Development of capillary column packed with thiol-modified gold-coated polystyrene particles and its selectivity for aromatic compounds.

Three types of thiol compounds (n-octadecanethiol, thiophenol, and 2-phenylethanethiol) were used to modify the gold-coated polystyrene particles (dp. 5microm) to prepare a stationary phase for capillary liquid chromatography through the formation of self-assembled monolayer. The column with n-octadecanethiol-modified gold-coated polystyrene particles (C18-Au) demonstrated the higher affinity to phenanthrene and anthracene than small aromatics compared to the ODS column. In addition, the shape selectivity between phenanthrene and anthracene in the C18-Au column was much higher than that in the ODS column (separation factors: 1.82 and 1.14, respectively). The relationship between the retention factor and acetonitrile content in the mobile phase revealed that the retention behaviors in the C18-Au column was more sensitive on the acetonitrile content than those in the ODS column. Relatively higher affinity for phenanthrene and anthracene was commonly observed in all the three thiol-modified Au columns than that for the conventional ODS column, whereas separations of benzene and nitro- and chlorobenzenes were quite different among the three thiol-modified Au and ODS columns.

Retention behavior of neutral solutes in pressurized flow-driven capillary electrochromatography using an ODS column.

Several alkyl benzenes are separated by pressurized flow-driven capillary electrochromatography using a temperature-controlled capillary column packed with octadecyl siloxane-modified silica gel, and the effect of applied voltage on the retention is investigated. The van't Hoff plot shows good linearity at the column temperature between 305 and 330 K under applications from -6 to +6 kV. The applied voltage causes a relatively large variation in the enthalpy and the entropy of transfer of the solute from the mobile phase to the stationary phase (> 20%). However, the direction of variation in the enthalpy is almost opposite to that in the entropy, both of which might compensate each other. Therefore, the retention factor is not significantly varied (< 4%) by the application of voltage.

Instrumentation of high temperature and pressure reaction system with continuous flow and its hyphenation to a liquid chromatograph.

A continuous flow-type hydrothermal reaction system, in which both temperature and pressure were rapidly and dynamically controlled, was constructed. It is called a real time high temperature and pressure reaction system (RT-HighTP). The RT-HighTP was hyphenated with liquid chromatograph (RT-HighTP-LC) to achieve prompt analysis of the reaction products. The RT-HighTP system produced stable pressure under the supercritical and subcritical phases (%RSD < 3%). The RT-HighTP-LC was able to change the hydrothermal condition rapidly. The hydrothermal products under four conditions were analyzed within 70 min. Further, the hydrothermal products of lignin was easily taken out using the sample collecting loop and analyzed by an external instrument of GC/MS.

Effect of the net surface charge density of heptakis-6-bromo-6-deoxy-beta-cyclodextrin bonded silica gels on the retention behaviors of neutral cresol isomers in HPLC.

The effect of the surface charge density of heptakis-6-bromo-6-deoxy-beta-cyclodextrin (beta-CD-BR) bonded silica gels, which was used as the stationary phase of a packed capillary column for HPLC, was investigated concerning the retention behaviors of neutral cresol isomers. On the whole, the retention factors of the cresol isomers increased with an increase in the pH values of the mobile phase, although they were slightly smaller at pH 6.1 than at pH 4.7. An investigation on the retention variation using a van't Hoff plot revealed that the increase in the retention factor (k) at a higher pH region could be mainly attributed to the increase in DeltaS, while a partial decrease in k around pH 5 - 6 was caused by a decrease in the -DeltaH/T value. On the other hand, a measurement of the electroosmotic flow velocity under various pH of the mobile phase solutions revealed that the retention variations of the neutral cresol isomers were strongly correlated with the surface charge on the packing materials. The positive charge of secondary ammonium functional groups to bind beta-CD-BR inhibit the insertion of the cresol isomers into the cavity of beta-CD-BR while reducing the retention factor, whereas the negative charge of silanol group enhanced it through a local change in the mobile phase composition.

Development of dual gradient column in liquid chromatography.

The dual gradient column, in which both the chemical property of the stationary phase and the flow velocity in the mobile phase are heterogeneous longitudinally along the column, is developed to obtain the mobile phase gradient-like elution in an isocratic condition. Here, the step-wise dual gradient columns were prepared by connecting an inlet column (I.D. 50 microm, packed with ODS) serially to an outlet column (I.D. 100-200 microm, packed with the mixture of ODS and C1 [9:1]). The retention behavior of alkylbenzenes was able to be controlled in the dual gradient column depending on the variation in the flow velocity. Moreover, the change in retention behavior induced by the flow velocity variation for the dual gradient columns was quite different from that by the variation in organic modifier content of the mobile phase in isocratic elution for a single gradient column and can induce the similar effect with an ordinary gradient elution in a mobile phase composition.

Effect of the silanization conditions on chromatographic behavior of an open-tubular capillary column coated with a modified silica-gel thin layer.

The performance of an open-tubular capillary column coated with a modified silica-gel thin layer was investigated, particularly concerning the effect of the silanization process on it. Although the increase in the octadecyltriethoxysilane (ODTES) concentration in the silanization process could enhance the retention factor of naphthalene, its theoretical plate number was significantly reduced (ODTES, 5 to 50%; k, 0.2 to 4.3; N, 79600 to 2600 m(-1)). Namely, the increase in the retention factor was accompanied by a decrease in the theoretical plate number. A similar phenomenon was also observed when octadecyldimethylchlorosilane (ODCS) was used as the silanization regent. However, increases in both the retention factor and the theoretical plate number could be achieved (sample, naphthalene; k, 0.05 to 0.09; N, 149000 to 220000 m(-1)) by a NaOH treatment to the fabricated thin porous silica-gel layer before silanization with ODCS. The electrochromatographic separation of proteins and peptides by using the NaOH-treated column could obtain more peaks than electrophoretic separation.

Study of flow rate in pressurized gradient capillary electrochromatography using splitter and separation of peptides using an Amide stationary phase.

A pressurized gradient capillary electrochromatograph using a splitter was constructed. The variation in flow rate during gradient elution was investigated and separations of peptides using an Amide stationary phase were demonstrated. The flow rate, which is one of the important factors to control chromatographic behavior, was increased during the gradient elution, and the mismatching of mobile phase between the column and the resistance tubing derived three variation patterns in the flow rate. The electrophoretic migration in electrochromatography could enhance in separation of peptides. The separated peak number of tryptic digest of bovine serum albumin was increased from 30 to 40 by the application of +5 kV.

Effect of pressure on electrophoretic mobility of polystyrene latex particles.

A capillary electrophoresis system that can apply arbitrary helium gas pressures at both inlet and outlet reservoirs was constructed. The system was used to investigate the effect of pressure on electrophoretic behavior of polystyrene latex particles. The electrophoretic mobility of latex particles was increased with the application of pressure (< 3.0 kgf/cm2). The shrinkage of particle diameter under pressurization was observed using a microscope, however, the magnitude of shrinkage was not enough to explain the increase in electrophoretic mobility. Therefore, the application of pressure might increase the electric charge of the latex particle. Since methanol inhibited the enhancement in the electrophoretic mobility of the latex particles, water might play an important role in increasing mobility.

Preparation of a new type of fiber adsorbent attached with silica microparticles.

A new type of fiber adsorbent attached with silica microparticles was prepared. The silica microparticles were formed by the polymerization of silica oligomers on glass fibers, which were woven into a glass filter. The surface of the silica microparticles was chemically modified by bonding C18-ligands. SEM images indicated that the diameter of the uniform and spherical silica microparticles fixed on glass fibers was on the order of micrometers. It was confirmed that the glass filter adsorbent was effective for the adsorptive removal of toluene of low concentrations.

Alternating voltage capillary electrochromatography.

A phenomenon was found whereby the application of an alternating voltage to a capillary column can vary the capacity factor of a sample solute. The alternating voltage-induced variation in the capacity factor was studied using an anion exchange mini-bed (a short capillary column 12 mm long). The capacity factor varied according to both the amplitude and frequency of an applied alternating voltage. The variation greatly depended on the kinds of sample solutes and packing materials. A new separation mode for capillary electrochromatography using an alternating voltage, that is, alternating voltage capillary electrochromatography (AV-CEC), was proposed as an application of this phenomenon to control the retention of a sample solute. The chromatographic behavior of three organic acids (benzoic acid, phthalic acid, and salicylic acid) was studied in AV-CEC using an anion exchange column.

Behavior of neutral solutes in pressurized flow driven electrochromatography using a mixed stationary phase of ODS and anion-exchange.

The behavior of neutral sample solutes in pressurized flow driven electrochromatography using a mixed stationary phase, which consisted of ODS and anion-exchange (ODS-SAX), was studied. Applications of both positive and negative voltage on a column induced increases in retention factors of sample solutes. The direction of an electroosmotic flow under applications of positive and negative voltage were the same, therefore, the sign of the surface charge density under positive and negative voltage was opposite. We proposed a new equation for the relationship between applied voltage and surface charge density, and the practical electroosmotic flow conformed to this equation. Studying the electroosmotic flow using our proposed equation revealed that the applied negative voltage accelerates the protonation of the quaternary ammonium group and dissociation of the silanol group on packing materials. The retention behavior of a neutral solute was affected by the existence of the charged functional groups. We propose that this phenomenon is applicable to the control of the retention behavior of a sample solute using an electric field.

Estimation of electrophoretic mobilities of red blood cells in 1-G and microgravity using a miniature capillary electrophoresis unit.

Electrophoretic mobilities of red blood cells (RBCs) were measured in microgravity using a home-made capillary electrophoresis unit, which consisted of two small reservoirs of 0.6 mL and a fused-silica capillary tubing with 2 cm in length and 50 num in inner diameter. Migration of RBCs was observed by a microscope at 1000 times magnification and recorded on a videotape. The experiments were performed during stays in microgravity (about 0.01 G), which lasted 20 s and were attained by parabolic flights of an aircraft. On average, the electrophoretic mobilities of RBCs determined in microgravity were about 30% higher than those measured at 1-G condition irrespectively whether the cells were suspended in saline or serum during measurements. This difference might be explained as being mainly due to the cell floating in microgravity. Morphological changes of RBCs may contribute partly to the difference, while the variation in viscosity of the medium under microgravity could play only a minor role.

Alcohol ingestion stimulates mosquito attraction.

Mosquito bites should be avoided because of the risk of contracting parasitic and viral diseases such as malaria, dengue fever, and several encephalitides. Although humans have been said to suffer more mosquito bites after ingesting liquor, little is known about whether that is true. Thirteen volunteers (12 men from 20 to 58 years old and a 24-year-old woman) were chosen as test hosts and a 30-year-old man was established as a control. We measured ethanol content in sweat, sweat production, and skin temperature before and after ingestion of 350 ml of beer (ethanol concentration 5.5%) by volunteers and compared them with a control subject. Our study demonstrated that percent mosquito landing on volunteers significantly increased after beer ingestion compared with before ingestion, showing clearly that drinking alcohol stimulates mosquito attraction. However, ethanol content in sweat and skin temperature did not show any correlation between alcohol ingestion and mosquito landings. This study shows that persons drinking alcohol should be careful about their increased risk to mosquito bites and therefore exposure to mosquito-borne diseases.
