differences were found when comparing either the before breakfast or after breakfast treatment regimens with the fasting regimen for the estimated mean area under the plasma concentration-time curve (AUC). The maximum plasma concentration was not found to differ significantly among any of the treatment regimens. However, the lower limit of the CI for the comparison of fasting/before breakfast was not contained within the limits of bioequivalence. The time to reach maximum plasma concentration was significantly different when fasting and after breakfast regimens were compared. Thus, under repeated dose conditions, food has no influence on the bioavailability (expressed as AUC) of omeprazole given as the enteric-coated tablet formulation.

Combined therapy for undifferentiated giant and spindle cell carcinoma of the thyroid.

Undifferentiated giant and spindle cell carcinoma is an unusual and high-grade malignant type of cancer of the thyroid. The results of surgical and/or radiotherapeutic treatment have been discouraging. Only a few patients respond to therapy, usually for a short time. Recently, a few reports have been published on the effect of chemotherapy or a combined treatment of surgery, radiotherapy and chemotherapy. In the present paper the results of combining radiotherapy and chemotherapy, 5-FU and cyclophosphamide, are reported. For the future a more aggressive approach is discussed. The therapy should include surgical resection of the main bulk of the tumour, followed by radiotherapy and concomitant chemotherapy and thereafter prolonged adjuvant chemotherapy.
