Isoelectric focusing of bacterial DNA methylases.

The multiplicity of bacterial DNA methylases has been shown for new microorganisms, Mycobacteria and Shigella, by a double-step procedure including column chromatography followed by isoelectric focusing of the total methylase fraction. The profiles of the DNA methylating activity of Sh. sonnei 47 and M. butyricum strains were studied. Sh. sonnei 47 cells were found to contain five different proteins responsible for DNA methylation and having pI 4.2, 5.3, 6.2, 8.4 and 9.2. Four M. butyricum methylases were represented by proteins with pI 4.2, 6.0, 8.0 and 9.0.

[Phenomenon of DNA-abzyme cross-reactivity and its significance for the mechanisms of cytotoxicity and apoptosis].

The physiological role of DNA-abzymes and their involvement in pathogenesis of different autoimmune disorders is still unknown. At the same time, a variety of properties and features of DNA-hydrolyzing autoantibodies have been studied. Here, the phenomenon of the cross-reactivity of DNA-abzymes with the nuclear matrix proteins was studied. The possible value of the phenomenon for the cytotoxic activity of DNA-hydrolyzing autoantibodies was debated as well. A new hypothesis is put forward regarding the DNA-abzymes formation based on the phenomenon of the cross-reactivity of polyclonal DNA-abzymes with nuclear matrix proteins free of native DNA. Preliminary results suggest that there are mechanisms of cytotoxicity mediated by DNA-abzymes and independent from the system of complement and cytotoxic T-lymphocytes.

[EHF-therapy of herpetic keratouveitis].

A device BIK for active suppression of infection has been used against Herpes virus infection type 1 and 2 in 72 patients with keratouveitis. The immunological and genotypic monitoring show that patients with herpetic keratouveitis after BIK treatment have lower titers of antiherpetic antibodies, normal cellular and humoral immunity, weaker viremia and reduced quantity of viral material in corneal tissue. This positive effect persisted for 8-12 months. Thus, BIK device provides a definite, prolonged and stable therapeutic effect in patients with herpetic keratouveitis.

[The connection between the decomposition of mature collagen and the level of natural cytotoxicity in patients with focal scleroderma].

Increased rate of pyridinoline excretion with urine was detected in patients with focal sclerodermia: the ratio pyridinoline/creatinine was equal to 0.094 +/- 0.049 as compared with that of 0.073 +/- 0.049 in controls. Concentration of mature collagen cross-links involving pyridinoline structure in urine of patients with focal sclerodermia did not depend on the patients age and the disease duration. However, this pattern correlated distinctly with clinical form of the disease (0.073 +/- 0.049 in local form and 0.013 +/- 0.017 in generalized form of sclerodermia) as well as correlated reversely with the rate of activity of natural killer cells (r = -0.977).

[Possible role of natural cytotoxicity in impairment of collagen catabolism in focal scleroderma].

Relationship between the state of natural cytotoxicity and collagen catabolism were studied in patients with focal sclerodermia. In the impairment activity of natural killer cells (NK cells) was markedly altered depending on such clinical parameters as duration, severity, activity and distribution of the disease as well as it was related to definite transformations in catabolism of collagen proteins. Low rate of collagen production and intensive collagenolytic processes correlated with the high level of hydrolytic activity of NK cells at the early steps of systemic morphea guttata accompanied by highly progressive course of the disease. Under conditions of subacute forms of focal morphea guttata activity of NK cells was distinctly decreased down to 50% of that of normal state and was accompanied by alterations in the spectrum of collagenolytic enzymes as well as by considerable variations in collagen production and in posttranslational modification of collagen fibrils. During the chronic step of sclerodermia activity of NK cells was normalized and correlated with the lowered rate of collagen production as well as with the subsequent alterations in collagen proteins catabolism. Alterations in the level of natural cytotoxicity appear to correlate with the state of collagen metabolism in focal sclerodermia.

[Tissue-specific retinal proteins in vertebrate evolution].

Studies have been made on water soluble antigens of the retina from man and some animals. In the bovine retina, immunochemical analysis reveals, apart from antigens with a broad and narrow interorganic specificity, organospecific alpha 1- and rho-globulins. Immunochemically, the bovine alpha 1-globulin is partially identical with the same protein of the human retina and completely identical to retinal antigens from cattle; rho-globulin is characterized as an interspecific antigen in man and mammals. Molecules of organospecific alpha 1-globulins from the retina of man and some animals (sheep, camel, horse, cow, pig) do not contain the determinants related to the retinal antigens from fishes, reptiles and birds. In human and mammalian retina, acid neurospecific alpha 1-glycoprotein was found which is topical of the cerebral tissue. Organospecific alpha 1-globulin of the bovine retina is located in the pigment epithelium, in the zone of outer and inner photoreceptor segments; organospecific rho-globulin is distributed in the outer synaptic layer of the retina.

[Isoelectric focusing of DNA-methylases from Shigella sonnei 47].

Fractionation of bacterial methylases was studied by means of isoelectrofocusing technique. The method exhibited high accuracy, efficiency and reproducibility in studies of methylases from Shigella sonnei 47. High column capacity and distinct focusing effect increased the experimental advantages of work with various amounts of protein as compared with ion exchange chromatography. By means of the isoelectrofocusing technique three methylases were detected in the strain S. s o n n e i 47, which were dissimilar in their isoelectric points at pH 6.5, 8.4 and 9.8. These findings indicate the heterogeneous pattern of enzyme methylation in the strain studied. Complete removing of endogenous DNA was the main precondition for correct fractionation of methylases by means of the isoelectrofocusing technique. A procedure is described for purification of S. s o n n e i 47 methylases from traces of endogenous DNA by affinity chromatography on heparin-Sepharose.
