Carnosol, a component of rosemary (Rosmarinus officinalis L.) protects nigral dopaminergic neuronal cells.

Carnosol, a major component of Rosmarinus officinalis, is a phenolic diterpene that has potent antioxidant and anti-inflammatory activities. In this study, we investigated the protective effects of carnosol on rotenone-induced neurotoxicity in cultured dopaminergic cells. Results showed that cell viability was significantly improved with carnosol through downregulation of caspase-3. Furthermore, carnosol significantly increased the tyrosine hydroxylase, Nurr1, and extracellular signal-regulated kinase 1/2. These results suggest that carnosol may have potential as a possible compound for the development of new agents to treat Parkinson's disease.

Enhanced antioxidant enzymes are associated with reduced hydrogen peroxide in barley roots under saline stress.

Antioxidant enzymes are related to the resistance to various abiotic stresses including salinity. Barley is relatively tolerant to saline stress among crop plants, but little information is available on barley antioxidant enzymes under salinity stress. We investigated temporal and spatial responses of activities and isoform profiles of superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), non-specific peroxidase (POX), and glutathione reductase (GR) to saline stress in barley seedlings treated with 200 mM NaCl for 0, 1, 2, 5 days, respectively. In the control plant, hydrogen peroxide content was about 2-fold higher in the root than in the shoot. Under saline stress, hydrogen peroxide content was decreased drastically by 70% at 2 d after NaCl treatment (DAT) in the root. In the leaf, however, the content was remained unchanged by 2 DAT and increased about 14 % at 5 DAT. In general, the activities of antioxidant enzymes were increased in the root and shoot under saline stress. But the increase was more significant and consistent in the root. The activities of SOD, CAT, APX, POX, and GR were increased significantly in the root within 1 DAT, and various elevated levels were maintained by 5 DAT. Among the antioxidant enzymes, CAT activity was increased the most drastically. The significant increase in the activities of SOD, CAT, APX, POX, and GR in the NaCl-stressed barley root was highly correlated with the increased expression of the constitutive isoforms as well as the induced ones. Hepatitis B virus X protein sensitizes UV-induced apoptosis by transcriptional transactivation of Fas ligand gene expression.

Hepatitis B virus X protein (HBx) is a promiscuous transcriptional transactivator of many viral and cellular promoters. HBx plays an important role in hepatitis B virus pathogenesis related with liver diseases including hepatocellular carcinoma (HCC). HBx is also involved in the signal transduction and the apoptosis of HBV-infected cells. However, the exact mechanism of apoptosis by HBx is still controversial. To demonstrate the mechanism of apoptosis by HBx, we induced the apoptosis of HBx-expressing liver cells, HepG2-X, by UV irradiation. We found that HepG2-X was much more sensitive to the UV-induced apoptosis than normal liver cells by analyzing the DNA fragmentation and the cell viability. Very interestingly, when the Fas-associated death domain (FADD)-dominant negative mutant protein was present in HepG2-X, the sensitized apoptotic response of HepG2-X to UV was completely abolished suggesting that there is a close relationship between HBx and Fas pathway in apoptosis. Therefore we examined the transactivation of Fas receptor (Fas) promoter and Fas ligand (FasL) promoter by HBx. We found that HBx strongly transcriptionally transactivated FasL promoter, but not Fas promoter. In addition, it also turned out that the mRNA levels of FasL are higher than those of Fas in HepG2-X. Taken together, HBx sensitizes the apoptosis of UV-irradiated liver cells by transcriptional transactivation of FasL gene.

Dicer is essential for mouse development.

To address the biological function of RNA interference (RNAi)-related pathways in mammals, we disrupted the gene Dicer1 in mice. Loss of Dicer1 lead to lethality early in development, with Dicer1-null embryos depleted of stem cells. Coupled with our inability to generate viable Dicer1-null embryonic stem (ES) cells, this suggests a role for Dicer, and, by implication, the RNAi machinery, in maintaining the stem cell population during early mouse development.

Isolation of the enhanced neurovirulent HSV-1 strains from Korean patients.

Herpes simplex virus type 1 (HSV-1) is a neurotropic DNA virus which has latency in human. In this study, we isolated various HSV-1 strains, named KHS, from the skin lesions of Korean patients and characterized the specific features of each strain. We found that KHS strains produced small, cell associated and nonsyncycial plaques in Vero cells. We classified KHS strains into two substrains, KHS 1 which had highly condensed plaques and KHS 2 which had less condensed plaques. Since gD protein of HSV-1 plays important roles in viral plaque formation, we determined the nucleotide sequences of gD genes of KHS strains. According to deduced amino acid sequences of gD protein in KHS strains compared with prototype strains KOS and F, we found that gD of KHS strains have more putative O-glycosidic sites, serine in KHS 1 and threonine in KHS 2, respectively. To find out the establishment of viral latency, we infected each virus strain into eyes of mice and carried out trigerminal ganglia explanting experiment. We found that both KHS strains established latent infections stably just as did the prototype KOS and F strains. The eye swab experiments were carried out to check the viral replication in vivo. KHS 1 exhibited a longer shedding time in eyes of mice. We also found that KHS 1 has a higher neurotropic affinity by determining the time it took for the virus to reach the trigerminal ganglia from the eyes. Currently, we are studying the possible mechanism of high neuroinvasiveness of KHS 1 strain.

Synthesis of cationic polysaccharide derivatives and their hypocholesterolaemic capacity.

High-capacity bile acid-sequestering biopolymers with cationic polysaccharide derivatives have been developed. Cholestyramine, the bile acid sequestrant most widely used for treating hypercholesterolaemia, has unpleasant side effects because of the hydrophobic nature of its backbone. Methylan, a high-viscosity polysaccharide produced by Methylobacterium organophilum, was selected as a model polysaccharide for derivatization. Methylan was aminated to add dialkylaminoalkyl and free amino groups at hydroxyl sites in the methylan backbone, and these derivatives were quaternized to produce pH-independent cationic polyelectrolytes. The in vitro bile acid-binding capacity of quaternized DEAE-methylan was approximately 50% higher than that of cholestyramine. The bile acid-binding capacity of methylan derivatives increased with the number of carbons in the alkyl groups, indicating that hydrophobic interaction is a secondary factor for the sequestration of bile acids.
