The N6-methyladenosine reader IGF2BP2 in T-cell lymphoma.

Peripheral T-cell lymphoma (PTCL) represents a highly heterogeneous and aggressive lymphoid neoplasm that lacks pathogenic biomarkers of RNA modification with therapeutic potential. IGF2BP2 is recognized as an N6-methyladenosine reader critically involved in oncogenesis. In this study, we observed consistently high expression of IGF2BP2 across common nodal PTCL subtypes in 3 independent external cohorts, which was further confirmed in our RNA-sequencing (RNA-seq) data set of 196 patients with newly diagnosed PTCL. Both in vitro and in vivo, IGF2BP2 promoted tumor cell growth and inhibited CD8+ T-cell infiltration within the tumor microenvironment. Mechanistically, IGF2BP2 bound to endosome-related genes (RAB4, VPS35, RAB9, and STAM) to maintain their stability, which resulted in enhanced endocytic activity and increased internalization of membrane proteins, and ultimately induced tumor cell proliferation and inhibition of CD8+ T-cell-mediated tumor cytotoxicity. The relationship between IGF2BP2 and endocytosis-associated genes was confirmed using RNA-seq data from patients with PTCL. IGF2BP2 as an upstream regulator of both tumor growth and immune suppression was further demonstrated in patient-derived xenograft models and a coculture system established using tumor samples from patients with PTCL and peripheral blood mononuclear cells. Notably, therapeutic targeting of IGF2BP2 with CWI1-2 suppressed endocytosis and impeded tumor growth in both cell lines and patient-derived xenograft models. Collectively, our findings highlight IGF2BP2 as a clinically relevant oncogenic driver in PTCL that integrates tumor-intrinsic growth signals with immune evasion through endocytosis-centered regulation, providing a novel therapeutic rationale for RNA modification-based strategies that concurrently target tumor cells and the tumor microenvironment.

Brain perivascular macrophages regulate endothelial cell function via a cMAF-dependent transcriptional program in mouse and human.

Brain perivascular macrophages maintain brain physiology, yet their transcriptional regulators and functions in health and disease remain unclear. Using single-cell multi-omics and functional experiments, we identify cellular musculoaponeurotic fibrosarcoma oncogene (cMAF) as a key transcription factor for brain perivascular macrophages, and conditional deletion of cMAF disrupts their phenotype in vivo. Functionally, cMAF drives insulin-like growth factor-1 (IGF1) expression in perivascular macrophages, enabling communication with endothelial cells. Consistently, cMAF deletion in perivascular macrophages causes transcriptional alterations in cerebral arteries, affecting vascular functions. Notably, cMAF emerges as the main transcription factor for human perivascular macrophages, suggesting conservation of this transcriptional module. During Alzheimer's disease (AD), human perivascular macrophages upregulate cMAF and IGF1 to enhance communication with vascular cells, and this response is abrogated in APOE4 carriers. Lastly, we explore an uncharacterized polymorphism in cMAF, providing evidence that the cMAF program is protective against AD. Targeting cMAF in perivascular macrophages may offer new therapeutic strategies for neurodegenerative and cerebrovascular diseases.

Association of human serum apolipoprotein A5 with lipid profiles affected by gender.

BACKGROUND: Apolipoprotein A5 (ApoA5) is present in human serum at a very low concentration. We developed a new method to determine ApoA5 concentration in human serum, and to investigate the correlation between serum ApoA5 and the lipid profiles in healthy subjects, and to analyze whether the correlation was affected by gender. METHODS: All the subjects (total 92, male 50, female 42) were healthy subjects without any medication. Lipids were measured enzymatically. An ELISA performed by a couple of monoclonal antibodies was used to measure serum ApoA5. RESULTS: The average ApoA5 concentration was 182.7+/-104.7 ng/ml ranging from 5.4 to 455.6 ng/ml. Serum ApoA5 concentration was negatively correlated with TG in female (r=-0.496, P=0.001). In all subjects, ApoA5 concentration was positively correlated to HDL-C (r=0.453, P<0.001). This correlation was more predominant in female (r=0.617, P<0.001) than in male (r=0.289, P=0.042). ApoA5 concentration was negatively correlated to body mass index (BMI) with more significance in female than in male (r=-0.345, P=0.001 for all; r=-0.456, P=0.002 for female; r=-0.198, P=0.167 for male). CONCLUSIONS: The serum concentration of ApoA5 was very low. The concentration of ApoA5 was negatively correlated with TG and BMI, but positively correlated with HDL-C. The correlations were affected by gender.

[Microbiochemical analyzer based on continuous spectrum and its test for clinic use].

A microbiochemical analyzer based on continuous spectrum and its test for clinic use are introduced. The principle of splitting behind was adopted to design the micro biochemical analyzer, which has the characteristics of little disorder light, real time collection of continuous spectrum signal (340-770 nm), small volume, light weight, small wastage of sample and reagent and rapidness and direct view of check. It has wide application in the emergency treatment, middling and small hospitals, rescue of battleground, and medicinal research. Representative items for clinic test were selected, such as uric acid, total cholesterin and albumin in our experiments. Comparative test and analysis of the daily check-up samples were made respectively by our microbiochemical analyzer and by the standard one of the hospital (Olympus AU2700, RT-1904C, Beckman LX-20). The test results show that our micro biochemical analyzer can meet the requirement of clinic use.

[Impact of chronic schistosomiasis japonica on the protective immunity induced by vaccine against hepatitis B virus].

OBJECTIVE: To study the impact of chronic schistosomiasis on the protective immunity induced by vaccine against hepatitis B virus. METHODS: 24 patients with chronic or advanced schistosomiasis (experimental group) and 26 healthy volunteers (control group) all without hepatitis B virus infection were selected for the study. Sera of the subjects in the two groups were collected before inoculation and on the 35th day after inoculation with yeast-derived recombinant hepatitis B vaccine. The level of anti-Hbs, IL-2 and TNF-alpha in sera was examined by ELISA respectively. RESULTS: Anti-Hbs in both groups were negative before inoculation, with an average absorbance (A value) of 0.134 and 0.150 respectively. After inoculation, positive rate of anti-Hbs was 17% (4/24, average A value 0.145 ) in experimental group and 92% (24/26, average A value was 1.210) in control group. The vaccine against hepatitis B induced significantly higher level of anti-Hbs in healthy volunteers compared with that in schistosomiasis patients (P < 0.01). The level of IL-2 and TNF-alpha increased in both groups after inoculation without significant difference compared with the level before inoculation. CONCLUSION: The results suggest that the protective immunity of patients with chronic schistosomiasis is deficient to the stimulation of hepatitis B virus and it may involve in a higher incidence of hepatitis B among schistosomiasis patients.

Variants of the KCNMB3 regulatory subunit of maxi BK channels affect channel inactivation.

The steady-state and kinetic properties of the KCNMB3 regulatory subunits associated with calcium-activated potassium channels (BK channels) are presented. BK channels containing four sequence variants (V1-V4) in the four different isoforms of the beta-subunit (beta3a, beta3b, beta3c, and beta3d) were expressed in Xenopus oocytes. Reconstituted BK channel inactivation ranged from none to around 90% inactivation. In particular, channels expressing the beta3b-V4 variant displayed a right shift in the potassium current voltage-dependence of activation and inactivated to about 30% of the maximum conductance, compared with wild-type beta3b channels that showed no inactivation. When the membrane potential was depolarized, BK channels inactivated with a very rapid time course (approximately 2-6 ms). This same variant was previously demonstrated to show subtly higher incidence in patients with idiopathic epilepsy (IE) compared with controls, especially when combined with variant V2 (combined heterozygotes). Furthermore, the gene maps to a region containing a susceptibility factor for this disorder. Taken together, these data suggest that neurons expressing BK channels composed of the beta3b-V4 variant subunit may experience reduced levels of inhibition and may therefore play permissive roles in high levels of neuronal activity that is characteristic of epilepsy.
