Effects of Allergina on the treatment of otitis media with effusions.

The number of pediatric patients with recurrent otitis media with effusions (ROMEs) is increasing because of the frequency of recurrence. The herbal combination Allergina is used for inflammatory-disease treatment in the Republic of Korea; in our study, the patients with ROME were treated with either Allergina (11 ears) or antibiotics (13 ears). We analyzed the levels of cytokines in middle-ear effusions (MEEs) and compared these levels in the Allergina-treated patients with those in the antibiotics-treated patients. The mean levels of interleukin (IL)-2 and IL-4 in MEEs were significantly higher in the Allergina-treated patients than in the antibiotics-treated patients, whereas levels of IL-1beta, IL-6, IL-8, and tumor necrosis factor (TNF)-alpha in MEEs were significantly lower in the Allergina-treated patients than in the antibiotics-treated patients. Clinical signs of ROME disappeared markedly after all the patients were given an oral administration of Allergina. Our experimental studies provide evidence that Allergina may be beneficial in the treatment of ROME by regulating cytokine production.

Determination of the site of origin of Pinellia ternata roots based on RAPD analysis and PCR-RFLP.

Analyses of randomly amplified polymorphic DNA (RAPD) and PCR-restriction fragment length polymorphism (PCR-RFLP) were performed in an effort to distinguish between two different origins of Pinellia ternata. To determine whether the origin of Pinellia ternata is in China or Korea. RAPD analysis was carried out using ten 20-mer random primers. Although the coefficients of similarity between the DNA of Chinese and Korean accessions of Pinellia ternata were high, distinguishable band patterns were observed in the reaction performed using primer numbers 3, 6 and 10. Primer 3 produced one (410 bp) and primer 6 produced four (410 bp, 350 bp, 300 bp, 250 bp) Chinese Pinellia-specific fragments. Primer 10 produced one (900 bp) Korean Pinellia rhizome-specific fragment. In addition, using PCR-RFLP analysis, different fingerprints were obtained from Korean and Chinese Pinellia ternata respectively. These results suggest that the analyses with RAPD and PCR-RFLP can be used to authenticate the relevant Chinese and Korean herbal medicines.

Inhibitory effect of Artemisia capillaris on ethanol-induced cytokines (TNF-alpha, IL-1alpha) secretion in Hep G2 cells.

A human hepatoma cell line, Hep G2 cell, is reliable for the study of alcohol-induced hepatotoxicity. In this study, we investigated the effect of an aqueous extract of Artemisia capillaris Thunb (Compositae) plant (AC) on ethanol (EtOH)-induced cytotoxicity in Hep G2 cells. AC (0.5-5 microg/mL) inhibited the secretion of EtOH-induced interluekin-1alpha (IL-1alpha) and tumor necrosis factor-alpha (TNF-alpha). AC also inhibited the EtOH-, IL-1alpha-, and TNF-alpha-induced cytotoxicity. Furthermore, we found that AC inhibited the EtOH-induced apoptosis of Hep G2 cells. These results suggest that AC may prevent the EtOH-induced cytotoxicity through inhibition of the apoptosis of Hep G2 cells.

Induction of nitric oxide synthase by Oldenlandia diffusa in mouse peritoneal macrophages.

Oldenlandia diffusa (OD) has been used to treat malignant tumors. In this study using mouse peritoneal macrophages, we have examined the mechanism by which OD regulates nitric oxide (NO) production. When OD (1 mg/ml) was used in combination with 10 U/ml of recombinant interferon-gamma (rIFN-gamma), there was a marked cooperative induction of NO production (36.13+/-7.12 microM) by the Griess method (nitrite). Treatment of macrophages with rIFN-gamma plus OD (1 mg/ml) caused a significant increase in tumor necrosis factor-alpha (TNF-alpha) production (4.49+/-1.43 ng/ml) by enzyme-linked immunosorbent assay. The increased production of NO and TNF-alpha from rIFN-gamma-plus OD-stimulated cells was almost completely inhibited by pretreatment with 100 microM of pyrrolidine dithiocarbamate (PDTC), an inhibitor of nuclear factor kappa B (NF-kappaB). PDTC also inhibited phosphorylation of IkappaB in rIFN-gamma-plus OD-stimulated cells. These findings demonstrate that OD increases the production of NO and TNF-alpha by rIFN-gamma-primed macrophages and suggest that NF-kappaB plays a critical role in mediating these effects of OD.

Ixeris dentata green sap inhibits both compound 48/80-induced aanaphylaxis-like response and IgE-mediated anaphylactic response in murine model.

We studied the inhibitory effect of the green sap of Ixeris dentata (IXD) on compound 48/80-induced anaphylaxis-like response in a murine model. IXD dose-dependently inhibited the anaphylaxis-like response induced by compound 48/80 in mice. IXD inhibited the anaphylaxis-like fatal response at the dose of 0.1 g/kg by 75%. IXD had a significant inhibitory effect on compound 48/80-induced ear swelling response at the doses of 0.05 and 0.1 g/kg. IXD (0.1 g/kg) also inhibited passive cutaneous anaphylaxis mediated by anti-dinitrophenyl IgE by 81.45%. When IXD was given as a pretreatment at concentrations ranging from 0.005 to 0.1 g/l, the histamine release from rat peritoneal mast cells induced by compound 48/80 was reduced in a dose-dependent manner. These results indicate that IXD may possess antianaphylactic activity.
