Ozone impacts on allometry and root hydraulic conductance are not mediated by source limitation nor developmental age.

O(3)could reduce growth and carbohydrate allocation to roots by direct inhibition of photosynthesis and source strength. Alternatively, O(3) could reduce growth indirectly by inhibition of root hydraulic development through a primary lesion in carbohydrate translocation. Another alternative is that O(3) could slow the rate of plant development, only apparently altering carbohydrate allocation at a given plant age. Pima cotton (Gossypium barbadense L.) is used to address these possibilities, and four hypotheses were tested/ (1) O(3) exposure reduces leaf pools of soluble sugars; (2) pruning leaf area and reducing source strength to match that of O(3)-treated plants reproduces O(3)-effects; (3) pruning lower leaf area more closely reproduces O(3) effects than pruning upper leaf area; and (4) manipulating plant age and thereby plant size to match O(3)-treated plants reproduces O(3)-effects. All were falsified. Soluble sugars did not decline. Pruning upper and lower leaves and manipulating plant age all reduced biomass and leaf area similarly to O(3)-exposure, but neither reproduced O(3) effects on biomass allocation nor root function. It is concluded that O(3) induces an allometric shift in carbohydrate allocation that is not mediated by photosynthetic inhibition nor by alteration of developmental age. Effects of O(3) could be mediated by direct effects on phloem loading, with consequent inhibition of translocation to roots and root system development.

The impact of disease management on outcomes and cost of care: a study of low-income asthma patients.

An asthma disease management program designed specifically for low-income patients experiencing significant adverse events can improve health outcomes substantially, while lowering costs. The Virginia Health Outcomes Partnership aimed to help physicians in a fee-for-service primary care case management program manage asthma in Medicaid recipients. Approximately one-third of physicians treating asthma in an area designated as the intervention community volunteered to participate in training on disease management and communication skills. This large-scale study discovered that the rate of emergency visit claims for patients of participating physicians who received feedback reports dropped an average of 41% from the same quarter a year earlier, compared to only 18% for comparison community physicians. Although only a third of the intervention community physicians participated in the training, emergency visit rates for all intervention community physicians nonetheless declined by 6% relative to the comparison community among moderate-to-severe asthma patients when data for participating and nonparticipating physicians were combined. At the same time, the dispensing of some reliever drugs recommended for asthma increased 25% relative to the comparison community. A cost-effectiveness analysis projected direct savings to Medicaid of $3 to $4 for every incremental dollar spent providing disease management support to physicians. The results of this study demonstrate the potential this program offers, especially for Medicaid programs in other states that want to improve the care of their primary care case management networks and, at the same time, manage costs.

Clinical observations on 109 cases of vocal nodules treated with acupuncture and Chinese drugs.

The treatment of vocal nodules with the combined use of needling on Kaiyin Yihao Xue [symbol: see text] Kai Yin Point No. 1 for Voice-Regaining) and medicinal spray and tea made according to Xie Qiang's empirical prescription, obtained a total effective rate of 93.6%, obviously superior to that of the group treated with western medicine and the group simply treated by Chinese drugs (P < 0.01). This suggests that the combined use of acupuncture and Chinese drugs in the treatment of vocal nodules has better therapeutic effect and requires shorter course of treatment.

Hepatotropism of nonenveloped DNA virus in rhesus monkey infected by transfusion-transmitted virus.

OBJECTIVE: To study wether the nonenveloped DNA virus transmitted via blood transfusion is hepatotropic. METHODS: Total DNA was extracted from tissues of 5 experimentally infected Rhesus monkeys. A dot hybridization was done with virus double DNA strand probe or single antisense strand probe. RESULTS: Both single- and double-strand probes were hybridized with DNA of the liver, spleen, stomach, small intestine and colon. The virus was conformed present in most of all the organs when double-strand probe was used. The positive was noted only in the liver and small intestine when single-strand antisense probe was used, which showed that in liver and small intestine might have replicative intermediates of the virus. CONCLUSION: It suggests that nonenveloped DNA virus replicate in the liver and small intestine, so it might be hepatotropic.

Biphasic manner of melittin on isolated guinea pig atria.

AIM: To investigate the effect of melittin (Mel) on isolated guinea pig atria. METHODS: The effect of Mel on the contraction and heart rate of isolated guinea pig atria at different concentrations was determined. RESULTS: Mel at a lower concentration (0.1-0.8 mumol.L-1) enhanced the contraction of left atria in a concentration-dependent manner; but at a higher concentration (1.6-12.8 mumol.L-1) it exerted an inhibitory effect. At 0.1-30 mumol.L-1 it was found to increase heart rate of right atria. In addition, verapamil (Ver) 0.3 mumol.L-1 was found to depress the effect of Mel. CONCLUSION: Mel possesses a biphasic effect on left atria and a positive chronotropic effect on right atria. Its mechanism might be related with Ca2+ channel.

Blocking effects of heteroclitin D and gomisin J on L-type calcium channels in ventricular cells of guinea pig.

AIM: To study the effects of heteroclitin D (HD) and gomisin J (GJ), two lignans from Kadsura medicinal plants, on L-type calcium channels in ventricular cells of guinea pig. METHODS: The calcium currents were measured by whole-cell patch-clamp recording technique. RESULTS: HD 1 and 10 mumol/L decreased the L-type calcium current from (770 +/- 155) to (482 +/- 104) and (384 +/- 85) pA, respectively. GJ 10 mumol/L inhibited calcium current from (822 +/- 169) to (436 +/- 143) pA. Neither HD nor GJ affected the steady-state activation curve. But they had impact on steady-state inactivation curve. HD 10 mumol/L changed the half inactivation voltage (V0.5) from -22.7 to -40.9 mV, and slope factor (kappa) from 10.2 to 20.6 (n = 4 cells from 3 guinea pigs, P < 0.05). GJ 10 mumol/L changed the V0.5 from -17.7 to -33.3 mV, and kappa from 15.9 to 27.8 (n = 5 cells from 3 guinea pigs, P < 0.05). CONCLUSION: HD and GJ inhibited L-type calcium channels.

[Inhaled budesonide for severe asthma at high altitude].

OBJECTIVE: To determine the benefit of inhaled high dose budesonide combined with terbutaline in patients with severe asthma at high altitude. METHODS: 42 patients with severe asthma at high altitude were assigned in a randomized, double-blind fashion to receive either budesonide combined with terbutaline (budesonide group, 21 cases) or terbutaline with placebo (control group, 21 cases). Both groups received terbutaline delivered by a metered-dose inhaler in a dose of 2.5 mg. Budesonide delivered by a metered-dose inhaler in 1.2 mg in budesonide group and placebo managed by a specially prepared metered-dose inhaler in control group were administered after terbutaline treatment above procedures. Repeated once after 10 min. Before and after therapy, the scores of the activity of accessory respiratory muscles, dyspnea, wheezing (clinical index) and lung function were documented. RESULTS: At 1, 2, 4, 6 h after therapy, FEV1% [(43 +/- +/- 5)%, (50 +/- 5)%, (57 +/- 5)%, (67 +/- 6)%], PEF% [(47 +/- 5)%, (55 +/- 6)%, (62 +/- 7)%, (69 +/- 7)%], clinical index (5.1 +/- 0.8, 4.3 +/- 0.6, 3.5 +/- 0.6, 2.5 +/- 0.4) in budesonide group and FEV1% [(42 +/- 5)%, (44 +/- 5)%, (45 +/- 5)%, (45 +/- 5)%], PFE% [(46 +/- 5)%, (47 +/- 5)%, (49 +/- 6)%, (49 +/- 6)%], clinical index (5.3 +/- 0.7, 5.0 +/- 0.5, 4.9 +/- 0.7, 4.8 +/- 0.7) in control group were difference markedly compared with before therapy [budesonide group was (35 +/- 5)%, (38 +/- 5)%, 8.3 +/- 1.0, and control group was (33 +/- 5)%, (38 +/- 5)%, 8.3 +/- 1.1, respectively], all (P < 0.01). CONCLUSIONS: High dose of inhaled budesonide combined with terbutaline should be an effective therapy for patients with severe asthma at high altitude. Budesonide inhaled in high dose produces therapeutic effects as soon as 2 h after therapy.

[Experimental infection of a novel nonenveloped DNA hepatitis virus in Rhesus monkey].

OBJECTIVE: To test the susceptibility of the Rhesus monkey to the TT virus and to establish its transmission route. METHODS: Rhesus monkeys were administered orally and intravenously with the inoculum that was prepared with feces collected from a patient at the incubation period. Passage study was done with monkey' s positive feces. The blood, bile and feces were tested with polymerase chain reaction (PCR), and the liver, the jejunum tissues with in situ hybridization. RESULTS: The viremia occurred 4-7 days after intravenous inoculation and 7-10 days after oral administration. The virus was also excreted in the feces in a few days after oral infection and simultaneously with viremia after intravenous inoculation. The virus was also detected in the bile during the viremic phase. There was a prolonged carrier state that the viremia and fecal virus excretion persisted for more than 6 months. No serum transaminase elevation was found during the infection. There were virus signals in hepatocytes in columnar epithelium and lamina propria cells of jejunum villi, but no significant pathology was demonstrated in both sites. CONCLUSIONS: The liver infection of Rhesus monkey was established hb either oral or parenteral virus Inoculation. The virus may be released from liver into blood and intestine vial the bile or just from the gut wall into feces, and hence it may be transmitted by both routes.

Effect of melittin on potassium currents and action potential in ventricular myocytes of guinea pig.

AIM: To examine the effects of melittin (Mel), the major component of bee venom, on delayed rectifier K+ current (Ik), inward rectifier K+ current (Ikl) and action potential (AP) in guinea pig ventricular myocytes. METHODS: Ik, Ikl, and AP were recorded using the whole-cell patch-clamp technique. RESULTS: The action potential duration (APD) was shortened by Mel in a concentration-dependent manner. Mel 0.05, 0.1, 0.2 mumol.L-1 shortened APD50 from (520 +/- 55) to (459 +/- 91) (n = 5, P > 0.05), (385 +/- 102) (n = 5, P < 0.01), and (281 +/- 81) ms (n = 5, P < 0.01), respectively; and APD90 from (613 +/- 96) to (536 +/- 93) (n = 5, P > 0.05), (467 +/- 96) ms (n = 5, P < 0.01), and (354 +/- 95) ms (n = 5, P < 0.01), respectively. Mel increased the amplitude of Ik also in a concentration-dependent manner. Mel 0.05, 0.1, 0.2 mumol.L-1 increased Ik from (295 +/- 109) to (371 +/- 142) (n = 5, P < 0.05), (467 +/- 180) (n = 5, P < 0.05), (552 +/- 248) pA (n = 5, P < 0.05), respectively at testing potential of + 40 mV. But no significant effect of Mel on Ikl was observed at these concentrations. CONCLUSION: Mel significantly increased Ik in a concentration-dependent manner which contributed to shortening of APD.

[The arboviruses of genus alphavirus family togaviridae were isolated from mosquitoes captured from Yantai].

OBJECTIVE: To isolate arbovirus from mosquitoes caught from Yantai. METHODS: The isolated viruses were tested for their physico- chemical properties, examined by electron microscopy and specific immuno-reactivity. RESULTS: Fifteen strains of virus were isolated from mosquitoes in 1994 from Yantai. Three of them were further assayed. The results showed that the viruses could multiply on C6/36 cell and produce typical cytopathogenic effect. The viruses couldn't cause regular sickness and death of suckling mice by intracerebral inoculation. The viruses were sensitive to ether, but resistant to 5, -Idu. One sample was examined with electron microscope, and spherical virus particles were observed. The diameter of the virus particles is about 55 +/- 2. 3nm. It did not react with the JBE virus and Bunyaviridae group specific immuno-ascitic fluids but cross-reacted with the group A Togaviridae specific immuno-ascitic fluid. CONCLUSIONS: The results showed that the viruses isolated from Yantai belong to alphavirus of togaviridae.

Preclinical trials of human erythrocyte superoxide dismutase injection.

OBJECTIVE: To assess the quality of human erythrocyte superoxide dismutase (SOD) injection reaching the official standard for its clinical uses. METHODS: Human erythrocyte SOD injection prepared by McCord-Fridovich's method without column chromatography but with some modifications was used in preclinical trials, to observe the general pharmacology and pharmacodynamics of the product. RESULTS: The quality of human erythrocyte SOD injection conformed to the official standard of a biological product, which was found to be non-toxic and did not have any effects on the central and autonomic nervous systems as well as cardiovascular and respiratory systems. The efficacy of anti-inflammation and promotion of immuno-regulation especially on carrageenan and adjuvant-induced polyarthritis were shown in animals. CONCLUSION: Human erythrocyte SOD injection is appropriate for prophylactic and therapeutic uses in clinical trials.

[A comparison of polymerase chain reaction with other two widely used methods in detection of Porphyromonas gingivalis].

OBJECTIVE: To compare the efficiency of polymerase chain reaction (PCR) with cultural method and indirect immunofluorescence (IF) in detection of Porphyromonas gingivalis (P.g) in oral plaque samples. METHODS: 92 subgingival plaque samples were collected from patients of three kinds of periodontal disease. A program using PCR to detect Fimbrilin gene (Fim A) was designed to detect P.g. The efficiency of this PCR method was compared with cultural method and IF. RESULTS: The positive-rate of PCR was 94.6%, while that of cultural method and IF was only 48.9% and 68.5% respectively. The difference was significant in statistical analysis (P < 0.001). CONCLUSION: PCR is more sensitive than culture method and IF in detecting pathogens of periodontal diseases. The study provides a simplified and efficient method for oral clinical use.

[Thoracolumbar injuries: pathomorphology of and a new classification].

OBJECTIVE: To introduce a new classification of thoracolumbar injuries. METHODS: Three modes were used to classify the injuries according to the mechanisms of injuries: Type A, flexion-compression fractures; Type B, vertical compression fractures; Type C, distraction injuries. Two groups were used to distinguish the fractures in one type according to the pathomorphological differences and the clinical incidence: Group A, with a more clinical incidence, and Group B, with a less clinical incidence. Three subgroups were used to divide the injuries in one group in consideration of the severity of the fracture instability and the fracture displacement: fracture, subluxation, and dislocation. RESULTS: A new classification of thoracolumbar fractures was established. CONCLUSION: The new classification is more systemic and more convenient.

[Hyperpolarized cardiac arrest with ATP-sensitive potassium channel opener on myocardial protection during CPB].

OBJECTIVE: To investigate the myocardial protective effects of pinacidil-induced hyperpolarized arrest and compare them with those induced with depolarized hyperkalemic arrest. METHODS: 18 dogs were equally divided into three groups. In the hypothermic hyperpolarization group (LH group), after aortic cross-clamping, a single dose of 4 degrees C pinacidil containing St. Thomas cardioplegic was infused through the aortic root. Temperature during CPB was kept between 26 - 28 degrees C and warmed to 37 degrees C before aortic declamping. Global ischemia lasted 60 min and then reperfusion started for 30 min. In the normothermic hyperpolarized group (WH group), the same procedure was set as in the LH group, except maintaining temperature of 35 - 37 degrees C for CPB and pinacidil solution. In the control group (group C), no pinacidil in St. Thomas solution was the only difference to the other 2 groups. Cardiac arrest and its recovery, the ultrastructure of the myocardium and hemodynamic during ischemia and after reperfusion were observed in the 3 groups. RESULTS: (1) The percentages of normal mitochondria and glycogen were not changed significantly during ischemia and after reperfusion in the LH group, but declined markedly in the group C at ischemic 30, 60 min, and reperfusion for 20 min (P < 0.01). In the WH group, they were lower than those of the group LH, but higher than those of the group C before ischemia. (2) The recoveries of CO, SV, CI, LVSW, RVSW, MAP in the LH group were significantly better than those in the other two groups after reperfusion for 15 minutes and 30 minutes (P < 0.05 or 0.01). However, they were much better in the WH group than in the group C (P < 0.05 or 0.01). (3) The time from cardioplegic infusion to cardiac arrest was shorter in the group C and the group LH than in the group WH. CONCLUSIONS: Myocardial protection for global ischemia during CPB could be well achieved with hyperpolarized cardiac arrest induced by ATP-sensitive potassium channel opener, pinacidil, especially in the hypothermic state. The protection is weaker in normothermia but is still stronger than that with traditional depolarized arrest.

[Analysis of clinical epidemiology for 208 patients of measles and some lessons learned].

OBJECTIVE: To survey the epidemic patterns and clinical characteristics of a relatively large scale measles epidemic in Shenzhen, 1998. METHODS: Morbidity among permanent and temporary residents in 4 different age groups, related to their history of measles vaccination among the infant cases of 8 month olds among the two types of residents. Using data originated from the hospital documents of 208 measles cases in 1998. RESULTS: Among the total 208 cases: (1) Of 107 cases of temporary residents in age > 8 mon., 79.4% were unvaccinated while 41.7% were unvaccinated among 72 cases of permanent inhabitants in age > 8 mon.; (2) The two peaks fell on 9 month to 5 year olds (32.7%), in which predominant cases were permanent inhabitant, while among the 14 - 36 year group (33.7%) temporary resident accounted for 71.4%; (3) Among 208 cases, there were 29 in 3 - 7 mon. olds, taking up 14.0% of the whole, who all did not receive the vaccinations. Seventeen of the 29 cases (58.6%) of the said children had mothers with positive histories of measles while the infants manifested atypical mild measles. The rest 12 of the 29 cases (41.4%) whose mothers had an negative histories of contracting measles, 4 infants manifested typical measles and 8 atypical mild measles. CONCLUSIONS: (1) One of the main causes of this epidemic was the unsound history of vaccination in the population; (2) In Shenzhen, preventive measures should focus on infants and young children among permanent inhabitants while focusing on adolescents and youths on temporary residents; (3) It was suggested that in Shenzhen, infants born to mothers without measles history or infants at the age borderline for measles immunization but without vaccination should receive it; Women in reproductive ages without measles history or vaccination should be considered as vaccination receivers.

[The effects of cyclin E on the growth and other cell cycle related genes of breast carcinoma cells MCF-7].

OBJECTIVE: To investigate the effects of cyclin E in high and low expression on the cell growth and other cell cycle associated genes of MCF-7 cells. METHODS: Eukaryotic expression vehicles of sense and antisense cyclin E were constructed and transferred into the MCF-7 cell line using lipofectAMINE. The integration and expression of cyclin E were conformed by Southern and Western blot. The cells growth was observed and the changes in cell cycle were analyzed by flow cytometry. The expression of other cell cycle associated genes was assayed using Western blot. RESULTS: A high expression of cyclin E enabled to promote the cell growth and DNA synthesis and accelerated the proceeding of G(1) phase to S phase, It also promoted the phosphorylatin of pRB and up-regulate the expression of p27 while a low expression of cyclin E showed an opposite effect. CONCLUSION: Different expression of cyclin E enables to affect growth of MCF-7 cell by the changes of cell cycle related genes.
