[The expression of Fas antigen on lymphocytes in the patients with asthma].

OBJECTIVE: To study the expression of Fas antigen on lymphocytes in patients with asthma. METHOD: The peripheral blood lymphocytes (PBL) isolated from patients with asthma were cultured with dexamethasone 10(-5) mol/L, Fas antigen on lymphocytes was detected by flow cytometry at 0, 2nd, 8th, 24th hour respectively. RESULT: The native expression of Fas antigen of PBL isolated from the patients with asthma (5.3% +/- 2.0%, P < 0.05) was lower than that from healthy controls (8.5% +/- 1.8%). Increased expression of Fas antigen in the patients with asthma was observed with induction of dexamethasone (P < 0.01). The percentages of Fas positive cell were 5.3% +/- 2.0%, 33% +/- 5%, 63% +/- 11%, 76% +/- 7% at 0, 2nd, 8th, 24th hour respectively in asthmatic group. While 8.5% +/- 1.8%, 31% +/- 11%, 49% +/- 10%, 20% +/- 4% respectively in normal control group. CONCLUSION: Decreased native expression of Fas on lymphocytes may be related to airway inflammation in the patients with asthma. Increased expression of Fas induced by Dex may be one of the mechanism of lymphocytes apoptosis.

[The growth inhibition of anti-EGF receptor monoclonal antibody to human lung adenocarcinoma cells].

OBJECTIVE: Epidermal growth factor (EGF) and its receptor play a critical role in the growth and regulation of many type of malignant cell. The high level of expression of the EGFR on normal cell lung cancer(NSCLC) and the important role of EGFR in signal transduction make it potentially an excellent target for antibody directed therapy. The purpose of this study was to evaluate the anti-tumor effects of anti-EGFR monoclonal antibody on lung carcinoma cells. METHOD: Preparation of hybrid cell producing anti-EGFR monoclonal antibody using EGF receptors partially purified from A431 cells as immunogen. The effect of anti-EGFR monoclonal antibody to human lung cells was investigated in vitro. RESULT: The anti-EGFR monoclonal antibody was shown to inhibit the proliferation of human lung adenocarcinoma cells in vitro, but no growth-inhibitory effect was observed in the human small cell lung cancer cells. The concentration, 15 ug/ml, of anti-EGFR antibody could inhibit growth rate by 94% and 88% for human lung adenocarcinoma cell lines SPC A-1 and A549, respectively. CONCLUSION: Our initial results indicate that the anti-EGF receptor monoclonal antibody may be potentially useful for the biotherapy of lung adenocarcinoma.

[A reversed-phase high performance liquid chromatographic method for the determination of pinoresinol diglucopyranoside in Eucommia Ulmoides Oliv].

A reversed-phase high performance liquid chromatographic method for the determination of (+)-pinoresinol-di-O-beta-D-glucopyranoside in the bark of Eucommia Ulmoides was established. The air-dried bark of E. Ulmoides was ground into powder and extracted continuously with methanol. The extract was evaporated under reduced pressure, then suspended in water. This suspension being subjected to column chromatography was eluted successively with water, 30%, 50% and 90% aqueous solution of ethanol. The 30% EtOH eluate was concentrated and then injected into HPLC system. The conditions of HPLC were: YWG-C18 column (250 mm x 4.6 mm i.d. 10 microns); 28%(V/V) methanol mobile phase and UV detector at 232 nm. The calibration curve was linear in the range of 0.068 microgram-0.68 microgram of(+)-pinoresinol-di-O-beta-D-glucopyranoside with correlation coefficient of 0.9999. The coefficient of variation and average recovery were 0.50%-0.74% (n = 5) and 99.22% (n = 3) respectively.

[Study on the analysis of zinc 5-aminosalicylate and related compounds by high performance liquid chromatography].

In this paper, the separation and determination conditions of zinc 5-aminosalicylate (ZASA) and related compounds by high performance liquid chromatography(HPLC) have been studied systematically. The baseline separation was achieved at 20 degrees C on a CLC-ODS column(150 mm x 6.0 mm i.d., 10 microns) within 10 minutes by using 1% (V/V) HAc-MeOH(4:6) as mobile phase at a flow-rate of 1 mL/min. The RSDs of rention time within day and between days were less than 2% and 5% respectively. The injection volume was 20 microL, and relationships between the peak area with 280 nm detection and the analyte concentration were linear within certain ranges. This method was used for determination of ZASA, impurities and thermal decomposition compounds. The results showed that the impurities in the crude ZASA were 5-aminosalicylic acid and salicylic acid. The thermal decomposition compounds in acid medium were 5-aminosalicylic acid, 5-aminophenol and 5-benzeneazosalicylic acid.

[Separation and determination of carotenoids in Fructus lycii by isocratic non-aqueous reversed-phase liquid chromatography].

High performance liquid chromatography using a non-aqueous reversed phase with isocratic elution of C18 columns is a powerful tool for investigating the carotenoid composition of Fructus Lycii. This paper compared the effect of different eluents on the separation of carotenoids. An optimum solvent system consisting of acetonitrile and methylene chloride is suitable for simultaneous separation of compounds from polar xanthophylls to nonpolar carotenoid esters. Ten carotenoids were separated from Fructus Lycii by using Shim-pack CLC-ODS column(150 mm x 6 mm i.d.) and mobile phase of acetonitrile/methylene chloride(60:42, V/V) with a flow rate of 1 mL/min. The eluates were monitored with UV/VIS detector at 456 nm. The method which permit direct analysis of carotenoid extracts without saponification in 30 min provides a true information of carotenoids composition in Fructus Lycii as they exist. Fructus Lycii contains 2952 micrograms/g of total carotenoids, but 98.6% of carotenoids are existed as esterified forms, zeaxanthin dipalmitate are accounted for 77.5% of the total carotenoide.

[Effect on life span and membrane protein in red blood cells by integrated medicine therapy on chronic aplastic anemia].

OBJECTIVE: To explore the mechanism of integrated traditional Chinese and Western medicine (TCM-WM) therapy on chronic aplastic anemia (CAA). METHODS: The RBC life span of 30 normal human subjects and 30 patients with CAA were measured by 51Cr labelled technique before and after TCM-WM therapy. The morphology and distribution of RBC membrane protein granules were observed by freeze fracture etching and transmission electron microscope. RESULTS: The half life of erythrocytes (RBC T1/2) was shortened in CAA cases and there was a significant difference compared to healthy control (P < 0.01). After therapy, the RBC life span prolonged and approached the normal level. Before treatment, there existed abnormal in morphology, decrease in amount and uneven in distribution of protein granules in protoplasmic face (PF) and extracellular face (EF) of RBC membrane. After treatment, the protein granules of RBC membrane was improved and approached to control. CONCLUSIONS: The morphology, amount, quality and distribution of RBC membrane protein granule were closely related to its life span. The therapeutic effect of TCM-WM was better than that of WM alone and it had a function both in stabilizing membrane protein and extending the RBC life span.

[Psammomatoid ossifying fibroma].

OBJECTIVE: To clarify the histopathological, clinical behavior biological characterics and the relationship among the psammomatoid ossifying fibroma (POF), ossifying fibroma and fibrous dysplasia of bone. METHODS: Thirty-one cases of POF in the ear and sinonasal region were studied retrospectively. The sections, clinicopathological records from June, 1956 to November, 1996 were reviewed. The reticulum fiber and Masson's stain and alkaline phosphatase immunostain were done in 10 cases, Vimentin, Desmin, EMA and smooth actin immunohistochemical stain were done in 5 cases. One case was studied by transmission electron microscope. RESULTS: The specimens were broken tissue with the sand-like felling, eleven specimens with intact thin shell consisted of bone tissue were used to study the changes in the area between tumor and normal bone. Histopathologically they were rich in mesenchymal tumor cells and a lot of calcifying psammomatoid-like osteoid and a few of calcified bodies similar to those described by Shafer, and the bony trabecula was not found in the center of the tumor. In addition, three zones were noted at the junction of normal bone and tumor, namely, the normal bone-reactive proliferative bone-tumor. In some cases, the reactive bony proliferation may be not marked. The normal bone adjacent to the tumor may be absorbed and disappeared, and were then replaced by the tumor, and finally led to tumor tissue invasion into the soft tissue, but the infiltration appearance is not present. CONCLUSION: POF is the slowly growing benign tumor and is different from ossifying fibroma and fibrous dysplasia of bone.

[Pathologic changes of palatopharyngeal soft tissue in obstructive sleep apnea syndrome].

OBJECTIVE: To study the pathologic changes of palatopharyngeal soft tissues in patients with OSAS. METHODS: Different analyses have been done on uvular muscles (UM) and/or palatopharyngeal muscles (PPM) in patients with OSAS(n = 27), SS(n = 17) and controls (n = 15). The transverse sections were sequentially stained for HE, myofibrillar ATPase, NADH-TR, Lillie Oil Red-O. Ten specimens were evaluated by transmission electron microscopy(TEM). RESULTS: Patients with OSAS have a significantly smaller percentage of muscle in the uvula than that of SS patients (15.0 +/- 3.0 vs 20.3 +/- 5.5), The percentage of muscle is inversely related to the percentage of fibrous tissue in OSAS group (r = -0.75, Y = 42.6 - 0.64X); The total number and area of muscle fibers, the number and area of type I muscle fibers are significantly lower in OSAS than those in SS (800 +/- 345 vs 1200 +/- 360n; 1.10 +/- 0.56 vs 1.70 +/- 0.60 micron 2; 156 +/- 95 vs 360 +/- 225 n; 920 +/- 350 vs 1365 +/- 540 micron 2 respectively); PPM: The percentage of type I muscle fibers within total muscles and its area are significantly lower in OSAS than those in SS and normal controls(20.5 +/- 4.0 vs 34.5 +/- 4.5 n; 20.5 +/- 4.0 vs 35.5 +/- 4.7; 860 +/- 360 vs 1260 +/- 340 micron 2; 860 +/- 360 vs 1350 +/- 370 micron 2; respectively); the muscles from OSAS patients seemed to show neurogenic alterations. Structurally altered fibers shows myofibrillar loss, increased number of vesicles, and rough Z line and so on. CONCLUSION: Our finding suggest that the neurogenic functional disturbance of pharyngeal dilator muscles may play an important role in causing the airway collapse in OSAS.

[The effect of detrusor instability secondary to benign prostatic hypertrophy on the density of acetylcholinesterase-containing nerves].

OBJECTIVE: To study the effect of detrusor instability secondary to benign prostatic hypertrophy (BPH) on the density of cholinesterase-containing nerve in detrusor samples. METHOD: Our present study included 3 groups confirmed by urodynamic evaluation. They were control group (8 cases), obstructive detrusor stability group (7), and obstructive instability group (12). The specimens were obtained from the dome of bladder. The AchE-containing nerves were demonstrated by the method of Karnovsky-Roots staining and AchE silver staining. The density of AchE containing detrusor nerves in specimens was examined with sterologic techniques. RESULT: The density of AchE-containing nerves in the obstructive detrusor instability group and the stability group was significantly decreased as compared with the control group (P < 0.01). The density of AchE positive nerves were also significantly decreased in the obstructive instability group as compared with the obstructive stability group (P < 0.05). CONCLUSION: Detrusor instability secondary to BPH obstruction is related to the decrease of cholinergic innervation density in the detrusor muscles.

[Studies of mechanism of primary angle closure glaucoma using ultrasound biomicroscope].

OBJECTIVE: To investigate the mechanism of primary angle closure glaucoma (PACG) especially the role of the changes of ciliary body using ultrasound biomicroscope (UBM) combined with general A scan. METHOD: 99 eyes with PACG, including 50 eyes with acute and 49 eyes with chronic PACG, were evaluated and 58 normal eyes were in the control group. RESULTS: Compared with the normal eyes the PACG eyes had their anatomic features. They had a smaller corneal diameter, shorter axial length, shallower anterior chamber, thicker lens, more relative anterior location of lens, swelling of ciliary process and anterior rotation of ciliary body. All these differences were significant statistically (P < 0.05). Compared with the chronic PACG eyes the acute PACG eyes had a shallower anterior chamber and a more anteriorly situated lens position. The differences were significant statistically (P < 0.01). Compared with the normal eyes the acute PACG eyes had a shorter ciliary body - lens distance and the difference was significant (P < 0.05); the chronic PACG eyes also had a shorter ciliary body-lens distance, but the difference was not significant (P > 0.05). CONCLUSIONS: The PACG eyes have their anatomic features. Among these factors the pupillary block caused by the anterior displacement of lens and the narrow angle induced consequently are the common mechanism. The swelling ciliary process, the positional changes of the ciliary body and age-related thicker lens are possibly the initial causes causing the forward movement of lens, and the pupillary block plays a more important role in the pathogenesis of acute PACG.
