Hypnotic action of N3-substituted arabinofuranosyluracils on mice.

Methyl (2), ethyl (3), propyl (4), butyl (5), allyl (6), benzyl (7), o-, m-, p-xylyl (8-10), and alpha-phenylethyl (11) derivatives of arabinofuranosyluracil (1) were synthesized and their pharmacological effects in mice were examined by using hypnotic activity and synergism with pentobarbital as indices for the CNS depressant effects. At a dose of 2.0 micromol/mouse by intracerebroventricular injection, the values of mean sleeping time induced by 7-11 were 144, 154, 117, 33, and 34 min, respectively, whereas the alkyl (2-6) derivatives did not cause any hypnotic activity. N3-o-Xylylarabinofuranosyluracil (8) displayed the most potent hypnotic activity among the derivatives tested. Certain derivatives (6-11) significantly prolonged the pentobarbital-induced sleeping time compared to control. The present study indicated that substitution with benzyl and/or related groups on the N3-position of arabinofuranosyluracil produced CNS depressant effects.

A non-potentiometric sensing method for the determination of pentoxyverine with PQC sensors based on ion-pair complexes.

The construction and general performance characteristics of three piezoelectric quartz crystal sensors responsive to the pentoxyverine are described here. This kind of non-potentiometric sensing method is based on use of ion-pair complexes of the pentoxyverine cation with three counter anions, namely, tungstophosphate, tetraphenylborate and picrolonate. The complexes were embedded in a PVC matrix. Adsorption of the pentoxyverine ion on the complex caused a frequency decrease of the crystal. The frequency decrease was proportional to the amount of adsorbed analyte. The influencing factors were investigated in detail, and then optimized. The proposed sensors exhibit reasonable selectivity and a higher sensitivity than the potentiometric sensors. For a sensor modified with pentoxyverine-phosphotungstate, the calibration graph was linear over concentration of 1.0 x 10(-7) - 5.0 x 10(-5) M with a detection limit of 6 x 10(-8) M at pH 5.4.

An electrochemical quartz crystal impedance study on the rising of an aqueous solution meniscus for a partially immersed gold electrode during the electrochemical reduction of oxygen.

An electrochemical quartz crystal impedance system (EQCIS) was used to study the resonance behavior of an AT-cut 9-MHz piezoelectric quartz crystal (PQC) with its Au electrode partially immersed in KCl, Na2SO4 and NaClO4 aqueous solutions, respectively. An in situ determination of the immersed area and the height of the electrode was achieved by simultaneous measurements of the PQC electroacoustic admittance and the electrochemical impedance. The rising of the solution meniscus for a gold electrode partially immersed in aqueous solutions was found at oxygen reduction potentials and evaluated versus the electrolyte, electrolyte concentration, solution pH and oxygen concentration. The solution meniscus rising was explained based on a lowering of the contact-angle hysteresis and a continued collection of the water product at the solid-gas-solution interface during oxygen reduction.

A comparative study on the viscoelasticity and morphology of polyaniline films galvanostatically grown on bare and 4-aminothiophenol-modified gold electrodes using an electrochemical quartz crystal impedance system and SEM.

The equivalent-circuit parameters of the 9-MHz piezoelectric quartz crystal (PQC) resonance were measured in situ during the galvanostatic polymerization of aniline on 4-aminothiophenol(4-ATP)-modified and bare Au electrodes for ca. 2000 s, respectively. Two polymerization media, 0.100 mol L-1 aniline in 1.0 mol L-1 H2SO4 and in 2.0 mol L-1 HClO4 aqueous solutions, and two values of the current density, 12 and 36 microA cm-2, were used. At identical levels of the resonant frequency shifts in the solutions, obviously greater increases in the motional resistance (R1) were found after aniline polymerization on bare Au electrodes, though the absolute values of delta f0/delta R1 were all large; also, the resonant frequency shifts in air (delta f0g) were considerably smaller for PANI films grown on bare Au electrodes. It is thus concluded that, under identical polymerization conditions, (1) the PANI film grown on a bare gold electrode is rougher, less compact, and can entrap solution more notably; (2) the deposition efficiency of PANI is higher on a 4-ATP-modified Au electrode, owing to a significantly greater observed "dry" frequency shift, and thus a greater "net" mass value of the polyaniline backbone. SEM observations have confirmed that PANI films on 4-ATP-modified Au electrodes were smoother and more compact than those grown on bare Au ones under identical polymerization conditions. In addition, a technique of simultaneous measurements of the electroacoustic admittance of the PQC resonance and the electrochemical impedance was used to monitor the adsorption of 4-ATP onto a PQC gold electrode.

[Clinical efficacy of double autologous hematopoietic stem cell transplantation in the treatment of adult acute lymphoblastic leukemia].

OBJECTIVE: To evaluate the clinical efficacy of double autologous hematopoietic stem cell transplantation (DAHSCT) in adult acute lymphoblastic leukemia and analyse the affecting factors. METHODS: Thirteen adult acute lymphoblastic leukemia (ALL) patients received the first AHSCT in 12 months after complete remission (CR). The first conditioning regimen was Vp16,600-1000 mg or Ara-C 2-4 g/m2, CTX 120 mg/kg, total body irradiation(TBI) 7.5-9.0 Gy and in 6 patients BCNU 125 mg was added. All the patients received the second AHSCT in 4 to 10 months after the first AHSCT. The second conditioning regimen was Vp16,600-1000 mg or Ara-C 2-4 g/m2, Mel 140 mg/m2, CTX 120 mg/kg. RESULTS: All engrafted patients had rapid hematopoietic reconstitution. There was no AHSCT related death. The median follow-up duration was 837 days. Seven of the 13 DAHSCT patients were alive. The 3 year disease-free survival (DFS) was (53.8 +/- 7.7)%. Those who had more lymphoblasts in bone marrow at the second AHSCT than that at the first AHSCT had greater probability of relapse. CONCLUSION: It suggested that DAHSCT could be used as an important treatment owing to its low AHSCT related death and relatively long DFS.

Regional glucose metabolic increases in left auditory cortex in tinnitus patients: a preliminary study with positron emission tomography.

OBJECTIVE: To investigate the relationship between tinnitus and glucose metabolism in auditory cortex and whether positron emission tomography (PET) can be an objective tool in measuring tinnitus. METHODS: Eleven right-handed patients with severe tinnitus and ten right-handed control subjects participated in the 18F-FDG/PET study. Analysis with regions of interests was used to calculate asymmetry indices according to the formula: [(L-R) x 100/[(L + R) divided by 2]]. RESULTS: Glucose metabolism in the auditory cortex of tinnitus patients was asymmetric between the left and right auditory cortices, with that of the left being much higher than that of the right. The asymmetry indices of tinnitus patients was significantly higher than that of the control group (unpaired t test, P < 0.001). This revealed that the increased metabolic activity was present in the predominant left hemisphere with a significant focus on the superior and transverse temporal gyri (Brodmann areas 41 and 42, respectively corresponding to primary and secondary auditory cortex), and the results were independent of the subjective localization of the tinnitus sensation. CONCLUSION: It is suggested that the increased metabolism in the left auditory cortex is related to the tinnitus sensation. PET is capable of providing objective evidence for tinnitus and may be used as a potential tool in measuring tinnitus.

[The relationship among plasma soluble vascular cell adhesion molecule-1 level, airway responsiveness and blood eosinophil cationic protein in asthmatics with remission at least 3 years].

OBJECTIVE: To study the mechanism of asthma remission. The plasma soluble vascular cell adhesion molecule-1 (sVCAM-1), airway responsiveness (PC20) and other relative factors were measured in asthmatics with remission at least 3 years and compared with healthy controls. METHODS: Group I (asthmatics with remission at puberty): 20 cases including 11 males and 9 females, mean age of 21 years. All of the patients were definitely diagnosed as asthmatics in childhood and got natural remission for at least 3 years(with median duration of 6 years). Group II (normal controls): 21 healthy college students including 10 males and 11 females, mean age of 22.5 years, had no history of asthma or other allergic diseases. The intravenous blood was taken from all subjects with EDTA as anticoagulant for study. The airway responsiveness were measured by methacholine bronchial provocation test(PC20). The sVCAM-1 was measured with ELISA, according to the manual of the kit. Methacholine was atomized by Wright atomizer [Devilbiss AP-50, USA, driven by air compressor, with a constant atomizing volume of (0.13 +/- 0.02) ml/min, diameter of atomizes particles 1.3-3.0 microns]. Its concentration was redoubled from 0.03 to 128 g/L until a certain dose as PC20 value triggering forced expiratory volume in one second (FEV1) decreased by 20% reached. The eosinophil cationic protein (ECP) levels were measured with enzyme-linked immuno-CAP system. Blood eosinophils(> 330/mm3 as being above normal) and basophils (> 33/mm3 as abnormally increased) were counted using Acian blue dyeing. RESULTS: (1) The mean value of sVCAM-1 of group I (63.7 +/- 31.7) micrograms/L was significantly higher than that of group II (41.9 +/- 11.7) micrograms/L. (2) The percentage of the cases with increased eosinophils in group I was 15.0% (3/20), had no significant difference compared with that in group II (4.8%, 1/21). The percentage of the cases with increased basophils of group I was 45.0%(9/20), significantly higher than that of group II, 4.8%(1/21), P < 0.01. (3) The blood ECP level of group I was (2.7 +/- 1.3) micrograms/L, simillar to that of group II (2.18 +/- 0.5) micrograms/L. (4) The PC20 value of group I (M = 2.6 g/L) was significantly lower than that of group II (M = 128 g/L). (5) The basophil count negatively correlated with the PC20(r = -0.66), however, the sVCAM-1 did not have significant correlation with the basophil count or with the PC20. CONCLUSION: The quantity and activity of eosinophils in asthmatics with remission at least 3 years are droped to the normal level, but the expression of VCAM-1, quantity of basophils were still higher than normal. The airway responsiveness had negative correlation with the basophil count.

[A study of computer-aided drawing system for individualized ideal arches].

OBJECTIVE: The aim of this study is to devise a computer aided drawing system for individualized ideal arches, to resolve the discommode of manual drawing of Bonwill-Hawley arch charts and and to avoid the non-matching problem when using the universal arch charts. METHODS: On the basis of drawing principle of Bonwill-Hawley individualized ideal arches, the computer program was written with Visual Basic version 6.0. RESULTS: The new computer-aided drawing system of individualized ideal arches was based on Windows9X/NT. After the data of mesiodistal diameters of maxillary anterior teeth crown (from mesial surface to distal surface of bracket for canine) were input, a matching individualized ideal arch chart would be drawing using the designed program immediately. CONCLUSION: The computer-aided drawing system of individualized ideal arch can help to draw a matching individualized ideal arch accurately and quickly. This system eliminates the complex and cockamamie process of manual drawing.

Aromatase-deficient (ArKO) mice have a phenotype of increased adiposity.

The aromatase-knockout (ArKO) mouse provides a useful model to examine the role that estrogens play in development and homeostasis in mammals. Lacking a functional Cyp19 gene, which encodes aromatase, the ArKO mouse cannot synthesize endogenous estrogens. We examined the adipose depots of male and female ArKO mice, observing that these animals progressively accumulate significantly more intraabdominal adipose tissue than their wild-type (WT) littermates, reflected in increased adipocyte volume at gonadal and infrarenal sites. This increased adiposity was not due to hyperphagia or reduced resting energy expenditure, but was associated with reduced spontaneous physical activity levels, reduced glucose oxidation, and a decrease in lean body mass. Elevated circulating levels of leptin and cholesterol were present in 1-year-old ArKO mice compared with WT controls, as were elevated insulin levels, although blood glucose levels were unchanged. Associated with these changes, a striking accumulation of lipid droplets was observed in the livers of ArKO animals. Our findings demonstrate an important role for estrogen in the maintenance of lipid homeostasis in both males and females.

Retrovirus vector silencing is de novo methylase independent and marked by a repressive histone code.

Retrovirus vectors are de novo methylated and transcriptionally silent in mammalian stem cells. Here, we identify epigenetic modifications that mark retrovirus-silenced transgenes. We show that murine stem cell virus (MSCV) and human immunodeficiency virus type 1 (HIV-1) vectors dominantly silence a linked locus control region (LCR) beta-globin reporter gene in transgenic mice. MSCV silencing blocks LCR hypersensitive site formation, and silent transgene chromatin is marked differentially by a histone code composed of abundant linker histone H1, deacetylated H3 and acetylated H4. Retrovirus-transduced embryonic stem (ES) cells are silenced predominantly 3 days post-infection, with a small subset expressing enhanced green fluorescent protein to low levels, and silencing is not relieved in de novo methylase-null [dnmt3a-/-;dnmt3b-/-] ES cells. MSCV and HIV-1 sequences also repress reporter transgene expression in Drosophila, demonstrating establishment of silencing in the absence of de novo and maintenance methylases. These findings provide mechanistic insight into a conserved gene silencing mechanism that is de novo methylase independent and that epigenetically marks retrovirus chromatin with a repressive histone code.

Detection and analysis of Bacillus subtilis growth with piezoelectric quartz crystal impedance based on starch hydrolysis.

A piezoelectric quartz crystal (PQC) impedance method based on the alpha-amylase-catalyzed hydrolysis of starch present in a culture medium has been developed for in situ monitoring of the whole growth process of Bacillus subtilis and the variation in the activity of alpha-amylase during bacterial growth. An S-shaped response behavior was observed for Deltaf(0), and simultaneously inverse S-shaped responses were found for DeltaR(1) and DeltaL(1). The ratio of DeltaR(1) to Deltaf(0) or DeltaL(1) coincided well with that calculated from Martin's equations reflecting the solution density-viscosity effect, suggesting that the continuing change in liquid loading onto the PQC surface causes significant variation in Deltaf(0), DeltaR(1), and DeltaL(1). Bacterial growth equations were derived from the kinetics of the enzyme-catalyzed hydrolysis of starch, which fit well with the experimental responses of Deltaf(0), DeltaR(1), and DeltaL(1). Kinetic parameters of bacterial growth, including the asymptote (A), the maximum specific growth rate (microm), and the lag time (lambda), were obtained and were in good agreement with those obtained from the pour plate count method. The variation in the activity of alpha-amylase exhibited peak-type behavior with its maximum value at the later stage of the log phase. In addition, the influence of initial bacterial concentration was also investigated.

Expression of cyclooxygenase-1 and cyclooxygenase-2 in the human prostate.

OBJECTIVES: To determine the cell-specific expression of the two major isoforms of cyclooxygenase (COX-1 and COX-2) in human noncancerous and cancerous prostatic tissues. METHODS: Thirty-one specimens of prostate carcinoma (CaP) and 10 specimens of benign prostatic hyperplasia (BPH) were stained with mouse antihuman COX-1 and COX-2 monoclonal antibodies. The stained specimens were analyzed both descriptively and in a semiquantitative manner by assigning an immunoreactive intensity score (0 to 4). The averaged results were compared for different histologic tissue types, including luminal and basal epithelium of BPH, the peripheral zone, high-grade prostatic intraepithelial neoplasia (PIN), and CaP of varying Gleason grades. RESULTS: COX-1 expression in noncancerous prostatic tissue was seen predominantly in the basal epithelial cells of BPH (90% positive staining). COX-1 expression was minimal in noncancerous luminal epithelial cells (0% to 10%) but was upregulated in CaP (63% of CaP specimens). Strong COX-2 expression was demonstrated in the smooth muscle cells of the prostate. COX-2 was also expressed in the basal epithelial cells (60% BPH, 94% peripheral zone, 75% PIN). Luminal epithelial cells derived from BPH, the peripheral zone, and PIN expressed COX-2 in 0%, 26%, and 86% of samples, respectively. COX-2 expression in CaP was intense and uniform, with 87% of samples demonstrating immunoreactivity. CONCLUSIONS: The results of the present study indicate that expression of both COX-1 and COX-2 in human CaP is increased. COX-2 expression is also increased in the basal and luminal epithelial cells of PIN. These data indicate that COX-1 and COX-2 (and/or their prostaglandin products) may play a role in the malignant transformation of the prostate.

Study of the Adsorption of Glutathione on a Gold Electrode by Using Electrochemical Quartz Crystal Impedance, Electrochemical Impedance Spectroscopy, and Cyclic Voltammetry.

Adsorption of a biological peptide, glutathione, on a gold electrode was studied by using electrochemical quartz crystal impedance (EQCI), electrochemical impedance spectroscopy (EIS), and cyclic voltammetry (CV) techniques. The time courses of responses of piezoelectric and electrochemical impedance parameters were simultaneously obtained during the adsorption processes of the two forms of peptide, oxidized and reduced glutathione. It was found that the frequency curve due to the oxidized glutathione (GSSG) adsorption exhibited a character of a sum of two exponential functions. For reduced glutathione (GSH), the frequency adsorption curve could be expressed by a first-order reaction kinetic model and the corresponding kinetic parameters at different amounts of GSH were obtained. The heterogeneous charge-transfer rate constants of ferricyanide/ferrocyanide before and after the peptide adsorption were determined by CV and EIS methods, respectively. According to the simple equivalent electric network of the electrochemical interface, the electrochemical impedance parameters were also obtained. The results showed that the proposed method should be found in wider applications in interfacial biochemistry studies since these combined techniques have advantages in real time multidimensional information including electrochemical and electrochemical impedance parameters. alpha-Amylase immobilized on bulk acoustic-wave sensor by UV-curing coating.

A new method for immobilization of alpha-amylase by UV-curing coating is proposed in this paper. The immobilization procedure of UV-curing coating on piezoelectric quartz crystal is simple and convenient, and causes less loss of enzymatic activity. The activity of the immobilized alpha-amylase is monitored by a technique based on bulk acoustic-wave (BAW) sensor. The frequency shift of BAW sensor can reflect the degree of hydrolysis of starch by the immobilized alpha-amylase. It is appropriate for the immobilized alpha-amylase to hydrolyze the soluble starch under pH 7.0 condition, which is similar to that of the free alpha-amylase. Kinetic parameters (the Michaelis constant, K(m), and the maximum initial rate V(max)) of the enzymatic hydrolysis of starch by the immobilized alpha-amylase are estimated by using a linear method of Lineweaver-Burk plot. K(m)=12.7mgml(-1) and V(max)=15.9Hzmin(-1). And the experimental results show that the immobilized alpha-amylase entrapped by the UV-curing coating retains adequate enzymatic activity and can be reused more than 50 times under certain experimental conditions.

Molecular cloning, expression, and functional characterization of a novel member of the CD38 family of ADP-ribosyl cyclases.

We report the molecular cloning and functional characterization of a novel member of the CD38 family of cyclic ADP-ribose (cADPr)-generating cyclases. We cloned a cDNA insert that encoded a 298-amino-acid-long protein (M(w) approximately 39 kDa). The predicted protein displayed 69, 61, and 58% similarity, respectively, to mouse, rat, and human CD38. Rabbit CD38 was also 28% homologous to Aplysia ADP-ribosyl cyclase and leukocyte CD157 (another ADP-ribosyl cyclase); the three cyclases shared 10 cysteine and 2 adjacent proline residues. We then transfected CD38-negative NIH3T3 cells with cDNA encoding a CD38-EGFP fusion protein. Epifluorescence microscopy showed intense EGFP fluorescence confirming CD38 expression. We finally confirmed the ADP-ribosyl cyclase activity of the expressed CD38 by measuring its ability to catalyze the cyclization of the nicotinamide adenine dinucleotide (NAD(+)) surrogate, NGD(+), to its fluorescent nonhydrolyzable derivative, cGDPr.

Catalytic enantioselective aza-Diels-alder reactions of imines--an approach to optically active nonproteinogenic alpha-amino acids

A catalytic enantioselective aza-Diels-Alder reaction of imines has been developed. The reaction of N-tosyl alpha-imino ester with different dienes including activated, non-activated, cyclic, and acyclic dienes has been investigated in the presence of various chiral Lewis acids. A series of phosphino-oxazoline ligands have been synthesized and evaluated for the reaction. It was found that the combination of phosphino-oxazoline ligands with copper(I) salts gives the best results for the activated dienes, while BINAP-copper(I) complexes are good catalysts for all the dienes studied. In the case of activated acyclic dienes the aza-Diels-Alder products can be obtained in higher than 80% isolated yield and 96% ee, while for the unactivated cyclic dienes the exo diastereomer is formed as the major product in up to 95 % ee. For an activated cyclic conjugated diene, 2-trimethylsilyloxy-1,3-cyclohexadiene, the reaction proceeds as a Mannich-type addition reaction giving optically active gamma-oxo alpha-amino acid derivatives in good yields and up to 96% ee. The reaction of an unactivated acyclic diene, 2,3-dimethyl-1,3-butadiene, with the N-tosyl alpha-imino ester gives both the aza-Diels-Alder and aza-ene products, in a ratio of 9:1 favoring the aza-Diels-Alder product. Furthermore, a series of different imines have been synthesized and investigated as possible substrates for the present catalytic enantioselective aza-Diels-Alder reaction in order to obtain mechanistic insight. All imines studied gave moderate to high ee. Particularly, the reaction of the N-phenyl and N-p-methoxyphenyl substituted glyoxylate imines with Danishefsky's diene proceeded well affording the corresponding aza-Diels-Alder product in high yield with up to 91% ee at room temperature. The present catalytic enantioselective reaction of imines provided an effective route to optically active nonproteinogenic alpha-amino acids. The products of the catalytic enantioselective aza-Diels-Alder reaction of the cyclic dienes can be used for the preparation of key compounds such as natural products and compounds of pharmaceutical interest. The absolute configurations of five products have been solved by X-ray structural analysis, and it is found that the absolute configuration of the aza-Diels-Alder adduct is dependent on the substituent on the imine nitrogen atom. It turned out that the N-tosyl glyoxylate imine and N-p-methoxyphenyl glyoxylate imine give the aza-Diels-Alder adduct with opposite absolute configuration using the same enantiomer of the catalyst. On the basis of the results the mechanistic aspects for the reactions are discussed.

Reaction of reducing hydroxyl radical adducts of pyrimidine nucleotides with riboflavin and flavin adenine dinucleotide (FAD) via electron transfer: a pulse radiolysis study.

Using the techniques of pulse radiolysis with time-resolved spectrophotometric detection, it has been demonstrated that the interaction of reducing OH radical adducts of dCMP, TMP and UMP with riboflavin (RF) and flavin adenine dinucleotide (FAD) does proceed via an electron transfer reaction. From buildup kinetics of radical species, the rate constants of electron transfer from reducing OH adducts of pyrimidines to RF and FAD have been determined, respectively. It could be deduced that RF and FAD would reduce the probability of repair of the damaged DNA in the presence of enzymes and antioxidants, accordingly RF and FAD might have a radiosensitization effect on DNA damage.
