Isolation and characterization of proteohyaluronic acid from human umbilical cord.

Proteohyaluronic acid was extracted from human umbilical cord with 0.2% NaCl, then purified by cetylpyridinium chloride(CPC)-precipitation, DEAE-cellulose column chromatography, gel-filtration on Sephadex G-200 and on Sepharose 4B, in succession. The purified preparation contained glucosamine (40.1%), glucuronic acid (45.7%) and protein (2.6%). Glutamic acid, glycine, alanine, serine and aspartic acid were the major amino acids of the protein moiety. The result of the gel-filtration suggested that an average molecular weight was more than 1 x 10(6). Although analytical data for the constituent sugars and infrared spectra were similar before and after pronase digestion of this proteohyaluronic acid, the mobilities in electrophoresis and the elution patterns of gel-filtration differed remarkably from one another. Since the protein content decreased from 2.6% to 0.3% by the proteolytic digestion, it is evident that the degraded protein moiety played an important role in maintaining the macromolecular structure of this proteohyaluronic acid.

Augmentation of delayed-type hypersensitivity and resistance against allogeneic or syngeneic methylcholanthrene-induced tumors in mice preimmunized with the tumor extracts.

Delayed-type hypersensitivity (DTH) responses against methylcholanthrene-induced fibrosarcomas in C3H/He and BDF1 mice were developed in BDF1 mice by sc injection of the respective mitomycin C-treated tumor cells. The DTH responses to the allogeneic and the syngeneic tumor cells were accelerated and enhanced tumor-specifically by priming 7 days previously with KCl extracts of the respective tumors. The ability in the mice primed with the tumor extracts enhancing the DTH response against the tumor cells could be transferred to recipient mice by the spleen cells, but not by the T-cell-depleted spleen cells. Rejection of allogeneic tumor was accelerated under the development of accelerated and enhanced DTH response against the allogeneic tumor antigens. Moreover, resistance to syngeneic tumor growth increased significantly with the development of accelerated and enhanced DTH response against the syngeneic tumor antigens. Thus, the augmentation of DTH response by preimmunization with tumor extracts was accompanied by the increased resistance to tumor growth, suggesting that T cells involved in the augmentation of tumor-specific DTH response play some role in increasing the resistance to tumor growth.

Cytochemical localization of complex carbohydrates in the thyroid gland of normal and TSH-treated mice.

The silver methenamine method for the ultrastructural localization of carbohydrates and glycoproteins was applied to the thyroid glands of normal and TSH-treated mice. The majority of the cisternae of the rough endoplasmic reticulum showed a weak, but apparently positive reaction. These findings support the opinion that glycosylation of thyroglobulin occurs initially in the rough endoplasmic reticulum. By this method the Golgi apparatus was observed to display a staining gradient. The intermediate to inner saccules were intensely stained, whereas the outer saccules were not so heavily stained. This phenomenon indicates that the Golgi apparatus has a functional polarity for the addition of carbohydrates to thyroglobulin and other proteins. In the inner and/or the peripheral regions of the Golgi apparatus and in the apical cytoplasm, a large number of globules of various sizes, considered to be colloid droplets, lysosomes and apical secreting vesicles, showed a positive reaction. The luminal colloid was also positive with silver methenamine staining, with almost the same intensity as the globules and vesicles.

Characterization of multiple forms of histone phosphatase in rat liver.

By using chromatography on DEAE-cellulose, aminohexyl-Sepharose 4B and Sephadex G-200, rat liver extract was shown to contain at least three fractions, IA, IB and II, of histone phosphatase. Fractions IA and II are probably the same enzymes as the previously described glycogen synthase phosphatase and phosphorylase phosphatase, respectively, but IB exhibits noticeable activities only with phosphohistone as substrate. Approximate molecular weights of 69 000, 300 000 and 160 000 were determined by gel filtration on Sephadex G-200 for IA, IB and II, respectively.

[Metabolism of arsenic (Report 22). Effect of arsenic on the spontaneous motor activity, avoidance conditioning, extinction and swimming record in rats (author's transl)].

The experiments were conducted to determine the effect of arsenic on the pharmacological actions as seen in the behavior of Wistar albino rats. Nutrition during the suckling period has some apparently influence on the pharmacology behavior, we found no significant difference between the heavy (26 approximately 35g) and light (under 25g) groups. Also rats in group B1, were fed a diet which included As2O3 2 mg/kg, and rats in group B2 were fed a diet which included As2O3 15 mg/kg during the suckling period. There were no significant differences in spontaneous motor activity, avoidance conditioning and swimming record between the controls and group B1. However in group B2, the spontaneous motor activity increased as did the response on avoidance conditioning.

[Effect of arsenic on the developing brain of rat (author's transl)].

An experiment was conducted in an attempt to determine the effect of arsenic on body weight, brain weight and brain chemical composition in suckling rats. Four hundred and ninety-nine young rats from Wistar albino mothers in a closed colony (TAMURA, 1950) were grouped into four according to the treatment given. Group 1 served as control. Rats in Group II were treated with As2O3 1.5 mg/kg, those in Group III 15 mg/kg and those in Group IV arsenic compounds (corresponding 15 mg/kg of As2O3) for 7 to 21 days after birth. Measurements were recorded for 3 to 15 weeks. The mortality of groups I and II was approximately 10%, while that of groups III and IV was approximately 50%. Regarding body weight, brain weight and brain chemical composition of provision of the same, there was no significant difference between controls and the arsenic treated males and females under conditions Oriental feed diet and powdered milk.

Synthetic identification as a hexapeptide of alpha substance-IB inducing sexual agglutination in Saccharomyces cerevisiae.

For the identification of a peptidyl principle inducing sexual agglutination in the yeast, 2 supposed hexapeptides (1a, b) were synthesized by the conventional method. The 1a (H-Arg-Gly-Pro-Phe-Pro-Ile-OH) revealed complete identity with the natural peptide in TLC, MS and biological property on agglutination. The 1b showed the sexual agglutinability in the same degree as 1a, though distinct differences were observed in the chemical data. Both 1a and 1b had a strong bitter taste.
