[Mechanism of the action of baclofen on the contractile activity of the myocardium and blood vessels].

The cardiovascular effects of GABA-analogous-baclofen (10(-5) M) have been investigated in the experiments on isolated myocardium and vessels of the rats. The spontaneous contraction of vena porta segments and contraction of papillary muscle and arterial rings segments induced by electrostimulation were registered. Baclofen induced the growth of arterial tonus, decreased the amplitude of phase contraction as well as resulted in positive inotropic effects in myocardium and vena porta. The obsidan did not influence the baclofen effects, but alpha-adrenolytic dihydroergotoxine inhibited the inotropic effects of the drug. It is supposed that hypertensive baclofen effect which is observed during system injection may be caused not only by central, but also by peripheral regulation of calcium change in the myocytes. The latter (possible) kind of regulation may consist in the drug binding with adrenoreceptor and/or direct influence on the calcium transport across sarcolemma.

[Variability of Bacillus thuringiensis bacteriophages with C2-morphology].

Biological as well as physicochemical properties of Bacillus thuringiensis bacteriophages "17" and "7/13" having C2-morphology and isolated from factory phagolysates were studied. The bacteriophages are identical in the lytic spectrum++, morphology, size, GC-content, have the same buoyant density. The physical map for restriction endonucleases EcoRI, HindIII, SalGI and MvaI has been constructed of the bacteriophages DNA. Heteroduplex analysis has revealed the nonhomologous region of the deletion-insertion type as a 0.8 Md loop. The bacteriophages "17" and "7/13" are concluded to be closely related but not identical.

[Homologous and heterologous transcription of the Cry+-plasmid in Bacillus thuringiensis].

The possibility of homologous and heterologous transception of Cry+ plasmids in Bacillus thuringiensis is demonstrated. Cry+ plasmids from crystal bearing strain of Bacillus thuringiensis were transferred into acrystalline strain belonging to H5 serotype by mutual incubation. The donor strain was previously marked by the transmissive plasmid pAM beta 1 coding for erythromycin and lincomycin resistance. The transcipients having acquired the ability to synthesize delta-endotoxin were referred to H5 serotype due to their phenotype. By analogous method Cry+ plasmid was transferred from Bacillus thuringiensis to Bacillus cereus. Bacillus cereus strain GP7 was used as a recipient strain resistant to tetracycline. The presence of delta-endotoxin in transcipients was confirmed by bioprobes and immunoenzyme assay. To prove the transfer of Cry+ plasmid the plasmid profiles of the parent strains and transcipients have been analyzed. The formation of cellular contacts during mutual incubation of Bacillus thuringiensis and Bacillus cereus strains was demonstrated by electron microscopic study of ultrafine cuts.

[Internucleosome interaction: detection of dinucleosome fragmentation of chromatin by micrococcal nuclease. Analysis of the products of cleavage of chromatin from rat liver nuclei and L cells by micrococcal nuclease].

In murine L-cell nuclei micrococcal nuclease causes chromatin fragmentation with predominant liberation of dinucleosomes. Analysis of dynamics of rat liver nuclear chromatin cleavage by micrococcal nuclease revealed that the "dinucleosomal" mode of fragmentation is due to the pretreatment of nuclei with the non-ionic detergent Triton X-100 in the course of the isolation procedure. The set of particles detected in nuclease hydrolysates of nuclear chromatin pretreated with Triton X-100 and those isolated by the standard procedure was shown to be significantly different. In Triton X-100 treated nuclei the dichromatosome is the main hydrolysate component under various experimental conditions of nuclease hydrolysis and the sole component under "mild" conditions, whereas sucrose-treated nuclei contain three types of dinucleosomes. In Triton-treated nuclei prolongation of hydrolysis results in the liberation of the chromatosome which is absent in chromatin hydrolysates of sucrose-treated nuclei. Hydrolysis of Triton-treated nuclear chromatin by micrococcal nuclease is unaccompanied by the liberation (up to the stage of "deep" hydrolysis) of the core particle, the major component of the "sucrose" nuclear hydrolysate under the conditions used. The sharp differences in the accessibility of various types of dinucleosomes observed during pretreatment of nuclei with Triton X-100 are interpreted in terms of the localization of histone H1. The non-random type of the histone H1 molecule orientation along the nucleosome fibril is postulated.

[Internucleosome interactions: detecting the dinucleosome fragmentation of chromatin using micrococcal nuclease. Heterogeneity of nucleosomes and localization of histone H1].

The content of histone H1 (H1/H4 ratio) in dinucleosomes with the DNA of various length liberated from L-cell nuclear chromatin by micrococcal nuclease was analyzed. It was found that the histone H1 content in the dichromatosome is two times as low as that in the largest dinucleosome and in the complete mononucleosome. The set of chromatin fragments liberated from the Triton X-100 pretreated nuclei differs considerably from that of chromatin sites devoid of histone H1 (the de novo replicating chromatin and the chromatin formed on the undermethylated DNA). A scheme for asymmetric distribution of histone H1 with molecules oriented along the nucleosomal fibril, which reflects the peculiarities of chromatin fragmentation by micrococcal nuclease with predominant liberation of the dichromatosome, is proposed.

[Characteristics of Bacillus thuringiensis phages with circular permutation in the DNA molecule].

Bacteriophages Tm2 and Tg27 of different origins but identical in biological properties have been compared. Physicochemical characteristics of bacteriophages have revealed the existence of end repeats and circular permutation of phage DNA. Phages Tm2 and Tg27 share the same dimensions of incapsulated DNA, differing in the sizes of phage genome and end repeats. Bacteriophage Tm2 genome is 45.2 kb. long with the end repeats containing 5.7%. The genome of Tg27 is 42.53 kb and 11.8% of end repeat. The bacteriophages relation has been confirmed by heteroduplex and restriction analysis. Tm2 and Tg27 share 84% of homology. Two regions of nonhomology are found representing a single-stranded loop and equishouldered vesicle with the sizes 2.19 kb and 5.03 kb, respectively.

[Recovery after prolonged muscular work].

Increased protein, tyrosine and 3-methylhistidine content has been observed in the skeletal muscles of rats 2-24 h after a 10-hour swimming period. This was accompanied by a significant rise in 3-methylhistidine excretion during the second day of the recovery period. Such combination of alterations suggests simultaneous augmentation of both protein synthesis and decomposition in the muscles after active work. The start of the alterations coincides with post-exercise increase of blood corticosterone level (2-6 h after work) and with the achievement of glycogen supercompensation in the liver and muscles.

Pseudomonas bacteriophage phi KZ contains an inner body in its capsid.

Electron microscopical examination of the new virulent bacteriophage phi KZ, specific for Pseudomonas aeruginosa, has revealed an unusual structure in its capsid. In the center of the phage head is a cylinder of low electron density ("inner body"), surrounded by fibrous material which is packed around the inner body in a spoollike manner. The inner body itself has a springlike appearance. These structures disappear after adsorption of phage particles to bacteria. Various morphological forms, which can be interpreted as intermediate steps in phi KZ DNA condensation, have been seen in ultrathin sections of phi KZ-infected cells.

[Isolation of nonintegrated forms of proviral DNA of oncogenic viruses using preparative electrophoresis].

Nonintegrated forms of avian sarcoma virus (ASV) D6 proviral DNA have been analyzed in transformed chick embryo fibroblasts (CEF) 72 h after the initial infection. It is shown that nonintegrated proviral DNA in CEF cultures are represented by circular (form I) and linear (form III) forms. The molecular mass of the linear form of ASV D6 proviral DNA is 5.4-5.6 mD. The new method of preparative electrophoresis is proposed for isolation and separation of the nonintegrated forms of ASV D6 proviral DNA. The advantages of this method are considered as compared to the previously used one.

Effects of weightlessness on body composition in the rat.

Five male rats were exposed to 18.5 days of weightlessness in the Soviet mission COSMOS 1129 (flight group) and killed after reentry. They were immediately dissected into three major body subdivisions: musculoskeletal system, skin, and pooled viscera analyzed for fat, water, solids, and six elements. These results, expressed as percentages of the fat-free body or its components, were compared with two groups of terrestrial controls: one subjected to a flight simulation in a spacecraft mock-up and the other under standard vivarium conditions. Relative to the control groups the flight group showed 1) a reduced fraction of total body water, 2) a net shift of body water from skin to viscera, 3) a marked diminution in fraction of extracellular water in the fat-free body, 4) a marked reduction in fraction of bone mineral, 5) no change in the quantity of stored fat or adrenal masses, and 6) a net increase in total muscle mass as indicated by total body creatine, protein, and body cell mass.

[Electron microscope study of the intracellular development of the Pseudomonas aeruginosa bacteriophage phi KZ].

The study of the ultrathin sections of cells infected with virulent phage phi KZ has confirmed the presence of a specific cylindrical formation, an inner body, in the head of this phage and revealed the spiral structure of this inner body. The formation of DNA condensates whose structure resembles a spring wound around the core (the inner body) has been shown to occur in the cells in the process of the ultracellular development of phage phi KZ. This development leads to characteristic changes in the cellular structure, and in particular in the cell walls and the nucleoid.

Nucleomeric organization of chromatin.

In the nuclei fixed in situ, as well as in nuclei in low-ionic-strength solutions containing magnesium ions, chromatin is represented by globular nucleomeric fibrils 20-25 nm in diameter. Staphylococcal or endogenous nucleases cleave chromatin fibrils to nucleomers and multinucleomers. On removal of firmly bound magnesium, the nucleomers unfold into chains of four, six or eight nucleosomes. Mild staphylococcal nuclease digestion of nuclear chromatin releases mononucleomers, dinucleomers and trinucleomers that sediment in the sucrose density gradient in the presence of EDTA as 37-S, 47-S and 55-S particles, respectively. The mononucleomers in the sucrose density gradient with MgCl2 sediment as 45-S particles. The determination of the length of staphylococcal-nuclease-digested DNAs contained in the chromatin fragments showed that a nucleomer is composed of 8, and a dimer and trimer of 14-16 and 21-24 nucleosomes, respectively. When deprived of Mg2+ ions, the monomers lose their compactness (45 S) and become loose particles (37 S). This transition is completely reversible if nucleomers contain histone H1. Removal of this histone or dialysis of the nucleomer against EDTA at low ionic strength results in the complete unfolding of the nucleomer into a chain of nucleosomes. A structural model of a nucleomer fibril is suggested where the helicity of the nucleosome chain in a nucleomer (two turns of four nucleosomes each) is periodically discontinued. Such an organization of chromatin apparently provides additional hindrances for site-specific recognition of DNA in chromatin but permits local changes (within a single nucleomer) in chromatin when a hindrance is abolished.
