[A study of the maximal mouth opening movement of normal subjects].

OBJECTIVE: The aim of this paper was to study condyle movement and the relation between it and incisor movement when the subjects performed maximal open-closed movement. METHODS: 38 normal students were tested by a six degrees of freedom mandibular movement recorder MT-1602. The traces and velocity curves of the condyle movement were analized on the horizontal plane. RESULTS: (1) The horizontal traces of condyle movements included straight line and smooth curve. (2) The velocity curve was like triangle. Acceleration was constant before and behind the peak respectively. (3) Unequal velocity of the bilateral condyles was common. (4) The appearances of traces correlated with the beginning relative horizontal velocity of bilateral condyles. The tracing vector was inclined to the rapid side. (5) The incisor pathway was affected by both the relative horizontal velocity and the corresponding horizontal trace of the bilateral condyles. CONCLUSION: The movement of condyle were adaptable to the stomatognathic system.

[Local expresion of proinflammatory cytokines in maxillofacial gunshot wound].

OBJECTIVE: To study the relationship between proinflammentory cytokines and wound healing in local area. METHODS: Interleukin(IL)-6, IL-8 and tumor necrosis factor-alpha (TNF-alpha) expression in local soft tissues after lower jaw gunshot wound were assessed by using immunohisto-chemistry, and pathological methods. RESULTS: High expressions of these cytokines were observed in local soft tissues of wounds after the gunshot. The cytokines expressions were different between the area near the bullet path and that far from the path. There was only one peak of the cytokines concentrations in the area far from the bullet path, while there were two peaks of the cytokines concentrations in the area near the bullet path, specially IL-6, IL-8. There were time differences for some different cytokines to reach the peaks of the their concentrations. In wound situ, the fibroblasts, mononuclear phagocytes and endothelial cells expressed IL-6, IL-8 and TNF-alpha after the firearm. CONCLUSION: These results suggest that proinflammatory cytokines play an important role in regulating inflammatory cytokine reactions of local injury tissues. They might improve the functions of fibroblasts, mononuclear phagocytes and endothelial cells and stimulate wound cells to produce and release other cytokines.

[Observation of preventing and treating effect of Salvia miltiorrhiza composita on patients with ischemic coronary heart disease undergoing non-heart surgery].

OBJECTIVE: To observe the preventing and treating effect of Salvia miltiorrhiza composita (SMC) on patients with ischemic coronary heart disease (CHD) undergoing non-heart surgery. METHODS: The 108 patients were randomly divided into the control group and treatment group. In the treatment group, each patient of 54 were administered SMC intravenously during the operation. ECG, HR, MAP and SpO2 were observed and bleeding quantity was measured perioperatively. RESULTS: Effective rate of ECG in treatment group was 90.7% (49/54 cases) and the control group was 35.2% (19/54 cases). There was significant difference between the two groups (P < 0.01). HR of the treatment group was no great flutter but control group became faster (P < 0.05). MAP and SpO2 of the two groups were no significant difference perioperatively. There was no significant difference to bleeding quantity between the two groups. CONCLUSIONS: SMC could effectively improve and protect myocardial ischemia in patients with CHD undergoing non-heart surgery, and no side effect was found.

[The detection of T-cell receptor gene rearrangement in myelodysplastic syndromes].

OBJECTIVE: To analyse the T cell receptor (TCR) gene rearrangement in myelodysplastic syndromes (MDS). METHODS: Polymerase chain reaction (PCR) was used to detect the TCRVgammaI-Jgamma gene rearrangement in 36 cases of MDS. RESULTS: 8 cases (22.2%) were found to have TCRVgammaI-Jgamma gene rearrangement. The TCR gene rearrangement positive rate was higher in RAEB, CMML and RAEBT subtypes than that in RA and RAS subtypes (P < 0.05). The average time MDS evolving to acute leukemia was shorter in the MDS patients with TCRVgammaI-Jgamma for gene rearrangement than that in patients without (P < 0.01). CONCLUSION: TCR gamma gene rearrangement can also be found in some MDS patients and the detection of TCRgamma gene rearrangement might be helpful to identify the prognosis and the development of MDS.

[Single-catheter approach to radiofrequency current ablation of accessory pathway in Wolff-Parkinson-White syndrome with recurrent rapid atrial fibrillation].

Twelve patients with WPW were treated by single-catheter ablation. Eight patients had the histories of recurrent rapid atrial fibrillation(Af) and refractory to antiarrhythmic agents. Ventricular rate varied between 250-300 bpm. Defibrillation was needed in three patients, and one had syncope. All patients were treated successfully by ablation of accessory pathway with single-catheter approach and no Af occurred during the procedures. This result suggests that the single-catheter ablation may be an effective and feasible treatment for these patients with WPW and may be helpful in avoiding Af in multiple-catheter introducing procedure.

[Study on combined clinical manifestation and myocardial perfusion imaging with 99mTc-MIBI SPECT for diagnosing myocarditis].

In 568 cases suffering from upper-respiratory tract infection with sore throat and more than one of following clinical manifestations: arrhythmia, heart failure and chest pain or oppressive sensation over the chest(Group A), and another 108 cases without above clinical manifestations(Group B), myocardial perfusion imaging with 99mTc-MIBI SPECT was studied. The results revealed that the imaging in 404 cases (71.12%) of Group A supported diagnosis of myocarditis, but only 6 cases(5.56%) did so in Group B. Because of lacking the clinical manifestations of myocarditis but positive finding of CVB-IgM antibody was detected in their sera, we considered that the positive SPECT imaging in these six cases of Group B belonged to myocardial reaction due to virus infection. It was possible that some of them were suffering from subclinical type of myocarditis.

[Immunohistochemical detection of hepatitis C virus antigen from hepatocellular carcinoma].

To investigate the infection of HCV and the expression of HCV antigen (HCAg) in HCC, immunohistochemical protocol was performed on tumorous liver tissues from 40 patients with HCC to detect HCAg and hepatitis B virus surface antigen (HBsAg) simultaneously. The results showed that the positive rates for HCAg and HBsAg were 17.5% (7/40) and 70% (28/40) respectively. HBsAg and HCAg were simultaneously detected in six of 40 cases and only one case had HCAg positive alone in the liver tumorous tissue. The positive signals were localized in diffuse cytoplasm of hepatocytes and the positively stained cells were mainly distributed in local pattern. The results suggested that HCV infection did exist in some cases of HCC, so HCV might be a viral risk factor in addition to HBV in the genesis of HCC in China.

[Experimental study on head and neck hemodynamics after ligation of jugular veins].

OBJECTIVE: To study alteration of neck hemodynamics after ligation of jugular veins during radical neck dissection. METHODS: 4 rhesus monkeys were selected to simulate the human bilateral radical neck dissection. After operation, the jugular veins were ligated or loosen and the blood stream quantities and blood pressure of jugular veins and arteries were measured simultaneously. RESULTS: The hemodynamic index would changed after jugular vein was ligated, the more the jugular veins were ligated, the greater effects the hemodynamics index of jugular veins were occurred, but there was little effect on arteries. CONCLUSION: The external jugular veins have the effect of compensating cerebral venous outflow after removing the internal jugular veins, and the external jugular veins should be conserved when bilateral or unilateral radical neck dissection was done.

[Enantiomeric resolution of trans-1,2-disubstituted cyclopropane by high performance liquid chromatography].

The enantiomeric resolution of racemic mixtures of thirteen trans-1,2-disubstituted cyclopropane was achieved with HPLC by using Chiralcel OD and Chiralcel OJ as chiral stationary phase and hexane/2-propanol mixtures with different concentrations as eluent. The chromatographic parameters of these racemates on Chiralcel OD and Chiralcel OJ columns were examined. The results showed that enantiomeric resolution ability of the racemic mixtures of trans-1,2-disubstituted cyclopropane on Chiralcel OD and Chiralcel OJ was strongly dependent on the site and the property of substituents on the benzene cycle. On the other hand, some racemated of trans-1,2-disubstituted cyclopropane with aliphatic group were also separated on the columns. The results showed that the chiral recognition process was the sum of all the possible chiral interaction (the dipole-dipole, hydrogen bond-hydrogen bond and pi-pi interactions) between the chiral stationary phase and trans-1,2-disubstituted cyclopropane. This method has been applied to identify the optical purity of trans-1,2-disubstituted cyclopropane.

[A study of time-dependent blood flow distribution in ischemic myocardium and its dependence on perfusion pressure with modeling and emulation].

A mathematical model was proposed for studying the time-dependent blood flow distribution in ischemic myocardium with different developments of collateral circulation. The model includes the blood perfusion of normal myocardium and ischemic myocardium. There are three classical myocardial vascular nets in each part and each net is represented by a branch of time-dependent nonlinear resistance and compliance. Experimentally measured aortic pressure and left ventricular pressure from six male dogs were taken as input data for emulation. The results demonstrate that raising perfusion pressure with external counterpulsation can improve the blood perfusion condition in ischemic myocardium.

[Induction of protective immunity in mice against Schistosoma japonicum by nucleic acid vaccine encoding the full-length paramyosin].

AIM: To investigate the immune efficacy of nucleic acid vaccination in mice against full-length paramyosin of Chinese Schistosoma japonicum. METHODS: C57BL/6 and BALB/c mice were vaccinated intramuscularly with the nucleic acid vaccine (pCMV-SjC97) encoding the full-length gene of paramyosin of Chinese S. japonicum. Each group was immunized three times at weeks 0, 3 and 6. Mice vaccinated with pCMV blank vector served as negative control. Mice were challenged three weeks after final DNA boosting by percutaneous infection with cercariae. Six weeks after infection the mice were perfused, worm burden and eggs in the livers, spleens and intestines were counted. Sera from vaccinated mice were collected from the tail vein at weeks 0, 3, 6 and 9, respectively. RESULTS: C57BL/6 mice vaccinated with pCMV-SjC97 produced predominantly IgG2a and IgG2b; whereas in BALB/c mice, IgG1, IgG2a and IgG2b antibodies. Immunization with the pCMV-SjC97 in C57BL/6 mice could confer significant worm reduction rate (35.5%-41.4%, P < 0.05) and egg reduction rate (liver: 44.5%-59.6%, P < 0.05; spleen: 56.7%-82.4%, P < 0.05; intestines: 57.9%, P < 0.05), but not in BALB/c mice. CONCLUSION: The nucleic acid vaccine, pCMV-SjC97, could induce protective immunity in C57BL/6 mice significantly.

[Cloning, sequencing and expression of the full-length gene encoding paramyosin of Schistosoma japonicum in vivo].

AIM: To clone and sequence the gene encoding paramyosin of S. japonicum (Chinese strain) and to study the expression of the DNA-based vaccine encoding the full-length paramyosin of S. japonicum in vivo. METHODS: Total RNA was isolated from adult S. japonicum using TRIzol reagent. The full-length cDNA encoding paramyosin of S. japonicum was amplified by RT-PCR and cloned into pGEM-T vector and sequenced by the method of dideoxy-mediated chain-termination. The cDNA encoding paramyosin of S. japonicum was subcloned into the expressive plasmid vector pCDNA/AMP(pCMV-Sjc97), and the recombinants were identified by restriction enzyme digestion and sequencing. The immunofluorescence assay was used to study the expression of Sjc97 in vivo in mice. RESULTS AND CONCLUSION: The 2.6 kb cDNA encoding the full-length paramyosin of Chinese S. japonicum has been successfully cloned and sequenced for the first time. The full-length sequence of paramyosin of S. japonicum was determined. Comparison of the nucleotide sequence and the deduced amino acid sequence of Sjc97 with that of S. japonicum paramyosin (Philippine strain) (Sjp97), S. japonicum paramyosin(Japanese strain) (Sjj97), S. mansoni (Sm97), B6 and Y6 clone (the partial cDNA encoding paramyosin of Chinese strain) showed that Sjc97 differed from Sjp97 by 16/2,601 nucleotide and 3/866 amino acid substitutions (99.4% on nt-level and 99.7% on aa-level in homology); from Sjj97 by 20/2,601 nucleotide and 2/866 amino acid substitutions (99.2% on nt-level and 99.8% on aa-level in homology); and from B6 by 11/1,329 nucleotide and 1/443 amino acid substitutions (99.0% on nt-level and 99.8% on aa-level in homology); from Y6 by 13/1,329 nucleotide and 1/443 amino acid substitutions (98.9% on nt-level and 99.8% on aa-level in homology); Sjc97 differed from the Sm97 by 2,235/2,601 nucleotide and 34/866 amino acid (91.0% on nt-level and 96.0% on aa-level in homology). The plasmid expression vector encoding the full-length paramyosin of Chinese S. japonicum has been successfully constructed. The pCMV-Sjc97 vaccine could express Sjc97 protein in vivo in mice after intramuscular immunization.

[Studies on the features of protective immune response induced by recombinant Sjc26GST of Schistosoma japonicum].

AIM: To compare the differences in the protective immune response induced by reSjc26GST between C57BL/6 and BALB/c mice. METHODS: Mice were immunized subcutaneously with reSjc26GST emulsified with Freund's adjuvant. The specific antibody isotypes elicited by reSjc26GST, in vitro lymphocyte proliferation and cytokine responses to reSjc26GST were examined. RESULTS: The level of anti-reSjc26GST IgG was higher in immunized C57BL/6 mice than in BALB/c mice, and examination of the IgG subclasses demonstrated that reSjc26GST-immunized C57BL/6 mice resulted in predominantly IgG1 and IgG2a/IgG2b antibody responses, whereas in BALB/c mice, IgG1 antibody. Cytokine production assays revealed that the quantity of IFN-gamma and IL-2 and IL-5 released from the reSjc26GST-primed splenocytes were significantly higher in C57BL/6 mice than in BALB/c mice. These results showed that the immunization with reSjc26GST in C57BL/6 mice elicited a mixed immune reaction of Th1 and Th2 cell responses as demonstrated by the elevated levels of IgG1, IgG2a/IgG2b and IL-2, IFN-gamma, IL-5, whereas in BALB/c mice, reSjc26GST immunization elicited Th2 response as demonstrated by the elevated levels of IgG1, IL-2 and IFN-gamma. CONCLUSION: Immunization with reSjc26GST could elicit higher humoral and cellular immune responses in C57BL/6 mice than in BALB/c mice.

[Clinical significance and changes of fibrillation potential amplitude following denervation of laryngeal muscles].

OBJECTIVE: To evaluate the value of clinical application of examination of fibrillation potential amplitude of laryngeal electromyogram. METHOD: Fifty-four patients (65 sides) with recurrent laryngeal nerve paralysis were examined and only the maximum fibrillation potential amplitudes were recorded in the posterior cricoarytenoid muscles and thyroarytenoid muscles. RESULT: There was no significant difference between sexes, ages and sides. The fibrillation potential amplitude was maximum at 3 to 4 months after denervation of the posterior cricoarytenoid muscle or at 5 to 6 months after denervation of the thyroarytenoid muscle and still remained at certain level for years in some patients. Significant difference was showed in complete and incomplete nerve injuries groups, but changes of amplitude with time in incomplete nerve injuries group were small. CONCLUSION: The changes of fibrillation potential amplitude might indicate that the changes from muscle denervation was still reversible and might be more accurate than traditional method of examination.
