Endothelial-pericyte interdigitations in rat subcutaneous disc implanted angiogenesis.

Cytoplasmic interdigitations between the endothelial cells and pericytes (CIDEP) consist of a pericyte cytoplasmic projection and corresponding endothelial indentation. These are frequently present at the angiogenic sites. We have ultrastructurally examined CIDEP numbers at angiogenic sites induced by a polyvinyl alcohol foam disc implanted subcutaneously in rats. On Day 3 after implantation, the CIDEP number per endothelial cell and pericyte was similar to that of normal subcutaneous tissue, and then increased sharply with time to peak on Day 10. After this, it decreased, and on Day 30 was similar to that of normal subcutaneous tissue. The capillary density on Day 3 was significantly lower than that of normal subcutaneous tissue, increasing to peak on Day 7, and then decreasing with time. Until Day 7, capillary size was significantly smaller than in normal subcutaneous tissue, then increased with time. On Day 10, the number of endothelial cells significantly decreased, and that of pericytes significantly increased. The present study indicated that the predominant CIDEP distribution was observed on Day 10 following the active vascular proliferation stage on Day 7 and was concomitant with a decline in the number of the endothelial cells and an increase in the number of the pericytes.

Distribution of carbonic anhydrase isozyme III (CA-III)-positive cells in duct segments of the bovine submandibular gland.

The distribution of carbonic anhydrase III isozyme (CA-III)-positive cells in bovine submandibular glands was studied by immunohistochemistry. CA-III showed strong immunoreactivity in basal cells and some lining cells of the intercalated ducts as well as in striated and interlobular ducts cells. No significant immunoreactivity could be found in other portions of the glands. Electron microscopically, the immunoreactive basal cells contained scattered tonofibrils in their cytoplasm. The distribution of CA-III suggests that the CA-III-positive basal cells may play a special physiological role, and that they do not only represent undifferentiated lining cells.

Branching mode of the middle rectal artery from the prostatic artery in the dog.

The branching mode of the middle rectal artery from the prostatic artery was studied by gross dissection in 50 male dogs. The prostatic artery arose at the level of the 1st-3rd sacral vertebra from the internal pudendal artery, and gave off the ductus deferential and the middle rectal arteries, and supplied the prostate. The middle rectal artery ran caudally along the pelvic peritoneum, and supplied the rectum ampulla. The branching mode of the middle rectal artery could be classified into four types. Nineteen cases on the right side and 22 cases on the left side showed that the middle rectal artery was a first visceral branch of the prostatic artery. Fifteen cases on the right side and 11 cases on the left side showed that it was a second visceral branch of the prostatic artery which gave off the ductus deferential artery as its first visceral branch. Nine cases on the right side and 7 cases on the left side showed that it arose at the prostatic artery ramifying at the surface of the prostate. Seven cases on the right side and 10 cases on the left side showed that it arose at the urethral branch of the prostatic artery.

Tumour angiogenesis in prostatic carcinoma with and without bone marrow metastasis: a morphometric study.

One hundred and one cases of clinical prostatic carcinoma (PCa), primary site, were analysed to define the interrelationship between tumour angiogenesis, histological grade, and bone marrow metastasis. Tumour angiogenesis was determined by the blood capillary density ratio (BCDR; a/b), defined as the ratio between the area of the blood capillaries (a) and the area of the tumour (b). The BCDR was evaluated by a colour image analysis system employing a computerized morphometrical method. A total of 43 cases of PCa with bone marrow metastasis (stage D2) and 58 cases of PCa without metastasis (stage B, C) were utilized. The prostatic carcinomas were classified into three groups (low, intermediate, and high) using Gleason's grading system. The BCDR of the primary PCa with bone marrow metastasis was similar in each of the three histologically graded scores. On the other hand, in the cases of PCa without metastasis, the BCDR of high score PCa was higher than those of the low and intermediate score PCa (U-test; P < 0.001). The BCDR of the high score PCa without metastasis was similar to that of the PCa with bone marrow metastasis. The BCDR may provide help in predicting tumour progression with regard to bone marrow metastasis of PCa with low and intermediate Gleason's scores.

Epidermal growth factor receptor at endothelial cell and pericyte interdigitation in human granulation tissue.

Angiogenic immature capillaries in human granulation tissue possess many cytoplasmic interdigitations between endothelial cells and pericytes (CIDEP). Epidermal growth factor (EGF) is a potent mitogenic polypeptide which accelerates angiogenesis in vivo and in vitro. We have recently demonstrated in immature capillaries that EGF is present at the CIDEP and proposed that the CIDEP may be involved in a signaling pathway for EGF (Wakui et al., 1990c. Microvasc. Res. 40, 285-291). This study follows that previous report. In the present study, I have investigated the ultrastructural localization of the EGF receptor (EGF-r) at CIDEP. Immunoreactivity for the EGF-r in immature capillaries was located strictly at the CIDEP in large numbers, at the coated pits and vesicles of endothelial cells, and at some lysosome-like structures of the endothelial cells and the pericytes. On the other hand, immunoreactivity for EGF-r was absent in mature capillaries. At the CIDEP, EGF-r immunoreactivity was present at the cell membrane and a few cytoplasmic elements at the tip of pericyte cytoplasmic projections, but it was completely absent at the corresponding endothelial membrane indentation. The present results support our proposed hypothesis that the CIDEP in immature capillaries act as a pathway for EGF transportation from the endothelial cell to the pericyte by a receptor-mediated process.

Two cases of adrenal myelolipoma.

We report myelolipoma found in two patients, of whom one had hormonal abnormalities related to adrenal function. The first patient was a 36-year-old woman, who was found incidentally to have a left adrenal tumor by CT scan during admission for treatment of Guillain-Barré syndrome. Obesity, hirsutism and osteoporosis were also evident, and the patient was forwarded for additional endocrine function analysis, which revealed elevation of serum cortisol, urine 17-OHCS and 17-KS, and a decreased level of ACTH. These abnormalities returned to normal after excision of the tumor. Pathologically, the tumor was composed of mature fat cells and hematopoietic components, and was diagnosed as myelolipoma. The second patient was a 63-year-old woman, who was receiving follow-up care for hyperthyroidism. A right adrenal tumor was noted incidentally in a routine examination by CT scan. Endocrinologically, she was found to have no abnormalities of adrenal function. The tumor was excised, and diagnosed pathologically as myelolipoma, being composed of mature fat cells and hematopoietic components. Generally, although most myelolipomas have no endocrine function, our first patient showed features of Cushing's syndrome. Thus it is suggested that an interrelationship may exist between myelolipoma and endocrinological alteration.

Lectin histochemical study of the prostate gland of the rhesus monkey (Macaca mulatta).

Glycoconjugates in the secretory epithelial cells of the rhesus monkey (Macaca mulatta) prostate gland were investigated using lectin histochemistry. The monkey prostate possesses cranial and caudal lobes. Histochemical staining with a battery of 8 lectins demonstrated differences in lectin binding patterns of the secretory epithelial cells between the 2 lobes. BS-1 bound exclusively to the cranial lobe, and PNA bound exclusively to the caudal lobe. DBA bound exclusively to a number of the secretory epithelial cells of the caudal lobe. In addition, both lobes showed binding with Con-A, RCA-1, WGA and S-WGA, but were negative for binding with SBA. Specific lectin binding patterns suggest a differing carbohydrate composition for each region, and a biological difference between the cranial and caudal lobes of the rhesus monkey prostate.

Subepithelial elastic system fibers of oblique mucosal folds in the rat proximal colon.

The proximal colon of the rat is characterized by a 'herring bone' pattern of oblique mucosal folds (OMF) which are arranged in a parallel array. By light and electron microscopy the OMF exhibited rich subepithelial elastic system fibers which bound the epithelial basement membrane and the smooth muscle cells of the lamina propria together. The elastic system fibers usually consist of elastic, elaunin and oxytalan fibers. However, the subepithelial elastic system fibers of the OMF were composed of relatively thin elastic fibers with a few microfibrils, and elaunin and oxytalan fibers which were almost indiscernible. Areas other than the OMF were quite poor in subepithelial elastic system fibers. The interpositions between each of the OMF were composed of typical components: elastic, elaunin and oxytalan fibers. The composition of the subepithelial elastic system fibers of the OMF does not correspond to that of any other organs previously reported. The present study suggests that the OMF of the rat proximal colon might be equipped in such a way to resist to distension or compression.

Nuclear bodies in human autopsy prostate with special reference to appearance rate.

Nuclear bodies were observed in the nuclei of human prostatic epithelial cells at 2 to 3 hours post mortem. Although the epithelial cells of the autopsy cases revealed autolytic necrosis, the general feature of nuclear bodies were almost similar to those of fresh biopsied prostatic tissue. The nuclear body appearance rate was statistically analyzed in both secretory epithelial and basal cells of 3 different cellular condition areas (the hyperplastic nodule, non-nodule and atrophic). It was significantly high in the secretory epithelium of the hyperplastic nodule, but not in the other 2 areas. There was no significant result in the basal cells. The nuclear body appearance rate was statistically compared with that of human fresh biopsy cases previously reported by us (Furusato et al. 1986). The nuclear body appearance rates obtained in these 2 groups were not statistically different in each cell type and in 3 areas. The result suggests that the nuclear body appearance rate is reliable at 2 to 3 hours post mortem.

Arising patterns of the bovine diaphragmatic crura.

The arising patterns of the bovine diaphragmatic crura which consists of right and left crura in the lumbar portion of diaphragm, and the relative muscle weight of each crus were investigated. Diaphragm was obtained from 61 just before birth stage bovine fetus. The arising formation of the diaphragmatic crura in this study achieved its adults form. The arising patterns of the diaphragmatic crura were classified into 3 types according to the pattern of the cranial end of attachment of tendons in the right and left crura. The cranial end of the attachment of tendon is cranial in the right crus than in the left, shows the highest relative muscle weight of the right crus. The cranial end of the attachment of tendon is cranial in the left crus than in the right, shows the highest relative muscle weight of the left crus.

Studies on the Caldwell-Luc operation with or without counteropening at the inferior meatus.

The Caldwell-Luc operation was performed on 38 patients with and without the counteropening at the inferior meatus in order to investigate whether the opening is necessary or not. The blood loss with the opening was 108 g more on average than that on the side without it. The operation time was 11 min longer in the group with a counteropening. The differences in these two items were statistically significant. The difference in cheek swelling with both methods was not statistically significant. Postoperative conditions followed up to over 2 years showed no significant differences on both sides. The establishment of the counterdrainage at the inferior meatus is not necessary considering operation time, blood loss, possibility of nasolacrimal duct injury and early postoperative conditions.

Ultrastructural localization of fibronectin and laminin in human granulation tissue in relation to capillary development.

The distribution of fibronectin (FN) and laminin (LM) at developing capillaries during various developmental stages, from capillary sprouts to relatively developed capillaries, was studied by light- and electron-microscopy immunocytochemistry. By light-microscope, FN immunoreactivity was diffusely distributed throughout the stroma of the granulation tissues, while for LM it was preferentially distributed at the perivascular region with the various developmental stages of the immature capillaries. Ultrastructural study revealed that capillary sprouts were closely surrounded by plentiful deposits of immunoreaction with the FN, but only faintly for LM. Relatively developed capillaries with large and tall endothelium were surrounded by plentiful immunoreactive products with both FN and LM, and immunoreactivities in the cisternae of rER of the endothelium and/or the pericytes were also shown. Cytoplasmic interdigitations between the endothelium and the pericyte of developing capillaries were recognized without an immunoreaction for FN and LM. These results mean that the capillary sprouts are associated with a prepatterned FN-rich and LM-poor perivascular matrix, whereas relatively developed capillaries are associated with a FN- and LM-rich perivascular matrix which would have been produced by the capillary endothelium and/or pericytes.

Three-dimensional ultrastructural distribution of cytoplasmic interdigitation between endothelium and pericyte of capillary in human granulation tissue by serial section reconstruction method.

Two- and three-dimensional electron microscopic observations in the capillaries with angiogenetic sprout of human granulation tissue revealed the cytoplasmic interdigitations (CID) between the endothelium and the pericyte, which were composed of unit of cytoplasmic projection and an indentation. Two types of interdigitation were observed: the interdigitation composed of cytoplasmic projection from the pericyte plugging into the endothelial indentation, CID(P-E); the interdigitation which had a reverse mode of composition of the cytoplasmic projection and indentation to the former one, CID(E-P). We investigated three-dimensional distribution of the two kinds of interdigitation in two cases of the capillaries with angiogenetic sprout. The two kinds of interdigitation exhibited a remarkable difference in the three-dimensional distribution. The CID(P-E) was rather uniformly distributed over the parent capillary endothelial area (6.4 per endothelium in average) and was not observed at all in the angiogenetic area; the other type of cytoplasmic interdigitation, CID(E-P), was located in only the angiogenetic sprouting endothelial area (3.5 per endothelium in average). The cytoplasmic interdigitation between the endothelium and the pericyte in vivo was the only site for cell contact and seemed to be involved in a conducting function which is biochemically proved to exist in vitro between the two cells.

Simple mucodermal tracheoesophageal shunt method for voice restoration.

A newly devised simple primary tracheoesophageal shunt method, involving a mucodermal tunnel, was performed on 14 patients. Aspiration was prevented by percutaneous digital pressure on the shunt, with silicone or collagen injection around it, or the use of a voice prosthesis. The overall success rate was 79% (11/14). This method was an easy one-stage procedure that could be performed in about 15 minutes.

Crystalloids in latent prostatic carcinoma.

Intraglandular crystalloids have been studied in 108 cases of latent prostatic carcinoma collected at autopsy. The morphological features of these crystalloids are essentially the same as reported by previous authors. The crystalloids were identified in 62.0% of cases over age 50 years. They were present in 76.9% of cases in the seventh decade, the highest prevalence among the decades. The number of crystalloids relative to the number of malignant glands tended to be inversely proportional to the size of the carcinoma. We conclude that crystalloids are related to the early development of prostatic carcinoma.

Ototoxicity of carboplatin in guinea pigs.

Since carboplatin (CBDCA) is a platinum compound of second generation, its ototoxicity was investigated. In order to assess the ototoxicity, hearing acuity was measured based on the compound action potential (AP), and the organ of Corti was morphologically studied using scanning electron microscopy (SEM) and succinate dehydrogenase (SDH) staining in guinea pigs treated with CBDCA. CBDCA induced morphological changes in the organ of Corti when the guinea pigs received 50 mg/kg daily for 2 or 3 days. The threshold value of AP was elevated 10 to 25 dB in treated animals, as compared with the control value. The SEM study and SDH staining revealed that the administration caused scattered degeneration of the outer hair cells (OHCs), and the inner hair cells remained intact. These lesions were milder than those of OHCs treated with cisplatin (CDDP). The rapid elimination of CBDCA from the kidney and the inner ear seems to account for the low ototoxicity of this compound. Although CBDCA is less toxic than CDDP, it may affect hearing function if patients suffer from renal dysfunction or if the organ of Corti is vulnerable.
