Comparative bronchodilatory activity of cetiedil citrate monohydrate, theophylline, orciprenaline and placebo in adult asthmatics.

In 40 patients, a seven day observation period in which oral bronchodilators and corticosteroids were eliminated, cetiedil (100 mg, t.i.d.), theophylline (200 mg, t.i.d.) or orciprenaline (20 mg, t.i.d.) and placebo (100 mg Lactose, t.i.d.) were given for one week each, according to a double blind, crossover randomized code. Wheezing scores were improved by all three active drugs. Twenty five of these patients undertook a moderate degree of exercise. Cetiedil was the most effective drug in blocking exercise-induced bronchospasm of the drugs studied. At rest, FEV1/VC was improved by theophylline, while PEFR improved after orciprenaline. Cetiedil improved both parameters, significantly. In another 21 patients, cetiedil (100 mg, t.i.d.) was given, according to the 'patient blinded' method for two weeks. PEFR and Asthma Disability Scores steadily improved. Clinical effectiveness was observed as early as the 3rd or 4th day of cetiedil therapy, with maximum improvement detected on the 14th day. No major side effects or abnormalities in clinical laboratory tests were noted. Cetiedil also had a 'liquidifying effect' on bronchial mucous in these asthmatics.

Diffusion coefficients of single bile salt and bile salt-mixed lipid micelles in aqueous solution measured by quasielastic laser light scattering.

Diffusion coefficients of single bile salt micelles and mixed binary micelles consisting of single bile salts and phosphatidylcholine in the molar ratio 3 : 1 have been measured using quasielastic laser light scattering spectroscopy. Among single systems, marked concentration dependence of the diffusion coefficient was observed only in those containing sodium taurodeoxycholate. Micellar size correlates with bile salt molecular structure in a manner similar to that observed by earlier workers with some minor differences to towards a more consistent ordering. Phosphatidylcholine produces small increases in micellar size in most systems depending on bile salt structure, and seems most plausibly interpreted as simple swelling of the single species micelles. In some systems, when phosphatidylcholine is added, size or shape changes are indicated. Diffusion coefficients were also studied in the ternary systems bile salt-phosphatidylcholine-cholesterol which were saturated and metastably super-saturated with respect to cholesterol. The Stokes radii in both systems were the same within experimental error, 35 A. Our results indicate that intermicellar interactions significantly contributed to the measured diffusion coefficients in the ternary systems as well as in the single sodium taurodeoxycholate and binary sodium taurodeoxycholate-phosphatidylcholine systems in the concentration range studied.

The activity of avian rhodanese.

A survey of rhodanese activity (thiosulphate: cyanide sulphur transferase) in the tissues of the domestic fowl revealed that the highest activity occurred in the kidney, approximately twice that in the liver, 316 and 141 mumol SCN formed/min g protein, respectively. 2. In sparrows, pigeons and ducks, liver and kidney activities tended to be similar and increased with body size. 3. Relationships were found between enzyme activity and body weight to a fractional power, indicating relatively higher activity in smaller birds.

Simultaneous accumulation of Evans blue dye and albumin in the canine aortic wall.

The simultaneous transport of Evans blue dye (EBD) and 125I-labeled canine albumin (IALB) across normal and injured endothelial surface was measured by determining the intimal accumulation of EBD (ME) by a reflectometric method and accumulation of equimolar concentrations of EBD and IALB, ME was related to MI by ME = 0.092 + 5.04 MI - 0.508 MI2 for injured and ME = 0.002 + 0.435 MI (nmol-cm-2) for normal surfaces. Both ME and MI varied nonlinearly with exposure time for injured and linearly with time for normal surfaces, giving apparent permeability coefficients of 0.92 X 10(-8) cm-sec-1 for EBD and 2.00 X 10(-8) cm-sec-1 for albumin. These results show that the transvascular flux of EBD can be related to that of albumin by empirical equations, which is of practical importance but also implies unique underlying physical laws. Study of these physical laws will provide deeper insight into transvascular macromolecular transport processes.

Transmission electron microscopy of biliary mixed lipid micelles.

One potential artifact problem in election microscopy of micellar systems is that structural changes and aggregation could be induced by high vacuum dehydration in sample preparation prior to viewing and photomicrography. This could account for an earlier report on "macroassemblies" of biliary lipid, a finding which has been discrepant with the predominance of other evidence on the size, configuration and behavior of biliary micelles. In the present work, this possibility was explored and no evidence for such macroassemblies demonstrated when freeze-drying sample preparation was used; whereas, with conventional technique, the "macroassemblies" were again observed. It is concluded that freeze-drying or freeze-etching sample preparation is required to avoid this form of artifact in electron microscopy of micellar systems. The present findings are compatible with a spherical-equivalent or oblate ellipsoid configuration for biliary mixed micelles.

Kinetic analysis of cholesterol turnover in rat tissues.

A number of studies have been conducted on serum cholesterol disappearance following a single injection of tracer. They have shown 2 or 3 component curves depending on the length of time of observation. They have been interpreted to indicate pools of cholesterol in fast or slow equilibrium with serum and generally depicted as representing groups of tissues. A cholesterol kinetic study was conducted using rats so that each tissue could be analyzed for appearance-disappearance of cholesterol from 30 minutes to 75 days. Serum and liver appeared to die-away for 30 days in a 2 component configuration, then exhibited a repetition of the same curve in the next 30 days. Skeletal muscle and kidney had a slow buildup for 2 weeks, then a plateau for 2 weeks then a 2 component die-away. Adipose showed a high plateau from 30 minutes to 30 days then a 2 component die-away, with specific activity higher than serum at all times. The data are interpreted to indicate that adipose tissue sequestered part of the tracer dose and all tissues reached equilibrium at about 30 days. After that time all tissues had a statistically significant 2 component die-away curve. The mathematical models tested suggest that in the steady state, cholesterol enters and exits the slow pool of serum, kidney, muscle and adipose and in liver enters both pools. Interpretation of the complete die-away is that the tracer dose first equilibrates with membrane free cholesterol, then with the intracellular cholesterol, and finally dies-away at the rate of release by the slow intercellular pool.

A randomized comparison of prism adaptation and augmented surgery in the surgical management of esotropia associated with hypermetropia.

OBJECTIVE: To compare both motor and sensory surgical outcomes for patients with esotropia associated with hypermetropia managed with an augmented surgery based on the average of the near deviation with and without correction vs preoperative prism adaptation. METHODS: Forty-three patients with esotropia associated with hypermetropia entered a randomized prospective evaluation of augmented surgery (group A, 27 patients) versus prism adaptation (group P, 16 patients). The formula for augmenting the amount of the rectus muscle recession was based on the average of the near deviation with and without correction in group A, and the prism-adapted angle of deviation in group P. RESULTS: In group A, postoperative deviations of 8 prism diopters (delta) or less at distance and at near were achieved in 24 patients (88%), and in 22 patients (81%), respectively. In nine prism responder patients, postoperative deviations of 8 delta or less at distance and at near were achieved in eight patients (89%). In seven nonresponder patients, postoperative deviations of 8 delta or less at distance and at near were achieved in six patients (86%). No significant difference between the two groups in terms of near and distance deviation at 1 and 3 months postoperatively were noted. CONCLUSION: No significant difference existed in the motor outcomes between the two groups, or between the prism responder and prism nonresponder groups. Further detailed studies must be made in a large number of patients.
