Expression of autotaxin mRNA in human hepatocellular carcinoma.

OBJECTIVE: To investigate the expression of autotaxin (ATX) mRNA existed in the human hepatocellular carcinoma (HCC), and whether there is relation between the level of ATX expression and clinicopathological features of HCC. METHODS: Five normal liver tissues and 32 histologically verified HCC specimens were obtained. Semi-quantitative reverse transcription polymerase chain reaction was used to detect mRNA expression of ATX. RESULTS: ATX was expressed in all 32 HCC and 5 normal liver tissues. The mean expression level of ATX gene in HCC samples was higher than that in normal liver tissues (68.23% +/- 15.31% vs. 31.97% +/- 8.05%, P < 0.001). Patients were divided into two groups: low ATX HCC (15 cases) and high ATX HCC (17 cases) by the cutoff point of median value. Intrahepatic metastasis, vascular invasion and poor differentiation were more frequently noted in HCC patients with high ATX expression than in patients with low ATX expression. CONCLUSION: ATX gene was found to be overexpressed in some HCC and correlated with HCC development and metastases.

[Effects of intralesional injection of kenalog or interferon alpha-2b on PDGF BB gene expression in situ of hypertrophic scars].

OBJECTIVE: The study is to determine whether kenalog or interferon alpha-2b inhibits fibroblast growth in hypertrophic scars through decreasing PDGF. METHOD: Six patients with hypertrophic scars received intralesional injection of kenalog or interferon alpha-2b. On the 3rd and 7th day after injection, scar samples from each patient were collected and PDGF mRNA expressions in situ were studied. RESULTS: 1. On the 7th day after intralesional injection of kenalog or interferon alpha-2b, expressions of PDGF BB mRNAs decreased significantly (P < 0.01). However, on the 3rd day the expressions of PDGF BB mRNAs of the intralesional-injected area and non-injected area were not significantly different (P > 0.05). 2. Gene expressions of PDGF BB in situ were higher in hypertrophic scars than in normal skin. CONCLUSION: The mechanism of kenalog or interferon alpha-2b in ameliorating hypertrophic scar is to reduce mitosis and prohibit proliferation of fibroblasts in hypertrophic scars by inhibiting PDGF gene expression.

[WT1 gene expression in leukemia patients and its correlation with prognosis and multidrug resistance].

OBJECTIVE: To evaluate the value of expression of WT1 gene in predicting the prognosis of leukemia patients, and explore the relationship between WT1 gene expression and multidrug resistance and cell apoptosis. METHODS: Expressions of WT1, MRP and mdr1 were measured in 68 leukemia patients by RT-PCR method. Expression of bcl-2 was measured in 32 AML patients by immunofluorescence flow cytometry. RESULTS: Expression of WT1 was revealed in 36 of 68 leukemia patients and none of 23 normal controls. Complete remission rate (59.46%) was lower in WT1 positive patients than that (87.10%, P = 0.011) in WT1 negative patients. The rate of MRP expression was also higher in patients with WT1 expression (58.33%), than in those without WT1 expression (32.26%, P = 0.033). Thirty-two AML patients were divided into high-, intermediate-, and low-risk groups according to the expression of WT1 and bcl-2. CR rates were significantly different among these 3 groups (33.33% for high-, 47.37% for intermediate-, and 100% for low-risk group, P < 0.05). CONCLUSION: The expression of WT1 can predict the treatment outcome and the prognosis for leukemia patients. Expression of WT1 is the most important risk factor, and the coexpression of WT1 and bcl-2 predicts poor prognosis of AML patients.

[Quantitative study of vascular endothelial growth factor expression in primary lung cancer].

OBJECTIVE: To study the expression levels of vascular endothelial growth factor (VEGF) and its relation with intratumoral microvessel density (MD), cellular proliferation activity (PI), and lymph node metastasis of primary lung cancer. METHODS: Surgical samples of 53 patients with primary lung cancer were measured. The VEGF was immunofluorescence stained with VEGF polyclonal antibody. The propidium iodide was used to stain cellular DNA. VEGF expression levels and DNA content were analyzed quantitatively using flow cytometry. Intratumoral microvessel density was detected by factor VIII polyclonal antibody using immunohistochemistry. RESULTS: VEGF expression levels (FI) were high in lung cancer (1.3 +/- 0.6, positive ratio 73.58%). The cellular proliferation activity (PI) in VEGF-positive tumors was significantly higher than that in VEGF-negative tumors (29.8% +/- 9.2% vs 18.7% +/- 6.5%, P < 0.01). The intratumoral microvessel density (MD) in VEGF-positive tumors was higher than that in VEGF-negative tumors (19.4 +/- 11.3 vs 12.3 +/- 8.0, P < 0.05). No association was found between the MD and PI (r = 0.169, P > 0.05). The VEGF expression levels in lung cancer tissues with lymph node metastasis (n = 33) were significantly higher than that in those without lymph node metastasis (n = 20) (1.5 +/- 0.6 vs 1.0 +/- 0.5, P < 0.01). CONCLUSION: VEGF plays an important role in the growth and metastasis of lung cancer.

Quantitative assessment of Parkinson's disease deficits.

OBJECTIVE: To quantitatively analyze the tremor and rigidity due to Parkinson's disease. METHODS: 38 patients with Parkinson's disease (PD) ranging in age from 45 to 72 years and 211 normal subjects aged from 16 to 76 years were investigated. The frequency and range of tremor, the muscle tone of the upper limbs in elbow were detected by a computerized video motion detecting system and a new invented apparatus which can detects skeletal muscle tone. RESULTS: For the PD patients, the frequency of resting tremors was detected in 4 to 6 per second. For extensor and flexor in the PD patients, the value of muscle tone was higher than that of normal subjects and the value of muscle tone in flexor was higher than that of extensor. The rigidity increased gradually with repeat passive movement. The curves of rigidity were shown on computer screen or printed out. The data of rigidity were compared with the M-A Scale. A patient who was suspected to suffer from PD above by the equipments and found the muscle tone was higher than normal. In another PD patient the rigidity was obvious at one side and the muscle tone in "normal side" was also high. These equipments were used to record changes of rigidity and tremor in one more PD patient taking with different drugs in order to see the drug effect. CONCLUSION: Quantitative methods are useful to analyse the motion disorders due to PD.

[The study of E-cadherin expression on injury and repair of epithelium of respiratory tract in smoking mice].

OBJECTIVE: To investigate effects of smoking on E-cadherin (E-cd) expression of injury and repair of respiratory tract epithelium by observation of dynamic expression of E-cd on epithelial cells of respiratory tract in smoking mice. METHODS: Immunohistochemistry SP technique was used to examine E-cd expression on epithelial cells of respiratory tract in smoking mice of different stages and dosages and the morphological changes observed by light and electron microscopy respectively. RESULTS: Four and twelve weeks after smoking, the E-cd expression in the epithelial cells was significantly down-regulated compared with the control group (P < 0.01). Morphologically, the intercellular junction was loose, and the epithelial cells were injured and proliferated on the same time. Eight weeks after smoking, however, the E-cd expression was not different from that of the control group (P > 0.05). CONCLUSIONS: Smoking is able to cause wavy change of the E-cd expression on the epithelial cells which may play an important role in the injury and repair of the epithelial cell of the respiratory tract.

[Replacement of the superior vena cava with vascular prosthesis in surgical treatment of lung carcinoma].

OBJECTIVE: To investigate the effect, surgical technique and prognosis of replacement of the superior vena cava(SVC) with vascular prosthesis in surgical treatment of advanced lung carcinoma. METHODS: Twenty six patients with right lung carcinoma invading the SVC (central lung carcinoma in 21 cases, peripheral lung carcinoma in 5 cases) underwent radical resection of tumor with concomitant replacement of SVC with vascular prosthesis. All patients took a low dose of anticoagulant in short-term after operation. Long-term follow-up was conducted. RESULTS: All the cases had no intraoperative or postoperative early death, and no early or late obstruction of SVC. Follow-up was conducted in 23 cases. The overall survival rates at 1, 3 and 5 years were 85%(22/26), 39%(7/18) and 36%(4/11) respectively. CONCLUSIONS: Combination of resection of tumor and replacement of SVC enabled some cases of advanced lung carcinoma to achieve complete resection. This surgical technique extended the indications of surgical treatment of lung carcinoma, and increased the survival rate of advanced lung carcinoma. Further clinical applications are recommended.

[Effect of kidney deficiency caused by ovariectomy on serum osteocalcin level and tumor necrosis factor in mice with collagen induced arthritis].

OBJECTIVE: To explore the effect of ovariectomy caused Kidney Deficiency on the metabolism of bone in mice with collagen induced arthritis. METHODS: The mice for experiment were immunized with subcutaneous injection of type II collagen to induce arthritis after ovariectomy. Severity of joint swelling, radioimmunoassay of serum estradiol (E2), osteocalcin (OC) and tumor necrosis factor (TNF), and pathological changes of joint, including changes on synovia, articular cartilage and bone, were observed once weekly. RESULTS: The E2 level of ovariectomized mice dropped down obviously, while the contents of OC and TNF increased significantly. Severe pathological changes can be seen in synovial tissues, cartilage and bone. CONCLUSION: Kidney Deficiency caused by ovariectomy exacerbated the pathological changes of collagen-induced arthritis in mice.

[Clinical analysis of 12 cases of Chinese patients with adult T-cell leukemia/lymphoma].

OBJECTIVE: To define the clinical characteristics and differential diagnosis in Chinese patients with adult T-cell leukemia/lymphoma (ATLL) and its relation to human T lymphotropic virus type-1 (HTLV-1) infection. METHODS: Lymphocyte morphology in peripheral blood was examined in smears by Cytospin II. Lymphocyte immunophenotyping was carried out by indirect immunofluorescence method. HTLV-1 antibody was detected by indirect immunofluorescence method, proviral DNA detected by PCR method and PCR products demonstrated by Southern Blot analysis. RESULTS: The clinical manifestations in these 12 patients were fever (10/12), lymphadenopathy (8/12), skin changes (7/12), spleen and liver enlargement (7/12), jaundice (3/12), pleural effusion (1/12) and lung invasion (1/12). Patul-like cells were not easily found in routine blood smears, but easily found with Cytospin II (10/10). There was increase in lactic dehydrogenase (8/9) and soluble interleukin 2 receptor (9/9). Anemia (7/12), thrombocytopenia (6/12), pancytopenia (2/12), myelodysplasia (4/12) and eosinophilia (8/12) can be found, but no hypercalcemia. HTLV-1 antibody and proviral DNA were positive in 6/10 cases and 9/12 cases respectively. Nine cases were classified as acute type and three as lymphoma type. All patients were misdiagnosed as other diseases at the beginning. The initial presentations were skin changes (4/12), lymphadenopathy (3/12), myelodysplasia (3/12), progressive liver enlargement (1/12) and refractory pleural effusion (1/12). The complications were infections by undetermined pathogens (6/12), Streptococcal infection (1/12), gastrointestinal bleeding (3/12), acute renal failure (3/12), acute respiratory failure (2/12) and cerebral hemorrhage (1/12). Seven patients died, three still survive, and two are out of contact. CONCLUSION: ATLL may not be very rare in China. It is necessary to deepen the recognition of ATLL in Chinese.

[The mechanism and effect of erythromycin in development of pulmonary fibrosis].

OBJECTIVE: To observe the effect of erythromycin on the development of pulmonary fibrosis and explore the new therapy in pulmonary fibrosis. METHODS: By evaluating the pathologic changes, the synthesis of collagen protein, the level of activity and expression of tumor necrosis factor-alpha (TNF-alpha), the effect of erythromycin was investigated on the rat model of pulmonary fibrosis induced by bleomycin. RESULTS: The alveolitis alleviated in the first week in treatment group comparing with the control group (142.5 +/- 13.9 mg/g), the synthesis of collagen protein decreased in the fourth week treatment group (96.8 +/- 4.8 mg/g) (q Test P < 0.01). CONCLUSION: Erythromycin may alleviate the process of rat pulmonary fibrosis induced by bleomycin.

[Value of thoracoscopy in undiagnosed pleural effusion].

OBJECTIVE: To assess the value of thoracoscopy in undiagnosed pleural effusion. METHOD: By thoracoscopy, 114 cases of unknown origin pleural effusion were examined. Abnormal pleurae were biopsied in sight of thoracoscopy and compared with blind needle biopsy for pleurae. RESULTS: Positive rate was 91.2% (104/114) by thoracoscopy and 28.1% (32/114) by blind needle biopsy for pleurae. Significant difference between the groups was found (P < 0.005). In sight of thoracoscopy the morphology of pleural mesothelioma, metastatic carcinoma and tuberculous pleurisy was different. The rate of malignancy of sanguineous pleural effusion was 77.1% and yellow pleural effusion was 53.0%. PPD test showed that intense positive reaction was 42% in the benign cases, but negative response was 48% in the malignant cases. No serious complication occured in all cases. CONCLUSIONS: Thoracoscopy is an safe and effective measure in diagnosing patients with undiagnosed pleural effusion.

[Effects of metalloproteinases on pulmonary fibrosis by bleomycin in rats].

OBJECTIVE: Matrix metalloproteinases (MMPs) are the mediators for dynamic equilibrium of extracellular matrix. The purpose was to investigate the effects of metalloproteinases on the development of pulmonary fibrosis. METHODS: The expression and activity of metalloproteinases of alveolar macrophage, and fibroblasts were evaluated on rats with pulmonary fibrosis induced by bleomycin. RESULTS: The activity of collagenase was low in the macrophage and fibroblast in control group, 20.4 x 10(-7) U/cell and 30.6 x 10(-7) U/cell respectively. There was an increase in the first and fourth week groups of bleomycin rats. The highest level of activity of alveolar macrophages was 258.0 x 10(-7) U/cell in first week group. A peak of enzymatic activity was found in 4th week group (289.0 x 10(-7) U/cell). The mRNA of MMP-1, MMP-2 were expressed at lower levels in the normal lung tissue and there was an increase in first week, and fourth week group of bleomycin rats with a peak in one week group. CONCLUSIONS: The regulation of metalloproteinases might play an important role in the development of pathological processes in pulmonary fibrosis.

[Cervical spinal cord injury treated by expanded hemilaminectomy].

OBJECTIVE: To treat cervical spinal cord injury (SCI) accompanied with narrowing spinal canal by expanded hemilaminectomy. METHODS: From 1995 January to 1998 April 51 patients of cervical SCI were treated by expanded hemilaminectomy. Spinal injury classified in to 3 types: no fracture-dislocation (39 patients) fracture dislocation at the lower cervical spine (11), and burst fracture (1). The types of SCI included central cord injury (18 patients) incomplete cord injury (19), and complete cord injury (14). MR imaging in 23 patients showed degenerative changes with normal intensity of the cord in 14 patients, multiple level hyperintensity in 3, cystic changes in 3, myelomalasia in 3, and cord brocken in 1. Expanded hemilaminectomy was performed in 24 hours in 3 patients, in 48 hours in 9, in one week in 2, after one week in 35, and after one year in 2. The left or right laminae were removed from C(7) to C(3) in 42 patients, C(3) - T(1) in 3, C(2) - C(7) in 2, C(3) - C(6) in 3 and C(4) - C(7) in 3. Hemilaminectomy was expanded lateral to the inner of apophyseal joint and medial to the inner lamina beneath the spinal process. RESULTS: Follow-up lasted for 1 year and 7 months. Six patients with complete cord injury had of the no recovery lower extremity but recovery of the brachialis and extensor radial longus. 12 patients of central cord injury had full recovery except intrinsic muscles of the hand (5). They operated were on 2 weeks after injury. 17 patients of incomplete cord injury recovered to Frankel IV. CONCLUSIONS: Expanded hemilaminectomy is indicated for patients of cervical SCI with narrowing spinal canal or without fracture dislocation. Best results can be obtained in patients of central cord injury, and incomplete cord injury. Even in complete cord injury, 1 - 2 forearm muscle may recover (24.8%), securing a pinch grip reconstruction.

[Development of plasmid-based transgenic mice carrying with target gene xylE].

OBJECTIVE: To develop a transgenic mouse model carrying with plasmid pUC118NX integrated into its genomic DNA for detecting mutagenesis. METHODS: DNA of plasmid UC118NX in target gene xy1E was injected microscopically into male protonucleus of 376 mouse spermatova, and 225 survival spermatova were transferred into the oviducts in both sides of 11 pseudopregnant female mice to develop their offspring. The genomic DNA in survival young mice were analyzed with polymerase chain reaction (PCR)-Southern blot, plasmid transformation test and endonuclease-digestion. RESULTS: Seven pseudopregnant mice got pregnant, and 29 offspring were delivered and 25 survived of which 18 were identified with positive for PCR-Southern blot (72%). The two stout male mice with intact integration of plasmid pUC118NX in their genome were finally chosen as founders to detect gene mutation in vivo and establish transgenic mouse lineages. CONCLUSION: Transgenic C57BL/6J mice integrated with plasmid pUC118NX into their genomic DNA have been successfully developed.

[Experimental study on ultrashort wave therapy on the healing of fracture].

The models of fracture in the rabbit radii were made. The rabbits in the experimental group were treated--with ultrashort wave at the fracture sites. The changes in the x-ray, histology, and biomechanics were observed dynamically. The results were that the speed and quality of the fracture healing in the experimental group were much better than in the control group.

[Determination of sulfacetamide sodium and sulfanilamide in shao tang ling ointment by high performance liquid chromatography].

An HPLC method for the determination of sulfacetamide sodium and sulfanilamide in Shao Tang Ling ointment was developed. The chromatographic conditions were: column, Spherisorb C18, mobile phase, methanol-0.25% (V/V)-acetic acid solution (7:93); detector UV-257 nm. The average recoveries were 98.9% (n = 5), RSD = 0.64% for sulfacetamide sodium and 99.3% (n = 5), RSD = 1.58% for sulfanilamide. The analytical method is simple, quick, accurate and reproducible.

[A study on adhesive properties of transformed human embryonic tendon cells to PLA and PLGA].

To study the adhesive properties of transformed human embryonic tendon cells (THETCs) to polylactic acid(PLA) and polylactic-co-glycolic acid 85/15 (PLGA 85/15), the adhesive mechanical properties of single THETCs to PLA films (average thickness 18.6 microns) and PLGA 85/15 films (average thickness 17.3 microns) were measured by means of micropipet aspirating technique. The rsults showed that the attachment rate and adhesive force for two polymer groups(PLA precoated with bovine serum albumin (BSA) and PLGA 85/15 precoated with BSA) were significantly lower than those for two control groups(PLA and PLGA 85/15), respectively, while the attachment rate and adhesive force for other two polymer groups (PLA precoated with poly-D-lysine (PDL) and PLGA 85/15 precoated with PDL) were significantly higher than those for two control groups, respectively, and that the adhesive properties of THETCs to PLGA 85/15 films were better than those of THETCs to PLA films. These demonstrate that BSA can inhibit THETCs attachment to polymer, and PDL can promote THETCs attachment to polymer, and suggest that PLGA 85/15 films were better than those of THETCs to PLA films. These demonstrate that BSA can inhibit THETCs attachment to polymer, and PDL can promote THETCs attachment to polymer, and suggest that PLGA 85/15 coated with PDL may be used as a new type attachment substrate for THETCs.
