[Experimental study on early development of rat ear in vitro using whole embryo culture].

Rat embryos were explanted on late 11 day of gestation and cultured for 24 hours in rotating bottles with the yolk sac opened. Rat serum was used as culture medium and culture bottles were filled with 5% CO2 + 95% O2 gas mixture as gas phase. At the time of explantation and the end of the culture period, differentiation and growth of the embryos were monitored by counting somites and measuring crown rump length. About part of the embryos, protein determinations were made to measure growth. These data were compared with values found for 12 and 13 day embryos. The results for cultured embryos showed slight retardation in their differentiation and slight depression in the growth. At the same time to study early inner ear development in vitro, 9 of the other cultured embryos were serially sectioned and observed by light microscopy. In the cultured embryos endolymphatic duct elongated and acoustico-facial ganglion enlarged remarkably. Otocysts became flattened and elongated ventrally. Vestibular and cochlear portion were identifiable in the otocysts. Nerves arose from the ganglion reached to brain centrally and wall of pharynx peripherally. There was no significant difference in inner ear development between 13 day and the cultured embryos. This culture system should prove useful for studies on early inner ear development of mammalian embryos.

Automatic generation of a retinal sensitivity map using infra-red fundus television images.

We have already developed an eye movement analyzing system using an infra-red television fundus camera. Although the quality of the fundus TV images is not good, the system can measure eye movement with an accuracy order of 0.1 degree. We try to apply this system to compensating for eye movements in producing a retinal sensitivity map. A target with variable brightness is presented to various positions on the retina of a subject through a CRT assembled in the infra-red TV fundus camera. The subject is supposed to respond if he can perceive the target. The TV image of the eye fundus, together with the overlaid target and response sign, is recorded on a U-matic (3/4 in) VTR. The image of the video tape is then analyzed by a computer. We have obtained retinal sensitivity maps which show good agreement with the results obtained using the Goldmann perimeter.

Interspecies crossreactive antigen of the pancreatic duct cell prepared by monoclonal antibody. A preliminary study of its immunological significance in patients with chronic pancreatitis associated with Sjögren's syndrome.

An interspecies cross-reactive antigen (mol wt 60,000) for pancreatic duct cells was prepared by a monoclonal antibody (SP3-1; IgM). Homogenized pig pancreatic tissues were separated by gel filtration chromatography with Sephadex G-100 and ion-exchange column chromatography with DEAE-Sephacel and used as immunogens. Immunohistochemically, antigens against SP3-1 were distributed in duct cells of the pancreas, salivary gland, lacrimal gland, esophageal gland, bile duct, and the distal renal tubule in the pig, monkey, and human. The antigenic determinant contained protein, but not sialic acid or monosaccharide. Preliminary leukocyte migration inhibition test (n = 5) showed positive in four patients with definite Sjögren's syndrome (SS), whereas one patient without autoantibodies similar to SS showed negative. These findings suggest that this antigen may be involved in the pathophysiology in these conditions.

[A case of myelolipoma originating from bilateral adrenal glands; a comparison of CT and pathological findings].

We experienced a case of bilateral retroperitoneal tumor incidentally on CT which was carried out on suspicion of either cholelithiasis or choledocholithiasis. In company with other radiologic studies, US, angiography etc. we were able to make a correct diagnosis of adrenal myelolipoma preoperatively. Among these studies, CT coincided well with the structure of pathological findings and is thus the most powerful method for the diagnosis of myelolipoma. The case was discussed in addition to the studies of other literatures.

[A case of multiple cavities on chest film with high titer of serum cryptococcal antigen].

A 26-year-old man admitted to Nishinomiya Municipal Hospital for further evaluation for abnormal shadows on the chest film in a mass examination. He had no subjective complaints; for example, cough, sputum or dyspnea. His past history and physical examinations yielded no significant findings. The chest film revealed the multiple cavities accompanied with a little infiltration throughout several lobes bilaterally. The inflammatory reactions; such as, CRP and ESR, were all intact, but the titer of the serum cryptococcal antigen was high. He was not immunocompromised. Although bronchoscopy and transbronchial lung biopsy were performed twice during admission, they revealed no pathogenic findings both bacteriologically and histologically. The elevation of the serum cryptococcal antigen titer suggested that this disease is the primary pulmonary cryptococcosis. The titer decreased with spontaneous disappearance of the abnormal shadows on the chest film. This test has a sensitivity of 90% for cryptococcal infection, but in rheumatoid arthritis, a small percentage of false positive results have been reported. Therefore, if the rheumatoid factor is eliminated, the test is more specific for cryptococcal infection. On the other hand, some authors have reported the false positive results in Trichosporon beigelii infection by this method. But this disease is negligible for its frequency in clinical features in our country. We emphasized that this non-invasive test is more useful for the diagnosis of the primary pulmonary cryptococcosis.

[Effects of gestrinone on serum lipid and lipoprotein levels in women with endometriosis].

Gestrinone (G) was given to 12 females with endometriosis in weekly doses of 5 or 10mg for 4 to 6 months, and the change in serum lipids and lipoproteins was analysed. G decreased total cholesterol by 20% (p less than 0.05), triglycerides by 36% (p less than 0.05), phospholipids by 28% (p less than 0.01) and lipid peroxides by 34% (p less than 0.05), among which reductions in them were statistically significant when compared with the pretreatment levels. Levels of high density lipoproteins (HDL) also fell: HDL-cholesterol by 41% (p less than 0.01), HDL-triglycerides by 49% (p less than 0.05) and HDL-phospholipids by 38% (p less than 0.01) which were significant. Concurrently apolipoproteins (Apo) and lecithin-cholesterol acyltransferase activity (LCAT) decreased: Apo A-I by 31% (p less than 0.01), Apo A-II by 13% (p less than 0.05) and LCAT by 53% (p less than 0.05), which were significant. In contrast, there were few changes in the levels of low density lipoproteins (LDL) and Apo B. There was also little effect on very low density lipoproteins (VLDL) except VLDL-triglycerides which decreased by 52% (p less than 0.05). Meanwhile free fatty acids increased by 61% (p less than 0.05). Therefore, the atherogenic index defined as the ratio of LDL-cholesterol to HDL-cholesterol rose as much as 92% (p less than 0.01) of the initial value in 24 weeks of medication. When these results were examined with respect to the 5 and 10mg administration group, dose-dependent effects were observed, but these were not marked.(ABSTRACT TRUNCATED AT 250 WORDS)

Serum pseudouridine as a biochemical marker in patients with hepatocellular carcinoma.

The concentration of serum pseudouridine, a degradation product of transfer ribonucleic acid, was determined by high-performance liquid chromatography in patients with hepatocellular carcinoma, liver cirrhosis, other benign hepatobiliary diseases, and healthy controls. The serum pseudouridine concentration in patients with hepatocellular carcinoma was significantly higher than that in patients with cirrhosis or the controls. Twenty-seven (51.9%) of 52 patients with hepatocellular carcinoma had serum pseudouridine concentrations higher than the mean value for healthy controls plus 2 SD. Fourteen of the 36 patients who had serum alpha-fetoprotein levels below 400 ng/ml, had elevated serum pseudouridine concentration. In total, 36 of the 52 patients (69.2%) could be detected by combination of these two markers. Two patients who had developed hepatocellular carcinoma during the course of cirrhosis and were continuously negative for alpha-fetoprotein, had higher levels of the pseudouridine concentration when hepatocellular carcinoma occurred. Furthermore, 4 of the 7 patients who had a very small cancer and were negative for alpha-fetoprotein, had elevated serum pseudouridine concentration. These results indicate that serum pseudouridine is a useful biochemical marker and that serum pseudouridine and alpha-fetoprotein in combination are considered to serve as complementary markers, for the diagnosis of hepatocellular carcinoma.

The factor V-activating enzyme (RVV-V) from Russell's viper venom. Identification of isoproteins RVV-V alpha, -V beta, and -V gamma and their complete amino acid sequences.

The complete amino acid sequences of two isoproteins of the factor V-activating enzyme (RVV-V) isolated from Vipera russelli (Russell's viper) venom were determined by sequencing S-pyridylethylated derivatives of the proteins and their peptide fragments generated by either chemical (cyanogen bromide and 2-(2-nitrophenylsulfenyl)-3-methyl-3-bromoindolenine) or enzymatic (trypsin, alpha-chymotrypsin, and lysyl endopeptidase) cleavages. Both enzymes, designated RVV-V alpha and RVV-V gamma, consist of 236 amino acid residues and have a N-linked oligosaccharide chain at Asn229. The six amino acid substitutions between RVV-V alpha and -V gamma are: Thr22(alpha)-Ala22(gamma), Gly29(alpha)-Ala29(gamma), Gln191(alpha)-Glu191(gamma), Ile192(alpha)-Met192(gamma), Gln193(alpha)-His193(gamma), and Asn224(alpha)-Ser224(gamma). The molecular weights were calculated as 26,182 for RVV-V alpha and 26,167 for RVV-V gamma. The sequences of the RVV-V isoproteins exhibited 62% identity with that of batroxobin, a thrombin-like enzyme present in Bothrops atrox venom, and 33% identity with that of human thrombin B chain. The most interesting difference between the structures of RVV-V and other trypsin-type serine proteases is that the conservative Ser214-Trp215-Gly216 sequence (chymotrypsinogen numbering), considered as the site of antiparallel beta-sheet formation between the protein substrate and most serine proteases, has been replaced by the corresponding sequence Ala-Gly-Gly.

Production of monoclonal antibodies against human erythropoietin and their use in the purification of human urinary erythropoietin.

Several murine monoclonal antibodies (MAbs) specific for human erythropoietin (HuEpo) were produced by hybridomas obtained from the fusion of murine myeloma cells, P3X63-Ag.8-653, with the splenocytes of mice immunized with recombinant human Epo (rHuEpo). Based on epitope analysis by a competitive binding assay, these MAbs could be classified into at least three groups: (1) 1E10, (2) 1H7, (3) 2D6, 3D6 and 3D8. In a sandwich enzyme-linked immunosorbent assay (ELISA), using these MAbs as the solid-phase antibodies, MAb-bound HuEpo was detected with rabbit anti-HuEpo sera. Some combinations of two different classes of MAbs, such as 1H7 and 3D8, were found to capture much more HuEpo than each MAb used individually. Urinary HuEpo (U-HuEpo) was highly purified from the urine of patients with severe aplastic anemia with about 50% final recovery using an immunoaffinity column on which a mixture of 1H7 and 3D8 was immobilized. The purified U-HuEpo had a specific activity of 77,340 U/mg in a radioimmunoassay (RIA) and of 76,673 U/mg using an in vivo bioassay.

Direct determination of urinary pseudouridine by high-performance liquid chromatography.

A reversed-phase high-performance liquid chromatographic method was developed for the determination of pseudouridine in urine. This method does not need pretreatment by boronate affinity gel. Therefore, it can be used in screening patients with malignant disease and for monitoring clinical response to chemotherapy with other tumor markers.
