Three lysozymes (DLs-1, -2, and -3) were purified from Peking-duck egg white by adsorption on CM-Sephadex C-25 resin, followed by CM-Sephadex C-25 and Sephadex G-50 column chromatographies. The three enzymes each moved as a single band, but showed different electrophoretic mobilities, on disc-polyacrylamide gel electrophoresis at pH 4.1. Final yields of DL-1, DL-2, DL-3 were 18.2%, 22.0%, and 6.0%, respectively, from the crude material adsorbed on CM-Sephadex resin. The enzymatic activities of DL-1, DL-2, and DL-3 were 1.53, 1.52, and 1.34 times that of hen egg white lysozyme, respectively, using Micrococcus lysodeikticus cell wall as a substrate at pH 6.2 and 37 degrees C. All the DLs lacked histidine and their amino acid compositions differed from each other by a few amino acid exchanges. The amino acid sequences of DL-2 and DL-3 differed from that of DL-1 by two displacements (Ser-37 to Gly and Gly-71 to Arg) and three displacements (Pro-79 to Arg in addition to the same substitutions), respectively. In comparison with Duck II and Duck III lysozymes from Kaki-duck (Hermann and Jollès (1970) Biochim. Biophys. Acta 200, 178-179; Hermann et al. (1971) Eur. J. Biochem. 24, 12-17) as regards amino acid sequences, Duck II is identical to DL-1 except for one displacement of Gln-57 in DL-1 to Glu in Duck II, while Duck III is rather different from our three lysozymes. All DLs gave single precipitin lines with any rabbit antisera against DLs and each precipitin line fused completely with that of any of the DLs in Ouchterlony double diffusion tests. However, the radiobinding inhibition assay showed that some anti-DL antisera clearly discriminated fine differences among the three DLs. The displacement at residue 79 (Pro in equilibrium Arg) gave the clearest immunological difference among the three kinds of displacements found in DLs.
Cerebral blood flow (CBF) measurements were combined with sleep polysomnography in nine patients with complex partial seizures. Two methods were used: the 133Xe method for measuring regional (rCBF) and the stable xenon CT method for local (LCBF). Compared to nonepileptic subjects, who show diffuse CBF decreases during stages I-II, non-REM sleep onset, patients with complex partial seizures show statistically significant increases in CBF which are maximal in regions where the EEG focus is localized and are predominantly seen in one temporal region but are also propagated to other cerebral areas. Both CBF methods gave comparable results, but greater statistical significance was achieved by stable xenon CT methodology. CBF increases are more diffuse than predicted by EEG paroxysmal activity recorded from scalp electrodes. An advantage of the 133Xe inhalation method was achievement of reliable data despite movement of the head. This was attributed to the use of a helmet which maintained the probes approximated to the scalp. Disadvantages were poor resolution (7 cm3) and two-dimensional information. The advantage of stable xenon CT method is excellent resolution (80 mm3) in three dimensions, but a disadvantage is that movement of the head in patients with seizure disorders may limit satisfactory measurements.
Programs have been developed, using a single compartmental analysis model, that provide rapid computer derivation for values of both local cerebral blood flows (LCBF) and local tissue: blood partition coefficients (L lambda) using inhaled stable xenon gas (Xes) as the indicator. These programs are planned for utilization of raw data points derived from serial computed tomography scans made between the 3rd and 9th min after 35% Xes inhalation, while concentrations of end-tidal Xes (PEXes) are concurrently monitored. Double integration and least squares fitting permitted estimation of corresponding arterial (assumed from PEXes) and tissue Xes concentrations at each scanning interval during Xes saturation. Derived estimates for L lambda and LCBF values for each region of interest mathematically approximate values saturated to infinity. The method described can be utilized for any freely diffusible indicator. Cross-sectional analysis of results obtained in 13 normal healthy volunteers between 20 and 80 years of age showed no age-related changes in L lambda values but progressive decreases in blood flow of cortex, basal ganglia, and white matter with advancing age. In senile dementia of Alzheimer's type (SDAT, n = 8) L lambda values were likewise found to be unchanged, but cortical and thalamic gray matter LCBF values were significantly reduced compared to age-matched normal volunteers.

The spatial relationship for T-B cell cooperation between antigenic epitopes and carrier epitopes on the antigen molecule was studied. Two mono-DNP substituted derivatives of hen egg-white lysozyme (HEL), DNP1-33HEL and DNP1-96HEL were used as antigens; the former is dinitrophenylated only at lysine-33 and the latter only at lysine-96. Fragment peptides of HEL were used to induce specific T cells to the respective sites of the antigen. Adoptive cell-transfer experiments of site-specific T cells and DNP-primed B cells directly showed that multiple distinct carrier epitopes for T cells could help the antibody responses to the single antigenic epitope for B cells and that a single carrier epitope could help antibody responses to multiple antigenic epitopes. T cells primed with a synthetic peptide SP34-54 (corresponding to sequence 34-54 of HEL) co-operated with DNP-primed B cells on challenge with DNP1-96HEL, but not with DNP1-33HEL.
For the purpose of verifying the effectiveness of peplomycin, one of the derivatives of bleomycin, against carcinoma of the esophagus and the safety of it, the analysis of the data for total 113 cases collected from 25 institutions in Japan was made. The results are as follows. It was effective in 19 out of 74 evaluable cases of carcinoma of the esophagus (25.7%). In case of treatment with peplomycin alone, it was effective in 6 out of 39 cases (15.4%). In case of the combination treatment with peplomycin and some other therapy, it was effective in 13 out of 35 cases (37.1%). As for the side effects, the incidence of fever was the highest in both the cases of peplomycin alone and the combination treatment such as 39.6% and 37.0%, respectively. Anorexia, nausea, vomiting, respiratory symptoms and tiredness were found in relatively many cases. In the clinical laboratory tests, the vital capacity after the treatment tended to be lower than that before the treatment, but there was little change in the hematological tests, pulmonary function test and renal function test.

A new hybrid cell line, designated NG115-401, produced a significant amount of immunoreactive (IR) substance P-like material. The production was measured by radioimmunoassay using specific antisera raised to substance P. The production rate was dependent upon cell growth, and maximum production occurred at the same stage (late logarithmic stage) as the time of appearance of choline acetyltransferase activity, one of the characteristic neuronal properties of the cells. On the other hand, no IR-substance P was found in hybrid cells other than NG115-401, i.e. NG115-301 and NG115-303 cells, which were derived from the same parent mouse neuroblastoma N115TG-2 cells. Furthermore, in an attempt to survey IR-substance P production in several clonal cells, consisting of some mouse neuroblastoma cells and a rat glioma cell, and several of their hybrid cells, we could find no cell line producing a significant amount of IR-substance P except for NG115-401 hybrid cells. The cellular production of IR-substance P in NG115-401 hybrid cells was also confirmed by detecting the material in all 14 subpopulations of this hybrid cells. Partial characterization of IR-substance P-like material produced by NG115-401 hybrid cells was carried out by its application to gel-filtration column chromatography. Only high molecular weight material (approximately 26,000) appeared in the column eluates.

A sex difference in alpha-bungarotoxin binding capacity in the mouse amygdala has been demonstrated by quantitative light microscopic autoradiography. The difference persisted even under widely different steroid-hormonal environment. In addition, it was observed that the binding capacities in both sexes were reversibly activated by administration of either testosterone or estradiol. Neonatal castration, on the other hand, permanently altered the toxin binding capacity in the adult male mouse. These data suggest the possibility that neonatal sex steroids irreversibly modify the cholinergic nicotinic mechanism in the developing mouse amygdala, while the hormones reversibly modulate the mechanism when applied in adulthood.

Delayed-type hypersensitivity (DTH) can be demonstrated in A/J mice immunized with hen egg-white lysozyme (HEL) in complete Freund's adjuvant (CFA) by challenging primed animals in the ear with aqueous HEL. Normal A/J mice receiving soluble HEL derivative peptide, P-Ib, sequence 29-54 and 109-123 linked by a single S-S bond, 7 days prior to immunization with HEL showed much lower DTH response specific to the protein. The inhibition of DTH reactivity is due to active suppression and involves the generation of suppressor T-cells (Ts). Thus, the suppression induced with a single i.v. injection of P-Ib solution was able to be transferred into syngeneic recipients by the spleen cells from mice pretreated with P-Ib. These suppressor cells are T-cells, since their ability to suppress DTH is completely abrogated by treatment wit anti-Thy 1.2 and complement. Amongst HEL derivative peptides tested in the present study, only P-Ib could induce the tolerance.
Two derivatives of hen egg-white lysozyme (lysozyme) with single substitutions of a 2,4-dinitrophenyl (DNP) residue were prepared. The reaction of lysozyme with a 10-fold molar excess of 2,4-dinitrobenzene sulfonic acid provided one mono-DNP substituted lysozyme (DNP1-33lysozyme), which was purified by ion-exchange chromatographies. The other one (DNP1-96lysozyme) was prepared as follows. After maleylation of lysozyme in the presence of a 7-fold molar excess of maleic anhydride, the derivative with one free amino group was purified on DE-52. This material was dinitrophenylated with 2,4-dinitrobenzene sulfonic acid and the mono-DNP substituted derivative was purified on DE-52. DNP1-96lysozyme was finally purified on SE-Sephadex C-25, after demaleylation at pH 3.5, at 37 degrees C, for 5 days. DNP1-33lysozyme and DNP1-96lysozyme each migrated as a single band with slower mobility than that of native lysozyme. On reduction, carboxymethylation and chymotrypsin digestion, both mono-DNP substituted lysozymes yielded a single yellow peptide. The amino acid compositions or partial sequence of these peptides indicated that lysine-33 and lysine-96 were the only dinitrophenylated residues in DNP1-33lysozyme and DNP1-96lysozyme, respectively. DNP1-33lysozyme and DNP1-96lysozyme showed antigenic reactivities equal to that of native lysozyme with antisera to lysozyme. The DNP residues on the protein were accessible to anti-DNP antibodies, but the affinities of DNP1-33lysozyme to anti-DNP antibodies were lower than those of DNP1-96lysozyme. This result is discussed with respect to the local environments of the DNP residues in these proteins.
