Angiocardiographic estimation of left and right ventricular volume characteristics in normal infants and children.

Left (LV) and right ventricular (RV) volume data were obtained from biplane cineangiocardiograms in 31 patients with Kawasaki disease who showed normal coronary arteries in selective coronary arteriograms. LV and RV volumes were calculated by using Simpson's rule method. Both of end-diastolic (EDV) and stroke volume (SV) were excellently expressed as a function of body surface area (BSA) with exponential relationship: LVEDV = 104.3 (BSA)1.61 ml (r = 0.93, p less than 0.001), RVEDV = 109.7 (BSA)1.68 ml (r = 0.90, p less than 0.001), LVSV = 68.1 (BSA)1.60 ml (r = 0.89, p less than 0.001) and RVSV = 66.8 (BSA)1.76 ml (r = 0.86, p less than 0.001). LV ejection fraction (LVEF) averaged 66 +/- 1% (mean +/- S.E.) and RVEF 58 +/- 1%. Both of them showed no significant correlations with BSA. The normal values of EDV and SV in infants and children can be predicted from their BSA by using the exponential equations demonstrated by the present study. Thus, these values in pediatric patients with heart diseases could be evaluated in terms of percentage of normal.

Intracellular distribution of fumarase in various animals.

The subcellular distribution of fumarase was investigated in the liver of various animals and in several tissues of the rat. In the rat liver, fumarase was predominantly located in the cytosolic and mitochondrial fractions, but not in the peroxisomal fraction. The amount of fumarase associated with the microsomes was less than 5% of the total enzyme activity. The investigation of the intracellular distribution of hepatic fumarase of the rat, mouse, rabbit, dog, chicken, snake, frog, and carp revealed that the amount of the enzyme located in the cytosol was comparable to that in the mitochondria of all these animals. The subcellular distribution of the enzyme in the kidney, brain, heart, and skeletal muscle of rat, and in hepatoma cells (AH-109A) was also investigated. Among these tissues, the brain was the only exception, having no fumarase activity in the cytosolic fraction, and the other tissues showed a bimodal distribution of fumarase in the cytosol and the mitochondria. The mitochondrial fumarase was predominantly located in the matrix. About 10% of the total fumarase was found in the outer and inner membrane, although it was unclear whether this fumarase was originally located in these fractions. No fumarase activity was detected in the intermembranous space.

Virological classification of autopsy livers with hepatic disorders.

Forty six selected autopsy livers with hepatic disorders were classified histologically into three groups, i.e., hepatitis 8, liver cirrhosis 16 and hepatocellular carcinoma 22, chiefly by histological findings. These groups were subdivided into two categories after determining the presence or absence of three HBV associated markers, i.e., the hepatitis B surface antigen (HBsAg), the antibody to hepatitis B surface antigen (anti-HBs) and the antibody to hepatitis B core antigen (anti-HBc) in the sera and liver homogenates. At least one of these markers was found to be present in the sera and homogenates of 38% of the livers with hepatitis, 69% of the livers with liver cirrhosis and 77% of the livers with hepatocellular carcinoma. When the liver tissues were examined for the presence of HBV DNA using Southern blot technique, the majority of them (10 out of 11) which proved to be positive for at least one HBV associated marker were also positive for HBV DNA. However, HBV DNA could not be detected in the other 10 livers which contained no HBV associated markers. These results showed that a HBV associated serological marker was always expressed, when liver tissue HBV DNA was demonstrable. The results of the two detection methods we used in this study were found to be almost equivalent. These results showed no evidence of nucleic acid homology between DNA from the liver of patients with non-B type liver disorders and HBV DNA.

[Biological properties of two newly established cell lines (SKG-3a,3b) from a human uterine cervical epidermoid carcinoma].

This is an initial descriptive report of two morphologically different cell lines (SKG-3a and SKG-3b) obtained from a human uterine cervical epidermoid carcinoma. These sublines were found to be derived from a moderately differentiated epidermoid carcinoma partially mixed with clear cell components. Both sublines had already been subcultivated more than 80 times since the initial separation. SKG-3a cells were much smaller in size and volume than 3b. The numbers of chromosomes were almost identical. Their cytology revealed anaplastic and pleomorphic features. SKG-3a was positive with periodic acid-Schiff stain and this changed to negative after an amylase digestive test, suggesting that the cells contained glycogen. An electron-microscopic examination confirmed the presence of many glycogen particles in 3a cytoplasm, but few in 3b. Tonofilaments and desmosomes were observed in both cytoplasms, suggesting epidermoid origin. In nude mice, SKG-3a produced clear cell epidermoid carcinoma with much glycogen, while 3b grew as a moderately differentiated epidermoid carcinoma with little glycogen. It is concluded that SKG-3a is derived from the clear cell epidermoid carcinoma and 3b is from the moderately differentiated epidermoid carcinoma. It is also clear that both were more heat sensitive at and over 39 degrees C than normal cells.

Effects of fibrinolytic treatment on rabbit Masugi nephritis.

We investigated the effects of administration of urokinase (UK) and trans-4-aminomethylcyclohexane carboxylic acid (t-AMCHA) on the development of experimental rabbit Masugi nephritis. Treatment with 1000 U/kg of UK, 5000 U/kg of UK and 100 mg/kg of t-AMCHA twice a day intravenously was initiated on the 7th and continued for 7 consecutive days. Concomitantly with the nephrotoxin injection, 5000 U/kg of UK was also given twice a day intravenously for 14 days. The administration of 5000 U/kg of UK twice a day for 7 days and 14 days suppressed the increases in BUN values and the decreases in urinary fibrinolytic activity. Fibrin-fibrinogen degradation products (FDP) were detected more frequently in the urine of rabbits given UK than in the control. Immunofluorescence microscopy showed a decrease in intra- and extra-glomerular fibrin deposition, and a decrease in fibrin or fibrinoid deposits in Bowman's space by 5000 U/kg of UK was demonstrated electron microscopically. Light microscopy revealed that the accumulation of fibrin in glomeruli, crescent formation, and progressive glomerular disorganization by 5000 U/kg of UK were prevented. On the other hand, treatment with t-AMCHA enhanced the increase in BUN values and there was a decrease in urinary fibrinolytic activity. Dense fibrin deposits in the glomeruli, were observed immunopathologically by treatment with t-AMCHA and histopathologic changes were found to be even more severe.

Calculation of right and left ventricular ejection fraction in infants and children by first pass radionuclide angiocardiography using self-synchronization method.

First pass radionuclide angiocardiography was utilized to calculate right and left ventricular ejection fraction in 74 infants and children. For the synchronization of radionuclide imaging with the cardiac cycle, the peaks and valleys of corrected ventricular time activity curve were adopted as the time reference points instead of R wave of electrocardiogram. Left ventricular ejection fractions obtained by the radionuclide technique correlated well with those derived from the contrast angiographic technique (r = 0.90), but right ventricular ejection fractions correlated less well (r = 0.74). This noninvasive technique appeared useful for evaluation of right and left ventricular ejection fraction.

Dual contrast echocardiography following peripheral venous injection.

Contrast echocardiography was carried out using a dual method in which two ways of ultrasonic beam were recorded on a cross-sectional plane of the heart. The advantage of this method was to make it possible to demonstrate the right and left cardiac cavities simultaneously and to catch the flow of contrast echoes from many-sided viewpoints, whereas single beam echocardiography could not make it. Dual contrast studies following peripheral venous injection were performed on patients with congenital heart diseases. These studies revealed accurate hemodynamics in 26 of 27 patients.

Evaluation of prostaglandin D2 (PGD2) as an anticoagulative agent for haemodialysis in comparison with prostaglandin E1 (PGE1).

Prostaglandin D2 (PGD2), a potent antiplatelet agent, was evaluated as an antithrombotic agent for haemodialysis in comparison with prostaglandin E1 (PGE1) and prostaglandin I2 (PGI2). When antiplatelet action was evaluated, taking the degree of associated hypotension into consideration, PGD2 was found to be superior to PGE1 and PGI2 as the latter two had the negative effect of inducing hypotension, while PGD2 had a less hypotensive effect. The suppression of the platelet function by PGD2 was observed to have only a slight influence on platelet function in systemic blood.
