Raf-1 physically interacts with Rb and regulates its function: a link between mitogenic signaling and cell cycle regulation.

Cells initiate proliferation in response to growth factor stimulation, but the biochemical mechanisms linking signals received at the cell surface receptors to the cell cycle regulatory molecules are not yet clear. In this study, we show that the signaling molecule Raf-1 can physically interact with Rb and p130 proteins in vitro and in vivo and that this interaction can be detected in mammalian cells without overexpressing any component. The binding of Raf-1 to Rb occurs subsequent to mitogen stimulation, and this interaction can be detected only in proliferating cells. Raf-1 can inactivate Rb function and can reverse Rb-mediated repression of E2F1 transcription and cell proliferation efficiently. The region of Raf-1 involved in Rb binding spanned residues 1 to 28 at the N terminus, and functional inactivation of Rb required a direct interaction. Serum stimulation of quiescent human fibroblast HSF8 cells led to a partial translocation of Raf-1 into the nucleus, where it colocalized with Rb. Further, Raf-1 was able to phosphorylate Rb in vitro quite efficiently. We believe that the physical interaction of Raf-1 with Rb is a vital step in the growth factor-mediated induction of cell proliferation and that Raf-1 acts as a direct link between cell surface signaling cascades and the cell cycle machinery.

The effect of estradiol on output of adrenocorticotropin and prolactin by fetal sheep anterior pituitary cells.

We examined the hypothesis that estradiol (E2) would affect fetal anterior pituitary corticotroph and lactotroph function in vitro, and that any effects would be influenced by gestational age. Anterior pituitary cells from fetal sheep at day 129 (n = 4) and at day 139 (n = 5) of gestation were cultured. After 96 h in culture, cells were treated for 18 h with E2 concentrations ranging from 0 to 1000 nM, in the presence or absence of 100 nM of corticotropin-releasing hormone (CRH), cortisol, arginine vasopressin (AVP), or CRH and cortisol, to examine their effects on corticotroph function. Cells were also treated with bromocriptine or increasing concentrations of E2 to study their effects on lactotroph function. Immunoreactive (ir) adrenocorticotropin (ACTH) and prolactin in the culture medium were measured by radioimmunoassay. Levels of cellular pro-opiomelanocortin (POMC) mRNA and prolactin mRNA were determined by in situ hybridization. Immunohistochemistry was used to determine the percentage of cells that were immunopositive for ACTH (corticotrophs) or prolactin (lactotrophs). ACTH output was stimulated by CRH treatment at day 139 but not at day 129 of gestation, and cortisol attenuated this response. ACTH output by cells cultured with 10 nM E2 and 100 nM CRH, at 139 days of gestation, was greater than with CRH alone (p < 0.05). E2 did not affect basal ACTH output or ACTH output with any other treatment or levels of POMC mRNA. Prolactin output was not affected by E2 treatment. Bromocriptine significantly decreased prolactin output but not levels of prolactin mRNA. We conclude that E2 may affect CRH-stimulated fetal sheep pituitary corticotroph function late in gestation, but only within a narrow, physiological range of concentration.

Anatomical structures in the Leksell pallidotomy target.

OBJECTIVE: Because interruption of pallidal outflow signals by pallidotomy is believed to play an important role in the motor improvement in Parkinson's disease, the anatomical relationship of the two major pallidofugal tracts, namely the ansa lenticularis (AL) and the fasciculus lenticularis (FL) to the Leksell pallidotomy target (LPT) were studied. METHODS: Magnetic resonance (MR) scans of 20 patients with Parkinson's disease were studied. The scans were obtained in the inversion recovery and fast spin echo sequences and viewed in the reverse video mode. RESULTS: The pallidofugal tracts appeared as bands of high intensity on MR scan. FL was wide and extended from the posterior half of the globus pallidus interna (GPi) medially into the internal capsule. AL was narrow, parallel and posterior to the anterior commissure, parallel to and 3 mm above the optic tract, and just beneath the FL. The part of the FL immediately lateral to the internal capsule overlapped the lateral end of the AL. The center of this overlapping area was termed the 'operlapping point' (OP). In all cases, the OP was within the range of LPT. CONCLUSION: It is concluded that the structures in the LPT are FL and AL; and therefore, this procedure could also be known as 'fasciculoansatomy'.

Treatment of intrahepatic cancers with radiation doses based on a normal tissue complication probability model.

PURPOSE: To attempt to safely escalate the dose of radiation for patients with intrahepatic cancer, we designed a protocol in which each patient received the maximum possible dose while being subjected to a 10% risk of radiation-induced liver disease (RILD, or radiation hepatitis) based on a normal tissue complication probability (NTCP) model. We had two hypotheses: H1; with this approach, we could safely deliver higher doses of radiation than we would have prescribed based on our previous protocol, and H2; the model would predict the observed complication probability (10%). PATIENTS AND METHODS: Patients with either primary hepatobiliary cancer or colorectal cancer metastatic to the liver and normal liver function were eligible. We used an NTCP model with parameters calculated from our previous patient data to prescribe a dose that subjected each patient to a 10% complication risk within the model. Treatment was delivered with concurrent hepatic arterial fluorodeoxyuridine (HA FUdR). Patients were evaluated for RILD 2 and 4 months after the completion of treatment. RESULTS: Twenty-one patients completed treatment and were followed up for at least 3 months. The mean dose delivered by the current protocol was 56.6 +/- 2.31 Gy (range, 40.5 to 81 Gy). This dose was significantly greater than the dose that would have been prescribed by the previous protocol (46.0 +/- 1.65 Gy; range, 33 to 66 Gy; P < .01). These data are consistent with H1. One of 21 patients developed RILD. The complication rate of 4.8% (95% confidence interval, 0% to 23.8%) did not differ significantly from the predicted 8.8% NTCP (based on dose delivered) and excluded a 25% true incidence rate (P < .05). This finding supports H2. CONCLUSION: Our results suggest that an NTCP model can be used prospectively to safely deliver far greater doses of radiation for patients with intrahepatic cancer than with previous approaches. Although the observed complication probability is within the confidence intervals of our model, it is possible that this model overestimates the risk of complication and that further dose escalation will be possible. Additional follow-up and accrual will be required to determine if these higher doses produce further improvements in response and survival.

Resazurin reduction assay for ram sperm metabolic activity measured by spectrophotometry.

Resazurin (7-hydroxy-3H-phenoxazin-3-one 10-oxide) is a redox dye that can be reduced by metabolically active spermatozoa to resorufin and manifested as visual color change from blue (resazurin) to pink (resorufin). This study combined the resazurin reduction test with spectrophotometric methods and investigated the correlation between metabolic activity and fertilization potential of spermatozoa, using ram semen as a model. The absorbance at specific wavelengths of resazurin and resorufin was determined by scanning photo spectrometer (600 nm for resazurin and 570 nm for resorufin, respectively). The absorbance at wavelengths of 600 nm (A600) and 570 nm (A570) was measured by spectrophotometry and used to evaluate sperm metabolic activity. A600 decreased and A570 increased in relationship to the increased concentrations of motile spermatozoa and increased resazurin reaction times. We observed, upon using a retrospective experimental design, that fertile rams had greater relative absorbance values than rams with lower fertility. Also, we observed a wide range variation of absorbance between the fertile rams, which is highly correlated to the sperm motility. We conclude that the spectrophotometric measurement of resazurin reduction for sperm activity might be a good assay for ram fertility.

Evaluation of a safety educational programme for the prevention of pesticide poisoning.

The causal factors of pesticide poisoning in spraymen usually include sloppy handling, leakage of sprayers and a lack of personal protection, as well as of a knowledge about pesticide toxicity. This paper outlines the results of a safety educational programme to prevent pesticide poisoning among Chinese village spraymen in 1991 and 1992. The programme consisted of two parts: an interview of 3,286 trained subjects who were surveyed before and after the education courses, and a survey on the prevalence of pesticide poisoning among spraymen in 10 villages in 1991 and 1992, respectively. The results showed a general improvement in the knowledge about pesticide toxicity and safe use among the surveyed subjects. The prevalence of pesticide poisoning among the surveyed spraymen decreased from 1.05% in 1991 to 0.25% in 1992. Moreover, a 68.2% reduction of pesticide poisoning cases was observed in 10 villages in 1992. The success of the safety educational programme proved that education is an effective measure for preventing pesticide poisoning.

[A canonical correlation analysis on the traffic conditions and the road injury in China in 1996].

In order to expound the impact of traffic condition to road injury, the relations of road injury with motorization, road situation e and highway capacity were studied, using Canonical Correlation analysis method. Results showed that the Canonical Correlation Coefficient A(0.9486) was representing the direct correlation of the motorization with the injury and death rates, referring the higher the motorization, the greater the death and injury rates. The Canonical Correlation Coefficient B(0.9220) indicated the direct correlation between the highway capacity and the frequency of road injury, as well as the larger the highway capacity, the more frequency of road injury occurrance. The Canonical Correlation Coefficient C(0.6446) revealed the relations between levels of road situation and the accident rates per 10,000 vehicles. Results showed that the higher the quality of roadway, the lower was the accident rate. In view of this, traffic condition was closely related to road injury. Thus, the frequency of accident, death rate and injury rate could be reduced through improving the road quality and traffic conditions. Relations between two sets of random variables, the ties of the canonical variables and the original variables could all be well analysed with Canonical Correlation analysis. Canonical Correlation is useful to analyse two sets of random variables in epidemiological studies since it provides a great amount of information to be applied and operated widely and easily.

[Epidemiological studies on legionella infection in Taiyuan area of Shanxi Province].

OBJECTIVE: To study the prevalence and epidemic features of legionella infection in Taiyuan area of Shanxi Province. METHODS: IgM and IgG antibodies against 15 types of nine species of legionella were determined with enzyme-linked immunosorbent assay for out-patients, in-patients and healthy population. RESULTS: Positive IgM to legionella accounted for 17.4% in adult patients and 13.2% in child patients, with the highest infection rate both in the adults and children with acute respiratory infection and prevalent in the summer. The peak age of infection was at 30 to 50 years old for adults. Incidence of acute legionella infection in children increased with age. Acute infection occurred in six patients two weeks after their hospitalization with positive IgM against legionella. In healthy population, infection rate was the highest in employees of coal mines (36.7%) and the lowest in urban children (9.5%). Legionella micdadei and legionella pneumophila were the two most common causes of infection in adults and children patients and Legionella pneumophila in healthy population. CONCLUSION: Legionella are common pathogen of infections, possible to cause nosocomial infection. Legionella infection is also common in healthy population, especially in those often exposed occupationally to water and soil.

[Changes in trace elements contents of renal cells in cadmium poisoning].

OBJECTIVE: To understand the possible role of trace elements in renal damage caused by cadmium poisoning and its mechanism. METHODS: An experimental animal model with renal damage caused by cadmium poisoning was prepared, and trace elements contents in subcellular components in renal cells, lipid peroxidation reaction, renal function and its ultrastructural changes were determined. RESULTS: Uptake of cadmium could cause transfer of copper, zinc and manganese mainly distributed in the mitochondrion to cell nuclei and cytoplasm, and content of selenium and activity of glutathione-peroxidase (GSH-px) in cytosol declined and content of propandiolal increased. CONCLUSION: It suggests that changes in trace elements contents, especially in selenium content, during renal damage caused by cadmium poisoning, could correlate with the increase of lipid peroxidation, and abnormal subcellular distribution of trace elements was one of the important roles in renal damage caused by cadmium poisoning.

Erythrocyte and plasma Ca2+, Mg2+ and cell membrane adenosine triphosphatase activity in patients with essential hypertension.

OBJECTIVE: To assess the relationships between plasma and intracellular Ca2+, Mg2+ and blood cell membrane adenosine triphosphatase (ATPase) activity in normotensive and hypertensive subjects. METHODS: Plasma and intracellular Ca2+, Mg2+ were measured with atomic absorption spectrophotometry, and red blood cell membrane Na(+)-K+ ATPase and Ca(2+)-ATPase activities were determined with colorimetric method in 55 patients with essential hypertension and 32 normotensive controls. RESULTS: The results showed that the hypertensive group consistently demonstrated a significant decreased activity of ATPase studied, with significantly lower plasma Ca2+ and higher cytosolic Ca2+ levels when compared with those in normotensive group (P < 0.01 or P < 0.05, respectively). No significant differences were found in either plasma Mg2+ or intracellular Mg2+ level between the two groups. CONCLUSIONS: This study suggests that patients with essential hypertension have widespread depression of cell membrane Na(+)-K(+)-ATPase and Ca(2+)-ATPase activities with plasma Ca2+ depletion and cytosolic Ca2+ overload, which may reflect an underlying membrane abnormality in essential hypertension. The cellular abnormalities may be related to the defective transport mechanisms that in turn may be aggravated by plasma Ca2+ depletion.

Analysis of the characteristics of folate binding proteins and its relationship with expression of multidrug resistance P-glycoprotein in myelodysplastic syndromes.

OBJECTIVE: To observe the characteristics of folate binding proteins (FBP) in myelodysplastic syndromes (MDS) and leukemia and to study the clinical significance of reduced folate carrier (RFC) present in MDS and its relationship with multidrug resistance (MDR). METHODS: The features of FBP on bone marrow cells were analyzed using radiolabeled 3H-folic acid (3H-FA) binding membrane proteins and SDS-polyacrylamide gel electrophoresis (SDS-PAGE). In the same time, P-glucoprotein and mRNA of MDR gene were detected using immunocytochemistry and reverse transcription polymerase chain reaction (RT-PCR) respectively in patients with MDS and leukemia. RESULTS: Two kinds of FBP, folate receptor (FR) and reduced folate carrier (RFC), were found on the leukemic cells. The same results were presented on mononuclear cells of bone marrow in 5 out of 14 MDS patients, and MDR positive was seen in 4 patiens of them. In normal control and other 9 cases of MDS FRs were only found on the mononuclear cells of bone marrow. CONCLUSION: Reduced folate carrier, which is present in the leukemic cell, is a product of neoplastic cell. It might reveal preleukmic state and have the same significance with MDR that RFC is found in MDS patients.

Dynamic study of nitric oxide and endothelin-1 during endotoxin shock and effects of their antagonists on hemodynamics.

OBJECTIVE: To examine the relationship between the profound hypotension in endotoxic shock and the dynamic changes of nitric oxide (NO) and endothelin-1 (ET-1), so as to figure out which of the NO or ET-1 was more involved in the pathogenesis of endotoxic shock. And to investigate whether an offset of their opposite vasoactive effects would occur during endotoxic shock. METHODS: 24 rabbits were anesthetized and instrumented for recording hemodynamics. Endotoxin (E. coli 026: B6, 600 micrograms/kg) was bolus injected intravenously and the animals were randomly divided into four groups. Group I was control without any more intervention, and Group II, III, IV received bolus injections of L-NMA (10 mg/kg), phosphoramidon (2 mg/kg) or dexamethasone (2 mg/kg) respectively at 30 min post-endotoxin. Plasma NO3-, ET-1 and hemodynamics were measured at regular intervals. Their relationships were compared and analysed. RESULTS: Plasma ET-1 achieved its peak level at 60 min post-endotoxin, and then waned. Plasma NO3- started rising at 120 min post-endotoxin, then progressive increase continued till the last measurement at 180 min post-endotoxin. The decrease of blood pressure was significant at about 120 min post-endotoxin and further went down until death. The changes of hemodynamics and NO showed a quite close temporal correspondence between the increase of NO and the decrease of blood pressure. L-NMA and phosphoramidon obviously reduced the plasma levels of NO and ET-1 to below their respective baseline levels, and showed transient effect of increase on blood pressure. Soon afterwards, however, the status of hemodynamics was aggravated. Dexamethasone just inhibited the excessive increase of NO and ET-1 during endotoxic shock without interfering their baseline levels and showed most beneficial effects on hemodynamics. CONCLUSIONS: Both NO and ET-1 increase during endotoxic shock, but only the increase of NO has a close temporal correspondence with the decrease of blood pressure. It suggests a more important role of NO in pathogenesis of endotoxic shock. The increase of NO and ET-1 is different in time-process, which indicates that an offset of their opposite vasoactive effects would not occur. Intreference against the increase of NO and ET-1 during endotoxic shock is most beneficial when their baseline levels are maintained.

[Primary microsurgical repair of multi-structural defects of hand].

Forty-eight cases of multi-structural defects of hands were primarily repaired or reconstructed from July 1989 to 1997. The structural defects included: the defects of radial or ulnar aspect of hands involving fingers and skin, multiple fingers defects and the fingers and skin defects of whole hand. In this series, there were 32 males and 16 females with age ranged from 17 to 46 years old. The composite tissue grafts were obtained from wrap-around flap or 2nd toe skin flap of the foot. The result showed that composite 108 tissues transplantations, or 48 cases, were all survived. After a follow-up of 38.5 months (ranged from 5 months to 6 years), the grasp, pinch and opposition function of the reconstructed finger were restored, the two-point discrimination sensation was 4 mm-12 mm. Most of the patients had resumed their original works. So that the primary repair of multi-structural defects of hands by composite tissues transplantation was feasible and valuable, but thorough debridement and skilled microsurgecal technique were required.

[The clinical application of bridge by "Y" type vein].

UNLABELLED: In order to solve the defect of blood vessel in tissue transplantation and complicated palmar amputation, bridge by "Y" type vein had been used from Jan. 1990 to Jul. 1996. Twenty-three cases were treated. In this series, there were 16 males and 7 females, with ages ranged from 10 to 42 years old. Six cases were the defect of lower legs anterior skin and tibia, 3 cases were the femur fracture with injury of femoral artery and tissue's defect, 2 cases were defect of five fingers, 12 cases were complicated palmar amputation. RESULT: 15 cases with tissue transplantation and 12 cases with limb replantation were all survival without infection or necrosis. After the following-up for 3 years (ranged from 1 to 5 years), the function of injured limbs were satisfactory, 19 patients had resumed their original work. So, to bridge by "Y" type vein is a good method for repairing the defect of blood vessels in tissue transplantation and complicated palmar amputation, but skilled microsurgery technique is required.

Wiedendiol-A inhibits cholesteryl ester binding to its transfer protein.

AIM: To study the wiedendiol-A (W-A) inhibition mechanism of plasma cholesteryl ester (CE) transfer protein (CETP) on the transfer of CE. METHODS: Using gel filtration method. RESULTS: W-A at 30 mumol.L-1 inhibited association of CE with CETP by 76% and CETP transfer activity by 81%. In addition, W-A enhanced binding of TP2, a monoclonal antibody with a CETP C-terminal epitope which is involved in CE binding, to CETP, suggesting a W-A-induced conformational change at the epitope for increased TP2 binding. When CETP activity was measured by varying high-density lipoproteins (HDL) concentration, the apparent Vmax of CE transfer was inhibited by 74% and 83% in the presence of W-A at 14 and 25 mumol.L-1, respectively, while the apparent K(m) of HDL for CETP did not change. CONCLUSION: W-A action is mediated through interaction between W-A and CETP, but not through those between W-A and lipoproteins.

[Repair of peripheral nerve effect by direct suture after elongation of nerve by traction].

To find new technique for repair of peripheral nerve defect, the nerve elongation repair technique was adopted. Two cases with nerve defect were treated by this method. One was a 12 year old male, the defect length of right radial nerve was 7.2 cm at the elbow. The other one was a 28 year old male, the defect length of left ulnar nerve the was 5 cm at elbow. In this method, the nerve was elongated by slow stretch from distal and proximal end of the ruptured nerve. After a few days, the nerve was repaired by direct suture. After operation, the function of nerves were recovered in 119 days and 114 days respectively. Follow-up for 5 years, the function of the effected limbs were recovered to the normal side. It was concluded that: (1) the peripheral never can be elongated by slow stretch; (2) to stretch the nerve end in a rubber tube can prevent adhesion and connective tissue blocking; (3) strength and supporting point of stretching should be designed carefully.
