Construction of an ordered cosmid collection of the Escherichia coli K-12 W3110 chromosome.

A cosmid library of the Escherichia coli K-12 W3110 chromosome was constructed in which clones were assigned to locations on the chromosome map by hybridization and genetic marker complementation tests. Approximately 70% of the genome was represented by this library. The identified clones can be maintained in the homologous system and would facilitate genetic studies of E. coli.

[Effect of sevoflurane on respiratory impedance measured by the forced oscillation method with random noise input].

This study was performed to investigate the effect of sevoflurane, a new inhalation anesthetic, on the respiratory impedance measured by forced oscillation method with random noise input. Random noise (1 to approximately 25Hz) oscillated by a loudspeaker was imposed on the respiratory system of dogs through a tracheostomy and the oscillatory pressure wave and flow wave were monitored at the same point with a pressure transducer and a pneumotachograph. This procedure was repeated during inhalation of 100%-oxygen, 2.0%-then 4.0%-sevoflurane-oxygen mixture, respectively. Impedance was calculated with fast Fourier transform by spectrum analyzer and a personal computer. During inhalation of 4.0%-sevoflurane-oxygen mixture, respiratory impedance especially respiratory resistance decreased at the high frequency area (6to approximately 20Hz), but 2.0%-sevoflurane-oxygen mixture did not cause a significant change of respiratory impedance. This study suggests that inhalation of an anesthetic concentration of sevoflurane produces weak bronchodilation compared with halothane or enflurane.

[Radiation therapy of carcinoma of the hypopharynx and recurrence in the epipharynx and mesopharynx].

The role that radiation therapy plays in the prophylaxis of an epi-and mesopharyngeal recurrence of a hypopharyngeal cancer remains undefined. Because of this 48 patients with a hypopharyngeal cancer, who had undergone radiation therapy from 1975 through 1987 at Chiba University Hospital, have been analyzed retrospectively. The 5-year-survival was 17% and an epi- or a mesopharyngeal recurrence was found in 8 patients. A radiation dose of more than 1350 ret seemed to prevent a recurrence in these regions, although it must be further studied as to whether an increase of the radiation field to include the epipharynx can be effectively improved the recurrence-free survival rate.

[Partitioning and measurement of respiratory impedance into central and peripheral airways with catheter tip type transducer].

In this study, we have examined the availability of catheter tip type transducer for the partitioning of respiratory impedance. Total respiratory resistance and reactance from 1 to 20 Hz determined by forced oscillation method with random noise input in anesthetized dogs were partitioned by catheter tip type transducer into central airway resistance (Rc) and reactance (Xc) as well as peripheral resistance (Rp) and reactance (Xp). Intratracheal acetylcholine (0.1g) increased Rp at the wide frequency range (from about 1 to 20 Hz), although Rc did not change with it. The decreases of Rc at higher frequencies (over 12Hz) were produced by intratracheal salbutamol (10mg), whereas Rp did not change significantly. These results suggest that the predominant site of action of intratracheal acetylcholine and salbutamol is peripheral airway and central airway respectively. We have concluded that the respiratory impedance can be partitioned noninvasively with catheter tip type transducer without pneumothorax and hemorrhage caused with the retrograde catheter. This method can be applied clinically during general anesthesia or in intensive care.

Macrophage colony-stimulating factor is produced by human T lymphoblastoid cell line, CEM-ON: identification by amino-terminal amino acid sequence analysis.

A subclone, designated CEM-ON, derived from an azaguanine-resistant human leukemic T cell line, CEM-AG(R), constitutively secretes a colony-stimulating factor (CSF) which stimulates the production of macrophages from murine bone marrow progenitor cells. This CSF has been purified from serum-free conditioned medium. Highly purified CEM-ON CSF with a specific activity of 4.7 X 10(7) units/mg protein was obtained. Amino-terminal sequence analysis showed that the first 27 amino acids were identical to the amino-terminal sequence of the M-CSF (CSF-1) based on the cDNAs for human M-CSF. On SDS-PAGE analysis, CEM-ON CSF had an apparent molecular weight of 33,000-43,000; following reduction with 2-mercaptoethanol, this migrated as a 20,000-24,000 subunit, suggesting a homodimer structure. These results show that a human T cell line, CEM-ON, secretes M-CSF into its medium.

[The clinical evaluation of the serum SCC antigen levels with "SCC RIABEAD" by immunoradiometric assay].

The clinical significance of serum SCC antigen level was evaluated by the monoclonal antibody method (SCC.RIABEAD Dinabot Co. Ltd). Patients with squamous cell carcinoma showed a high positive SCC antigen level and positive rate elevated with the advance of the clinical stage. The serum SCC antigen level was decreased by treatment, and it increased again before obvious clinical recurrence was recognized. The results suggest that measurement of serum SCC antigen level is useful as a follow up of cancer treatment.

Photosensitive lichenoid reaction accompanied by nonphotosensitive subacute prurigo caused by carbamazepine.

A 58-year-old man developed a lichenoid reaction on light-exposed areas and subacute prurigo separate from the sun-exposed areas after receiving 200 mg of carbamazepine daily for one year. The difference action spectrum for erythema with or without ingesting the drug ranged from 250 to 390 nm with peaks at 260 and 290 nm. A positive photoingestion test was obtained after administering 2 mg of carbamazepine orally in one dose. Patch test with carbamazepine was negative, while photopatch test with carbamazepine was positive. Positive intracutaneous test was obtained with an irradiated mixture of human serum albumin and carbamazepine at 24 h and 48 h after injection. This patient appeared to be a case of photosensitive lichenoid reaction accompanied by nonphotosensitive subacute prurigo caused by carbamazepine.

[Non-Hodgkin's lymphoma in patients aged 70 or older].

Fourty-one patients, referred to the Department of Radiology, Chiba University Hospital from 1975 through July, 1987, have been analyzed to determine their prognostic factors. These patients showed a less favorable prognosis (2 and 5 year actuarial survival, 30% and 19% respectively) compared with the younger patients. The histological grade by WF and their clinical stage reflected the prognosis well. Although the achievement of complete remission seemed to affect prognosis favorably, aggressive regimens with chemotherapy and/or radiotherapy often caused their deaths. In senile patients with NHL, it is a prerequisite to consider the effects on the patient of curative therapy more carefully, especially when Adriamycin is used.

The syndrome of inappropriate secretion of antidiuretic hormone by rectal cancer--a case report.

The syndrome of inappropriate secretion of antidiuretic hormone (SIADH) is commonly observed in patients with bronchogenic carcinoma. We report here a rare case of SIADH induced by rectal cancer. Laboratory findings showed that the serum sodium concentration and serum osmolarity were low, and that the ADH level was high. Resection of the rectal cancer resulted in a complete correction of the associated inappropriate ADH. The titer of the rectal cancer tissue proved to be high (80 pg/g).

[Evaluation and comparison of high-sensitivity immunoradiometric assay kits for thyroid stimulating hormone].

Fundamental and clinical characteristics of 3 kinds of high-sensitivity immunoradiometric assay (IRMA) kits for thyroid stimulating hormone (TSH). i.e., RIA BEADS II (kit A), TSH kit Daiichi II (kit B) and Ab tube TSH 'Eiken' (kit C) and one conventional radioimmunoassay (RIA) kit, i.e., TSH kit Daiichi (kit D), were studied. In the recovery test and the reproducibility test, there was no significant difference between the 4 kits. The sensitivities of kits A, B and C were much higher than that of kit D, and those IRMA kits were sensitive enough to distinguish hyperthyroidism from normal samples. For low concentrations of TSH (less than 5 microU/ml), the data from kits D, B, C and A tended to show higher values in that order. The correlation between the data measured by kits B and D, and the tendency of kit A toward lower values agreed well with other reports.

Regions necessary for pH-dependent assembly of gizzard myosin rod.

Aggregated and disaggregated forms of gizzard myosin rod and its fragments in various concentrations of NaCl (0-0.30 M) at various pH (7.4-8.6) were distinguished from each other by their permeability through a Sepharose 4B column. The rod existed in three forms, namely: large aggregates impermeable to the column, small aggregates eluted at the void volume of the column and a disaggregated monomer which penetrated the column. The relative proportions of the three forms varied depending on the salt concentration and pH. The monomeric rod was detected in NaCl solutions above 0.20 M and its relative proportion at 0.25 M NaCl was larger than those of the small and large aggregates. The small aggregates of the rod were predominant at below 0.05 M NaCl and, upon decrease in pH from 8.6 to 7.4, these small aggregates in NaCl solutions between 0.10 M and 0.15 M were replaced by the large aggregates. Light meromyosin, which corresponded to the C-terminal two-thirds of the rod, existed exclusively as large aggregates in NaCl solutions below 0.15 M; increase of NaCl concentration to above 0.20 M resulted in the formation of its monomer, instead of the large aggregates. In contrast to the rod, no small aggregated form of the light meromyosin was detected. Truncated light meromyosin which had lost a small segment from either the C-terminal or N-terminal of light meromyosin was eluted only as a monomer in any NaCl concentration at any pH. It may be deduced from the above results that a small segment in the light meromyosin is requisite for the assembly of both rod and light meromyosin in the NaCl solutions below 0.15 M and that the relative proportion of small and large aggregates of the rod is determined in a pH-dependent manner by the subfragment 2 segment, the N-terminal third of the rod.

[A randomized trial with various combinations of cis-platinum (CDDP), mitomycin C (MMC), peplomycin (PEP) and ftorafur (FT) in the treatment of non-small cell lung cancer].

From March 1983 to February 1985, we treated 74 patients (pts) with inoperable non-small cell lung cancer. Twenty-seven pts with squamous cell carcinoma were randomized between regimen PMP (CDDP 60 mg/m2, d 1, MMC 6 mg/m2, d 3, d 10 and PEP 5 mg/m2, d 3-7) and regimen PM (CDDP 60 mg/m2, d 1 and MMC 6 mg/m2, d 3, d 10). Forty-seven pts with adenocarcinoma and large cell carcinoma were randomized between regimen PMF (CDDP 60 mg/m2, d 1, MMC 6 mg/m2, d 3, d 10 and FT 800 mg/body, d 3-21) and regimen PM. The response rates of evaluable cases (EC) were as follows: Squamous cell carcinoma; Regimen PMP 50% (1 CR + 4 PR/10 EC). Regimen PM 40% (4 PR/10 EC). Adenocarcinoma plus large cell carcinoma; Regimen PMF 13.6% (3 PR/22 EC). Regimen PM 11.1% (2 PR/18 EC). The median survival time (MST) was increased from 23 weeks in non-responders to 32 weeks in responders. However, the difference between the survival curves for responders and non-responders was not statistically significant. Of the toxic effects shown in all 74 registered pts, hematological (64.9%), gastrointestinal (60.8%) and renal (31.1%) toxicities were the common complications. We concluded that regimen PMP was more useful than regimen PM for pts with squamous cell carcinoma but that regimen PMF demonstrated no appreciable difference, compared with regimen PM for pts with adenocarcinoma and large cell carcinoma.

Expression of epidermal growth factor receptor gene in cultured human lung cancer cells.

The expression and organization of epidermal growth factor (EGF) receptor gene in cultured human lung cancer cell lines (5 adenocarcinomas, 3 squamous cell carcinomas, 2 small cell carcinomas and 1 large cell carcinoma) have been studied. Two (PC-8 and PC-9) of the adenocarcinomas overproduced EGF receptor mRNA and protein, and exhibited gene amplification, the magnitude of which was comparable to that of A431 cells. Six cell lines (3 adenocarcinomas, 2 squamous cell carcinomas and 1 small cell carcinoma) expressed EGF receptor gene and its product to a significant level without gene amplification, and the other three cell lines were found to be negative as regards expression.
