[Expression of fused luxAB gene of bacterial luciferase in liver carcinoma cells].

OBJECTIVE: This work was done to look into the expression of fused luxAB gene of bacterial luciferase as a reporter gene in liver carcinoma cells. METHODS: The mammalian expression vector pcDNA3-luxAB gene, constructed by the fusion of luxA and luxB genes, were amplified in the polymerase chain reaction (PCR) directed site mutagenesis from the vibrio harveyi 1600 strain and inserted into the plasmid of pcDNA3. This analysis was to confirm the fused luxAB gene and the positive clones obtained by the G418 resistant stable selection and transfected by lipofectin, when they were confirmed by the PCR. The growth curve of cell population and luminescence of bacterial luciferase was obtained through MTT and bioluminescence, respectively. RESULTS: The fused luxAB gene, being a monocistron, completely agreed with the design. No significant difference in the growth curves of cell population was observed between the transfected cells and untransfected ones. The recombinant plasmid was likely to be expressed in a stable fashion in the BEL7402 cell. Meanwhile, the maximum cellular level in terms of vitro bioluminescent strength reached the point of (8.71 +/- 1.21) mV/40 micrograms protein. CONCLUSION: Bacterial luciferase luxAB gene may become the first choice as a new, sensitive and non-invasive reporter gene in the research on liver cancer cells.

[Effects of copper mine tailings on growth and development and physiological function of wheat].

Through pot culture experiments, the effects of Tonling copper mine tailings on growth and development and physiological function of wheat were investigated in this paper. The results showed that the seeds of wheat could germinate in copper mine tailings, but seed germination percentage and germination rate were lower than that in control. High ratio tailings decreased seedling height. Tailings decreased chlorophyll contents, but didn't effect on chlorophyll a/b. Compared with control, tailings significantly inhibited root growth and decreased root number. Tailings increased root and shoot biomass and significantly increased root/shoot ratio of wheat. The permeability of cell membrane and the free proline content in leaves of wheat increased with the increase of the ratio of tailings, they were relative tolerance index of wheat to copper tailings stress.

[Study on variation and interrelations of characteristics at ginseng germplasms].

The variation and interrelations of some characteristics of Panax ginseng germplasms(lines), which were five selfmating generations of some single plants were studied. Methods of multivariate analysis, such as correlation, regression, or clustering, and coefficient of variation were used. The results were of benefit to the breeding and standardized planting of Panax ginseng.

Prospective survey on neurosurgical intensive care for patients with severe head injury.

OBJECTIVE: To prospectively compare the clinical outcome of intensive care therapy (ICT) with that of conventional care therapy (CCT) in severe head injured patients. METHODS: Patients with severe head injury were assigned randomly into Group ICT and Group CCT, 100 patients in each group. Patients in Group ICT accepted intensive care therapy in neurosurgical intensive care (NIC) unit for the first 2 weeks after admission, while patients in Group CCT accepted conventional care therapy in ordinary ward. The outcomes were evaluated 3 months after injury. RESULTS: There was a significant increase in good recovery (54%) (chi(2)=4.43, P<0.05) and significant decrease of death (25%) (chi(2)=4.50, P<0.05) in Group ICT compared to 39% and 39% in Group CCT respectively. The differences were also confirmed statistically in the following aspects: the patients under 50 years with good recovery pronounced a number increase (chi(2)=7.54, P<0.01), while the mortality in the same range of age was decreased in Group ICT (chi(2)=5.28, P<0.05). The mortality was reduced significantly in patients with GCS for 6-8 on admission (chi(2)=8.47, P<0.01) and in patients with the level of brain stem injured below mesencephalon (chi(2)=4.15, P<0.05). ICT would improve the outcome in patients undergoing conservative therapy only (chi(2)=13.13, P<0.01). CONCLUSIONS: NIC plays an important role in assessing the neurological state, guiding management, evaluating curative effect and estimating the outcome.

[Phalloplasty using an abdominal flap with two sources of blood supply (16 cases reports)].

OBJECTIVE: To improve the survival of a reconstructed penis and reduce injury of the donor site at the abdominal wall. METHODS: Sixteen cases of phalloplasty were performed using an abdominal flap based on two sources of blood supply i.e. the cutaneous branches of the inferior epigastric blood vessel and the superficial epigastric blood vessel. RESULTS: All of the operations were successful. CONCLUSIONS: With abundant blood supply and high survival rate and the advantage of easy transposition, the flap is a good option for phalloplasty.

[Evaluation the pathological diagnostic values of koilocyte in condyloma acuminatum].

OBJECTIVE: To evaluate the histological diagnostic values of koilocyte in histopathologic diagnosis of condyloma acuminatum. METHODS: Koilocyte was served as diagnostic criteria to scan 2 063 biopsies and compared the results with "goldan standard" of histopathologic diagnosis of condyloma acuminatum. RESULTS: Using koilocyte as diagnostic standard to diagnose condyloma acuminatum, the sensitivity and specificity were 77.93% and 95.26%, with positive predictive value 97.25%, negative predictive value 66.70%, positive likelihood rate 81.81, negative likelihood rate 23.17, under diagnostic rate 22.07%, misdiagnostic rate 4.74%. CONCLUSION: The koilocyte was proved to be one of classical histopathological morphologic features of human papillomavirus infection and other virus infection which served one of the most important histopathological markers for diagnosis of condyloma acuminatum, but not the only one.

[Study on the risk factors of psoriasis].

OBJECTIVE: To investigate psoriasis patients with family history and non-family history on their age and season of onset, disease severity and predisposing factors. METHODS: Two hundred and twenty psoriasis patients with family history and 547 psoriasis patients without, together with 647 normal controls have been investigated through questionnaires. EPI info 6.0 method was applied for data analysis. RESULTS: (1) In 220 psoriasis patients with family histories, morbidity rates of the first and second-degree relatives were 16.98% and 2.7% respectively. (2) Both mean age and age of onset were younger in females than that in males with family histories (t = 1.715 0, P < 0.05). The mean age of onset was younger in males with family histories than those without. However, there was no significant difference found in females (t = 0.739 2, P > 0.05). The mean age of onset seemed to be younger in females with family histories than that without, but with no significant statistical difference. (3) There were no significant differences in the age and season of onset, disease severity, and predisposing factors between groups with family histories and without. However, environment factors seemed to influence not only psoriasis patients with family histories but also those without, which showed significant difference (chi(2) = 0.23, P < 0.05). CONCLUSIONS: There was no significant difference in the following aspects as season of onset, disease severity and predisposing factors between the ones with family history and those without, except the age of onset and age of average. The results showed that genetic factor might play an important role in psoriasis patients with family history. It also demonstrated that psoriasis is a multifactor hereditary disease.

[Modulation of the activity of human alpha1 (I) procollagen gene promoter by basic fibroblast growth factor (bFGF)].

OBJECTIVE: To investigate the modulating effect of bFGF on the proliferation of human cutaneous fibroblasts and on the activity of human alpha1 (I) procollagen gene promoter. METHODS: Human cutaneous fibroblasts were cultured and subcultured by tissue block culture technique. The influence of different concentrations of bFGF on the proliferation of fibroblasts was determined by ELISA method with BrdU incorporated into fibroblast DNA. Three plasmids containing various lengths of 5prime prime or minute flank sequences of human alpha1 (I) procollagen gene and CAT as reporter gene were constructed and transfected into the fibroblasts by FuGENE transfection reagent. The quantitative expression of the fibroblast CAT was determined by ELISA after treatment with bFGF. RESULTS: After 24 hours of treatment of the fibroblasts by serial concentrations (0.25 ng/ml similar 64.00 ng/ml) of bFGF in DMEM containing 2% (v/v) or 10% (v/v) FCS, the BrdU incorporation into DNA was determined. The proliferating rate of the fibroblasts differed significantly from each other in all the groups (P < 0.05). After the fibroblasts were transfected with the three plasmids and treated thereafter by 4 ng/ml and 16 ng/ml of bFGF for 24 hours, the relative CAT expression values were determined. It indicated that the expression value was evidently different between bFGF processing and control groups (P < 0.05). CONCLUSION: bFGF might inhibit the proliferation of human cutaneous fibroblasts and exert negative regulating effect on the human alpha1 (I) procollagen gene promoter sequence in dose -- dependent pattern.

[Invasive rhinocerebral aspergillosis occurred during myelosuppressive phase after chemotherapy: a case report and literature review].

OBJECTIVE: To report a case of invasive rhinocerebral aspergillosis (IRA) during myelosuppressive phase after chemotherapy and the treatment outcome. METHODS: A patient with acute non-lymphoblastic leukemia (M(2a)) occurred IRA during myelosuppressive phase after chemotherapy and was treated with amphotericin B (AmB, including AmB-L), garlicin, 5-flucytosine (5-FC), fluconazole, itraconazole, ketoconazole, econazole and griseofulvin with twice of cerebrotomy lesion resections and cerebral decompressions. RESULTS: Though the survival time of this patient is a little longer than the others, she finally died of uncontrolled infection. CONCLUSION: For most of the IRA, there remains no effective treatment. The best treatment is AmB in combination with surgical excision.

[Investigation on method speliality in identifying Fructus Crataegi with vitexin rhamnoside].

The methanol extract of Fructus Crataegi was purified on macro-reticular resins(model DM130)-polyamide column. Vitexin rhamnoside(I) was identified from Fructus Crataegi by TLC on polyamide layer sheets. The false and the confused species of Fructus Crataegi and some traditional Chinese herbal medicines which contain vitexin were studied by the same method. The results indicated that compound I was special constitute of Fructus Crataegi and the identification method of Fructus Crataegi with I was special.

[The influence of surface roughness on bacteria adhesion on titanium].

OBJECTIVE: To investigate the correlation between surface roughness and bacteria adhesion. METHODS: Surface roughness of four abutments were determined by Talysurf/6p-120 topographer. Ra, Rz, Rp, Rv, Ry and Sm values were gathered. Commercial pure titanium and titanium alloy discs, S. mutans and A. viscosus were used in adhesion experiment. RESULTS: Ra values (in microns) of the tested abutments were: 0.1556, 0.2073, 0.6976, and 0.3811. No statistical difference was observed in bacterial quantity when Ra < 0.4528 microns and Rz < 3.3882 microns in adhesion experiment. CONCLUSIONS: The surface roughness of Ra < 0.4 micron and Rz < 3.4 microns is recommended for implant abutment.

A Web-based interactive education programme on vaccination for public health officials.

Health and Welfare Training Center of Korean National Institute of Health (KNIH) has developed various education curricula for officials who are involved in the public health or welfare sectors. Still almost all education programmes are off-line based. In off-line settings, both the lecturers and the students should come to NIH from their counties to join the education programmes, and it is impossible for the students to review or to re-practice the education contents. We developed a prototype on-line setting education programmes to provide a more convenient and more effective service to the officials. This web-based interactive education programme consisted of the specific objectives on vaccination for the public health officials who were working for the national immunization programmes. It became a prototype of web-based education and was evaluated by the participants in April, 2001. From the evaluation, we improved the contents and tried to develop more subjects for the officials' training.

[Comparison of the effect of fluoride and non-fluoride dentifrices on enamel demineralization and remineralization in vitro].

OBJECTIVE: The purpose of the experiment is to investigate the effect of fluoride dentifrices in China on the intact and artificial caries-like bovine enamels. METHODS: The artificial caries-like lesions were created by dipping the teeth into an acid buffered solution. Both the bovine intact enamels and the artificial caries-like enamels were assigned tooth brushing 20 times every half an hour with 3 kinds of fluoride dentifrices and one kind of non-fluoride dentifrices respectively. The amount of calcium dissolved by lactic acid was determined as the susceptibility of demineralization by atomic adsorption spectrometry (AAS). RESULTS: The group treated with fluoride dentifrice showed significantly less demineralization than the non-fluoride group (P < 0.02). The effect of inhibition of acid solubility in artificial caries-like enamel after tooth brushing is significantly greater than that in intact enamel. CONCLUSION: Fluoride dentifrices in this experiment can prevent both intact enamel and artificial caries-like enamel from acid solubility after tooth brushing, and it seems that the effect of the fluoride dentifrices on the redemineralization is greater than the reduction of enamel solubility.

[A sinorhizobium fredll strain that effectively nodulate Medicago sativa].

Sinorhizobium fredii 042BS was isolated from root nodules of alfalfa (Medicago sativa) from Xinjiang Region. Nodulation experiments showed that both soybean and alfalfa were nodulated by 042BS effectively. The 16S rDNA PCR-RFLP analysis was carried out by four restriction endonucleases, and the restriction maps of strain 042BS were identical with those of S. fredii USDA205. The DNA G + C mol% of strain 042BS was 60.0. The DNA homology between 042BS and S. fredii USDA205 and USDA191 were 84.9% and 89.6%, respectively. To prove the capability of 042BS to nodulate both soybean and alfalfa, constitutively expressed green fluorescence protein gene(gfp) was introduced to 042BS, and the recombinant strain 042BSG was obtained. The reisolates from nodules of the soybean and alfalfa inoculated with 042BSG were observed using the confocal laser-scanning microscope, and the expressions of gfp were detected, respectively. 042BS showed various nodulation capacities with different alfalfa cultivars used.

[Subcloning and sequencing of DNA fragment related to salt tolerance in Sinorhizobium meliloti 042B].

A 4 kb ClaI DNA fragment related to salt tolerance from S. meliloti 042B was digested by HindIII down 2.4 kb fragment, and a 1.6 kb ClaII-HindIII fragment was retained on plasmid pML122. Then, the 2.4 kb DNA fragment was ligated with plasmid pBBR1-MCS2, and the recombinant plasmid was transformed to E. coli DH5 alpha, and transformant GS2 was obtained. Three-parental mating experiments were carried out with transformant GS2 as donor, salt sensitive strains GZ17 as recipient and pRK2013 as helper plasmid, then the transconjugant GG2 was selected on FY plates containing kanamycin and 0.4 mol/L NaCl. The remaining DNA fragment was self ligated with pML122 and then transformed into E. coli S17-1 and transformat GS0 was obtained. Two-parental mating experiment was carried out with transformant GS0 as donor and salt sensitive strain GZ17 as recipient, but no transconjugant was obtained on the FY plates. Then, the 2.4 kb HindIII DNA fragment was ligated into sequencing vector pGEM-7Zf(+) for sequencing. The result of sequencing and analysis showed that the 2.4 kb DNA fragment contained three ORFs. According to the result of sequencing, further subcloning was conducted and 1.9 kb HindIII-Sac II DNA fragment related to salt tolerance was obtained.

[Introduction of Vitreoscilla hemoglobin gene in a recombinant E. coli for PHB production].

Vitreoscilla hemoglobin gene (vgb) was introduced into the recombinant Escherichia coli VG1(pTU14) for production of poly-beta-hydroxybutyrate (PHB), in order to enhance the oxygen uptake of strain from molecular level and to solve the problem of oxygen limitation in process of fermentation. The carbon monoxide difference spectra analysis of Vitreoscilla hemoglobin (VHb) showed that vgb could be successfully expressed in VG1 (pTU14), and this expression was under the regulation of dissolved oxygen (DO). The higher amounts of VHb could be got correspond to the lower level of DO. Because of the introduction of vgb, cell growth and PHB accumulation of VG1 (pTU14) were all strongly promoted during the process of batch culture.

[Irradiation of 235 uranium on the growth, behavior and some biochemical changes of brain in neonatal rats].

The effects of enriched uranium on growth, behavior and biochemical changes of brain in neonatal Wistar rats was studied. Neonatal rats were irradiated with a single injection of 2 microliters enriched uranium (containing 0, 1, 5 and 10 micrograms of 235U) into left lateral ventricle of brain at the first day (< 24 h) after birth. The growth and neurobehavior development of the rats was delay and abnormal. The microautoradiographic tracing technique showed that the radionuclide was mainly accumulated in the nucleus, also appeared in the cytoplasm of cells and between cells. The activity of superoxide dismutase (SOD) and the level of endothelin (ET) of cerebellum, cerebral cortex, hippocampus and diencephalons of brain after exposure to alpha irradiation were examined with radioimmunoassay. The data indicated that SOD and ET was elevated by low dose irradiation and distinctly inhibited by high dose irradiation. It was concluded that alpha irradiation could have damages on the developing brain of neonatal rats characterized by changing the sensibility, fragility and compensation of nerve cells.

[Study on immunomodulating mechanism of the active fraction of liuwei dihuang decoction].

OBJECTIVE: To investigate the immunomodulating mechanism of 3A, an active fraction extracted from the ethanol-soluble portion of Liuwei Dihuang Decoction (LWDHD). METHODS: Influence of 3A on T-lymphocyte subsets, IgG production of B cells, interferon-gamma (IFN-gamma) mRNA expression in cyclophosphamide (Cy)-treated mice, tumor bearing mice and senescence accelerated mouse-prone/8 (SAMP8) were assessed in vivo and in vitro. RESULTS: Oral administration of 3A could elevate the decreased T-lymphocyte subsets in all model mice, it also could promote the IgG secretion of splenic cells in SAMP8 and increase IFN-gamma mRNA expression in Cy-treated mice. CONCLUSION: The immunomodulating effect of 3A is related with its effects in regulating ratio of T and B cells or in improving their functions.
