Immunodetection of ralA and ralB GTP-binding proteins in various rat tissues and platelets.

Polyclonal antibodies were generated against a synthetic peptide corresponding to the C-terminal (amino acids 192-204) region of ralA and ralB GTP-binding proteins. The ralA and ralB antibodies recognized a 27 kDa protein in the human platelet particulate fraction. Incubation of ralA antibodies with ralB immunizing peptide and ralB antibodies with ralA immunizing peptide prior to Western blotting did not abolish the ability of antibodies to recognize the 27 kDa protein in human platelet particulate fraction. However, when antibodies were incubated with the respective immunizing peptide prior to Western blotting, the 27 kDa human platelet protein was no longer recognized by the antibodies. Incubation of nitrocellulose blots containing polypeptides separated using SDS-PAGE with [alpha-32P]GTP demonstrated the presence of GTP-binding proteins of molecular mass between 23-27 kDa in rat platelets and the various tissues tested. Analysis using subtype specific antibodies demonstrated that both ralA and ralB GTP-binding proteins were expressed in rat platelets and the various tissues tested. The protein recognized by the ralA and ralB antibodies in rat tissues and platelets had mobility on SDS-PAGE identical to that of the human platelet ral protein. Varying amounts of these proteins were detected in all the tissues tested except white muscle which contained very low level of ralB protein. The widespread distribution of ralA and ralB GTP-binding proteins suggests that they may participate in a common pathway in mammalian cells and tissues.

The expression of functional postsynaptic alpha2-adrenoceptors in the corpus cavernosum smooth muscle.

1. The purpose of this study was to determine if corpus cavernosum smooth muscle expresses functional postsynaptic alpha2-adrenoceptors (AR). 2. The alpha2-adrenoceptor agonist UK 14,304 elicited concentration-dependent contractions in rabbit corpus cavernosum smooth muscle (CCSM). The half-maximal response occurred at 0.32+/-0.03 microM and the maximum contraction at 10 microM UK 14,304. 3. Pretreatment of CCSM strips with selective alpha2-adrenoceptor antagonists, rauwolscine and RS-15385, produced rightward shifts in the dose-response curves to UK 14,304 (pA2 values 7.1 and 8.5, respectively). In contrast, these antagonists did not alter contraction induced by the alpha1-adrenoceptor agonist phenylephrine (PE) or oxymetazoline. UK 14,304-induced contractions were also inhibited by prazosin (pA2 = 9.08). 4. UK 14,304-induced contractions, unlike those to PE, were highly dependent on the presence of extracellular Ca2+. 5. [3H]-rauwolscine bound to CCSM membranes with high affinity (Kd = 1.5 nM). [3H]-rauwolscine binding was displaced by unlabelled rauwolscine, RS-15385, UK 14,304 and prazosin, but not by PE. 6. UK 14,304 inhibited forskolin and prostaglandin E1 (PGE1)-induced increases in intracellular cyclic AMP concentration in primary cultures of rabbit CCSM cells. 7. These results demonstrate that CCSM expresses Gi-coupled postsynaptic alpha2-adrenoceptors, and activation of these receptors causes contraction of trabecular smooth muscle.

Black tea and mammary gland carcinogenesis by 7,12-dimethylbenz[a]anthracene in rats fed control or high fat diets.

Epidemiological studies suggest that tea may reduce cancer risk, and in laboratory rodents, chemopreventive effects of tea or purified extracts of tea have been demonstrated in lung, gastrointestinal tract and skin. There is some evidence of chemoprevention by tea in the mammary gland, but the data are not conclusive. In order to evaluate more fully the possible influence of black tea on 7,12-dimethylbenz[a]anthracene (DMBA)-induced mammary gland tumors in the female S-D (Sprague-Dawley) rat, three large studies were performed: experiment 1, tumorigenesis in rats fed AIN-76A diet and given 25 mg/kg DMBA and 1.25 or 2.5% whole tea extract or water to drink; experiment 2, tumorigenesis in rats given 15 mg/kg DMBA and the same diet and fluids as in experiment 1; experiment 3, tumorigenesis in rats fed control or HF (high fat, corn oil) diet and given 15 mg/kg DMBA and 2% tea or water to drink. Tea was given throughout the experiment; DMBA was given by gastric gavage at 8 weeks of age. There was no consistent effect of tea on tumorigenesis in rats fed AIN-76A diet; there was, however, evidence in experiment 3 of a reduction of tumorigenesis by tea in rats fed the HF diet. In experiment 3, rats fed the HF diet and given water showed the expected increase in tumor burden (number and weight) compared with rats fed control diet. However, rats fed the HF diet and given 2% tea showed no increase in tumor burden; their tumor burden was significantly lower than in rats fed the HF diet and given water (P < 0.01) and was not different from rats fed control diet and given water or tea. In addition, in experiment 3, the number of malignant tumors per tumor-bearing rat was increased by the HF diet in water-drinking rats (P < 0.01) but not in tea-drinking rats. Therefore, it appears that tea partially blocked the promotion of DMBA-induced mammary tumorigenesis by the HF diet.

Modelling human immunodeficiency virus infection and acquired immune deficiency syndrome cases in Scotland: data sources, prior information and Bayesian estimation.

"The paper describes the methodology developed to carry out predictions of the acquired immune deficiency syndrome (AIDS) epidemic in Scotland. Information on the human immunodeficiency virus (HIV) epidemic comes from formal case reports of AIDS cases and HIV positive tests, reports from surveillance schemes and from special studies. These sources of information, up to the end of 1994, are reviewed.... A simple model of the HIV epidemic in Scotland is proposed and the information is summarized in terms of this model. Bayesian methodology, using Markov chain Monte Carlo methods, is described and used to predict future cases of AIDS in Scotland and people who will be living with AIDS in the years 1995-1999."

Prevalence of human melioidosis on Hainan Island in China.

Eight human cases of melioidosis were diagnosed at Hainan People's Hospital over a period of one year. Four of the cases were of septicemia form, while the rest were of chronic form with abscesses in different organs. All the isolates were intrinsically resistant to cefazolin, cefuroxime, and gentamicin, while being rather sensitive to ampicillin/sulbactam, ticarcillin/clavulanic acid, and imipenem. A serological survey of exotoxin antibodies at different farms showed that Xinglong was a farm seriously devastated by Burkholderia pseudomallei, while the mountainous farm of Licai had the lowest prevalence (P< 0.01). From the results of serological survey and melioidosis case distribution, it could be clearly seen that melioidosis predominantly exists in coastal plain regions around this island, where the altitude above sea-level is below 100 m, the annual rainfall is up to 2,300 mm and a rather warm climate in coldest months of December and January.

Consumer perspectives on genetic testing, research and services for ethnoculturally diverse populations.

A panel of individuals from diverse ethnocultural backgrounds and representing a variety of genetic disorders presented their consumer perspectives on genetic programs, testing and services. Their remarks emphasized how misunderstanding and miscommunication between health care professionals and many of the populations for whom they provide services can lead to unfilled genetic service needs. Panelists recommended that health care professionals become more aware and knowledgeable about the diversity of customs, beliefs and cultures of those receiving their services. Only by building a foundation of trust and mutual respect will genetic testing, research and services become more accessible to individuals from diverse populations, their families and their communities.

Radioulnar load-sharing in the forearm. A study in cadavera.

Custom-designed miniature load-cells were inserted into the distal end of the ulna and the proximal end of the radius in ten fresh-frozen forearms from cadavera. The forces transmitted through the bones at these sites were measured under 134 newtons of constant axial load that was applied through the metacarpals as the forearm was rotated from 60 degrees of supination to 60 degrees of pronation. The simultaneous measurements of these forces allowed the calculation of radioulnar load-sharing at the wrist and the elbow as well as the calculation of the amount of force that was transferred from the radius to the ulna through the interosseous membrane. With the elbow in valgus alignment (that is, with contact between the radial head and the capitellum), the main pathway for load transmission through the forearm was direct axial loading of the radius; measurements from both load-cells were unaffected by the angle of elbow flexion. When the forearm was in neutral rotation, the mean force in the distal end of the ulna averaged 2.8 per cent of the load applied to the wrist and the mean force in the proximal end of the ulna averaged 11.8 per cent; this indicated that only a small amount of tension developed in the interosseous membrane. With the elbow in varus alignment (that is, with no contact between the radial head and the capitellum), load was transmitted through the forearm by a transfer of force from the radius to the ulna through the interosseous membrane. When the forearm was in neutral rotation, the force in the distal end of the ulna averaged 7.0 per cent of the load applied to the wrist and the force in the proximal end of the ulna averaged 93.0 per cent; the force through the interosseous membrane decreased with supination of the forearm. Testing with the elbow in valgus alignment and shortening of the distal end of the radius in two-millimeter increments produced corresponding increases in force in the distal end of the ulna and decreases in force in the radial head. The forces through the interosseous membrane remained low after each amount of radial shortening.

Construction of engineering host by non-random method in Saccharomyces cerevisiae.

A yeast engineering host, GJ30, was constructed by a non-random method. GJ30 derivated from wild type yeast have leu2- and ura3- deficients which can be used as selection markers. The PCR analysis showed that the homologous recombinations occurred in the leu2 and ura3 locus of GJ30, respectively. The biological properties (cellular growth density, stability and efficiency for gene expression) of GJ30 were compared with other several yeast stains. The results showed that GJ30 is a suitable host strain for expression of foreign genes.

[Changes of blood supply from portal system after transcatheter arterial chemoembolization in huge hepatocellular carcinoma].

OBJECTIVE: Blood supply changes of the portal venous system to the tumor mass before and after transcatheterarterial chemoembolization(TACE) in a series of 22 patients with huge hepatocellular carcinoma were monitored to study the necessity of intervention via portal vein. METHODS: Twenty-two selected patients with huge hepatocellular carcinoma were enrolled in the study. Elscint double helical CT was used to scan liver during the maximum concentration of contrast medium in portal venous system. CT Angiography and MIP(Maximum Intensity Projection) were used to image formation of the portal venous system. RESULTS: The technique of scan and image formation were excellent to evaluate the attribution of portal venous blood supply to HCC. The portal venous blood supply was mainly the tumor periphery distribution (P < 0.01). After treatment of TACE, the portal venous blood supply to tumor periphery increased significantly (P < 0.05), and some did appear signs of enriched blood supply around the tumor. Negative correlation between the degree of reduction in tumor and portal venous blood supply to the tumor periphery before TACE was evident. CONCLUSION: The attribution of portal venous blood supply to HCC was in the tumor periphery, and regression of tumor after TACE was affected by portal venous blood supply to the tumor, thus it is reasonable to suggest interventional therapy via portal vein.

[Study on the power of O/E method].

This paper addresses the methodology of calculating the power of O/E method and introduces the relevant algorithm for estimating the sample size. Based on the principles of hyporthesis testing and Poisson distribution, the power can be worked out for given sample size, baseline rate(pi 0) and the factor of increasing (gamma). The power increases with the increase in sample size, pi 0 and gamma. In shape the power is not a continuous smooth line but a zigzag line. The method illustrated in this paper can be also used for estimaging the sample size of studies on the intervention of diseases.

[An endometrium morphometry study on ovariectomized rats subjected to nilestriol and estradiol replalcement therapies].

This study was designed to evaluate the effect of estrogen on rat endometrium and compare the results of Nilestriol and Estradiol replacement therapies for preventing osteoporosis in ovariectomized rats. Forty 4-month old SD female rats were randomly divided into four groups, namely sham operation, bilateral ovariectomy, ovariectomy plus supplementary ethinyl estradiol (0.2 microgram/100 g B. W. qd x 5), and ovariectomy plus supplementary Nilestriol (0.15 mg/100 g B. W. once a week). Computerized image analyzer was used to evaluate the endomitrial pathological changes 10 weeks later. The results showed an atrophied endometrium in ovariectomized group, a slightly proliferative endometrium in Nilestriol treated group, and a markedly proliferative endometrium with local atypical hyperplasia in estradiol treated group. The difference between group 3 and group 4 in morphometry was significant (P < 0.5). These suggest the relative safety of Nilestriol replacement therapy for preventing menopausal osteoporosis.

[A longitudinal study on growth model and velocity of term small for gestation age].

To probe into the vegetal pattern and find out the key period of promoting normal growth of term small for gestational age (TSGA), a longitudinal study on growth model and velocity of TSGA was conducted from Jan 1993 to June 1997. The body weight and length of 150 children of TSGA (57 boys, 93 girls) and 152 children as controls (58 boys, 94 girls) were measured from the 1st month to 36th month. The growth model was analysed by multilevel models. The result showed that there was a difference between the growth models of TSGA and controls. During 8 and a half months (center month), the weight, length of controls were 1.19 kg, 3.46 cm greater than those of TSGA; the weight, length of boys were 0.34 kg, 1.08 cm more than those of girls respectively. The growth velocity of TSGA was similar to that of controls. The maximal growth velocity was observed during the first 6 monthes after birth. The order of growth velocity from high to low was 1 mo. > 2 mo. > 3 mo. in length and weight, showing that children of TSGA have their own growth model. Their growth velocity should be monitored attentively. If low velocity appears one should search for the cause and adopt apropriate measures to ensure their growth in accordance with their "own track".

[Follicular fluid from patients with endometriosis decreases the binding of human sperm to zona pellucida in hemizona assay].

OBJECTIVE: To evaluate the effects of follicular fluid (FF) from women with endometriosis and tubal factor on sperm binding to zona pellucida under hemizona assay(HZA). METHODS: Follicular fluid was collected at IVF from patients with endometriosis and without endometriosis (tubal factor infertility). The HZA was used to study the effect of FF from patients with endometriosis and tubal factor on sperm binding to zona pellucida using media (EBSS/BSA) as control. RESULTS: The mean number of sperm binding to zona in endometriosis specimens and tubal factor infertility was significantly fewer than the number treated by culture medium (91 +/- 21 vs 239 +/- 35, P < 0.001; 109 +/- 22 vs 211 +/- 42, P < 0.005). The hemizona index was lower when the incubation was performed with FF from endometriosis patients compared with sperm incubated in FF from tubal factor patients (36 +/- 4 vs 54 +/- 4, P < 0.005). CONCLUSIONS: The inhibiting effect of FF from endometriosis patients is stronger than those without endometriosis. The follicular fluid of patients with endometriosis could contribute to infertility through impairment of gamete interaction.

[Telomerase activity in gastric cancer and premalignant lesions: determination and its clinical significance].

OBJECTIVE: To evaluate the role of telomerase activity in the development and progression of gastric cancer (GC) and if knowedge of telomerase activity may be an indicator for the early diagnosis and poteneial treatment of GC. METHODS: Using PCR-based TRAP (telomeric repeats amplification protocol) assay, we examined telomerase activity in 176 samples of gastric mucosa. Meanwhile, some tumor tissues were also analyzed by serial dilution. RESULTS: Telomerase activity detected in chronic atrophy gastritis (CAG), intestinal metaplasia (IM); dysplasia (Dys); and gastric cancer (GC) was 24.6% (14/57), 38.9% (7/18), 37.5% (3/8), and 92.3% (60/65) respectively; which was significantly higher than that in normal tissues (NT) (P < 0.01-0.05). Telomerase activity in group of GC was also significantly higher than that in group of CAG, IM and Dys (P < 0.01). Frequencye and levels of telomerase activity were not associated with patients' sex, tumor location, size gross type, depth of invasion, lymph node metastases, differentiation and clinical stage. CONCLUSION: Telomerase activity is detected not only in GC tissues, but also in some premalignant lesions or diseases.

[The pre-experiment on space crystallization of pig heart mitochondrial F1-ATPase].

ATP synthase (H(+)-ATPase) plays a key role in oxidative phosphorylation. Its catalytic moiety F1-ATPase is under intense structural study because of different crystallographic results and diverse hypotheses of its catalytic mechanism. To obtain high quality crystals, F1-ATPase sample is going to be carried to outer space by recovable satellite for crystal growth under microgravity. And to make it suitable for space crystal growth, F1-ATPase was purified from pig heart and the conditions were optimized for its maintenance, transportation, and crystallization. By using hanging drop vapor diffusion method and sitting drop vapor diffusion method, the crystals were obtained within 5 d and with a size of 0.1 mm x 0.1 mm x 0.l mm on the 7th day. The reproducibility of these experiments is also good.

[RP-HPLC determination of water-soluble active constituents and oleanolic acid in the fruits of Ligustrum lucidum Ait. collected from various areas].

The contents of p-hydroxyphenethyl-beta-D-glucoside, specnuezhenide and oleanolic acid in the fruits of Ligustrum lucidum collected from various areas in China were determined by reverse phase high performance liquid chromatography. The results could be used to evaluate the quality of the crude drug more comprehensively.

The puzzle of Wang Bing's life story.

Since Wang Bing made great contributions to the development of TCM, exploration on Wang Bing's life story is helpful to the analysis on the intention of his medical works. There are controversies on Wang's life story, some of which are still in debate. Through the elucidation of Wang's name, other names, sobriquet, dates of birth and death, official positons and scholarly lineage, this author holds that more relevant historical materials are needed so that the puzzle of his life story can be eventually revealed and uncovered.

[Maisonneuve fracture of the fibula: clinical and experimental study].

OBJECTIVE: To study the traumatic mechanism and clinical treatment of Maisonneuve fracture. METHOD: 19 of 25 patients (6 - 60 months, average 16.8 months) were followed up. 11 patients were operated on and 8 were treated conservatively. Of the 25 patients, 8 had MRI. Osteotomy and serial section of the interosseous membrane (IOM) on fresh cadaveric specimens were doc carried out to observe the diastasis on stress mortise view. RESULT: Functional rehabilitation of the ankle joint: in 11 operative cases, 8 showed good results and 3 fair. In 8 conservative cases, 3 showed good results, 2 fair and 2 poor. MRI: IOM was ruptured in a range 3 - 8 cm proximal to the tibial plafond in the 8 cases. No IOM rupture was found at the high fibular fracture level. Fresh cadaveric experiment: fixation of the medial side alone can not control the syndesmotic diastasis on the artificial Maisonneuve fracture model made on cadaveric specimen. CONCLUSION: The injury level of the IOM during Maisonneuve fracture may not be consistent with the high fibular fracture. The limited external rotation of the fibula during injury by the superior tibio-fibular syndesmosis comprise to the high level fibular fracture. ORIF with syndesmotic fixation is recommended for the treatment.
