Inspired oxygen concentrations with or without an oxygen economizer during ether draw-over anaesthesia.

An oxygen economizer tube is attached to draw-over vaporizers and acts as a reservoir of supplemental oxygen. The clinical importance of the presence or absence of the economizer tube (volume 130 ml) has not been adequately studied in manually ventilated patients using ether from an Ohmeda Cyprane Portable Anesthesia Complete (PAC) draw-over vaporizer. A total of sixteen patients ASA 1-2, undergoing elective surgery for peripheral orthopaedic procedures were studied with and without an economizer tube. Each patient acted as his or her own control. Standard procedures were used for anaesthetic induction with muscle relaxant, endotracheal intubation and anaesthetic maintenance. Supplemental oxygen was supplied by an oxygen concentrator. Using the draw-over vaporizer without an oxygen economizer tube, there was a slight increase in FiO2 of 20%, 23%, 27%, 30%, 33% and 33%, with increasing oxygen supplementation of 0 to 5 l/min, respectively. With an economizer tube, the FiO2 values increased to 20%, 26%, 35%, 46%, 54% and 66% at 0 to 5 l/min of oxygen respectively. The FiO2 values were significantly different at 3, 4, and 5 l/min (P < 0.05), showing the potential advantages of an oxygen economizer tube attached to a draw-over vaporizer in this setting. No significant differences were seen in the oxygen saturations of these healthy patients with or without an oxygen economizer.

Extracellular calmodulin-binding proteins in body fluids of animals.

The extracellular calmodulin-binding proteins (CaMBPs) were investigated in body fluids of animals by using the biotinylated calmodulin gel overlay method. Four major CaMBPs with molecular masses of 24, 31.5, 44/45 and 94 kDa were detected in serum, two of 24 and 63 kDa in bovine milk and three of 14, 24 and 52 kDa in human saliva. It suggested that extracellular CaMBPs exist commonly in body fluids of animals, and this result may provide a new clue for understanding the role of extracellular calmodulin.

A 120 kilobase resolution contig map of the rice genome.

92% of the rice genome (4.3 x 10(8) bp, 2n = 24) was covered by 631 contigs of various length, which were generated by fingerprinting from a representative and genetically stable bacterial artificial chromosome (BAC) library of the Guang Lu Ai 4 (a O. Sativa variety) genome with the average insert of 120 kb in length. To form the contig map, 565 molecular markers of RFLP, STS, cDNA and anchor set derived from two O. Sativa varieties were by colony hybridization mapped to the contigs, which were then assigned to and ordered along the particular chromosomes according to the marker colinearity. Being highly conserved DNA sequences shared among the genomes of rice, barley, wheat, oat, maize, sorghum and sugar cane, 89 anchor markers mapped help to identify the rice genes through the information provided by the maps of relative genomes, and vice versa. Numerous repeated DNA sequences of various length were identified and mapped to the chromosomes. Physical distances have been determined for hundreds pairs of adjacent markers, which would facilitate the identification by map-based cloning the rice genes of interest. The accuracy of clone overlaps in contigs was further confirmed by the existence in contigs of well fit stacks of marker-lodged clones independently identified by hybridization. Large scale DNA sequencing of individual chromosomes could now be initiated simply by selecting and sequencing the minimally overlapped BAC clones of the contigs.

[Study on p53 protein expression, cell proliferation and apoptosis in benign and malignant brain tumors].

OBJECTIVE: To explore the effect of p53 protein expression in brain tumors on the cell proliferation and apoptosis, and the relationship between above parameters and histological classification and malignant degree of these tumors. METHODS: The p53 protein expression, cell proliferation and apoptosis of 10 control brain tissues (CBT) and 80 brain tumor samples were studied by in situ cell death detection and immunohistochemistry. RESULTS: Sixty-nine of the brain tumors (86.3%) expressed p53 protein. The amount of tumor cells positively stained for p53 protein increased with the degree of malignancy. All the CBTs did not express p53 protein. The densities of cells positive for proliferating cell nuclear antigen and Ki-67 antigen staining in tumor groups were higher than that in CBT group and were elevated with the malignant degree and the level of p53 protein expression of the tumors. On the contrary, the density of apoptotic cell in tumor groups was lower than that in CBT group, reducing in accord with the increase of malignancy and the level of p53 protein expression of the tumors. CONCLUSIONS: These results suggest that above 4 parameters have a referential value for the evaluation of biological behavior of brain tumors. It is clear from the present study that the abnormality of expression and function of p53 protein correlates with the imbalance between cell proliferation and apoptosis. This may be one of the important factors in the development and progression of primary and metastatic brain tumors.

[Effect of pathogen-stimulated human CD4+ T cells on gamma delta T cells].

OBJECTIVE: CD4+ helper T lymphocytes and gamma delta T cells play important role in antimicrobial immunity, however, the mechanism of interactions in question still remains to be elucidated. METHODS: CD4+ T cells and gamma delta T cells separated from peripheral blood lymphocytes of healthy donors by panning technique were stimulated with intracellular pathogen-Mycobacterium tuberculosis and human herpes virus-6 as well as extracellular pathogen-Trichomonas Vaginalis respectively in vitro. RESULTS: The stimuli induced CD4+ T cells significantly to proliferate and producing Th1-type cytokine profile-IL-2 and IFN-gamma. With a semipermeable co-culture system in vitro, antigen-activated CD4+ Th1-like cells were found to promote the proliferation and cytotoxicity of gamma delta T cells via soluble factors. CONCLUSION: Functions of gamma delta T cells might be inhanced by CD4+ cells with Th1-type cytokines in antimicrobial immunity.

[The alveolar bone density in rats with experimental diabetes].

The alveolar bone turnover in rat with experimental diabetes was investigated. The serum osteocalcin concentration in diabetic rat was shown to increase as determined by radioimmunoassay; while alveolar and femur bone density decrease as determined by Dual Energy X-ray Bone Densitometer; histological examination showed that bone resorption was increased and bone formation decreased in the alveolar bone in diabetic rat. It is suggested that the alveolar bone in rats with diabetes mellitus have a tendency to develop osteoporosis.

[Significance of c-erbB-2 and PCNA expression in adenocarcinoma of uterine cervix].

OBJECTIVE: Significance of c-erbB-2 and PCNA expression in adenocarcinoma. METHODS: Expression of c-erbB-2 and PCNA in 74 cases of adenocarcinoma of the uterine cervix was examined by immunohistochemistry. RESULTS: The expression of c-erbB-2 was detected in 34 cases. The positive staining of c-erbB-2 was associated with increased lymph node metastasis (57.1% Vs 24.0%, P = 0.041) and lower 5-year survival rate (32.4% Vs 58.9%, P = 0.008). The average PCNA labelling index (PCNALI) was 40.6% (0.1%-91.4%). High PCNALI was associated with lymph node metastasis (56.4% Vs 38.5%, P = 0.016) and lower 5-year survival rate (28.7% Vs 64.4%, P = 0.005). Positive staining of c-erbB-2 were associated with high PCNALI (44.7% Vs 34.6%, P = 0.003). CONCLUSION: c-erbB-2 and PCNALI were associated with malignant biological behavior and poor prognosis. They may be potential prognostic factors in cervical adenocarcinoma.

[Diastase and reducing sugar in Fritillaria thunbergii Miq. during the process of dormancy relieving with low temperature].

The changes of diastase's activity and reducing sugar's content in Fritillaria thunbergii were studied during the process of relieving dormancy with low temperature. The results showed that the metabolism in bud and scale was changed, the diastase's activity and reducing sugar's content of the adaxial epiderm were higher than those of the inner store tissue in the scale, but were approximate to those of the bud.

[An experimental study on the effects of lingzhi spore on the immune function and 60Co radioresistance in mice].

Lingzhi spore (2 g.kg-1.d-1, 10 times) was orally administered to mice, and studies were carried out on the carbon clearance, delayed-type skin hypersensitivity (DTH), antibody hemagglutinin value and spleen weight. The results showed that Lingzhi spore could increase the immune function, inhibit the reduction of white blood cells and raise the survival rate of mice after they were radiated by 870 rad 60Co.

[Application of TLC to tannin measurement].

There has been little report on the use of in measuring tannin in traditional Chinese medicinal preparations by TLC. Gallic acid, the decomposd product of tannin, was in bright yellow and green color, showing a strong specificity. The measurement of gallic acid can provide reliable indexes for quality assay of TCM preparations.

[The development and application of computer assisted photogrammetric-system of facial soft tissue running under multiple facilities for taking image].

The photogrammetric-system was developed by using multimedia computer, colour television camera, SE100 video blaster, Newsketch 1212 HR digitizer and by using Visual Basic language. It ran under Windows 3.1 and Windows 95. The features of this system: 1. It had three kinds of functions: photogrammetric function for frontal and profile facial soft tissue, function for managing file and function for forming dynamic data base. 2. It ran under different facilities for taking image (television camera with video blaster, digitizer, scanner) speedily and specificly, and had very friendly interface, and was of multimedia. The application of this system: 1. assisting diagnosing, making plan for treatment and observing results of treatment in clinic of orthodontics, orthopedics and facial plastics, 2. assisting analysing the features of facial soft tissue about certain people, 3. assisting the multimedia teaching.

[The analysis of frontal facial soft tissue of normal native adult of han race of Guangdong province by using the computer assisted photogrammetric-system].

UNLABELLED: The authors analysed the feature of frontal facial soft tissue of 150 native adults of Guangdong province by using the computer assisted photogrammetric-system made by the authors, which contained several new mode parameters planed by the authors. THE RESULTS: 1. The mean measurements of six parameters (Sn-Mes, Gs-Mes, Exr-Exl, Enr-Enl. [symbol: see text]Zyr-Rc-Mes, [symbol: see text]Zyl-Lc-Mes.) of the male subjects were larger than those of the female subjects, and the mean measurements of two parameters (Zyr-Zyl/Gs-Mes, Rc-Lc/Gs-Mes) of the male subjects were smaller than the ones of the female subjects. 2. The four measurements (Tr-Mes, Gs-Sn, Zyr-Zyl, Chr-Chl) in both male and female subjects of Guangdong province were smaller than the ones of Sichuan and Shandong provinces, and one measurement (Tr-Gs) of Guangdong male subjects was also smaller than the one of Sichuan or Shandong province, and one measurement of Guangdong female subjects smaller than the one of Shandong province. The conclusions: 1. The normal native adults of han race of Guangdong province show sex-difference and region-difference in the feature of frontal facial soft tissue. 2. The new-mode parameters (such as: [symbol: see text]Zyr-RC-Mes, [symbol: see text]Zyl-Lc-Mes, Zyr-Zyl/Gs-Mes, Rc-Lc/Gs-Mes) are helpful in studying the feature of frontal facial soft tissue.

[Relationship between nm23-H1 expression and lymph node metastasis and prognosis in cervical cancer].

OBJECTIVE: To investigate the expression of nm23-H1 in cervical carcinoma and its significance. METHODS: Expression of nm23-H1 was examined by immunohistochemical method in 39 cases of adenocarcinoma and 39 cases of squamous cell carcinoma of the uterine cervix. The relationship between expression of nm23-H1 and clinic-pathologic factors and prognosis was analyzed by chi-square test. RESULTS: Positive staining rate of nm23-H1 was 44.6% in adenocarcinoma and 39.2% in squamous cell Carcinoma. The positive staining rate of nm23-H1 in stage I and II adenocarcinoma was 61.1% and 28.6% respectively (P = 0.044); in patients with recurrence nm23-H1 positive rate was lower than that in patients without recurrence (21.5% vs 56%, P = 0.39); in patients with lymph node negative, nm23-H1 positive staining was more than that in patients with lymph node positive (52% vs 28.6%), however, this difference was not statistically significant (P = 0.162). None of 14 cases of lymph node metastasis was strong positive stainig, whereas 7 of 25 without lymph node metastasis were demonstrated to have strong positive staining (P = 0.031). The 5-year survival rate in negative staining group was lower than that in the positive staining group (52.5% vs 82.4%, P = 0.042). In squamous cell carcinoma there was no statistically significant relationship between nm23-H1 expression and clinic-pathologic factors and prognosis. CONCLUSIONS: nm23-H1 expression was associated with biologic behavior in cervical adenocarcinoma.

[Alteration in regional blood flow in minipigs' femur under inadequate decompression studied by isotopes washout method].

We studied the alteration in regional blood flow in minipigs' femur under inadequate decompression after hyperbaric air exposure. The animals were placed in the hyperbaric chamber and exposed to the pressure of 0.5 MPa for 1.5 h, which was reduced to atmosphere at an ascent rate of 0.03-0.04 MPa/min. Regional blood flow in the femur was measured by the isotopes washout method of inhaling 133Xe. The radioactivity was monitored by a multifunctional blood flowmeter interfaced to a minicomputer. Before exposure, the values of average blood flow (F) of femur on the left and right were 15.4 +/- 1.8 and 16.9 +/- 2.0 ml/100 g.min respectively. The values of blood flow (f1) of hematopoietic marrow were 19.1 +/- 2.0 and 21.3 +/- 2.0 ml/100 g.min (n = 7). After inadequate decompression, F values were reduced to 10.3 +/- 1.8 and 11.1 +/- 1.6 ml/100 g.min and f1 values reduced to 13.9 +/- 1.4 and 13.8 +/- 1.0 ml/100 g.min (n = 7), which were obviously lower than those before exposure (P < 0.05). This experiment showed that significant reduction in blood flow in the femur region occurred under inadequate decompression after the exposure of minipigs hyperbaric environment, suggesting that ischemia is a causative factor of osteonecrosis.

Evolution of proneural atonal expression during distinct regulatory phases in the developing Drosophila eye.

BACKGROUND: Receptors of the Notch family affect the determination of many cell types. In the Drosophila eye, Notch antagonises the basic helix-loop-helix (bHLH) protein atonal, which is required for R8 photoreceptor determination. Similar antagonism between Notch and proneural bHLH proteins regulates most neural cell determination, however, it is uncertain whether the mechanisms are similar in all cases. Here, we have analyzed the sensitivity of atonal expression to Notch signalling using a temperature-sensitive Notch allele, by the expression of activated Notch or of the ligand Serrate, and by monitoring expression of the atonal-dependant gene scabrous and of the Notch-dependent Enhancer of split genes. RESULTS: The atonal expression pattern evolves from general "prepattern' expression, through transient "intermediate groups' to R8 precursor-specific expression. Successive phases of atonal expression differ in sensitivity to Notch. Prepattern expression of atonal is not inhibited. Inhibition begins at the intermediate group stage, corresponding to the period when atonal gene function is required for its own expression. At the transition to R8 cell-specific expression, Notch is activated in all intermediate group cells except the R8 cell precursor. R8 cells remain sensitive to inhibition in columns 0 and 1, but become less sensitive thereafter; non-R8 cells do not require Notch activity to keep atonal expression inactive. Thus, Notch signaling is coupled to atonal repression for only part of the atonal expression pattern. Accordingly, the Enhancer-of-split m delta protein is expressed reciprocally to atonal at the intermediate group and early R8 stages, but is expressed in other patterns before and after. CONCLUSIONS: In eye development, inhibition by Notch activity is restricted to specific phases of proneural gene expression, beginning when prepattern decays and is replaced by autoregulation. We suggest that Notch signalling inhibits atonal autoregulation, but not expression by other mechanisms, and that a transition from prepattern to autoregulation is necessary for patterning neural cell determination. Distinct neural tissues might differ in their proneural prepatterns, but use Notch in a similar mechanism.

T cells bound by vascular cell adhesion molecule-1/CD106 in synovial fluid in rheumatoid arthritis: inhibitory role of soluble vascular cell adhesion molecule-1 in T cell activation.

Elevated levels of soluble vascular cell adhesion molecule-1 (sVCAM-1)/CD106 have been reported in synovial fluid (SF) from patients with rheumatoid arthritis (RA). In the present study, VCAM-1-positive lymphocytes were found in SF from RA patients. The data strongly suggest that sVCAM-1 might be bound to lymphocytes in SF. rsVCAM-1 in the fluid phase can bind to both SF lymphocytes and IL-2-dependent T cell lines with up-regulated expression and binding activity of VLA-4. Furthermore, proliferative responses of SF mononuclear cells (SFMC) with PHA, immobilized anti-CD3, or anti-CD2 and PMA were inhibited to various extents in the presence of rsVCAM-1, but only PMA-induced proliferative response of PBMC from normal individuals was inhibited notably in the presence of rsVCAM-1. rsVCAM-1 also drastically reduced IL-2 production of Jurkat leukemic T cells possessing high affinity VLA-4 with the stimulation of anti-CD3 and PMA, suggesting that the T cell hyporesponsiveness induced by rsVCAM-1 might stem from impairment of IL-2 production. These results indicate that sVCAM-1 provides a negative signal to T cell activation, probably by affecting the pathway of protein kinase C activation. Thus, binding of sVCAM-1 to SF lymphocytes might partly explain the anergic state of these lymphocytes.
